Colonization and domestication of seven species of larval-prepupal and pupal native new world Hymenopterous fruit fly (diptera: Tephritidae) parasitoids
Techniques used to colonize and domesticate seven native New World species of hymenopterous parasitoids that attack flies within the genus Anastrepha (Diptera: Tephritidae) were developed: Doryctobracon areolatus (Szépligeti), Doryctobracon crawfordi (Viereck), Opius hirtus (Fischer), Utetes anastrephae (Viereck) (all Braconidae, Opiinae), Aganaspis pelleranoi (Bréthes) and Odontosema anastrephae Borgmeier (both Figitidae, Eucoilinae) (all larval-pupal parasitoids), and the pupal parasitoid Coptera haywardi (Ogloblin) (Diapriidae, Diapriinae) were reared successfully on Mexican fruit fly, Anastrepha ludens (Loew) larvae or pupae reared on artificial diet. Descriptions of the different rearing techniques used throughout the domestication process are described to help researchers to colonize local parasitoids. We also describe handling procedures such as number of hosts in parasitization units and compare optimal host and female age, differences in parasitism rate, developmental time, life expectancy and variation in sex ratios in each parasitoid species over various generations. In the case of D. crawfordi and C. haywardi we also provide partial information on mass-rearing techniques such as cage type, parasitization unit, larval irradiation dose and adult handling.Fil: Aluja, Martin. Evaluation and proposal for improvements in the design and manufacturing process of PSS 3-1100 rumen presses for Frigoríficos Guadalupe
En el marco de la pasantía realizada en Frigoríficos Guadalupe, empresa especializada en el procesamiento de carne, se desarrolló un proyecto orientado a la optimización de la prensa ruminal modelo PSS 3.3-1100, una máquina clave en el tratamiento del contenido ruminal bovino. El objetivo principal fue mejorar su eficiencia y funcionalidad, contribuyendo así a la productividad, sostenibilidad y competitividad de la empresa. Durante el proceso, se realizó un análisis comparativo entre las prensas ruminales PSS 3.3-800 y PSS 3.3-1100, identificando áreas críticas de mejora. Se desarrollaron planos técnicos detallados mediante software CAD, se ejecutaron simulaciones computacionales de capacidad operativa, y se diseñaron documentos esenciales como un Dossier de Calidad, Manual de Uso y Manual de Mantenimiento. Estas acciones permitieron optimizar la capacidad de procesamiento de rumen, reducir tiempos operativos, minimizar residuos y garantizar un mejor aprovechamiento de los subproductos. El proyecto integró conocimientos técnicos, normativas ambientales y criterios de mejora continua, brindando a Frigoríficos Guadalupe herramientas para consolidar sus estándares de calidad y sostenibilidad, al tiempo que fortaleció la toma de decisiones estratégicas sobre la actualización tecnológica de sus equipos industriales.As part of the internship carried out at Frigoríficos Guadalupe, a company specialized in meat processing, this project focused on optimizing the ruminal press model PSS 3.3-1100, a critical machine in the treatment of bovine rumen content. The main objective was to enhance its efficiency and performance, thereby contributing to the company's productivity, sustainability, and competitiveness. The project involved a comparative analysis between the PSS 3.3-800 and PSS 3.3-1100 ruminal presses to identify critical areas for improvement. Detailed technical drawings were developed using CAD software, computational simulations of processing capacity were conducted, and key documentation—including a Quality Dossier, User Manual, and Maintenance Manual—was created. These efforts helped increase rumen processing capacity, reduce operational times, minimize waste, and ensure better utilization of by-products. The project integrated technical knowledge, environmental standards, and continuous improvement principles, providing Frigoríficos Guadalupe with the tools to strengthen its quality and sustainability standards, while supporting strategic decision-making regarding the technological upgrade of its industrial equipment
Inducing The Holy Spirit through the Pentecostal Message
