    The Role of a Neuron-Specific V-ATPase in Synapse Specification, Function, and Maintenance

    The Vacuolar-type (V-) adenosine triphosphatase (ATPase) is a proton-pumping nanomachine consisting of two multi-subunit, reversibly associating protein sectors, the cytosolic V1 sector and the membrane-bound Vo sector. The V1/Vo holoenzyme hydrolyses ATP to translocate protons across biological membranes thereby modulating lumenal and extracellular pH. Additionally, accumulating evidence suggests that the Vo sector has a role in membrane fusion when dissociated from the V1 sector, its proton-pumping partner. Published evidence for this includes a null allele for the neuron-specific a subunit in the Drosophila Vo sector, v100, which leads to defects in synaptic function that are unrelated to pH regulation. My project emerged from the need to explain why v100 has two additional phenotypes that are absent in other synaptic function mutants: functional and structural degeneration in photoreceptor cells and patterning defects in the visual system neuropil. I proposed that v100 has a previously undocumented role on a neuron-specific endo/lysosomal pathway in addition to its documented role in neurotransmitter secretion. To test my hypothesis in the context of only one of the two purported v100 functions, I generated transgenic animals with v100 mutations designed to specifically disrupt either acidification or membrane fusion. Using these genetic tools, I discovered that v100 has an essential role in sorting cargo into an endo-lysosomal pathway that concomitantly requires v100 for the acidification-dependent maturation of degradation-competent organelles. This 'sort-and-degrade' mechanism for v100 defines a neuron-specific degradation pathway that is required for synaptic specification, function, and maintenance. In developmental stages, v100 is required to 'sort-and-degrade' guidance receptors as part of the synapse specification program. In the adult, the 'sort-and-degrade' mechanism provides additional degradative capacity to neurons, a cell type that must often maintain homeostasis for unusually long periods of time. Finally, I provide evidence that the role for V100 in membrane fusion requires a direct, physical interaction with Syntaxin-1, an interaction that can be specifically disrupted in vitro and in vivo. In brief, my results provide mechanistic insight into the acidification-independent role of v100 and reveal the existence of a neuron-specific endo-lysosomal pathway on which v100 functions to 'sort-and-degrade' cargo in order to meet the special needs of a neuron in development, function, and maintenance

    Functional Impact of Preleukemic Mutations on the Human Hematopoietic Stem and Progenitor Cell Compartment

