    Anti-tumor effect of docosahexaenoic acid : involvement of microRNAs and TNFα

    L’acide docosahexaénoïque (DHA) est un acide gras polyinsaturé oméga-3 avec des propriétés anti-inflammatoires et anti-tumorales. L’effet du DHA dans le cadre du cancer colorectal pourrait être la conséquence d'une action anti-proliférative directe sur les cellules cancéreuses et de sa capacité à réduire l’inflammation propice au développement de la tumeur. Le Tumor Necrosis Factor-alpha (TNFa) est une cytokine pro-inflammatoire et présente des effets paradoxaux. En fonction du contexte cellulaire, le TNFa activera une voie de signalisation dépendante de la kinase RIP1 engageant la cellule cancéreuse vers la prolifération ou la mort cellulaire. Notre objectif fut d'évaluer le rôle du TNFa dans l'effet anti-prolifératif du DHA sur des cellules cancéreuses coliques et de préciser les mécanismes moléculaires régulant l'expression de cette cytokine. Le DHA induit l'expression de TNFa et sa sécrétion par les cellules cancéreuses. Nous avons montré que des anticorps neutralisant l'action autocrine du TNFa sur les cellules cancéreuses prévenait l'effet pro-apoptotique du DHA et abolissait l'effet anti-cancéreux observé dans des souris nude avec tumeurs HCT-116 sous régime DHA. L’induction de l'expression de TNFa par le DHA prend son origine à un niveau post-transcriptionnel par la répression du microARN miR-21 perdant sa capacité à dégrader l'ARNm TNFa. Le DHA par l'activation des kinases AMPKa et RIP1 déclenche la translocation nucléaire du facteur de transcription FOXO3a se fixant sur le promoteur miR-21 et diminuant l’expression de ce microARN. Nos travaux mettent en évidence un nouveau mécanisme moléculaire soutenant l'action anti-tumorale du DHA.Docosahexaenoic acid (DHA) is an omega-3 polyunsaturated fatty acid with anti-inflammatory and anti-tumoral properties. The anti-tumor effect of DHA in colorectal cancer might be attributed to direct anti-proliferative action on cancer cells and to its ability to reduce inflammatory status involved in tumor growth. Tumor Necrosis Factor-alpha (TNFa) is an inflammatory cytokine with paradoxical effect in cancer biology. According to the cellular context, TNFa activates RIP1 kinase dependent signaling pathway leading to proliferation or cell death. Our aim was to evaluate the role of TNFa in anti-proliferative effect of DHA in colon cancer cells and to precise the molecular mechanisms regulating TNFa expression.DHA treatment increased TNFa expression and secretion by cancer cells. We have shown that neutralization of autocrine TNFa action prevented the pro-apoptotic effect of DHA colon cancer cells and abolished anti-cancer effect in tumor HCT-116 bearing nude mice fed a DHA-enriched diet. Induction of TNFa expression by DHA occured at post-transcriptional level through microRNA miR-21 repression reducing its ability to induce TNFa mRNA degradation. DHA activates AMPKa and RIP1 kinases triggering nuclear translocation of the transcription factor Foxo3a which bound to miR-21 promoter and repressed the microRNA expression. Our works highlight a new molecular mechanism supporting the anti-cancer action of DHA

    Going Beyond Counting First Authors in Author Co-citation Analysis

    The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed.     “Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship

    Appropriate Similarity Measures for Author Cocitation Analysis

    We provide a number of new insights into the methodological discussion about author cocitation analysis. We first argue that the use of the Pearson correlation for measuring the similarity between authors’ cocitation profiles is not very satisfactory. We then discuss what kind of similarity measures may be used as an alternative to the Pearson correlation. We consider three similarity measures in particular. One is the well-known cosine. The other two similarity measures have not been used before in the bibliometric literature.     Role of lingual CD36 in lipid oro-perception and feeding behavior

