C-Jun N-terminal kinases/c-Jun and p38 pathways cooperate in ceramide-induced neuronal apoptosis
Understanding the regulation of the apoptotic program in neurons by intracellular pathways is currently a subject of great interest. Recent results suggest that c-Jun N-terminal kinases (JNK), mitogen-activated protein kinases and the transcription factor c-Jun are important regulators of this cell death program in post-mitotic neurons following survival-factor withdrawal. Our study demonstrates that ceramide levels increase upon survival-factor withdrawal in primary cultured cortical neurons. Furthermore, survival-factor withdrawal or addition of exogenous c2-ceramide induces JNK pathway activation in these cells. Western blot analyses of JNK and c-Jun using phospho-specific antibodies reveal that JNK and subsequent c-Jun phosphorylation occur hours before the initiation of apoptosis, reflected morphologically by neurite retraction and fragmentation, cell-body shrinkage and chromatin fragmentation. Immunocytochemistry using the same antibodies shows that phospho-JNK are localized in the neurites of control neurons and translocate to the nucleus where phospho-c-Jun concurrently appears upon ceramide-induced apoptosis. To determine if ceramide-induced c-Jun activation is responsible for the induction of the apoptotic program, we performed transient transfections of a dominant negative form of c-Jun, truncated in its transactivation region. Our results show that DNc-Jun partially protects cortical neurons from ceramide-induced apoptosis. Treatment of dominant negative c-Jun-expressing neurons with the pharmacological inhibitor of p38 kinase, SB203580, completely blocked neuronal death. Thus our data show that p38 and JNK/c-Jun pathways cooperate to induce neuronal apoptosis

This very short cruise was primarily to deploy a novel moored observatory at PAP (NE Atlantic), a long term study site at which a variety of observations have been made over the past 20 years with increasing levels of intensity and sophistication. This new design was in collaboration with the UK Met Office.In addition to this, a benthic lander was to be deployed which would transmit data by an acoustic link to the surface buoy and hence to land by satellite link. Unfortunately this was lost due to an imploding sphere.A number of equipment trials were also carried out to good effect. In addition studies were made on the effects of high CO2 of calcifying phytoplankton. A team of photographers were present to in order to increase public awareness of observational oceanography and numerous interviews were completed. In addition a daily web diary was maintained by the EuroSITES outreach team
RV Pelagia Cruise 64PE372, 27 Jun - 11 Jul 2013. Flow dynamics and sedimentation in an active submarine channel: a process-product approach
Novel technologies are providing new opportunities to study the structure and dynamics of submarine sediment-gravity flows; these flows are the dominant process for transfer of sediment into the deep ocean, but are very hard to monitor due their destructive and unpredictable character. The primary aim of 64PE372 was to image the 3D structure of submarine flows passing though a rare example of an active submarine channel system in the southeast Black Sea. The channel system is maintained by through-flow of relatively dense, saline water coming from the Bosporus Strait outflow. ADCP mapping of internal flow structure within and outside the channel was achieved using the NERC Autosub3 Autonomous Underwater Vehicle (AUV), supplemented by vertical, vessel-based, CTD profiling, and a fixed CTD mooring in the proximal channel for the duration of the cruise. Sedimentary features associated with the channel were imaged using towed high-resolution sidescan sonar, supported by vessel-based multibeam bathymetry and backscatter, AUV subbottom profiler data, and gravity cores. The 64PE372 cruise built upon a previous cruise to the study area in spring 2010, but was far more successful due to the increased capability of the barter vessel and the improved performance of the AUV

Measured weights in PEI adults reveal higher than expected obesity rates
MacLellan et al seek to summarize the findings of the Prince Edward Island (PEI) Nutrition Survey. A random sample of 1,995 adults aged 18-74 were interviewed in their homes and weights, heights, and waist circumference measurements were obtained. Results show that almost one third of PEI adults are obese. Cruise CD158 to the Porcupine Abyssal Plain (PAP) maintained the long term observatory site on the Porcupine Abyssal Plain (PAP) at 49N 16.5W and continued a series of multidisciplinary occuaptions of the site. There were two objectives, A: to recover and redeploy various moorings and landers some of which were part of the EU funded ANIMATE project, and B: to measure particulate export from the surface layer using a variety of complementary approaches and to characterise the site from a biological and chemical perspective in order to interpret the export data. These all contribute to the NERC funded BICEP project
RRS Discovery Cruise 365, 11 May-02 Jun 2011. The Extended Ellett Line 2011
The Extended Ellett Line is a full-depth hydrographic section between Iceland, 60°N 20°W, Rockall and Scotland. The original Ellett Line across the Rockall Trough was first occupied in 1975 when measurements were attempted four times a year. In 1996 the line was extended to Iceland and since then has been occupied approximately annually. The data form a 35 year time series of the oceanic conditions west of the British Isles.The section monitors the characteristics of the warm water inflow into the Nordic Seas and thence to the Arctic, and observes part of the returning cold water outflow with easurements of the Iceland-Scotland Overflow and the overflow of the Wyville-Thomson Ridge into the Rockall Trough.The 2011 occupation, RRS Discovery cruise 365, was curtailed by both technical problems and bad weather. 45 of the 48 CTD stations were worked between the Iceland and Scotland shelf edges. Line G, part of the SAMS observation network of the Scottish continental shelf was partially completed, with 4 stations at the western end not worked. Samples were taken for CFC and SF6 analysis, DIC and alkalinity, inorganic nutrients, aluminium, POC, bacterial abundance and biomass, and for phytoplankton community structure. Plans to repeat stations, to collect validation data for the SAMS glider and to investigate eddies in the Rockall Trough had to be abandoned

RRS James Cook Cruise 87, 31 May - 18 Jun 2013. The Twilight Cruise to the Porcupine Abyssal Plain Sustained Observatory
The Twilight Zone is that depth zone in the ocean between 100 and 1000m depth where a tremendous amount of activity takes place. Much of the material containing carbon which sinks out of the upper sunlit or "Euphotic" zone is broken down in the twilight zone and then mixes back up to the surface in the winter. If it manages to sink further, this carbon is lost for periods of centuries.The main factor that affects this sedimentation process and the rate of destruction of the sinking particles is the structure and function of the biological community living near the sea surface and in the twilight zone beneath. This is because the planktonic plants and animals living there both generate and destroy particles. The Porcupine Abyssal Plain sustained observatory (PAP) is a heavily instrumented area of the open ocean 350 miles southwest of Ireland and in a water depth of 4800m. The instruments measure a wide variety of properties of the environment above the water, within it and on the seabed and much of the data is transmitted in real time to land via satellite