    Dissecting functional consequences of gene mutations contributes to understanding disease development and identifying potential treatment strategies. Mutations in epigenetic regulators (especially in DNMT3A, TET2, and ASXL1, termed DTA mutations) have been identified in acute myeloid leukemia patients, as well as individuals without history of hematologic malignancies. Mutations in these genes are hypothesized to confer a clone a competitive advantage, leading to preferential expansion of this specific clone. DTA mutations are proposed to be preleukemic events that are acquired before leukemia onset and are thought to elevate the risk for acquisition of additional mutations that in turn drive malignant transformation. Clones harboring these mutations are able to survive chemotherapy, potentially increasing the risk for disease recurrence. In this thesis, I investigated the impact of DTA mutations in the human hematopoietic stem and progenitor cell compartment, as well as the role of preleukemic events in patients with AML t(8;21). Using CRISPR/Cas9, site-specific mutations were introduced in CD34+ progenitor cells freshly isolated from umbilical cord blood samples. By delivering Cas9 nuclease together with an in vitro transcribed sgRNA as ribonucleoprotein, I was able to achieve editing efficiencies ranging from 40% for the sgRNA targeting exon 13 of ASXL1 to over 90% for the sgRNA targeting exon 6 of TET2. A known hotspot mutation in DNMT3A R882H was introduced with an efficiency of up to 50% in the cell bulk. After successful genetic modification, cells were subjected to in vitro assays to assess phenotype, self-renewal properties, differentiation capacities, and long-term culture-initiating potential. Clonal composition and expansion were assessed via deep sequencing and subsequent analysis of introduced specific insertions and deletions. To investigate the role and importance of preleukemic mutations in AML leukemogenesis, a comprehensive genetic in-depth characterization of AML t(8;21) was performed. To this aim, DNA from the time of diagnosis was analyzed via next generation sequencing with a targeted gene panel including exons and coding regions of 66 genes recurrently mutated in hematologic malignancies. From 56 patients, samples from clinical remission were analyzed for the previously identified mutations by targeted re-sequencing of amplicons spanning the target region. In this thesis, I was able to demonstrate that mutations in DNMT3A, TET2, and ASXL1 influence the behavior of hematopoietic stem and progenitor cells (HSPCs) in a gene-specific fashion. Mutations in ASXL1 had only little effect on HSPCs with no observed changes in expression of differentiation markers after culture for three weeks and no enhanced proliferation in long-term culture. The only hint that ASXL1 mutations have an impact on the fitness of HSPCs was increased colony forming ability after long-term culture, meaning that although progenitor cells did not proliferate during the long-term culture period, they were able to sustain and generate colonies later. DNMT3A mutations led to a longer retention of CD34 marker expression in differentiation culture experiments, but not enhanced serial replating capacity. TET2-mutated cells, in contrast, showed delayed myeloid marker expression in short-term culture and increased self-renewal of committed progenitor cells. Both, DNMT3A and TET2 mutations led to clonal expansion of distinct cell clones during long-term culture with a competitive advantage over wild type cells. The clonal composition of TET2- and DNMT3A-mutated samples differed, with TET2mut samples being more diverse. Collectively my data indicate that DNMT3A mutations preferentially influence primitive HSC clones, whereas TET2 mutations enhance the fitness of committed progenitor cell clones. ASXL1 mutations might need additional events to drive clonal expansion. Analysis of patient samples revealed that mutations in DNMT3A and TET2 are early events in AML t(8;21) and potentially acquired before the onset of the disease. ASXL1 mutations are neither early nor late events in these patients but are potentially cooperating events in leukemia transformation. In this thesis, I was able to demonstrate that DTA mutations enhance the fitness of distinct human hematopoietic stem or progenitor cell clones, in terms of self-renewal and proliferative capacity, leading to clonal expansion. The results obtained here highlight the importance of monitoring specific preleukemic events to prevent disease recurrence as well as development of severe hematologic conditions.Um die Entwicklung von Krankheiten aufzudecken und etwaige zielgerichtete Behandlungsstrategien zu entwickeln, ist es essenziell den funktionellen Einfluss bestimmter Genmutationen zu untersuchen. Mutationen in epigenetischen Regulatoren