Going Beyond Counting First Authors in Author Co-citation Analysis
The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation
counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings
are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that
only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into
account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed. Reasons for these effects are discussed
Variations on the Author
“Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship
Appropriate Similarity Measures for Author Cocitation Analysis
We provide a number of new insights into the methodological discussion about author cocitation analysis. We first argue that the use of the Pearson correlation for measuring the similarity between authors’ cocitation profiles is not very satisfactory. We then discuss what kind of similarity measures may be used as an alternative to the Pearson correlation. We consider three similarity measures in particular. One is the well-known cosine. The other two similarity measures have not been used before in the bibliometric literature. Finally, we show by means of an example that our findings have a high practical relevance.information science;Pearson correlation;cosine;similarity measure;author cocitation analysis
Design, synthesis, and biological evaluation of selective estrogen receptor modulator/histone deacetylase inhibitor hybrid molecules
Therapeutic options for the treatment of breast cancer have long been limited to invasive surgical procedures, or chemo- and radiotherapies that are plagued with detrimental side-effects. Selective estrogen receptor modulators (SERMs), such as tamoxifen and raloxifene, are a large subclass of molecules that have been used in adjuvant therapies and have provided substantial clinical benefits as preventative and long-term treatment options. Despite their success, endocrine therapies face several challenges including the development of endocrine resistance and, in the case of tamoxifen, the increased risk of endometrial cancer. Recent studies have shown that dual administration of SERMS and histone deacetylase inhibitors (HDACis) in vitro have led to cooperative effects such as increased drug potency, the resensitization of endocrine resistant cell lines, and an overall decrease in the risk of endometrial cancer development. In an effort to combine the cooperative effects of SERMs and HDACis, this project focuses on the design, synthesis, and biological evaluation of a series of hybrid SERM/HDACi molecules that combine the pharmacophores of both drug classes. The design of the hybrids was based on previous research projects investigating hybrid antiestrogens within the Gleason group. This project expanded on a promising subsetof 4-OHT based hybrids and began with in silico screening of a virtual library using the FORECASTOR docking platform. Seven compounds were chosen for synthesis and a previously devised route was unsuccessfully attempted using a McMurry cross-coupling as the key step. An alternate route to the triphenylethylene scaffold is hinged on a highly convergent and modular three-component coupling reaction was designed and successfully carried out. Four hybrids were synthesized and purified by preparatory HPLC prior to biological evaluation.Fluorogenic HDACi assays to determine the hybrid affinities of HDACs 3 and 6 were carried out by the author within the Gleason lab. Three of the four hybrids showed lowmicromolar HDAC 3 potencies (IC50) and the fourth showed submicromolar potency, and suggested that increased chain-lengths led to a higher degree of HDAC inhibition. HDAC 6 inhibition results trended similar to those of HDAC 3 and three of the four hybrids showed submicromolar potencies. Cell-based bioluminescence resonance energy transfer (BRET) and luciferase transactivation assays were conducted by the Mader lab at Université de Montréal to evaluate the ER affinity and antagonism profile of the hybrids. Each hybrid exhibited full antagonism against the ER and three of the hybrids showed submicromolar IC50 values with regards to their ER affinity. MCF-7 breast cancer cell growth curves were carried out and a single hybrid outperformed 4-hydroxytamoxifen (4-OHT), tamoxifen, and endoxifen. The ER data alongside the HDACi results are suggestive of a hybrid SERM/HDACi that was capable of eliciting a cooperative antiproliferative effect against the breast cancer cell line. The final chapter of this thesis presents a brief project regarding the design of a 3-acyl-1,5-diene substrate for the organocatalytic Cope rearrangement. A novel diazepanecarboxylate organocatalyst capable of catalyzing the Cope rearrangement of hindered aldehydes via iminium catalysis was recently reported by the Gleason group. DFT calculations suggested that iminium ion formation of 3-acyl-1,5-dienes would accelerate the Cope rearrangement. The final 3-acyl-1,5-diene substrate was shown to be capable of undergoing the Cope rearrangement under simple thermal conditions and future work will investigate the potential for organocatalytic rate acceleration.
