    Representative samples of drill core were collected from each of the five main areas drilled on the TAG (Trans-Atlantic Geotraverse) mound during Leg 158 (Humphris, Herzig, Miller, et al., 1996). In this report, we present the results of chemical analyses of 66 samples previously analyzed for Cu, Fe, Zn, Pb, Ag, and Cd by atomic absorption during Leg 158. Data are presented for an additional 38 elements plus total sulfur, loss on ignition, and the rare earth elements by a combination of optical emission spectrometry (ICP-ES), mass spectrometry (ICP-MS), and neutron activation (INAA). These data are discussed in detail in other chapters in this volume

    AbstractVitamin D is an essential micronutrient, playing a vital role in the regulation of calcium homeostasis and bone metabolism. In recent years, vitamin D has been increasingly recognized for its regulatory functions in immunomodulation, anti-inflammation, cellular proliferation and differentiation. The pandemic vitamin D deficiency has been associated with an increasing risk for several metabolic diseases. However, the effects of vitamin D deficiency and its supplementation on the underlying molecular mechanisms of these diseases are poorly understood. This thesis focuses on the effect of vitamin D on the regulation of molecular and metabolic signaling pathways involved in adipose tissue inflammation and hepatic insulin resistance.Study I examined the effect of vitamin D on the downregulation of inflammatory signaling pathways in adipose tissue. Vitamin D inhibited the translocation of the transcriptional factor NFκB as well as inhibiting the expression of inflammatory cytokines in human adipocytes differentiated from mesenchymal stromal cells (MSCs) and mature adipocytes isolated from intra-abdominal fat biopsies.In Study II, induction of vitamin D deficiency via a deficient diet impaired systemic glucose homeostasis and insulin sensitivity in lean and obese mice. Subsequent vitamin D re-supplementation restored the insulin sensitivity and ameliorated the hepatic tissue inflammation and oxidative stress in vitamin D deficient lean but not obese mice.In Study III, lower vitamin D levels were associated with a higher incidence of the metabolic syndrome in a cross-sectional cohort of older subjects living at latitude 65° North.In conclusion, vitamin D has an important role in modulating the molecular pathways involved in adipose tissue inflammation and insulin resistance in hepatic tissue. Adequate levels of vitamin D are necessary for the regulation of metabolic pathways to maintain body homeostasis. D-vitamiini inhiboi transkriptionaalisen tekijän NFκB:n siirtymistä ja tulehduksellisten sytokiinien ilmentymistä ihmisen mesenkymaalisista stroomasoluista (MSC) erilaistetuissa rasvasoluissa sekä sappirakkoleikkausten yhteydessä otetuista rasvanäytteistä eristetyissä kypsissä rasvasoluissa.Tutkimuksessa II havaittiin, että D-vitamiinin puutoksen aiheuttaminen puutteellisen ruokavalion avulla heikensi verenkierron glukoositasapainoa ja insuliiniherkkyyttä sekä hoikilla, että liikalihavilla hiirillä. Sitä vastoin D-vitamiini lisäravinteena palautti insuliiniherkkyyden ja lievitti maksakudoksen tulehdusta sekä oksidatiivista stressiä D-vitamiini puutteellisilla hoikilla hiirillä, mutta ei liikalihavilla hiirillä.Tutkimuksessa III selvisi, että kohortissa matalammat D-vitamiinitasot ovat yhteydessä suurempaan metabolisen oireyhtymän esiintyvyyteen iäkkäämmillä koehenkilöillä, jotka elävät pohjoisella pallonpuoliskolla (65° N).Yhteenvetona voidaan todeta, että D-vitamiinilla on tärkeä säätelevä rooli rasvakudostulehduksen ja insuliiniresistenssin molekyylireittien kontrolloinnissa maksakudoksessa.     Despite remarkable developments in diffraction unlimited super-resolution microscopy, in vivo nanoscopy of tissues and model organisms is still not satisfactorily established and rarely realized. RESOLFT nanoscopy is particularly suited for live cell imaging because it requires relatively low light levels to overcome the diffraction barrier. Previously, we introduced the reversibly switchable fluorescent protein rsEGFP2, which facilitated fast RESOLFT nanoscopy (Grotjohann et al., 2012). In that study, as in most other nanoscopy studies, only cultivated single cells were analyzed. Here, we report on the use of rsEGFP2 for live-cell RESOLFT nanoscopy of sub-cellular structures of intact Drosophila melanogaster larvae and of resected tissues. We generated flies expressing fusion proteins of alpha-tubulin and rsEGFP2 highlighting the microtubule cytoskeleton in all cells. By focusing through the intact larval cuticle, we achieved lateral resolution of <60 nm. RESOLFT nanoscopy enabled time-lapse recordings comprising 40 images and facilitated recordings 40 µm deep within fly tissues