wie DNMT3A, TET2 und ASXL1 (auch DTA-Mutationen genannt) wurden sowohl in Patienten mit akuter myeloischer Leukämie als auch in Patienten ohne hämatologische Erkrankung gefunden. Diese Mutationen führen zu einem Vorteil bestimmter Zellklone, die daraufhin expandieren. DTA-Mutationen werden auch als prä-leukämische Mutationen bezeichnet, die vor dem Ausbruch der Leukämie bereits in Stamm- und Vorläuferzellen vorkommen und ein erhöhtes Risiko für das Auftreten weiterer Mutationen bedingen. Zusätzliche Mutationen in einem prä-leukämischen Klon können wiederum zur malignen Transformation des Zellklons führen. Des Weiteren können Zellen mit prä-leukämischen Mutationen resistent gegen Chemotherapie sein und somit das Risiko eines Rezidivs erhöhen. In dieser Arbeit habe ich den Einfluss von Mutationen in DNMT3A, TET2 und ASXL1 auf humane hämatopoetische Stamm- und Vorläuferzellen untersucht, sowie die Rolle von prä-leukämischen Mutationen in Patienten mit AML t(8;21) erforscht. Mit Hilfe von CRISPR/Cas9 wurden spezifische Mutationen in CD34-positiven Vorläuferzellen eingeführt, die frisch aus Nabelschnurblut isoliert wurden. Die Cas9-Nuklease wurde zusammen mit in vitro-transkribierter sgRNA in Form von Ribonukleoproteinen in die Zellen gebracht. In Exon 13 von ASXL1 wurden Insertionen und Deletionen in mindestens 40% der Zellen generiert und in Exon 6 des Gens TET2 konnten in mehr als 90% der Zellen Insertionen und Deletionen entdeckt werden. Die spezifische Mutation DNMT3A R882H wurde in bis zu 50% der transfizierten Zellen eingeführt. Die erfolgreich mutierten Zellen wurden anschließend in in vitro-Experimenten untersucht in Hinsicht auf Veränderungen im Phänotyp, der Fähigkeit zur Selbsterneuerung, Differenzierungsfähigkeit und der Fähigkeit sich in Langzeitkulturen zu vermehren. Die klonale Zusammensetzung, sowie die Expansion bestimmter Zellklone wurden mit Hilfe von Deep Sequencing und anschließender Analyse der generierten Insertionen und Deletionen untersucht. Patienten mit AML t(8;21) wurden auf prä-leukämische Mutationen untersucht, indem DNA vom Diagnosezeitpunkt via Next-Generation Sequenzierung analysiert wurde. Kodierende Regionen von 66 Genen, die wiederholt in malignen hämatologischen Erkrankungen auftreten, wurden in die Analyse eingeschlossen. Von 56 Patienten wurden DNA-Proben vom Zeitpunkt der klinischen Remission erneut auf die bereits identifizierten Mutationen untersucht. In dieser Arbeit konnte ich zeigen, dass Mutationen in DNMT3A, TET2 und ASXL1 unterschiedliche Effekte auf hämatopoetische Stamm- und Vorläuferzellen haben. Mutationen in ASXL1 hatten den mildesten Effekt. Weder Unterschiede in der Expression von Differenzierungsmarkern noch erhöhte Replatierungsfähigkeit in Vergleich zu Wildtypzellen wurden beobachtet. Des Weiteren zeigten die Zellen kein vermehrtes Wachstum in der Langzeitkultur. Sie waren jedoch in der Lage Kolonien nach der Langzeitkultur zu bilden, was bedeuten könnte, dass diese Zellen trotz des unveränderten Proliferationsverhaltens einen Vorteil gegenüber Wildtypzellen haben und somit in der Lage sind die Langzeitkultur zu überdauern. Zellen mit DNMT3A Mutationen exprimierten den Vorläuferzellmarker CD34 länger als Kontrollzellen. Mutationen in TET2 führten zu einer verspäteten Expression von myeloischen Differenzierungsmarkern und einem erhöhten Selbsterneuerungspotential von Vorläuferzellen. Sowohl in Zellen mit DNMT3A- als auch mit TET2-Mutationen konnte ich die Expansion bestimmter mutierter Zellklone nachweisen. Die Proben mit eingeführten Mutationen in TET2 hatten jedoch eine größere Anzahl verschiedener Insertionen und Deletionen als Proben mit DNMT3A-Veränderungen. Die Daten dieser Arbeit zeigen, dass Mutationen in DNMT3A dazu führen, dass primitive Zellklone einen evolutionären Vorteil erhalten, wohingegen reifere Vorläuferzellen von Mutationen in TET2 profitieren. Mutationen in ASXL1 führen möglicherwiese nur in Anwesenheit weiterer genetischer oder zellulärer Veränderungen zu klonaler Expansion. In Patienten mit AML t(8;21) treten Mutationen in DNMT3A und TET2 wahrscheinlich früher auf als andere Mutationen und kommen möglicherweise in Vorläuferzellen vor, bevor sich die eigentliche Erkrankung entwickelt. Mutationen in ASXL1 scheinen weder frühe noch späte Ereignisse zu sein, sondern treten irgendwann dazwischen auf. Sie kooperieren möglicherweise mit anderen Mutationen, um Zellen zu transformieren. In dieser Arbeit konnte ich zeigen, dass DTA-Mutationen die Fitness von bestimmten Stamm- und Vorläuferzellen im menschlichen Stammzellkompartiment erhöhen und so zu einer Expansion der mutierten Zellklone führen. Die Ergebnisse dieser Arbeit bestätigen, dass die Untersuchung und Beobachtung prä-leukämischer Klone dazu beitragen könnte, ein Rezidiv oder sogar die Entstehung schwerer hämatologische Erkrankungen zu verhindern

