Large-pore mesoporous silica: template design, thin film preparation and biomolecules infiltration
Nanopores have been applied in the development of artificial biocatalytic systems, controlled drug delivery, and solid-state sensing devices. The interaction of biomacromolecules with surfaces show a dependence on the nanopore diameter, crucial in their ability to infiltrate porous materials. In this context, ordered mesoporous materials obtained by evaporation-induced self-assembly are model materials to test pore-biomolecule interactions. Nevertheless, these materials are generally restricted to pore diameters within the 2-10 nm range, therefore, new polymers as templating agents hold potential to provide an easy reproducible route for the synthesis of mesoporous silica thin films (MTF) with pore diameters above 10 nm without the use of swelling or additional structuring agents. Here, we present a novel and simple approach towards large pore MTF through the combination of supramolecular templating and phase separation with tailor-made block co-polymers. Accurate tuning of the oxide pore size distribution (with small mesopores between 13-18 nm diameter) is achieved by controlling the length and the nature of the hydrophilic polymer block used as a template through a simple reversible addition-fragmentation chain transfer (RAFT) polymerization approach. The importance of these features is highlighted by showing the capability that these new materials offer for biomolecule infiltration benchmarked against the widespread MTF prepared using pluronic F127 as a template. Effect of protein to pore diameter ratio, protein location and effect of pH and ionic strength is briefly tested and discussed.Fil: Alberti, Sebastián. Polymer biointerfaces for plasmonic nanostructures enabling fast, multiplexed, and sensitive detection of biomarkers
In der Gesellschaft besteht ein zunehmender Bedarf an einem schnellen, kosteneffizienten und zuverlässigen Nachweis von Biomolekülen, insbesondere in der frühen Diagnose von Krankheiten. In den letzten Jahren wurden dafür zahlreiche Biosensortechnologien entwickelt, bei denen mehrere zentrale Herausforderungen gelöst werden müssen, um eine genaue Analyse der in komplexen biologischen Flüssigkeiten in nur geringer Menge vorkommenden Analyten zu ermöglichen. Diese Herausforderungen betreffen die Empfindlichkeit und Spezifität von Sensormodalitäten, die einen direkten Nachweis der Spezies ermöglichen, ohne dass umfangreiche Probenvorbereitungsschritte und zeitaufwändige Amplifikationsstrategien erforderlich sind. Diese Dissertation kombiniert optische Biosensormethoden, die sich metallische Strukturen mit maßgeschneiderten plasmonischen Eigenschaften zunutze machen. Die Arbeit konzentriert sich insbesondere auf deren Nutzung zur Signalverstärkung, auf Multiplexing und auf die mögliche Implementierung in kompakte Auslesegeräte, mit der Fähigkeit Proteinmoleküle bis zu femtomolaren Konzentrationen zu detektieren. Zuerst wurden spezielle nanostrukturierte Oberflächen entwickelt, die zur Verstärkung von schwachen optischen Signalen verwendet werden können. Diese werden für das Auslesen von Affinitätsbindungen mittels Oberflächenplasmonenresonanz und oberflächenplasmonenverstärkter Fluoreszenzspektroskopie eingesetzt. Zweitens wurden Biointerface-Architekturen entwickelt, um als Bindungsmatrix an der Sensoroberfläche zu dienen. Dabei waren mehrere voneinander abhängige Aspekte der Post-Modifikation der Sensoroberfläche, der Minimierung unspezifischer Wechselwirkungen und der zusätzlichen (nicht-optischen) Signalverstärkung von Interesse. Schließlich wurde die Verwendung von responsiven Polyelektrolytbürsten und Hydrogelen auf Poly(N-Isopropylacrylamid)-Basis für den empfindlichen Nachweis von Biomarkern in klinischen Speichel- und Plasmaproben durchgeführt.There is an increasing need in society for rapid, cost effective, and reliable detection of biomolecules for the purpose of diagnosing diseases, including early diagnosis of non-communicable and infectious diseases. Over the last years, these needs have been addressed by research in numerous biosensor technologies, where several key challenges must be addressed in order to enable accurate analysis of the low abundant species present in complex biological fluids. These challenges concern sensitivity and specificity in modalities that allow direct detection of the species without the need for extensive sample preparation steps and time-consuming (mostly enzymatic) amplification strategies.The thesis combines optical biosensing methods that take advantage of metallic structures with tailored plasmonic characteristics. The work is particularly focused on their utilization for signal amplification, multiplexing with the ability to achieve femtomolar detection limits for proteins and implementation of the biosensor into compact readout devices.First, plasmonic nanostructured surfaces were specially designed to be manufactured by scaled-up lithography methods and to be employed for weak optical signal amplification in surface plasmon resonance and surface plasmon-enhanced fluorescence spectroscopy readout of affinity binding events. Second, advanced biointerface architectures were developed in order to serve as an affinity binding matrix at the sensor surface. Several interdependent aspects of the sensor surface post-modification, of minimizing non-specific interactions and of additional (non-optical) signal enhancement were investigated. Going Beyond Counting First Authors in Author Co-citation Analysis
