Metals and Parkinson's disease: mechanisms and biochemical processes

    No full text
Genetic background accounts for only 5 to 10% of the reported cases of Parkinson's disease (PD), while the remaining cases are of unknown etiology. It is believed that environmental factors may be involved in the causality of a large proportion of PD cases. Several PD genes are activated by xenobiotic exposure, and a link between pesticide exposure and PD has been demonstrated. Many epidemiological studies have shown an association between PD and exposure to metals such as mercury, lead, manganese, copper, iron, aluminum, bismuth, thallium, and zinc. This review explores the biological effects, the pathogenetic processes, genetic susceptibilities to metals as well as examining future strategies for PD treatment, such as chelation therapy

Molecular genetic and physiological studies to unravel the mystery of Sphingomonas wittichii RW1 dibenzo-p-dioxin degradation

Dibenzofuran and dibenzo-p-dioxin are ubiquitous environmental pollutants in soil and sediment. Sphingomonas wittichii RW1 is one of a few strains known for the ability to grow on the related compounds dibenzofuran (DBF) and dibenzo-p-dioxin (DXN) as the sole source of carbon. The genes for the initial steps in the DBF catabolic pathway (ring hydroxylating dioxygenase, ring cleavage dioxygenase, and a hydrolase) which result in the formation of salicylate and a five-carbon fragment have been localized to a mega-plasmid designated pSWIT02 in RW1. Plasmids highly similar to pSWIT02 have been found in other DBF degrading Sphingomonas strains. However, despite having the pSWIT02-encoded DBF degradation pathway these other bacteria are not capable of growth on DXN. This thesis describes involvement of chromosomally encoded genes in dibenzo-p-dioxin degradation by RW1. RW1 lacking the pSWIT02 dbfB gene grows extremely slowly on DBF and accumulates the ring cleavage substrate 2,2',3-trihydroxybiphenyl. The mutant grows normally on DXN as the sole source of carbon indicating that dbfB is not necessary for the DXN catabolic pathway and suggesting involvement of other ring cleavage dioxygenases in DXN pathway. Knockout of gene SWIT3046 resulted in a strain that grows normally on DBF but that does not grow on DXN. The double knockout does not grow on either DBF or DXN. These results prove that separate ring cleavage enzymes are necessary for DBF and DXN degradation. We then examined the third enzymatic step in RW1, the hydrolase. RW1 lacking the pSWIT02 encoded gene dxnB1 or the chromosome encoded gene dxnB2 grow normally on both DBF and DXN. A double knockout of both genes does not grow on DBF but still grows on DXN. We then examined previously published transcriptomic data that showed that the SWIT0910 encoded hydrolase is up regulated during growth on DBF and DXN. A knockout of SWIT0910 grows normally on DBF but does not grow on DXN. Our results demonstrate that a chromosomally encoded hydrolase, SWIT0910, is absolutely required for growth on DXN and that two different hydrolases (chromosomally and plasmid encoded) contribute equally to growth on DBF.Genes for three biphenyl ring cleavage dioxygenases from Burkholderia xenovorans LB400, Sphingomonas yanoikuyae B1, and Pseudomonas putida F1 were moved into a mutant lacking the RW1 DBF and DXN ring cleavage genes. All three ring cleavage dioxygenases allowed the mutant RW1 to grow on DBF at different rates. Interestingly, only bphC from Burkholderia xenovorans LB400 allowed RW1 mutant to grow on DXN.Ph.D.Includes bibliographical reference

Translating Brecht : versions of "Mutter Courage und ihre Kinder" for the British stage

    No full text
This study analyses five British translations of Bertolt Brecht's 'Mutter Courage und ihre Kinder'. Two of these translations were written by speakers of German, and three by well-known British playwrights with no knowledge of the source text language. Four have been produced in mainstream British theatres in the past twenty-five years. The study applies translation studies methodology to a textual analysis which focuses on the translation of techniques of linguistic "Verfremdung", as well as linguistic expression of the comedy and of the political dimension in the work. It thus closes the gap in current Brecht research in examining the importance of his idiosyncratic use of language to the translation and reception of his work in the UK. The study assesses the ways in which the translator and director are influenced by Brecht's legacy in the UK and in turn, what image of Brecht they mediate through the production on stage. To this end, the study throws light on the formation of Brecht's problematic reputation in the UK, and it also highlights the social and political circumstances in early twentieth century Germany which prompted Brecht to develop his theory of an epic theatre. The focus on a linguistic examination allows the translator's contribution to the production process to be isolated. Together with an investigation of the reception of each performance text, this in turn facilitates a more accurate assessment of the translator and director's respective influence in the process of transforming a foreign-language text onto a local stage. The analysis also sheds light on the different approaches taken by speakers of German, and playwrights creating an English version from a literal translation. It pinpoints losses in translation and adaptation, and suggests how future versions may avoid these