    Das MYCN-Onkogen als Marker für minimale Resterkrankung und therapeutisches Ziel beim Neuroblastom

    Neuroblastoma, the most common extracranial solid childhood cancer, arises from precursors of the developing sympathetic nervous system. MYCN oncogene amplification is a determinant of high risk and occurs in ~25% of neuroblastomas. Despite intensive treatment, more than half these patients succumb to their disease, implying persistence of therapy-resistant MYCN-amplified minimal residual neuroblastoma cells. This thesis proposes a comprehensive concept for the specific diagnostic detection of the MYCN amplicon and evaluates new treatment options for MYCN-amplified neuroblastoma. Disease-relevant nucleotide changes, structural gene rearrangements and copy number alterations were detected in tumor material by next-generation sequencing of a customized hybrid capture-based targeted panel. Unique MYCN amplicon breakpoints in the rearranged gene constitute a target sequence for a personalized minimal residual disease (MRD) PCR diagnostic. MYCN amplicon breakpoints in neuroblastoma cell lines and tumors were identified and recovered by individual, semi-quantitative PCR assays and Sanger sequencing. The assay was further developed for highly sensitive, real-time quantitative and droplet digital PCR detection for selected MYCN breakpoints in cell lines. MRD level detected in bone marrow aspirates collected during therapy outlined different disease courses in patients, including MRD persistence until relapse and good response to the first treatment course. Combining multi-agent chemotherapy in current high-risk protocols with indirect MYCN inhibitors provides a potential route to improve poor cure rates for MYCN-amplified neuroblastomas. Different hyperactive biological networks in MYCN-amplified neuroblastoma were tackled using small molecule inhibitors of the bromodomain and extra-terminal (BET) domain-containing protein BRD4, phosphoinositide 3-kinase (PI3K) and polo-like kinase 1 (PLK1). BET (JQ1, OTX015 and TEN-010) and kinase (alpelisib, volasertib and rigosertib) inhibitors demonstrated anti-cancer activity by diminishing viability in cell line-based drug screens at nanomolar to low micromolar concentrations. Rigosertib treatment altered PLK1 and PI3K signaling and strongly impaired the cellular ability for wound healing and colony formation. In line with in vitro observations, rigosertib reduced tumor growth in patient-derived neuroblastoma xenografts in mice. Combining OTX015 and volasertib produced synergistic anti-tumor responses in two MYCN-amplified neuroblastoma cell lines. To prevent MYCN-driven proliferation of tumor cells, further indirect MYCN targets are also being considered. This is exemplified by a substrate of PLK1, ASPM, which is elevated in MYCN-amplified primary neuroblastomas. Knockdown of ASPM, a microtubule-associated protein involved in mitotic spindle assembly, in MYCN-amplified neuroblastoma cell lines reduced viability and proliferation, accompanying a neuronal differentiation phenotype with neurite-like outgrowth, cytoskeletal changes and increased expression of differentiation markers. This study presents clinical implementable molecular diagnostics to pinpoint unique MYCN-amplified neuroblastoma cells within non-invasively accessible biopsy material, and proposes indirect small molecule-based MYCN therapies and potentially new drug targets for a personalized treatment.Das Neuroblastom, der häufigste extrakranielle solide Krebs im Kindesalter, entsteht aus Vorläuferzellen des sich entwickelnden sympathischen Nervensystems. Eine Amplifikation des MYCN-Onkogens ist ein bestimmender Faktor für ein hohes Risiko und tritt bei ~25% der Neuroblastome auf. Trotz intensiver Behandlung erliegt mehr als die Hälfte dieser Patienten ihrer Krankheit, was die Persistenz therapieresistenter, MYCN-amplifizierter minimaler Restneuroblastomzellen impliziert. Diese Arbeit stellt ein umfassendes Konzept für den spezifischen, diagnostischen Nachweis des MYCN-Amplikons vor und evaluiert neue Behandlungsoptionen für MYCN-amplifizierte Neuroblastome. Krankheitsrelevante Nukleotidveränderungen, strukturelle Genrearrangements und Kopienzahl-veränderungen wurden im Tumormaterial mit Hilfe eines maßgeschneiderten, zielgerichteten hybrid-capture-basierten Next Generation Sequencing (NGS) Assays nachgewiesen. Einzigartige MYCN-Amplikon-Bruchpunkte im rearrangierten Gen stellen eine Zielsequenz für eine personalisierte PCR-Diagnostik der minimalen Resterkrankung (MRD) dar. MYCN-Amplikon-Bruchpunkte in Neuroblastom-Zelllinien und Tumoren wurden durch individuelle, semi-quantitative PCR-Assays und Sanger Sequenzierung identifiziert und wiedererkannt. Der Assay wurde für den hochsensitiven, quantitativen Echtzeit- und digitalen Tröpfchen-PCR-Nachweis für ausgewählte MYCN-Bruchpunkte in Zelllinien weiterentwickelt. Die MRD Level, die in den während der Therapie gesammelten Knochenmarkaspiraten nachgewiesen wurden, skizzierten die verschiedenen Krankheitsverläufe bei den Patienten, einschließlich der MRD-Persistenz bis zum Rezidiv und des guten Ansprechens auf den ersten Behandlungsabschnitt. Die Kombination der Multi-Wirkstoff-Chemotherapie in den aktuellen Hochrisikoprotokollen mit indirekten MYCN-Inhibitoren stellt einen möglichen Weg dar, die schlechten Heilungsraten für MYCN-amplifizierte Neuroblastome zu verbessern. Verschiedene, hyperaktive biologische Netzwerke in MYCN-amplifizierten Neuroblastomen wurden mit niedermolekularen Inhibitoren der Bromdomäne und des extra-terminalen (BET) domänenhaltigen Proteins BRD4, der Phosphoinositid-3-Kinase (PI3K) und der polo-ähnlichen Kinase 1 (PLK1) behandelt. BET (JQ1, OTX015 und TEN-010) und Kinase-Inhibitoren (Alpelisib, Volasertib und Rigosertib) zeigten eine krebshemmende Wirkung, indem sie die Viabilität in zelllinienbasierten Wirkstoff-Screens bei nanomolaren bis niedrigen mikromolaren Konzentrationen verminderten. Die Behandlung mit Rigosertib veränderte die PLK1- und PI3K-Signalübertragung und beeinträchtigte die zelluläre Fähigkeit zur Wundheilung und Koloniebildung stark. In Übereinstimmung mit In-vitro-Beobachtungen reduzierte Rigosertib das Tumorwachstum in von Patienten stammenden Neuroblastom-Xenografts bei Mäusen. Die Kombination von OTX015 und Rigosertib erzeugte synergistische antitumorale Aktivität in zwei MYCN-amplifizierten Neuroblastom-Zelllinien. Um die MYCN-gesteuerte Proliferation von Tumorzellen zu verhindern, werden weitere indirekte MYCN-Targets in Betracht gezogen. Ein Beispiel hierfür ist ein Substrat von PLK1, ASPM, das in MYCN-amplifizierten, primären Neuroblastomen erhöht ist. Das Herunterregulieren von ASPM, einem Mikrotubuli-assoziierten Protein, das an der mitotischen Spindelanordnung beteiligt ist, führte in MYCN-amplifizierten Neuroblastom-Zelllinien zu einer verminderten Viabilität und Proliferation, was mit einem neuronalen Differenzierungsphänotyp mit neuritenartigem Auswuchs, zytoskelettalen Veränderungen und erhöhter Expression von Differenzierungsmarkern einherging. Diese Studie stellt eine klinisch umsetzbare, molekulare Diagnostik vor, um einzigartige MYCN-amplifizierte Neuroblastomzellen in nicht-invasiv zugänglichem Biopsiematerial zu detektieren, und schlägt indirekte, niedermolekular-basierende MYCN-Therapien und potenziell neue Zielmoleküle für eine personalisierte Krebsbehandlung vor

