Human iPSC-derived motoneurons harbouring TARDBP or C9ORF72 ALS mutations are dysfunctional despite maintaining viability

We report the use of human induced pluripotent stem cell (iPSC)-derived motoneurons (MNs) to study the pathophysiology of ALS. We demonstrate that MNs derived from iPSCs obtained from healthy individuals or patients harbouring TARDBP or C9ORF72 ALS-causing mutations are able to develop appropriate physiological properties. However, patient iPSC-derived MNs, independent of genotype, display an initial hyperexcitability followed by progressive loss of action potential output and synaptic activity. This loss of functional output reflects a progressive decrease in voltage-activated Na+ and K+ currents, which occurs in the absence of overt changes in cell viability. These data implicate early dysfunction or loss of ion channels as a convergent point that may contribute to the initiation of downstream degenerative pathways that ultimately lead to MN loss in ALS.Peer reviewe

Musikstädte as real and imaginary soundscapes: urban musical images as literary motifs in twentieth-century German modernism
No full textPhDThis study examines German literary images of musical life as part of the wider sound identity of the modern German city at the turn of the twentieth century. Focussing on a forty-year period from 1890 to 1930, synonymous with the emergence of the modern German metropolis as an aesthetic object, the project assesses, compares and contrasts how musical life in the Musikstädte was perceived and portrayed by writers in an increasingly noisy urban environment. How does urban musical life influence and condition city writings? What are the differences and similarities between the writings on various musical cities? Can an urban textual sound identity be derived from these differences and similarities? The approach employed to answer these questions is a new, cross-disciplinary one to urban sound in literature, moving beyond reading the key sounds of the urban soundscape using urban musicology, sensorial anthropology and cultural poetics towards a literary contextualisation of the urban aural experience.
The literary motifs of the symphony, the gramophone and urban noise are put under the spotlight through the analysis of a wide range of modernist works by authors who have a special relationship with music. At the centre of this analysis are the Kaffeehausliteratur authors Hermann Bahr, Alfred Polgar and Peter Altenberg, the then Munich-based author Thomas Mann and the lesser known René Schickele. The analysis of these particular works is framed in the music-geographical context of the Musikstadt and literary underpinnings of this topos, ranging from Ingeborg Bachmann to Hans Mayer and, once again, Thomas Mann. In analysing these texts, the methodological approach devised by Strohm, who identifies the blending of a range of urban sounds as a definition of urban space and identity, is applied. His ideas combine historical literary

Rare Variant Burden Analysis within Enhancers Identifies CAV1 as an ALS Risk Gene
No full textCooper-Knock et al. identify amyotrophic lateral sclerosis (ALS) risk variants within non-coding regulatory DNA linked to a known ALS gene, TBK1, but also CAV1 and CAV2. Disease-associated variants reduce CAV1/CAV2 expression and disrupt membrane lipid rafts with consequences for neurotrophic signaling. CAV1 coding sequence also contains ALS-associated mutations. © 2020 The Author(s) Amyotrophic lateral sclerosis (ALS) is an incurable neurodegenerative disease. CAV1 and CAV2 organize membrane lipid rafts (MLRs) important for cell signaling and neuronal survival, and overexpression of CAV1 ameliorates ALS phenotypes in vivo. Genome-wide association studies localize a large proportion of ALS risk variants within the non-coding genome, but further characterization has been limited by lack of appropriate tools. By designing and applying a pipeline to identify pathogenic genetic variation within enhancer elements responsible for regulating gene expression, we identify disease-associated variation within CAV1/CAV2 enhancers, which replicate in an independent cohort. Discovered enhancer mutations reduce CAV1/CAV2 expression and disrupt MLRs in patient-derived cells, and CRISPR-Cas9 perturbation proximate to a patient mutation is sufficient to reduce CAV1/CAV2 expression in neurons. Additional enrichment of ALS-associated mutations within CAV1 exons positions CAV1 as an ALS risk gene. No full textIn zorgwoongebouwen verblijven de meest kwetsbare mensen in grote eenzaamheid. Enerzijds is er de gezondheidseis van social distancing en anderzijds de broodnodige contactmomenten tussen mensen tijdens de lock down om niet te vervreemden. Nergens wordt dit dilemma zo pijnlijk gevoeld als in zorggebouwen. De COVID-19 epidemie zal niet de laatste crisis zijn waarbij besmetting met zoönotische virussen centraal staat en gezonde afstand enerzijds en verlangen naar contact anderzijds ons leven bepaald. Het is daarom heel waarschijnlijk dat deze epidemie de architectuur van zorggebouwen zal gaan veranderen naar gezonde gebouwen die letterlijk ruimte geven aan gebruikers. Alleen al het feit dat iedereen inmiddels de vaardigheid van videobellen en thuiswerken heeft aangeleerd leidt tot een heroriëntering op het type gebouwen dat wij gebruiken. In dit artikel is Peter Boerenfijn geïnterviewd die zich met ouderenhuisvesting bezighoudt en de te verwachte veranderingen van het gebouwtype als gevolg van COVID-19.<br/

No full textDigitale Editionsformen müssen auf dem Methodenstand der gedruckten Editionen aufbauen. Hier sind der Lachmannsche Ansatz der Textrekonstruktion und die Verfahren der historisch-kritischen Ausgabe im Laufe der Zeit zwar dominant geworden, aber nicht alternativlos geblieben. Die Kritik an diesen Methoden ist so alt wie diese selbst, und viele andere Schulen haben sich neben ihnen etabliert. Die verschiedenen Ansätze lassen sich als Produkt ihrer Zeit und als Funktion bestimmter theoretischer Grundbegriffe und Grundannahmen beschreiben. Vor allem aber sind sie nicht nur historisch und theoretisch, sondern immer auch "technisch relativ", weil Typografie und Druckkultur unsere Theorien vom Text und unsere Methoden der Edition beeinflussen. Ein Verständnis dieser Abhängigkeit unserer scheinbar "natürlichen" Editionsvorstellungen von einer bestimmten Technik ist ein wichtiger Ausgangspunkt für die Entwicklung neuer Editionsformen

The problems explored are focused firstly around the issues of the representation of Eastern Europe and contemporary Eastern European art, in terms of organizing exhibitions in the context of globalisation, and secondly the role of a contemporary art curator as compared to the role performed by a contemporary cultural manager.
