Heterozygosity for the Y701C STAT1 mutation in a multiplex kindred with multifocal osteomyelitis
Heterozygosity for dominant-negative STAT1 mutations underlies autosomal dominant Mendelian susceptibility to mycobacterial diseases. Mutations conferring Mendelian susceptibility to mycobacterial diseases have been identified in the regions of the STAT1 gene encoding the tail segment, DNA-binding domain and SH2 domain. We describe here a new heterozygous mutation, Y701C, in a Japanese two-generation multiplex kindred with autosomal dominant Mendelian susceptibility to mycobacterial diseases. This mutation affects precisely the canonical STAT1 tyrosine phosphorylation site. The Y701C STAT1 protein is produced normally, but its phosphorylation is abolished, resulting in a loss-of-function for STAT1-dependent cellular responses to IFN-γ or IFN-α. In the patient's cells, the allele is dominant-negative for gamma-activated factor-mediated responses to IFN-γ, but not for IFN-stimulated gene factor-3-mediated responses to IFN-α/β, accounting for the clinical phenotype of Mendelian susceptibility to mycobacterial diseases without severe viral diseases. Interestingly, both patients displayed multifocal osteomyelitis, which is often seen in Mendelian susceptibility to mycobacterial diseases patients with autosomal dominant partial IFN-γR1 deficiency. Multifocal osteomyelitis should thus prompt investigations of both STAT1 and IFN-γR1. Reflections on Postwar Japanese Society in Osamu Dazai’s Novel “No Longer Human”
Bakalaura darbā “Pārdomas par pēckara Japānas sabiedrību Osamu Dadzai romānā Diskvalificēts kā cilvēks”, pamatojoties uz tekstuāliem pierādījumiem, tiek pētīti literatūrā atspoguļotie indivīda centieni pielāgoties mainīgajai Japānas sabiedrībai. Galvenā problēma, kas sastopama pēckara laika autoru darbos, ir atstumtības sajūta, jauno tendenču nesaprašana un grūtības iederēties sabiedrībā. Darba mērķis ir apzināt Japānas pēckara sabiedrības problēmas, analizējot Osamu Dadzai romānā Diskvalificēts kā cilvēks paustās pārdomas. Darba pirmajā nodaļā ir sniegts pārskats par situāciju Japānā Otrā pasaules kara beigās, kā arī aplūkoti galvenie literāro darbu motīvi, atpazīstamākie rakstnieki un Osamu Dadzai dzīves gājums un literārā darbība. Ir pētīti vēsturiskie avoti, kas apraksta Japānu pēc kara beigām, kā arī raksti un periodiskie izdevumi, kas apkopo pēckara aktuālās tendences un virzienus literatūrā un izceļ ievērojamākos rakstniekus. Pētījumā ir izmantota zinātniskā literatūra, ar kuras palīdzību ir apkopota informāciju par pēckara Japānu, pēckara literatūras galvenajiem motīviem un Osamu Dadzai dzīves gājumu. Sākot ar darba otro nodaļu ir aprakstīts Osamu Dadzai dzīves gājums, analizētas romāna Diskvalificēts kā cilvēks galvenā varoņa pārdomas, literatūrā tēlotie notikumi ir salīdzināti ar reāliem notikumiem, kā arī ir apkopoti kritiķu vērtējumi par Osamu Dadzai rakstību. Analīze ir veikta, izmantojot romānu Diskvalificēts kā cilvēks, kā arī ir izmantoti raksti, grāmatas un citi avoti, lai izzinātu Osamu Dadzai dzīves gājumu un apkopotu kritiķu vērtējumu. Secinājumi sniedz skaidrojumu par Japānas pēckara literatūras galvenajām tendencēm un romāna Diskvalificēts kā cilvēks galvenā varoņa un Osamu Dadzai pieredzes salīdzinājumu, kā arī apkopo autores atziņas par Japānas sabiedrību.Bachelor’s thesis ‘Reflections on postwar Japanese society in Osamu Dazai’s novel No Longer Human’ explores individuals attempts to adjust to a changing Japanese society. Research is based on textual evidence. The main problem encountered among the authors of the post-war period is the feeling of exclusion, the misunderstanding of new trends and the difficulty of fitting into society. The purpose of the work is the cognition of the problems of post-war Japanese society through the reflections of the individual in the novel No Longer Human. The first chapter of the work provides an overview of the situation in Japan at the end of World War II, as well as the main motives of literary works, the most recognizable writers and the life course and literary activity of Osamu Dazai. Historical sources describing Japan after the end of the war are studied, as well as articles and periodicals that summarize post-wartime trends and directions in literature, highlighting the most prominent writers. The research uses scientific literature, with the help of which the author of the thesis collects information about post-war Japan, the main motifs of post-war literature and the life course of Osamu Dazai. Starting with the second chapter of the work, the life stories of Osamu Dadzai are described, the reflections of the main character of the novel No Longer Human are analyzed, events are compared with real events, and critics' ratings about Osama Dazai are compiled. The information is mainly collected through the novel No Longer Human, as well as articles, books and other Internet sources are used to describe the life course of Osamu Dazai and collect critics' ratings. The conclusions summarize what has been learned about the main trends in Japanese post-war literature and the similarities between the experiences of the protagonist of the novel No Longer Human and Osamu Dazai, as well as the author’s insights on Japanese society are summarized