    Pattern formation as a neural wiring strategy during development of the Drosophila visual system

    Natural patterns are everywhere around us and can greatly support the developmental progress of complex organs. The eye of a fruit fly (Drosophila melanogaster ) is such a perfectly patterned organ. The 800 small eyes, called ommatidia, contain six motion-vision photoreceptors (PRs) each. Via axons, PRs transfer environmental information to the first optic ganglion in the brain; the lamina. The six photoreceptors from neighboring ommatidia that see one point in the environment are connected to one post-synaptic target in the lamina, to form a retinotopic map of the surroundings. Neural superposition sorting systematically sorts PR axons during a series of distinct developmental steps. Initially, PR axons extend in bundles from the ommatidia to the lamina plexus (LP) and form a scaffold, or pre-pattern. Scaffold formation is an essential step in creating a functional visual map, yet, how it establishes is largely unknown. From this scaffold, all six PRs from one ommatidium extend their axonal growth cones in different angles during a lateral extension phase towards their different post-synaptic targets. How PR growth cones determine this sub type specific angle is largely unknown. Finally, extended growth cones adhere and connect with Lamina Neurons (LNs) at the target location to form synapses and a functional visual circuit. Whether LNs are required for axon extension and/or adherence at the target location is unknown. The aim of this thesis is to describe underlying mechanisms involved in pattern formation and neural superposition sorting during development of the visual map, as well as to investigate the role of LNs during superposition sorting. The results presented in this thesis show that PR growth cones can form a scaffold in absence of LNs. Furthermore, the extension of photoreceptors is independent of the presence of LNs the presentation of adhesion proteins on their cell membrane. Strikingly, it is possible for extending growth cones to find their target correctly and, in rare cases, form photoreceptor clusters where they would normally form a cartridge with LNs. This suggests that photoreceptor and LN dynamics are largely independent and that neural superposition sorting is remarkably robust. I conclude that LNs are required in late development of the visual map for robust circuit formation, but do not contribute to PR sorting. This is a novel finding for the wiring of the Drosophila visual system.Widerkehrende Strukturen und Muster sind überall in der Natur zu finden und können während der Entwicklung einen wichtigen Einfluss spielen. Ein perfekt strukturiertes Organ ist das Auge der Fruchtfliege, Drosophila melanogaster. Es besteht aus etwa 800 repetitiven Untereinheiten, den so genannten Ommatidien, welche jeweils sechs bewegungssensitive Photorezeptoren (PRs) besitzen. Diese leiten Umgebungsinformationen über Axone zum ersten optischen Ganglion, der Lamina, weiter. Die sechs Photorezeptoren von benachbarten Ommatidien, die denselben Raumpunkt wahrnehmen, sind mit dem selben postsynaptischen Zellen in der Lamina verbunden um eine retinotope Karte der Umgebung zu arrangieren. Die Axone von Photorezeptoren werden durch neuronale Superposition-Sortierung in einer Reihe von Entwicklungsschritten geordnet. Zunächst wachsen die Axone der Photorezeptoren in Bündeln von den Ommatidien zum Lamina Plexus (LP) und formen dabei ein eindeutiges Grundgerüst oder ‘pre-pattern‘. Die Bildung dieses Grundgerüsts stellt einen essenziellen Schritt in der Formierung einer funktionellen visuellen Karte dar. Wie genau das Gerüst etabliert wird, ist jedoch größtenteils unbekannt. Nach Bildung des Grundgerüsts wachsen alle sechs PRs eines Ommatidiums in einer lateralen Wachstums-Phase. In dieser wachsen die Wachstumskegel der PRs in unterschiedlichen, subtyp-spezifischen Winkeln zu deren jeweiligen neuronalen Partnern. Wie hierbei die Wachstumskegel der Photorezeptoren ihre spezifischen Winkel bestimmen ist weitestgehend unbekannt. Letztlich verbinden sich Wachstumskegel der Photorezeptoren mit Lamina Neuronen (LNs) an ihren Zielpositionen, formen Synapsen und so einen funktionellen visuellen Schaltplan. Ob LNs für axonales Wachstum und/oder Adhäsion von Wachstumskegeln an ihrer Zielposition nötig sind, ist nicht bekannt. Das Ziel dieser Studie ist die grundlegenden Mechanismen, die in der Entwicklung einer funktionellen visuellen Karte notwendig sind, zu untersuchen. Hierbei werden die Mechanismen, die für die Grundgerüstformierung und neuronale Superpositions-Sortierung notwendig sind, sowie die Rolle von LNs während dieser, analysiert. Die hier präsentierten Ergebnisse zeigen, dass Wachstumskegel von Photorezeptoren sogar in Abwesenheit von Lamina Neuronen ein Grundgerüst bilden können. Zudem ist das laterale Wachstum der Photorezeptoren unabhängig von der Laminar Neuronen Präsenz oder deren Membran-Adhäsionsproteinen. Überraschenderweise ist es ausdehnenden Wachstumskegeln möglich ihr Ziel korrekt zu finden und in seltenen Fällen sogar Photorezeptor-Cluster zu bilden, wo normalerweise “cartridges” mit LNs geformt würden. Het visuele systeem van een fruitvliegje (Drosophila melanogaster) is zo’n orgaan dat ontwikkelt met gebruik van patronen. Eén oog bevat ongeveer 800 individuele minioogjes, zogenaamde ommatidia, die elk zes bewegingssensitieve fotoreceptoren bevatten. Via axonen geven zij informatie door aan de eerste hersenlaag in het visueel systeem; de lamina. Zes fotoreceptoren van naburige ommatidia, die allen één punt in de ruimte zien, worden verbonen met één cluster van post-synaptische partners en vormen een visuele kaart. Het sorteren van axonen via “neuron superpositie sorteren”, doorloopt verschillende ontwikkelingsstadia. Eerst sturen fotoreceptoren hun axonen in bundels van het oog naar de lamina plexus (LP) waar ze een pre-patroon vormen. Deze stap is essentieel voor de vorming van de visuele kaart, maar hoe het wordt gevormd is grotendeels onbekend. Vanaf het gevormde pre-patroon groeien zes fotoreceptoren uit één ommatidium in zes verschillende richtingen naar hun verschillende post-synaptische targets. Hoe ze deze groeirichting bepalen is onbekend. Tenslotte komen axon extensies samen bij een Lamina Neuron (LN) target en vormen ze synapsen en daarmee een werkend visueel circuit. Of LNs een rol spelen tijdens extensie, of voor de vorming van connecties, is onbekend. Het doel van deze thesis is het beschrijven van onderliggende mechanismen in neuron superpositie sorteren tijdens de ontwikkeling van het visuele system van Drosophila en te onderzoeken of LNs nodig zijn voor dit proces. De resultaten laten zien dat photoreceptoren een pre-patroon kunnen vormen in afwezigheid van LNs en dat hun extensies onafhankelijk zijn van de aanwezigheid van LNs of van moleculen op het celmembraan. Ook is het voor fotoreceptoren mogelijk om vanuit correcte extensies in sommige gevallen een cluster te vormen met andere fotoreceptoren ipv. met LNs.     Brp which forms part of the CAZ functions in the release of neurotransmitters by establishing a close proximity of Ca2+ ion channels with synaptic vesicles and formation of T bars. Reducing Brp levels specifically in KCs via genetically targeted RNA-interference [RNAi; (Wagh et al., 2006) impairs the formation of aversive ARM (Knapek et al., 2011). Furthermore, Histone Deacetylase 6, which influences transmitter release from neuromuscular AZs by shaping the spatial arrangement of Brp also participates in ARM (Perry et al., 2017). This thesis however, performed circuit analysis of brp effect on ARM. Brp effect on Drosophila olfactory aversive ARM learning showed a broader effect than reported earlier on in a separate study (Knapek et al., 2011). Brp effect on the KC-DANs-MBONs synapses showed a recurrent loop activity that existed in the circuits. The recurrent activity in the loop required a concurrent activation of the glutamatergic MBONs and depolarization of the dopaminergic neuron in order to gate the activity of the presynaptic plasticity. Lastly, the presynaptic activity may involve two dopaminergic receptors i.e. D1 and D2. Equilibration between D1R and D2R are very pivotal in formation of ARM. The thesis laid down the foundation of possible mechanism of ARM. Henceforth, thorough research on the specificity of the Kenyon cells and MBONs synapses coupled with cross-talk between DANs- MBONs connections need to be addressed in relation to their memory valence.Brp, ein Teil der CAZ-Funktion, ist mitverantwortlich für die Freisetzung von Neurotransmittern, indem es eine enge Verbindung mit den Ca2+ Ionenkanälen herstellt und zusammen mit synaptischen Vesikeln T-Stäbe erzeugt.     Regulierung der sensorischen Mechanotransduktion zur Linderung neuropathischer Schmerzen

