    Improve OpenMVG and create a novel algorithm for novel view synthesis from point clouds

    This thesis presents work to improve open source 3D reconstruction software OpenMVG and to create a novel algorithm to render photorealistic images from new views given a photo collection and 3D point cloud. First, the original OpenMVG is parallelized using GPU and its data structure is optimized. Moreover, we integrated the MatchMiner algorithm into OpenMVG to further improve its efficiency. Last but not least, an initial pair selection formulation and a default focal length setting are introduced and implemented to automize OpenMVG. Then 3D sparse point clouds of construction sites are reconstructed by performing Structure-from-Motion (SfM) with the improved version of OpenMVG and source images (images that are used in SfM) are calibrated and registered to point clouds. Furukawa's Patch-based Multi-view Stereo(PMVS) algorithm is used to reconstruct dense point clouds using calibrated cameras as inputs. With known depth values of 3D points in the dense point cloud, we estimate depth maps of source images using optimization similar to Levin's colorization algorithm. For a novel view of the point cloud, we find source images that share some common elements of the construction site that are also visible to the novel view. Then we warp depth maps of these candidate images to the novel view. We estimate a depth map and label pixels for the novel view by solving a multi-label Markov Random Field (MRF) optimization problem using graph-cuts. We introduce a novel energy minimization formulation exploits both 2D and 3D information. Finally, a photorealistic image of the novel view is rendered by copying pixel colors from selected candidate source images using pixel labels computed with graph-cuts. We experimentally validate our approach on several challenging viewing angles of a point cloud model of a complicate construction site.     Ontological Duality in Rock Poetry by Victor Tsoi (on Material of Song “In Our Eyes”)

    The article investigates the ontological duality as one of the important philosophical aspects of rock poetry by V. Tsoi. The importance of a philosophical interpretation of the rock text that embodies a worldview of its author is emphasized. It is shown that the text of the song “In Our Eyes” reflects the concept of being, various components of which are sometimes filled with opposite meanings; and yet they are logically combined, creating a complex philosophy of the lyrical hero. The attention is focused on the fact that ontological duality is represented at all levels of the artistic world of the song, including the level of lyrical hero, addressee, composition, but first of all - at the content and ideological level. It is noted that the text of the song “In Our Eyes” is in relationship with other works of the album “Blood Group,” being an integral part of the overall metaphysical plot of the rock cycle. It is proved that the concept of ontological duality of human existence is formed in the text of the rock song; rock, the author seeks to solve the philosophical problem of choice, and a lyrical hero of the song is not afraid to confront the world and at the same time does not seek to destroy it, he is ready to take place in the world, while maintaining his uniqueness

    Notch signaling requiem : orchestral role of notch signaling in cancer and developmental disease

    Notch signaling is an evolutionary conserved contact-dependent cell-cell communication pathway. This “contact” spans from hydra to fruit flies to human; orchestrating development, homeostasis and cancer, thus the Requiem, a song of life and death. Upon the “contact” of Notch receptor and ligand, the intracellular domain NICD is released and translocates to the nucleus. NICD, together with the DNA binding protein CSL and other co-activators, activate downstream targets. In this thesis, I have investigated the role of Notch signaling in multiple contexts with a modular approach. This includes: the non-canonical role of CSL in breast cancer, crosstalk of Notch signaling with hypoxia signaling in cancer, canonical Notch signaling in blood development, a novel mouse model for Alagille syndrome, and the hyperactivated Notch during mammary development and tumourigenesis.Here I phrase them in five sections of a requiem (Mozart’s Requiem, 1791): Introitus: In Paper I, we found that ablation of CSL unleashed a hypoxic response in normoxic conditions and enhanced tumour growth in breast cancer. A large part of the deregulated genes in the CSL null cell line is Notch independent. We demonstrated a non-canonical role of CSL and the possible implication of loss of CSL in breast cancer. Kyrie: In Paper II, we established that Notch signaling can modulate hypoxia signaling in multiple cancer cell types. By siRNA knocked down of HIF2α, we found that Notch signaling requires HIF2α for regulating a subset of Notch targets in medulloblastoma cells. Differences in the effect of N1ICD and N2ICD were also shown in the medulloblastoma cells. Lastly, we presented evidence of Notch signaling contributing to the HIF1α-to-HIF2α switch. Dies Irae: In Paper III, we revealed that canonical Notch signaling is dispensable in adult steady-state and stress myelo-erythropoiesis by ablating CSL in the myeloid lineage. Some of the Notch targets were derepressed in some of the progenitor stages, indicating CSL could act as a repressor in some contexts. Rex tremendae: In Paper IV, we established and characterized a mouse model for Alagille syndrome in human, recapitulating defects in multiple organ-systems. We showed a mutation in Jag1 caused delay differentiation and structural abnormalities in the bile ducts. From transcriptomics of mice and patients samples, we also found some commonly affected genes across species. Lastly, we discovered that the mutated Jag1 failed to bind to Notch1 and reduced the extent of Notch2 and Notch3 activation. Lacrymosa: In Paper V, we observed that hyperactive Notch in the luminal lineage during lactation cause defect in ductal development and led to mammary tumour development. Furthermore, we showed that this lineage can contribute to a large part of the mammary tumour.List of scientific papersI.     Opportunistic infection is always a potentially life threatening complication in pediatric oncology patients and hematopoietic stem cell transplant recipients. With the advances in various disease treatment protocols, the overall and event-free survivals of this high risk population improve significantly. In this thesis, the author reported a number of original studies to discuss different strategies in prevention of this serious complication. Firstly, the author demonstrates that pediatric oncology patients are still vulnerable to various vaccine-preventable infectious diseases up to 18 months after stopping chemotherapy. For those vaccine-preventable infectious diseases, pediatric oncology patients can mount a significant and persistent immune response to common inactivated vaccine (namely diphtheria-tetanus-pertussis vaccine). For non-vaccine preventable infectious diseases, regular monitoring of plasma viral load and strategic use of antiviral agents as pre-emptive or prophylactic agent is an effective approach to prevent infection. In hematopoietic stem cell transplant setting, adoptive transfer of acquired immunity from donor to recipient and incorporation of this parameter in donor selection process can be considered. The findings of the studies can be applied to clinical setting. The future direction of our studies includes the immune responses of other common vaccines namely pneumococcal vaccine and pandemic influenza vaccine in high risk population. The role of transfer of donor's varicella zoster immunity in prevention of herpes zoster infection in transplant recipient can be further explored.     Failure of Blast-Loaded Reinforced Concrete Slabs

