3D fluorescence molecular tomography utilizing a novel SPAD camera
3D Fluorescence Molecular Tomography (FMT) is an important pre-clinical tool for quantifying and characterizing molecular markers for different diseases. Herein, we demonstrate, for the first time, 3D K-Space FMT reconstructions using a large time-gated SPAD camera, SwissSPAD2 (SS2). The 3D reconstruction results are obtained using traditional and Deep Neural Network-based inverse solvers. Moreover, the 3D reconstruction performance with SS2 is benchmarked against that obtained with a gated-ICCD camera. The reconstruction results obtained using SS2 are in good agreement with those obtained from the gated-ICCD.AQU
AI-enhanced rapid lifetime determination method for fast macroscopic and mesoscopic fluorescence lifetime imaging
Fluorescence lifetime imaging (FLI) offers several advantages, including the quantitative imaging of unique biosignatures in biological tissues, studies of drug-target interactions, and tumor margin identification. However, its adoption in surgical environments has been hindered by the time-consuming data acquisition and processing pipeline. In this work, we present an AI-enhanced rapid lifetime determination (RLD) method utilizing the SwissSPAD3 (SS3) detector, capable of acquiring a single frame to generate a large area FLI map. This method has been validated using fluorescence inclusions in tissue-mimicking phantoms, paving the way for complex in-vivo imaging. In conclusion, the AI-enhanced single-shot fluorescence lifetime estimation using the SwissSPAD3 detector represents a significant advancement in the RLD method, with promising potential for fluorescence-guided surgical applications.AQU
Mesoscopic light-sheet imaging set-up for 3D SWIR fluorescence intensity and NIR fluorescence lifetime imaging
Fluorescence lifetime imaging offers the unique ability to monitor the molecular status of the tissue microenvironment. However, the scattering nature of tissues remains a challenge for mesoscopic 3D high resolution imaging. We introduce the design of a mesoscopic light-sheet system with short-wave infrared (SWIR) intensity and near infrared (NIR) fluorescent lifetime imaging capabilities. This system enables 3D mesoscopic imaging of high-resolution probe biodistribution and fluorescent lifetime. The system was characterized and validated through phantom experiments, and further tested on tumor spheroids and ex vivo organ samples.AQU
Combining diffuse reflectance spectroscopy and ultrasound imaging for resection margin assessment during colorectal cancer surgery
Establishing adequate resection margins during colorectal cancer surgery is challenging. Currently, in up to 30% of the cases the tumor is not completely removed, which emphasizes the lack of a real-time tissue discrimination tool that can assess resection margins up to multiple millimeters in depth. Therefore, we propose to combine spectral data from diffuse reflectance spectroscopy (DRS) with spatial information from ultrasound (US) imaging to evaluate multi-layered tissue structures. First, measurements with animal tissue were performed to evaluate the feasibility of the concept. The phantoms consisted of muscle and fat layers, with a varying top layer thickness of 0-10 mm. DRS spectra of 250 locations were obtained and corresponding US images were acquired. DRS features were extracted using the wavelet transform. US features were extracted based on the graph theory and first-order gradient. Using a regression analysis and combined DRS and US features, the top layer thickness was estimated with an error of up to 0.48 mm. The tissue types of the first and second layers were classified with accuracies of 0.95 and 0.99 respectively, using a support vector machine model. Medical Instruments & Bio-Inspired Technolog
Going Beyond Counting First Authors in Author Co-citation Analysis
The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation
counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings
are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that
only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into
account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed. Variations on the Author
“Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship
Appropriate Similarity Measures for Author Cocitation Analysis
We provide a number of new insights into the methodological discussion about author cocitation analysis. We first argue that the use of the Pearson correlation for measuring the similarity between authors’ cocitation profiles is not very satisfactory. We then discuss what kind of similarity measures may be used as an alternative to the Pearson correlation. We consider three similarity measures in particular. One is the well-known cosine. The other two similarity measures have not been used before in the bibliometric literature.