    Neuropathic pain is caused by lesion or disease of the somatosensory nervous system. It is a multifactorial condition with common symptoms and disease mechanisms (allodynia and hyperalgesia) that causes severe suffering in patients and a high burden to society. We wanted to gain a deeper understanding of disease contributions of the peripheral nervous system with the goal to develop new analgesic substances. We performed chronic constriction injury (CCI) on experimental animals which produced robust mechanical hypersensitivity. Gait analysis was established to provide an additional experimenter-independent read-out for mechanical hypersensitivity following neuropathic injury. Light- and electron microscopy were used to quantify the damage subjected to the peripheral nerve. An ex-vivo skin nerve preparation from the glabrous skin of experimental animals was used to characterize mechanoreceptors and nociceptors in neuropathic skin and uncover possible sensitization effects linking behavioural data with nerve recordings and therefore compile an integrative dataset. STOML3 is upregulated after nerve injury, therefore I additionally investigated the effects of in vivo overexpression of STOML3 on sensory mechanotransduction, as well as stoml3 conditional deletion specifically in sensory neurons. I found that the physiological properties of low-threshold mechanoreceptors (LTMRs) are not altered following CCI, except a diminished static phase activity in slowly-adapting mechanoreceptors (SAMs). However, sensitization following neuropathic injury could be shown in C-fibers, displaying a significant reduction in transduction thresholds as well as emerging dynamic-phase coding properties that resulted from the injury. These alterations in physiological properties could explain in part the phenomenon of mechanical allodynia. Stimuli that would under normal circumstances not induce activity in the nociceptive C-fiber system, now do so following CCI. Following neuropathic injury proportionately more nociceptors are activated by low intensity mechanical stimuli therefore nociceptive input plays a more pronounced role in the overall sensory barrage into the spinal cord in animals with neuropathic pain. The in vivo overexpression of STOML3 has immense effects on the physiological properties of mechanoreceptors and nociceptors. LTMRs (RAMs and SAMs) were clearly sensitized and again C-fibers displayed prominent changes in their mechanosensitivity. Polymodal C-fibers displayed marked sensitization phenotypes: increased activity, lowered mechanical thresholds as well as dynamic-phase coding properties emerged after induced STOML3 overexpression. CMs increased their activity as well but only for the largest suprathreshold force stimuli. Overall, the overexpression of STOML3 in sensory neurons phenocopied the physiological changes emerging following CCI. Animals acquired mechanical hypersensitivity and the changes in the C-fiber nociceptive system were very similar to neuropathic injury. STOML3 is essential for the emerging sensitization of C-fibers following neuropathic injury proven by genetic ablation of stoml3 in sensory neurons. C-fibers recorded from conditional knockouts did not display elevated physiological activity (CMs) or showed intermediate phenotypes (polymodal C-fibers) following CCI. Interestingly, polymodal C-fibers showed increased activity immediately (<1s) after a mechanical stimulus, however in CMs this physiological abnormality was completely abolished. STOML3 seems to be essential for CM sensitization after neuropathic injury and for polymodal C-fibers partially responsible in producing the sensitization phenotype. Finally, I showed that sensitized C-fibers acquired the ability to follow low frequency (5Hz) vibrations. In some cases, to the extent of phase-locking, and in some cases with less accuracy. High stimulation strengths were necessary to induce activity in CMs in the conditional knockouts, polymodal C-fibers lacking STOML3 did not respond to this type of stimulation at all, further evidence that STOML3 is important for onset activity, that STOML3 is essential for peripheral sensitization following neuropathic injury and that the changes in C-fiber physiology might be the driver for mechanical hypersensitivity displayed by experimental animals after induced neuropathy

    Molecular mechanisms downstream of Neurod family transcription factors involved in mouse corticogenesis

    Die Erweiterung des grundlegenden Verständnisses der molekularen Mechanismen der Entwicklung, Organisation und Funktion des Neokortex ist ein Schlüssel zur Erforschung von Entwicklungsstörungen des Gehirns und zur Erarbeitung neuer Therapieansätze. Hier untersuchten wir die molekularen Mechanismen stromabwärts von Transkriptionsfaktoren der Neurod-Familie in der neokortikalen Entwicklung der Maus. Neurod1, Neurod2 und Neurod6 sind zentrale Regulatoren der neuronalen Differenzierung, Spezifikation und Axonführung. In dieser Arbeit zeigen wir, dass Neurod-Transkriptionsfaktoren über ein Zwischenmolekül den Tbr2-Transkriptionsfaktor unterdrücken, um eine ordnungsgemäße neuronale Migration und Differenzierung sicherzustellen. Die verlängerte ektopische Tbr2-Expression stört die neuronale Migration, Somagröße und dendritische Verzweigung in embryonalen und postnatalen Perioden. Wir untersuchen den genetischen Crosstalk zwischen Neurod1/2/3- und WWP1/2/miR-140-Wegen zu ihrem gemeinsamen nachgeschalteten Ziel Tbr2 und zeigten, dass diese Wege unabhängig voneinander auf Tbr2 konvergieren. Wir identifizieren neue nachgeschaltete Ziele von Neurod-Transkriptionsfaktoren wie Kcnq3, Bhlhe22 und Prdm8 und demonstrieren ihre entscheidende Rolle bei der richtigen Corpus Callosum-Etablierung. Darüber hinaus wird in dieser Forschung ein In-vitro-System zur Untersuchung der Callosom-Axon-Führung entwickelt und etabliert. Wir stellen fest, dass die Sh3gl2-Expression unter der Kontrolle von Neurod-Transkriptionsfaktoren steht und die Eliminierung von Sh3gl2 zu einer verzögerten Bestimmung des Zellschicksals führt. Wir nehmen an, dass die beeinträchtigte Sh3gl2-Expression in Neurod2/6 dKO- und Neurod1/2/6 tKO-Mutanten die Clathrin-unabhängige Endozytose und die anschließende Internalisierung von Membranrezeptoren stört, was zu einer Störung der Cortex-Zytoarchitektur führt.Expanding the fundamental understanding of the molecular mechanisms of neocortex development, organization, and function is a key to investigating brain developmental disorders and elaborating new therapeutic approaches. Here, we studied the molecular mechanisms downstream of Neurod family transcription factors in mouse neocortical development. Neurod1, Neurod2, and Neurod6 are pivotal regulators of neuronal differentiation, specification, and axon guidance. In this study, we demonstrated that Neurod transcription factors via an intermediate molecule, repress the Tbr2 transcription factor to ensure proper neuronal migration and differentiation. The prolonged ectopic Tbr2 expression disrupts neuronal migration, soma size and dendritic branching in embryonic and postnatal periods. We investigated the genetic crosstalk between Neurod1/2/3 and WWP1/2/miR-140 pathways to their common downstream target Tbr2 and showed that these pathways converge on Tbr2 independently of each other. We identified new downstream targets of Neurod transcription factors such as Kcnq3, Bhlhe22, and Prdm8, and demonstrated their crucial role in the proper Corpus Callosum establishment. Moreover, an in-vitro system for investigation of the callosal axons guidance was developed and established in this research. We detected that Sh3gl2 expression is under Neurod transcription factors control and the Sh3gl2 elimination results in the delayed cell fate specification. We hypothesize that impaired Sh3gl2 expression in Neurod2/6 dKO and Neurod1/2/6 tKO mutants disrupts the clathrin- independent endocytosis and subsequent membrane receptors internalization, that leads to disturbance of cortex cytoarchitecture