    La modification de notre comportement alimentaire a impacté les apports lipidiques alimentaires, qui sont aujourd'hui de plus de 40% dans les pays occidentaux. La consommation excessive de lipides favorise l'apparition de pathologies comme l'obésité. Cette préférence gustative viendrait du fait que le gras, comme les autres modalités gustatives, serait détecté par le biais de récepteurs spécifiques. L'équipe NUTOX a démontré que les récepteurs CD36 et GPR120 sont exprimés sur les papilles linguales. Ils jouent un rôle dans la perception oro-sensorielle des lipides alimentaires, respectivement dans la détection des acides gras lors de la mastication et dans la régulation post-ingestive du repas. De plus, les études menées par l'équipe ont confirmé que cette perception est diminuée chez des sujets obèses car CD36 est soit internalisé à l'intérieur des papilles gustatives, soit faiblement exprimé. En conséquence, la majorité des sujets obèses préfèrent un repas riche en lipides et en consomment en grande quantité. Dans ce contexte, l'équipe à synthétisé des leurres lipidiques (« Fat-Taste Modifiers »), agonistes de CD36 et GPR120, plus puissants que les acides gras naturels qui permettent de diminuer le degré d'obésité chez des souris nourries par un régime hyper-lipidique. L'objectif de cette thèse est de démontrer les mécanismes d'action de leurres lipidiques (NKS-3 & NKS-5) et leurs interactions avec les récepteurs lipidiques. Afin de démontrer l'importance de CD36 dans la perception orale des lipides alimentaires, un modèle de souris exprimant CD36 uniquement dans les papilles gustatives « CD36 KO lingual » a été développé. Ces dernières et des souris sauvages seront nourries soit avec un régime normo-lipidique, soit rendues obèses par un régime hyper-lipidique (35% d'huile de palme). De plus, l'implication de la libération de peptides/hormones de l'axe gastro-entérique chez ces souris sera étudiée. Les peptides satiétogènes CCK (cholécystokinine), PYY3-36, GLP-1 (glucagon-like peptide-1) et leptine seront dosés dans le sang de l'animal. Le microbiote (intestinal et buccal) ainsi Les paramètres métaboliques tel que test de tolérance au glucose, insulinémie, etc. seront également réalisés sur ces molécules. Les études sur le circuit de la récompense dans le cerveau et sur le dynamisme de l'interaction moléculaire des leurres lipidiques avec CD36 et GPR120 se feront en partenariat.The modification of our dietary behavior has impacted the dietary fat intake, which is now more than 40% in Western countries. Excessive consumption of lipids favors the appearance of pathologies such as obesity. This taste preference would come from the fact that fat, like other taste modalities, would be detected through specific receptors.The information on the quality of the food ingested (taste) is then sent to the brain via the central nervous system (CNS). The NUTOX team demonstrated that CD36 and GPR120 receptors are expressed on the lingual papillae. They play a role in the oro-sensory perception of food lipids, respectively in the detection of fatty acids during mastication and in the post-ingestive regulation of the meal. Moreover, studies conducted by the team confirmed that this perception is diminished in obese subjects because CD36 is either internalized within the taste buds or weakly expressed. As a result, the majority of obese subjects prefer a high-fat meal and consume large amounts of it. In this context, the team has synthesized fat-taste modifiers, agonists of CD36 and GPR120, more potent than natural fatty acids, which reduce the degree of obesity in mice fed a high-fat diet. The objective of this thesis is to demonstrate the mechanisms of action of lipid decoys (NKS-3 & NKS-5) and their interactions with lipid receptors. In order to demonstrate the importance of CD36 in the oral perception of food lipids, a mouse model expressing CD36 only in the taste buds "CD36 KO lingual" has been developed. These mice and wild type mice will be fed either a normo-lipidic diet, or made obese by a hyper-lipidic diet (35% palm oil). In addition, the involvement of gastroenteric axis peptide/hormone release in these mice will be studied. The satiety peptides CCK (cholecystokinin), PYY3-36, GLP-1 (glucagon-like peptide-1) and leptin will be measured in the blood of the animal. The microbiota (intestinal and oral) as well as metabolic parameters such as glucose tolerance test, insulinemia, etc. will also be performed on these molecules. Studies on the reward circuit in the brain and on the dynamics of the molecular interaction of lipid lures with CD36 and GPR120 will be done in partnership

    En dépit des efforts déployés, l'incidence de l'obésité est en augmentation dans le monde entier où la consommation d‘aliments moins coûteux et hyperénergétiquesriches en matières grasses, représente la principale cause. Des études récentes ont révélé l'existence d'un goût de gras et que l'envie de le satisfaire pourrait être responsable de l‘excès des prises alimentaires et de l'attirance pour les aliments riches en gras. Différentes stratégies ont été utilisées pour réduire la teneur engraisse des aliments ou pour la remplacer par des mimétiques et des substituts de graisse. Dans cette thèse, nous avons non seulement exploré des mécanismes de signalisation tels que le rôle des canaux TRPC3 Ca2+ dans les cellules du bourgeon gustatif, mais nous avons également tenté de rechercher de nouvelles classes de molécules artificielles et naturelles, d‘origines végétales, qui pourraient agir comme exhausteurs de goût ou comme activateurs du goût. Sur la base des résultats obtenus par des études biochimiques et comportementales au cours de la thèse actuelle, nous proposons que Zizyphine purifié à partir de Zizyphus lotus, d‘acide oléanolique et de TUG891 (un agoniste chimique de GPR120) puissent être un candidat novateur et prometteur pour le traitement et la prévention de l'obésité et du syndrome métabolique qui lui est associé.Despite efforts, the incidence of obesity is on a rise throughout the world andconsumption of less expensive fat rich and high energy diet remains the major cause.Recent studies have successfully unveiled the existence of a fat taste, and that theurge to satisfy it might be responsible for the overeating behaviour and attractiontowards fat rich foods. Different strategies have been used to reduce fat content offood or replace it with fat mimetics and fat replacers. In this dissertation, we have not only explored signalling mechanisms like role of TRPC3 Ca2+ channels in taste bud cells but also tried to search for newer classes of molecules from botanical and chemical origin that might either act as fat taste enhancers, or fat taste activators. Based on the result of various biochemical and behavioural studies during the current dissertation, we propose that Zizyphin purified from Zizyphus lotus,Oleanolic acid and TUG891 (a chemical agonist of GPR120), might be novel andpromising candidates for the treatment and prevention of obesity and associatedmetabolic syndrome