The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation
counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings
are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that
only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into
account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed. Reasons for these effects are discussed
Plasmonically amplified fluorescence assays for ultrasensitive detection of biomarkers
Heutzutage steigt der Bedarf an schneller, günstiger und zuverlässiger Detektion in verschiedensten Bereichen, einschließlich der Krebsfrüherkennung und der Detektion von Infektionskrankheiten. Diese Diplomarbeit beschäftigt sich mit der Untersuchung von Assays für den Nachweis von Biomarkern mit verbessertem Fluoreszenzsignal. Die Plasmonenverstärkung von Fluoreszenzlicht, welches durch Fluorophore, die als Markierungen in diesen Assays verwendet werden, emittiert wird, machen es möglich, sogar Spuren dieser Moleküle nachzuweisen. In dieser Arbeit wird die Verwendung von zwei Fluoreszenzassays auf verschiedenen plasmonischen Biosensorplattformen gezeigt. Das erste Sensordesign betrifft die Detektion des Sepsis-Biomarkers Interleukin 6 (IL-6) in medizinisch relevanten Konzentrationen im Serum auf nanostrukturierten Plasmonenchips. Zweitens wird die Analyse von Hepatitis B-Antikörpern in klinischen Serum- und Speichelproben unter Verwendung einer zwitterionischen Bürstenarchitektur mit verbesserten Anti-Fouling-Eigenschaften demonstriert. Die erhaltenen Ergebnisse wurden mit Ergebnissen von Standard-ELISA-Tests bestätigt. Die Möglichkeit, Analyten auch in komplexen biologischen Flüssigkeiten in Spuren nachzuweisen, kann als eine Grundlage für zukünftige Sensoranwendungen gesehen werden.Nowadays fast, cheap and reliable detection is of increasing need in various fields, including early cancer diagnosis and infectious diseases. This thesis deals with investigating assays for the detection of biomarkers with enhanced fluorescence signal. The plasmonic amplification of fluorescence light emitted by fluorophores used as labels in these assays make it possible to detect even trace amounts of these molecules. In this regard, the employment of two fluorescence assays on different plasmonic biosensor platforms is shown. The first sensor design concerns the detection of the sepsis biomarker Interleukin 6 (IL-6) at medically relevant concentrations in serum on nanostructured plasmonic chips. Secondly, the analysis of Hepatitis B antibodies in clinical serum and saliva samples utilizing a zwitterionic brush architecture with enhanced anti-fouling properties is demonstrated. The obtained results were validated against results from standard ELISA. Variations on the Author
“Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship
Appropriate Similarity Measures for Author Cocitation Analysis
We provide a number of new insights into the methodological discussion about author cocitation analysis. We first argue that the use of the Pearson correlation for measuring the similarity between authors’ cocitation profiles is not very satisfactory. We then discuss what kind of similarity measures may be used as an alternative to the Pearson correlation. We consider three similarity measures in particular. One is the well-known cosine. The other two similarity measures have not been used before in the bibliometric literature. Dispelling the Myths Behind First-author Citation Counts
We conducted a full-scale evaluative citation analysis study of scholars in the XML research field to explore just how different from each other author rankings resulting from different citation counting methods actually are, and to demonstrate the capability of emerging data and tools on the Web in supporting more realistic citation counting methods. Our results contest some common arguments for the continued
use of first-author citation counts in the evaluation of scholars, such as high correlations between author rankings by first-author citation counts and other citation
counting methods, and high costs of using more realistic citation counting methods that are not well-supported by the ISI databases. It is argued that increasingly available digital full text research papers make it possible for citation analysis studies to go beyond what the ISI databases have directly supported and to employ more