    Anthropocene is characterised by significant, and in many ways outcompeting natural processes, the impact of human activities on natural ecosystems. The excessive emission of greenhouse gases caused by global-scale use of fossil fuels, anthropogenic land-use change, and drastic decrease in pristine areas, have all contributed to global climate change. Although the Earth’s climate has historically fluctuated, the current situation is novel as climatic change is occurring at an unprecedented rate and has mainly been driven by human behaviour. The rapid pace of environmental change forces living organisms to respond fast in order to survive and reproduce. The magnitude of such responses might be especially prominent in migratory species due to their diverse and dispersed habitat requirements. Migratory species connect habitats across the globe, coupling biodiversity and ecosystem services. By travelling long distances, migratory species move biomass between habitats; transporting energy, nutrients, and other organisms. Also, migratory species alter the dynamics of resident communities that they connect on their journeys by foraging and becoming prey in numerous habitats during their annual cycle. Therefore environmental alteration leading to disruption of long-distance migratory behaviour will have far-reaching consequences which may drastically affect synchrony and unity in natural ecosystems. The aim of this dissertation was to reveal climate-driven changes to populations of migratory bats, from a historical overview through to future perspectives. To understand the magnitude of ongoing changes in populations of bats, we require adequate reference points from the past. Here, I had a unique opportunity to study recent changes in migratory behaviour and distribution of common European migratory bat species, Nyctalus noctula, based on long term dataset. In the presented work, I combined different approaches such as stable hydrogen isotopes to reveal migratory destinations; analysis of demographic data to evaluate sex and age population dynamics; respirometry measurements to establish physiological parameters and environmental modelling to predict suitable for hibernation areas under three climate change scenarios. This variety of applied technics allowed for complex investigation of animal responses to climate change. The obtained results are reported in two peer-reviewed publications and one manuscript, which is in preparation for submission. The research outcome aims to bridge the most demanding gaps in knowledge related to climate-driven changes in populations of migratory animals. The first chapter contributed to the improvement of the method for studying migratory animals by using stable isotopes. The second chapter was dedicated to climate change responses, particularly to an investigation of the mechanism of climate-driven range changes in populations of migratory bats. The third chapter described and evaluated the novel mechanistic approach in environmental modelling of species distribution under the impact of climate change. In chapter one, I have contributed to the validation of the assumptions for stable hydrogen analysis. Stable isotopes are broadly applied for the identification of the geographical origin of bats via transfer function, which is consists of the isotope signature of animals of know origin. However, there was no age validation performed before for stable isotope signatures in bat tissues. The stable isotopes approach was planned as the main method for the assignment of migratory bats in a long-term dataset. Therefore, in chapter one, I presented a validation approach for age differences in stable isotope signatures to better understand variation around the transfer function and increase the precision in the prediction of the geographic origin. In chapter two using a transfer function, which included individuals of both age classes, I investigated migratory behaviour and demographic structure of N. noctula under the distribution shift. The common noctule bats increased their winter range by more than 500 km during several decades. Apart from the observed biogeographic pattern little was known regarding the mechanism relevant for the range change. I hypothesized that the generation shift is the mechanism relevant to the range change in migratory bats. To test the hypothesis, I looked at the long-term data set regarding the demographic structure of winter colonies in the recently occupied winter range and I used stable hydrogen analysis to establish the geographic origin of bats. My finding supported the hypothesis regarding the generational shift in migratory behaviour of bats which resulted in winter range change. Obtained results, for the first time, demonstrated the relevance of generation shift in the regulation of species distribution in mammals. Chapter three was dedicated to the elaboration of a mechanistic physiologically-based model for the evaluation and forecasting of suitable hibernation areas in migratory bats. Climate change is expected to affect the cost of hibernation in a given area and might ultimately modify species winter distribution. With a novel approach, I aimed to combine fine-scale thermoregulation data of noctule bats with environmental variables from their past and present winter ranges to model species occurrence dynamics. Firstly, this joint approach improved our understanding of species physiological requirements to external conditions. Secondly, it enabled us to refine the modelling of future species distribution under different climate change scenarios. The presented work had revealed climate-driven changes in migratory behaviour and distribution of common noctule bats. By incorporation various technics and methodological approaches, I could tackle the magnitude of responses to climate change on different levels of ecological organisation: from an organismal physiological response to environmental conditions; to change in migratory behaviour on a population level and change in geographic distribution on a species level. The main findings elaborate on physiological and behavioural mechanisms underlying climate-driven range change and provide evidence for migratory bats being able to respond fast to the rapid pace of climate change. However, the consistency of responses is conditioned by species-specific traits such as physiological flexibility, high mobility and dispersion rate, short generation life, and diversity of migratory strategies; and climate velocity, as a metric of speed, and direction of climate displacement