Is dental amalgam safe for humans? The opinion of the scientific committee of the European Commission

Abstract It was claimed by the Scientific Committee on Emerging and Newly Identified Health Risks (SCENIHR)) in a report to the EU-Commission that "....no risks of adverse systemic effects exist and the current use of dental amalgam does not pose a risk of systemic disease..." [1, available from: http://ec.europa.eu/health/ph_risk/committees/04_scenihr/docs/scenihr_o_016.pdf]. SCENIHR disregarded the toxicology of mercury and did not include most important scientific studies in their review. But the real scientific data show that: (a) Dental amalgam is by far the main source of human total mercury body burden. This is proven by autopsy studies which found 2-12 times more mercury in body tissues of individuals with dental amalgam. Autopsy studies are the most valuable and most important studies for examining the amalgam-caused mercury body burden. (b) These autopsy studies have shown consistently that many individuals with amalgam have toxic levels of mercury in their brains or kidneys. (c) There is no correlation between mercury levels in blood or urine, and the levels in body tissues or the severity of clinical symptoms. SCENIHR only relied on levels in urine or blood. (d) The half-life of mercury in the brain can last from several years to decades, thus mercury accumulates over time of amalgam exposure in body tissues to toxic levels. However, SCENIHR state that the half-life of mercury in the body is only "20-90 days". (e) Mercury vapor is about ten times more toxic than lead on human neurons and with synergistic toxicity to other metals. (f) Most studies cited by SCENIHR which conclude that amalgam fillings are safe have severe methodical flaws.</p

Mutter matter:murmurings from the drawing room table

‘Mutter matter; murmurings from the Drawing Room Table’, a collaborative, site responsive installation by Jane Fox and Irene Mensah was developed and shown at Pitzhanger Manor House, London W5 in June/July 2012 at the invitation of curator Carol Swords. The work attends to the fragile and elusive nature of memory and the significance of domestic objects and materials in exploring and re-siting the past. It builds on the artists’ previous work ‘Mutter Matter - murmurings from the kitchen table’ selected by curator Celia Davies for ‘House’ festival, May 2011 at The Regency Town House, Brighton. The impetus for the work was two domestic books, each inherited from a grandmother: ‘Enquire Within Upon Everything’ 114th edition, 1929 inherited from Hilda Fox (Fox) and ‘Quotations and Recipes’ 1931, Wisbech, inherited from Lily Shaw (Mensah). The books provided a lens through which the artists explored their own and their grandmother’s lives via collisions of memory and material, and in conversation between themselves in an original re-working of recipes, texts and domestic convention, and in response to site. At Pitzhanger Manor, owned by the architect Sir John Soane from 1800 -1809, a new voice enters the dialogue – Dr William Kitchiner, Regency cook and author of ‘The Cook's Oracle’ (4th edition 1822). ‘Mutter matter; murmurings from the Drawing Room table’ draws on this and other traces from Soane family life to expand and hone the work, creating new collisions and resonances within its layers. The artists tread a careful collaborative path and test a range of shared creative strategies within which to hold and develop a multi faceted artwork. ‘Mutter matter; murmurings from the drawing room table’ was funded by Grants for the Arts, Arts Council England and supported by Wright’s Baking and PM Gallery and House

Ansatzpunkte für Nachsorgeprogramme für Reha-Aufenthalte bei Müttern mit Adipositas

    Get PDF
The development of Germany’s population continuously gaining weight over the last decades, lead to a problematic situation: Obesity is no longer just a medical but an important economical issue. Experts estimate the costs for German health insurances up to 214 billion Euros per year. Especially the share of women with children is notably represented among the group of obese people. Therefore this study examines the existing after-care following a rehabilitation treatment at mother-child-centers with its focus on obesity. According to the female patients a three week therapy has a positive effect on their general health situation. This short-term effect was verified by results of a study conducted by the author in June 2010. An average weight loss of 1.6 kg was noticed after a three week treatment with a sample of 21 women. Other studies have shown that long-term after-care programs can enhance these results and help patients and their families to adopt a healthier life style, including a healthy diet, physical activity or relaxation. However, despite these facts a survey conducted among 30 mother-child-rehabilitation-facilities revealed that only six of them offer a long-term mentoring or other forms of support following the three week program at the clinic and that a of the therapy’s result was neglected in many cases. The purpose of this study was to invest if. More over the author develops a list of key criteria for the success of the implementation of a long-term program for the mentoring of obese patients. Namely, these were among others “accessibility of the program”, “duration of mentoring” or “personal contact“