    Heterodimeric Pbx-Prep1 homeodomain protein binding to the glucagon gene restricting transcription in a cell type-dependent manner

    Homeodomain proteins specify developmental pathways and cell-specific gene transcription whereby proteins of the PBC subclass can direct target gene specificity of Hox proteins. Proteins encoded by nonclustered homeobox genes have been shown to be essential for cell lineage differentiation and gene expression in pancreatic islets. Using specific antiserum in an electrophoretic mobility shift assay and in vitro transcribed/ translated proteins, the nuclear proteins binding domain B of the G3 enhancer-like element of the glucagon gene were identified in the present study as heterodimers consisting of the ubiquitously expressed homeodomain protein Prep1 and the also widely expressed PBC homeoprotein Pbx (isoform 1a, 1b, or 2), These heterodimeric complexes were found to bind also to the glucagon cAMP response element and to a newly identified element termed G5 (from -169 to -140). Whereas the expression of Prep1 or Pbx forms alone had no effect, coexpression of Pbx1a/1b-Prep1 inhibited the glucagon promoter when activated by cotransfected Pax6 or another transcription factor in non-glucagon-producing cells. In contrast, in glucagon-producing pancreatic islet cells, Pbx-Prep1 had no effect on GAL4-Pax6-induced mutant glucagon promoter activity or on Pax6-dependent wild-type glucagon promoter activity. Furthermore, 5'-deletion of G5 enhanced glucagon promoter activity in a non-glucagon producing cell line but not in glucagon-producing islet cells. This study thus identifies a novel target and Hox-independent function of Pbx-Prep1 heterodimers that, through repression of glucagon gene transcription in non-glucagon-producing cells, may help to establish islet cell-specific expression of the glucagon gene

    A Jewish lawyer, Jakub Herzig (1887–1956), survived World War II together with his family, changing his place of residence many times and staying in hiding. After the war, he emigrated to Israel. Some of his works are inedita. This article aims to present the figure of Herzig, as well as some of his literary works, in the context of post-war emigration and the motif of Eretz Israel. Przeżył on Holokaust na ziemiach polskich, bardzo często zmieniając miejsce zamieszkania. Był świadkiem wielu dramatycznych wydarzeń, z których relacje starał się zdać już po wojnie. Pozostawił po sobie kilka obszernych utworów literackich, w tym – prawdopodobnie nigdy niepublikowany w języku polskim – dramat O honor…, podejmujący temat powstania w getcie warszawskim z perspektywy żydowskich bojowców. Utwór jest wprawdzie niedoskonały literacko, jednak niezwykle cenny i interesujący jako świadectwo Zagłady. W artykule opisano także inne utwory Herziga, przekazane w postaci maszynopisów do Jad Waszem. Część z nich ukazała się później w Nowym Jorku i Tel Awiwie.The paper presents the history of the play entitled O honor [For Honour] and the fate of its author, Jakub Herzig (1887–1956). Herzig survived Holocaust thanks to frequent relocations. He witnessed numerous dramatic events about which he tried to talk after the war. Herzig wrote numerous works of fiction, including the play O honor [For Honour] that probably was never published in Polish. The play tells the story of the Warsaw Ghetto Uprising from the point of view of the Jewish fighters. While the play is not a literary masterpiece, it is a valuable and interesting testimony of Holocaust. The paper also describes other works penned by Herzig, the manuscripts of which were given to Yad Vashem.