    Rab GTPases as mediators of neuronal robustness in Drosophila

    One common feature of all eukaryotic cells is the presence of various specialized organelles, separated by membranes, which necessitates a coordinated trafficking of materials between these subcellular membrane-bound compartments. Especially neurons, with their long lifespan, polarized and often complex morphology, as well as their specialized functions, have particular requirements for membrane trafficking. Not surprisingly, membrane trafficking is involved in all aspects of neuronal development, function, and long-term maintenance. The evolutionary conserved family of small Rab GTPases functions as key regulator of coordinated vesicular trafficking in the endomembrane system. Expression profiling efforts revealed that in Drosophila half of all Rab GTPases are enriched in or specific to the nervous system, in humans it is one-third. However, the exact functions of the majority of nervous system-enriched Rab proteins are still unknown. Thus, studying the individual roles of those Rab GTPases more closely provides a great opportunity to gain more insight into the membrane trafficking networks in neurons. Ultimately, this will surely contribute to the understanding of what keeps neurons and in particular synapses healthy and functional over extended periods of time. In the nervous system, Rab GTPases and the membrane trafficking events these mediate have been widely associated with many neurodegenerative diseases. However, the established relations are often more correlatively than causatively linked, as discussed in Manuscript 1. Regarding the importance of an intact intracellular trafficking machinery for the development as well as neuronal function and maintenance, I primarily focused on the systematic functional analysis of nervous system-enriched Rab GTPases in Drosophila during my doctoral work. Previously, no systematic rab mutant characterization in any multicellular organism had been performed. The analysis, presented in Manuscript 2, revealed that the homozygous mutants of all nervous system-enriched Rab GTPases, raised under laboratory conditions, are viable and fertile, whereas, null mutants of ubiquitously expressed Rabs are all lethal under homozygosity. Thus, suggesting that Rab proteins, with high expression in the nervous system, serve more modulatory, specialized functions which are not essential for the survival of the organism. Further, we could show that all viable rab mutants differentially affect the development or neuronal function under variable, more challenging environmental conditions, such as temperature and light. This highlights the evolved robustness of developmental processes and nervous system function towards varying conditions. Additionally, during the in-depth functional analysis of nervous system-enriched Rab26, we revealed a stimulus-dependent role in the trafficking of the single acetylcholine receptor subunit Dα4 at cholinergic synapses of outer photoreceptors. However, we could not verify a role for Rab26 in the autophagic turnover of synaptic vesicles in neurons. Additional assays, such as the RUSH system, can be useful to support functional analyses. While this acute, synchronous release system could be established for Rabs in developing photoreceptors and salivary glands, wing imaginal discs proofed to be more sensitive and no working conditions could be established. Using the RUSH assay, different release dynamics with ‘fast’ as well as ‘slow’ releasing Rab GTPases could be identified as shown in Manuscript 3. Further, two nervous system-enriched Rab proteins, namely Rab23 and Rab26, show a clear re-localization from the cell body to the axon terminals, which is in agreement with their predominant synaptic neuropil localization revealed by expression profiling. In conclusion, the findings made during my doctoral work will contribute to a better understanding of the functional requirements of neurons regarding Rab-mediated membrane trafficking. The complete rab null mutant collection as well as the RUSH and LAMA transgenic toolbox provide a strong basis for further investigations of individual Rab functions during development and homeostasis

    Die Rolle von Piccolo in der zerebellären Netzwerkorganisation und die Ätiologie der pontozerebellären Hypoplasie Typ 3

    Piccolo, a presynaptic active zone protein, is known for its role in the regulated assembly and function of vertebrate synapses. Genetic studies suggest a link to several psychiatric disorders as well as Pontocerebellar Hypoplasia Type 3 (PCH3). A Piccolo knockout rat model was recently generated by means of transposon mutagenesis (Pclogt/gt) [116]. During my doctoral studies, I sought to characterise this rat model. In doing so, I took an exploratory approach by beginning to examine overall changes in brain anatomy, narrowing the focus to the level of the network, synapse and the ultrastructure of the presynaptic terminal in the hope of elucidating some of the mechanisms involved in PCH3. I found that the loss of Piccolo leads to a dramatic reduction in brain size compared to wildtype (Pclowt/wt) animals, attributed to a decrease in the size of the cerebral cortical, cerebellar and pontine regions. Moreover, the maturation of mossy fibre (MF) afferents, expression of the α6 GABAA receptor subunit at mossy fibre-granule cell synapses and the innervation of Purkinje cells by cerebellar climbing fibres are all perturbed in Pclogt/gt cerebella. Ultrastructural and functional studies revealed a reduced size and complexity of MF boutons, with fewer synaptic vesicles and changes in synaptic transmission. These data imply that Piccolo is required for the normal development, maturation and function of neuronal networks formed between the brainstem and cerebellum. Consistently, behavioural studies demonstrated that adult Pclogt/gt rats display impaired motor coordination, despite adequate performance in tasks that reflect muscle strength and locomotion. Together these data suggest that loss of Piccolo function in patients with PCH3 could be involved in many of the observed anatomical and behavioural symptoms, and that the further analysis of these animals could provide fundamental mechanistic insights into this devastating disorder.Piccolo, ein Protein der präsynaptischen aktiven Zone, ist bekannt für seine Rolle im regulierten Aufbau und in der Funktion von Wirbeltier-Synapsen. Hierbei legte ich den Schwerpunkt auf die Ebene des Netzwerks, der Synapse und der Ultrastruktur des präsynaptischen Nervenendes, in der Hoffnung, einige der an PCH3 beteiligten Mechanismen aufzuklären. Fand ich heraus, dass der Verlust von Piccolo zu einer dramatischen Verringerung der Hirngröße im Vergleich zu Wildtyp-Tieren (Pclowt/wt) führt, was auf eine Abnahme der Größe der Hirnrinde, des Kleinhirns und des Pons zurückzuführen ist. Darüber hinaus sind die Reifung der Moosfaser-Afferenzen, die Expression der α6 GABAA-Rezeptor-Untereinheit an den Synapsen zwischen Moosfasern und Körnerzellen und die Innervation der Purkinjezellen durch cerebellare Kletterfasern in Pclogt/gt Cerebella gestört. Ultrastrukturelle und funktionelle Studien ergaben eine reduzierte Größe und Komplexität der Moosfaser-Synapsen, mit weniger synaptischen Vesikeln und Veränderungen in der synaptischen Übertragung. Diese Daten implizieren, dass Piccolo für die normale Entwicklung, Reifung und Funktion der neuronalen Netzwerke, die zwischen Hirnstamm und Kleinhirn gebildet werden, erforderlich ist. Übereinstimmend haben Verhaltensstudien gezeigt, dass erwachsene Pclogt/gt Ratten, trotz adäquater Leistung bei Aufgaben, die Muskelkraft und Fortbewegung widerspiegeln, eine beeinträchtigte motorische Koordination aufweisen. Zusammenfassend deuten diese Ergebnisse darauf hin, dass der Verlust der Funktion von Piccolo bei Patienten mit PCH3 an vielen der beobachteten anatomischen und verhaltensbezogenen Symptome beteiligt sein könnte und dass die weitere Analyse dieser Tiere grundlegende mechanistische Erkenntnisse über diese verheerende Erkrankung liefern könnte