I. A total synthesis of (-)-virosaine A and II. A direct approach for the synthesis of 1,3-disubstituted bicyclo[1.1.1]pentan-1-amines
Part I. The Securinega alkaloids are a fascinating class of bridged tetracyclic natural products that are produced exclusively by plants of the Phyllanthaceae family. These plants have a rich history in traditional folk medicine and the Securinega alkaloids themselves have been associated with a wide range of biological activities. Virosaine A is arguably the most structurally complex monomeric member of this alkaloid family. Specifically, it features an unprecedented caged pentacyclic skeleton that is comprised of a 7-oxa-1-azabicyclo[3.2.1]octane ring system, multiple bridged bicycles, and an isoxazolidine ring. The intriguing structural features and unique biosynthetic origin of virosaine A inspired us to pursue its total synthesis and Part I of this thesis details our efforts in accomplishing this goal. A novel bio-inspired cascade reaction sequence, involving sequential epoxide opening/[3+2] nitrone cycloaddition, was programmed into the synthesis as a strategy to rapidly construct the complex virosaine core. A robust route was developed to prepare the pivotal cascade precursor in enantiopure form, beginning with an enantioselective Diels-Alder reaction between furan and 2-bromoacrolein. Subsequent implementation of the key cascade reaction sequence facilitated access to the core of virosaine A in a total of only 5 steps. Several methods for C-H functionalization were applied to the virosaine core. These studies led to the formation of a diverse set of novel complex polycycles and provided valuable information on the differences in reactivity between several positions. Finally, a combination of NMR and computational analyses laid the groundwork for a successful directed lithiation/bromination strategy to selectively functionalize the desired position on the caged core. This selective late-stage functionalization enabled the total synthesis of virosaine A in a total of 10 steps and 9% overall yield. Part II. The bicyclo[1.1.1]pentane (BCP) motif is a unique bioisostere used in drug discovery for the replacement of phenyl rings, tert-butyl groups, and internal alkynes. Its incorporation into lead compounds has been shown to impart favorable physicochemical properties while also creating novel chemical space in the extremely competitive intellectual property landscape. Despite their utility, the synthesis of BCP-containing building blocks is, in many cases, quite challenging. This limits their potential use in drug discovery and highlights an important need to develop efficient methods for their preparation. While significant advances have recently been made in this area, there are currently no methods available to directly access 1,3-disubstituted bicyclo[1.1.1]pentan-1-amines. A collaboration between our group and Inception Sciences prompted the development of a novel approach for the synthesis of 3-alkyl bicyclo[1.1.1]pentan-1-amines, which is detailed in Part II of this thesis. This goal was motivated by their specific need to prepare a 1,3-disubstituted BCP building block for the preparation of a lead oncology clinical candidate. An efficient difunctionalization of [1.1.1]propellane was developed to access 3-allyl-N,N-dibenzylbicyclo[1.1.1]pentan-1-amine via a one-pot amination/allylation protocol. This reaction represented the first example of a 3-alkyl bicyclo[1.1.1]pentan-1-amine substrate being prepared from [1.1.1]propellane in a single step and its utilization enabled an efficient synthesis of Inception Sciences' building block. Finally, the scope of this reaction was extended to include the use of several alkyl iodide electrophiles. This enabled the synthesis of a diverse set of 3-alkyl bicyclo[1.1.1]pentan-1-amines.
