    Functional expression cloning reveals proapoptotic role for protein phosphatase 4

    © 2003 Nature Publishing Group All rights reservedFunctional expression cloning strategies are highly suitable for the analysis of the molecular control of apoptosis. This approach has two critical advantages. Firstly, it eliminates prior assumptions about the properties of the proteins involved, and, secondly, it selectively targets proteins that are causally involved in apoptosis control and which affect the crucial cellular decision between survival and death. The application of this strategy to the isolation of cDNAs conferring resistance to dexamethasone and italic gamma-irradiation resulted in the isolation of a partial cDNA for the catalytic subunit of protein phosphatase 4 (PP4). Cells transfected with this partial cDNA in an expression vector downregulated PP4 and were resistant to both dexamethasone and UV radiation, as demonstrated by both membrane integrity and colony-forming assays.     Communities of practice as a social theory of learning: A conversation with Etienne Wenger

    The aim of this article is to contribute to the understanding and use of the theory of communities of practice. In order to clarify terms, explore applications for education, and reflect on various critiques of the theory in the literature, two educational researchers conducted a series of interviews with the theorist Etienne Wenger-Trayner. The interviews have been thematically organised around key concepts from the theory. By relating the concepts to their uses in research and to other social theories, Wenger-Trayner clarifies key ideas of the theory including what constitutes a ‘community of practice’. He explains how he conceptualises identity and participation in order to develop a social theory of learning in which power and boundaries are inherent. The interviewers draw on these conceptual discussions with Wenger-Trayner to consider how the theory of communities of practice resonates with key debates and issues in education. By unpacking some key concepts of the theory from an educational perspective, we provide researchers with conceptual tools to support the complex decision-making that is involved in selecting the best and most appropriate theory or theories to use in their research

    Artiklen undersøger, hvordan sygeplejersker danner deres professionsidentitet gennem orienteringer ind og ud af forskellige praksisfællesskaber, som de møder igennem grunduddannelsen og de første møder med arbejdsmarkedet. Med inspiration fra Wenger-Trayner og Wenger-Trayner (2015) forstås profes-sionsidentitet som subjektive orienteringer i det landskab af praksisfællesska-ber, der tilsammen udgør sygeplejeprofessionen. Analysen er empirisk funderet i kvalitative, longitudinelle studier af en gruppe af sygeplejersker, der er blevet fulgt etnografisk i foreløbigt syv år. Analysen viser, hvordan sygeplejerskers professionsidentitet anno 2023 udvikler sig som bevægelser igennem et landskab af praksisfællesskaber. Analysen viser, hvordan professionsidentiteten formes af deltagelse inden for det enkelte prak-sisfællesskab, og at denne deltagelse rummer både engagerende og des-engagerende læreprocesser

    As comunidades de prática nas organizações da economia social e solidária em Portugal

    Fundamentado na sua experiência como gestora de formação profissional e voluntária em organizações portuguesas da economia social e solidária, a autora propõe que o desenvolvimento de uma abordagem estratégica para a aprendizagem contínua nestas organizações é incipiente ou inexistente. Com isto, estas organizações não estão a conseguir desenvolver o potencial humano e tecnológico necessário para uma ação concertada na sociedade do conhecimento. Esta dissertação explora a visão de que a aprendizagem contínua é inseparável do "modo de vir a estar" dos profissionais que trabalham nestas organizações. Baseada numa revisão da literatura relevante, a autora reflete sobre o facto das comunidades de prática serem uma abordagem adequada à forma de estar "aprendente" requerida no século XXI. O desenvolvimento de parcerias de aprendizagem e a criação de recursos coletivos são atividades organizacionais que poderiam ajudar a desenvolver as competências estratégicas das organizações, gerando um crescimento sustentável? Para investigar esta linha de pensamento foram realizadas treze entrevistas com gestores de organizações da economia social e solidária da região de Lisboa. Este grupo constituiu-se como uma comunidade de prática embrionária, dando um exemplo do potencial deste "modo de fazer as coisas". Através de um estudo de caso suportado na análise das entrevistas efetuadas aos membros daquele grupo, a autora reflete sobre se uma abordagem em comunidade de prática é adequada para criar competências estratégicas nas organizações da economia social e solidária em Portugal.Grounded in her experience as professional training manager and volunteer in Portuguese organizations, the author proposes that developing a strategic approach to continuous learning in organizations concerned with the social and solidarity economy in Portugal are weak or non-existent. What is more, these organizations are not developing the human or technological potential required for concerted action in a knowledge society. This dissertation explores the view that continuous learning is inseparable from a “way of becoming” for adults working in such organizations. Based on a review of the relevant literature, the author reflects on whether communities of practice are a strategy for the continuous learning required in the 21st century. Is the development of learning partnerships and creation of collective resources organizational activities that could help develop the organizational strategic competences and generate sustainable growth? To investigate this line of thought thirteen interviews were carried out with managers of organizations in the Lisbon region. This group was an embryonic community of practice, providing an example of the potential of this "way of doing things". Through a case study analyzing interviews of those in the group, the author reflects on whether a communities of practice approach is appropriate for creation of core competencies in social and solidarity economy organizations in Portugal

    The article explores how nurses form their professional identity through orientations in and out of different communities of practice that they encounter during their basic education and first encounters with the labour market. Inspired by Wenger-Trayner and Wenger-Trayner (2015), professional identity is understood as subjective orientations in the landscape of communities of practice that constitute the nursing profession. The analysis is empirically grounded in qualitative, longitudinal studies of a group of nurses who have been ethnographically followed for seven years so far. The analysis shows how nurses' professional identity in 2023 develops as movements through a landscape of communities of practice. The analysis shows how professional identity is shaped by participation within the individual community of practice, and that this participation involves both engaging and disengaging learning processes

    Isolation of genes controlling apoptosis through their effects on cell survival

    The identification of the most suitable molecular targets for gene and drug therapy is the crucial first step in the development of new disease treatments. The rational identification of such targets depends on a detailed understanding of the pathological changes occuring at the molecular level. We have applied forward genetics approaches to the identification of the critical genes involved in the control of apoptosis in mammalian cells, since defective control of apoptosis underlies many diseases, including cancer and neurodegenerative diseases. We have identified two groups of genes by their effects on cell survival using retroviral cDNA functional expression cloning and retroviral insertional mutagenesis. The identification of these novel genes opens up new areas for apoptosis research and subsequently for the development of new gene and drug therapies