    AbstractThe flexural response and failure of reinforced concrete rectangular slabs subjected to blast loading are investigated based on the rigid-plastic modelling. The effects of orthotropic reinforcement and asymmetric support bending restraints are incorporated to account for structural configurations and detailing. Two schemes of failure pattern are introduced and features of dynamic response under typical blast loading are outlined. Numerical studies are carried out and the results obtained from the present model and explicit finite element simulations are compared. It is shown that the proposed analysis is capable of capturing the essential features of dynamic response of reinforced concrete slabs and provides a simple and efficient means for blast design and assessment of concrete slabs

    Viktor Tsoi and his band KINO were some of the most well-known musicians of the late 1980s in the former USSR. Their influence on Russian rock-culture has been quite large and their output started being analyzed a few years ago. One of the main themes in Tsoi’s poems and songs is rain. This work is an attempt to interpret this motif in some of Tsoi’s most important songs.     本研究針對三個重要層面(論述、鬥爭和情感的過程)進行探索,討論社會運動中反對勢力框架的建構和發展。本文將先探討社會運動如何在論述層面上建構框架,令運動框架取得認受性並挑戰主流霸權論述;是次研究亦會討論互聯網時代如何影響框架爭奪戰的進行;最後,本文將分析框架美學化的現象如何影響社會運動的動員。本研究以香港的菜園村抗爭運動作為案例,並採用文本分析、訪問及參與式觀察等研究方法。本文旨在擴充框架分析的討論層面:除探討其認知層面外,本文更試圖討論框架建構中被忽略的面向:情感性層面。此外,本文亦會分析框架爭奪戰的場境如何在互聯網時代被擴大。在認知層面上,本文發現在論述性的框架建構過程中,若框架的理念能充分回應社會、政治及文化上的斷裂,框架動員力便會提高。此外,在運動過程中,策略性的框架轉化亦有助號召不同界別的群體參與。在鬥爭層面上,本文指出在互聯網時代,社會運動與政府、主流傳媒進行框架爭奪戰的場境已被伸延及擴大。而互聯網所造就的分散式競爭能量及沉積性網絡結構則對運動於框架爭奪戰過程中有利。最後,在情感層面上,本文認為近年社會運動中採用的美學化運動手段,有助運動在情感和道德價值上動員群聚,因美學化的框架不單有助社會運動鞏固其內部聯繫,對民眾整體都有吸引力。This study investigates the construction and development of opposition frames in a social movement by exploring three important dimensions: the discursive, contested, and affective processes. This study first elucidates how counter-frames can be discursively constructed to gain legitimacy in challenging the dominant hegemonic belief. Second, the study reviews how the framing processes have been influenced by the Internet in the digital era. Finally, it investigates how the aestheticization of frames relates to the affective mobilization of a movement. These questions were addressed through the case of the Tsoi Yuen resistance movement, a two-year struggle in Hong Kong. To this end, textual analysis, interviews, and participant observations are employed.This study seeks to elaborate upon the framing perspective by discussing not only the cognitive meaning alignment processes, but also expanding the theoretical discussion in the affective dimension of framing, which is an under-explored area. In addition, it contributes to the investigation of the expanded frame contestations in the Internet era.On the cognitive level, this thesis argues that the discursive framing strategies have worked best when meanings are activated by exploiting the social, political, and cultural cleavages in post-colonial Hong Kong. In addition, the transformation of framing strategies can facilitate mobilizations across a variety of groups of targets when different cleavages are echoed.For the contested framing processes, it is argued that the media ecology of the framing contest between the movement’s frames and the dominant frames constructed by the government and the mainstream media have been considerably enlarged by the Internet. The distributed contested power and sedimentary networks emerging in the Internet era are enabling the contested processes in the frame battle.Finally, on the affective level, this study argues that the series of aesthetic movement tactics employed in social movements enhance the affective dimension of framing processes through the mediation of the emotional and moral components.