Development of novel enantioselective diazepane carboxylate catalysts for the diels-alder reaction of α-substituted enals and studies investigating urea hydrogen bonding
Green chemistry has become an important topic due to current environmental concerns. Green chemistry is a broad term which generally refers to the development of chemical processes that are less toxic and more environmentally friendly. One important field of green chemistry, that has rapidly grown the last 20 years is organocatalysis, wherein the catalyst for a given chemical transformation is an organic molecule. This thesis describes two projects which have focused on organocatalysis and green chemistry. Part A involves investigations into urea hydrogen bond catalysis while Part B describes the development of a new type of enantioselective iminium ion organocatalyst for the Diels-Alder reaction. The first part describes our efforts into investigating optimal parameters for hydrogen bond catalyst systems that resulted from failed attempts to develop a new peptide coupling catalyst. It was hypothesized that a system capable of both activation of a carboxylate, through hydrogen bonding, and activation of an amine, via proximity to the carboxylate, might be competent in furnishing the desired bond. The screening for catalytic activity was unsuccessful and it was unknown what was limiting activity. A significant unknown was the optimal position of the nucleophile relative to the urea and we accordingly undertook investigations to probe this key parameter. A catalogue of general scaffolds consisting of a urea and linker to a hydroxyl nucleophile were synthesized and screened for their activity towards additions to an electrophile. Close proximity to the urea was found to be key and two acyl transfer agents were designed based on the results with one successfully employed as an additive in peptide coupling, affording activity equal to or slightly better than that of the current standard HOAt in combination with a coupling reagent. In the second part, we describe the development of an organocatalytic Diels-Alder cycloaddition using an achiral ethyl diazepane carboxylate catalyst developed in our lab. The catalyst showed good reactivity for alpha-substituted enals, a substrate class typically incompatible with iminium ion catalysts. We set out to investigate the reactivity and mechanism of this achiral diazepane for these challenging alpha-substituted enals, showing it was compatible with a wide range of enals which showed no activity with contemporary secondary amine catalysts. Efforts to reach high enantioselectivity proved challenging for this scaffold. The efforts focused on three areas: installation of bulky substituents on the ring, tuning the electron withdrawing group (including employing chiral electron withdrawing groups), and computational studies to provide insight into the origins of the selectivity. The conclusions of these studies led to a final diazepane carboxylate catalyst containing stereogenic centers on both the ring and electron withdrawing group. This catalyst displayed good reactivity and moderate to excellent exo-, regio- and enantioselectivity for a wide range of alpha-substituted alpha,beta-unsaturated aldehydes for the Diels-Alder cycloaddition.
Studies towards the total synthesis of palau'amine: Synthesis of trans-fused 5,5 azabicycles and the phakellin core
The pyrrole-imidazole alkaloids (PIAs) are a family of structurally related natural products isolated from several species of marine sponges. Palau’amine, a dimeric PIA, has been a target for total synthesis ever since its isolation in 1993 due to its significant biological activity and, more importantly, its complex molecular architecture. In 2007, the structure ofpalau’amine was revised to incorporate a highly strained trans-fused azabicyclo[3.3.0]octane bicyclic system. This thesis follows the progression of efforts that culminated in the development of a cyclization strategy for the formation of the strained trans-fused 5,5-bicyclic ring system. In our initial exploration, attempts to secure the trans-fused 5,5-ring system via previously established metal-mediated diamination protocols and amidyl radical cyclization were thwarted. Finally, the synthesis of the trans-fused core within a simple cyclic model systemwas enabled by the conjugate addition of a carbamate nitrogen to an ynoate which furnishes a trans-fused azabicyclo[3.3.0]octane bicyclic system containing an exocyclic α,β-unsaturated ester. The latter would act as a synthetic handle to construct the remaining right-hand side core of palau’amine. Two particular monomeric PIAs, phakellin and phakellstatin, greatly resemble palau’amine in that they share the same ABCD tetracyclic core. The application of our conjugate addition strategy afforded the pyrrolidine A-ring, containing an exocyclic α,β-unsaturated ester. To access the phakellstatin core, the elaboration of that synthetic handle to the vinyl urea was achieved via a Curtius rearrangement. A cyclofunctionalization reaction secured the pyrazinone B-ring affording the desired tricyclic carbinolamine precursor suitable for the synthesis of phakellstatin. To access the phakellin core, the exocyclic α,β-unsaturated ester was elaborated, via a Curtius rearrangement/aza-Wittig one-pot process, to an advancedintermediate containing the complete nitrogen and carbon framework of phakellin. The cyclofunctionalization reaction with the guanidine-containing intermediate secured the pyrazinone B-ring affording the desired tricyclic carbinolamine. Unfortunately, final attempts to close the guanidine C-ring and to complete the synthesis of the natural product werehowever unsuccessful.
Dispelling the Myths Behind First-author Citation Counts
We conducted a full-scale evaluative citation analysis study of scholars in the XML research field to explore just how different from each other author rankings resulting from different citation counting methods actually are, and to demonstrate the capability of emerging data and tools on the Web in supporting more realistic citation counting methods. Our results contest some common arguments for the continued
use of first-author citation counts in the evaluation of scholars, such as high correlations between author rankings by first-author citation counts and other citation
counting methods, and high costs of using more realistic citation counting methods that are not well-supported by the ISI databases. It is argued that increasingly available digital full text research papers make it possible for citation analysis studies to go beyond what the ISI databases have directly supported and to employ more
sophisticated methods