    Rôle de la leptine dans la modulation de la préférence gustative pour les corps gras chez la souris

    Malgré les efforts déployés, l'incidence de l'obésité est en augmentation dans le monde entier. La consommation d'aliments moins chers, riches en graisses et riches en énergie reste la principale cause d'obésité. Des études récentes ont dévoilé avec succès, l'existence d'un goût gras. L'envie de satisfaire, cela pourrait être responsable du comportement de suralimentation et de l'attirance pour les aliments riches en graisses. Différentes stratégies ont été utilisées pour réduire la teneur en matières grasses. Dans cette thèse, des études biochimiques et comportementales ont permis d'explorer le rôle de la leptine dans la modulation de la préférence gustative pour les corps gras chez la souris. Les tests de préférence double choix et de licking ont été utilisés pour évaluer la perception gustative lipidique chez la souris. La microscopie confocale a été utilisée pour démontrer la colocalisation des récepteurs du goût gras (CD36 et GPR120) et de la leptine dans les cellules des papilles gustatives de souris. Pour explorer le rôle de la leptine dans la perception oro-sensorielle des acides gras alimentaires, nous avons utilisé des techniques de RT-qPCR, de signalisation calcique, d'ELISA et de western blot. Nous avons émis le postulat suivant : la leptine, libérée du micro-environnement des papilles de la langue, a un rôle inhibiteur dans la prise alimentaire. De plus, l'injection intrapéritonéale de leptine augmente la sensibilité au goût gras chez les souris ob/ob, génétiquement obèses. L'augmentation de cette sensibilité chez ces souris utilisant de la leptine, pourrait être une nouvelle étude qui aide au traitement et à la prévention de l'obésité et du syndrome métabolique associé.La leptine réduit la phosphorylation de la signalisation ERK1/2 induite par l'acide linoléique et augmente la libération du calcium intracellulaire dans les cellules des papilles gustatives isolées des souris ob/ob.The incidence of obesity is on the rise throughout the world. The intake of a fat-rich and high-energy diet remains one of the major causes of obesity. Recent studies have successfully unveiled, the existence of a gustatory cue, i.e., taste for fat. This gustatory perception might result into overeating behavior. In this dissertation, we used several biochemical and behavioral approaches to explore the role of leptin, an anorectic peptide that controls eating behavior via its action on different areas in the brain. The two-bottle preference and licking tests were used to assess the fat taste perception in mice. Confocal microscopy was used to demonstrate the colocalization of fat taste receptors (CD36 and GPR120) with leptin receptor in mice taste bud cells. To explore the role of leptin in the oro-sensory perception of dietary fatty acids, we used RT-qPCR, calcium signaling, ELISA, and western blotting techniques. We conducted our experiments on isolated taste bud cells, TBC (ex-vivo studies), or on different mice models like genetically modified obese (ob/ob) or diet-induced obese (DIO) mice (in vivo studies) in order to explore the implication of leptin in the modulation of oro-sensory perception of dietary long-chain fatty acids. The in-vitro studies on cultured TBC show that leptin, released from these cells into the micro-environment of papillae of the tongue, exerts an inhibitory action on fat preference. Surprisingly, administration of exogenous leptin into mice increased preference for fat in ob/ob mice, but not in DIO mice. Our results show that leptin may control fat eating behavior via its local action on taste cells, probably in an autocrine manner, in the taste papillae, and also via its peripheral action where this hormone upregulates this phenomenon in oob/obce. Finally, we can state that leptin might regulate preference for fat at different levels

    Dispelling the Myths Behind First-author Citation Counts

    We conducted a full-scale evaluative citation analysis study of scholars in the XML research field to explore just how different from each other author rankings resulting from different citation counting methods actually are, and to demonstrate the capability of emerging data and tools on the Web in supporting more realistic citation counting methods. Our results contest some common arguments for the continued use of first-author citation counts in the evaluation of scholars, such as high correlations between author rankings by first-author citation counts and other citation counting methods, and high costs of using more realistic citation counting methods that are not well-supported by the ISI databases. It is argued that increasingly available digital full text research papers make it possible for citation analysis studies to go beyond what the ISI databases have directly supported and to employ more sophisticated methods
