intermediates, that is, partially degraded proteins, cannot be distinguished from those of intact proteins by global proteomics
analysis, which quantify total protein abundance levels. This study aimed to develop a tool for assessing the aspects of degradation
regulation via proteolytic processing through a new multiplexed Nterminomics method involving selective isobaric labeling of protein N-termini and immunoaffinity capture of the labeled N-terminal peptides. Our method allows for not only identification of
proteolytic cleavage sites, but also highly multiplexed quantification of proteolytic processing. We profiled a number of potential
cleavage sites by signal peptidase and provided experimental confirmation of predicted cleavage sites of signal peptide. Furthermore, the present method uniquely represents the landscape of proteomic proteolytic processing rate during early embryogenesis in Drosophila melanogaster, revealing the underlying mechanism of stringent decay regulation of zygotically expressed proteins during early stages of embryogenesis.11Nsciescopu
The use of information sources by Internet users in answering questions
The purpose of this study was to investigate what kinds of sources people prefer to use when they answer questions online, especially, in the context of social Q&A. Social Q&A is a Web-based service, that allows people to ask questions and receive answers from their fellow users. In social Q&A, people often cite sources of information when they answer questions. It could be a name, a short description, or hyperlinks to the original sources. Yahoo! Answers was chosen for this study due to its popularity as a top ranked social Q&A service as well as its capability for separately indicating sources for the answers in its format. We collected data with a crawler that used Yahoo! Answers APIs. A total number of 5,391 sources were identified and analyzed with the following three approaches: (1) source distribution by online accessibility, (2) source distribution by genre, and (3) source distribution by subjects. At the early stage of this study, it was expected that the results of source preferences heavily relied on sources online, since people ask and answer questions on the Web-based service. Nevertheless, it was found that human (56.4%) was the most frequently cited type of source, and it was followed by online (40%) and offline sources (4%). According to the source distribution by genre, human (56.4%) was followed by the Internet (38.1%), books (3.6%), and mass media (1.6%), and the sub-categories of these sources were analyzed. Additionally, the patterns of source distribution were shown differently across subjects. The categories of Health, Home & Electronics, and Society & Culture relied heavily on human sources, while Computers & the Internet included most of the Internet-based sources of information.Peer reviewedIssue published online June, 3, 2009; article first published online June 1, 2009, according to publisher's website. Intrinsically disordered region-mediated condensation of IFN-inducible SCOTIN/SHISA-5 inhibits ER-to-Golgi vesicle transport
Newly synthesized proteins in the endoplasmic reticulum (ER) are sorted by coat protein complex II (COPII) at the ER exit site en route to the Golgi. Under cellular stresses, COPII proteins become targets of regulation to control the transport. Here, we show that the COPII outer coat proteins Sec31 and Sec13 are selectively sequestered into the biomolecular condensate of SCOTIN/SHISA-5, which interferes with COPII vesicle formation and inhibits ER-to-Golgi transport. SCOTIN is an ER transmembrane protein with a cytosolic intrinsically disordered region (IDR), which is required and essential for the formation of condensates. Upon IFN-γ stimulation, which is a cellular condition that induces SCOTIN expression and condensation, ER-to-Golgi transport was inhibited in a SCOTIN-dependent manner. Furthermore, cancer-associated mutations of SCOTIN perturb its ability to form condensates and control transport. Together, we propose that SCOTIN impedes the ER-to-Golgi transport through its ability to form biomolecular condensates at the ER membrane. © 2023 The Author(s)11Nsciescopu

Coordinate regulation of the senescent state by selective autophagy
Cellular senescence is a complex stress response implicated in aging. Autophagy can suppress senescence but is counterintuitively necessary for full senescence. Although its anti-senescence role is well described, to what extent autophagy contributes to senescence establishment and the underlying mechanisms is poorly understood. Here, we show that selective autophagy of multiple regulatory components coordinates the homeostatic state of senescence. We combined a proteomic analysis of autophagy components with protein stability profiling, identifying autophagy substrate proteins involved in several senescence-related processes. Selective autophagy of KEAP1 promoted redox homeostasis during senescence. Furthermore, selective autophagy limited translational machinery components to ameliorate senescence-associated proteotoxic stress. Lastly, selective autophagy of TNIP1 enhanced senescence-associated inflammation. These selective autophagy networks appear to operate in vivo senescence during human osteoarthritis. Our data highlight a caretaker role of autophagy in the stress support network of senescence through regulated protein stability and unravel the intertwined relationship between two important age-related processes.11Nsciescopu

Accessing and extracting knowledge from online documents is crucial for therealisation of the Semantic Web and the provision of advanced knowledge services.
The Artequakt project is an ongoing investigation tackling these issues to facilitate the creation of tailored biographies from information harvested from the web.
In this paper we will present the methods we currently use to model, consolidate and store knowledge extracted from the web so that it can be re-purposed as adaptive content. We look at how Semantic Web technology could be used within this process and also how such techniques might be used to provide content to be published via the Semantic Web
A study on educational development cooperation: Case analysis in Guatemala
This paper outlines the status of demand for educational ODA (Official Development Assistance) in Guatemala. It includes reviews of ODA references of major donor countries & international organizations, and interviews of experts carried out during field study. The paper analyzes cooperative educational development projects of major donor countries & international organizations. Germany and the Us are analyzed on a bilateral level while UNICEF is analyzed on a multilateral level. Based on the results of this analysis, it draws specific implications of South Korea’s ODA policies for providing educational ODA in Guatemala. <br /