Pentecostalism (PC) is the most prolific branch of American evangelical Christianity (Jacobsen 2003) as it has been able to provide to its adherents, subcultures and collective identities that bestow meaning and belonging (Smith 1998). PC arose from Methodist and Baptist branches of Christianity at the end of the 19th century (Blumhofer 1993) and was involved in adjusting millions of migrants to U.S. city life (Synan 1997). Because PC has “straddled the race line in ways that most other American religious movements did not” (Jacobsen 2003: 260), it is significant to consider its wide appeal by analyzing how it manifests itself in local ways. Furthermore, if PC can distinguish itself while directly engaging the cultures it interacts with (Robbins 2004), how does it become appealing to the five million Latinos in the U.S. who consider themselves pentecostal (Sanchez 2003)? My attempt is to ascertain the type and extent of influence that Latino culture exerts on the general pentecostal model by considering the language, traditions, concepts, and practices of a local congregation mostly composed of firstgeneration migrants from Guatemala, Mexico, and various other Spanish-speaking nations. I seek to understand how PC transforms along different contexts while focusing on the relation of religion and culture through two pressing themes: Latino culture and gender

EVALUATION OF GENE EDITING APPROACHES IN HIGHER PLANTS
Advances in gene editing have facilitated new approaches to improve traits of interest in plants. Specifically, CRISPR-Cas9 and associated endonuclease systems allow for the targeting of agriculturally important genes such as disease resistance, improving yield and shelf life, and changing specific traits related to a final high-quality product. SpCas9 uses a tracrRNA: crRNA (gRNA) sequence to target the DNA and induce double-stranded breaks at targeted locations. SpCas9 is widely used for genome editing in mammals and plants; however, other promising endonucleases have been discovered with the potential for greater editing efficiency. Particularly, the nucleases Cas12a (also known as Cpf1) and Cms1 are promising for plant transformation and genome editing approaches. Cas12a, discovered in the bacteria Francisella novicida U112, and Cms1, discovered in the genera Microgenomates and Smithella, are smaller enzymes than SpCas9. Both utilize only a gRNA formed by a mature crRNA to target a T-rich PAM sequence and generate cohesive DNA ends. Although successful gene editing has been achieved in several plants, a single approach that is useful, economically efficient, and reliable to use across different crops has been challenging to develop. The first aim of this work was to answer this question among our candidates: Cas12a, Cms1, and SpCas9. We explore efficiency by targeting the GFP gene in N. benthamiana protoplast as an evaluation system.The generation of gene-edited plants requires identifying the most suitable method to deliver the CRISPR system into the plant cell. Therefore, recalcitrant crops to transformation is a challenge for gene editing in certain crops. Moreover obtaining lines without transgenic elements relies on outcrossing the transformed-edited obtained plant to non-transgenic plants. This outcrossing approach is incompatible with several high-value, obligated outcrossing crops. The second aim of this work is to present a novel method of delivering gene editing reagents across graft junctions using transgenic tobacco as the donor and wild-type tobacco as a scion. This innovation expands the capabilities of plant gene editing to generate non-transgenic edited plants.Implementing gene editing in agriculturally relevant cases is a promising tool for addressing challenges (such as generating climate-resilient crops and creating the next generation of elite cultivars). Grapes were reported to produce 74 million tonnes in 2021 (FAO); 70% of the grape production is used for the wine industry. A promising target for editing in wine grapes, an economically significant crop, is the o-methyltransferase-encoding gene VvOMT3. The VvOMT3 protein is involved in the synthesis of methoxypyrazine, which imparts an undesirable green pepper flavor in wines and produces low anthocyanin content, weak color, and poor tannin quality. Removal of this compound could result in important improvements to flavor. Therefore, the third aim of this work is to target the OMT3 gene, using gene editing, for expression knock-out in organogenic cultures from Vitis vinifera (cv. ‘Cabernet Sauvignon’).Knowledge of the efficacy of multiple nuclease systems, plant tissues in which they are most efficient, and their utility in both fundamental and applied systems, is expected to greatly improve the success of future gene editing projects. As new editing technologies improve and broaden to employ additional endonucleases, the success of inducing changes in genes of interest, such as the proposed OMT modification in grapes, becomes increasingly feasible.

Plan de comercialización para la venta de telas plásticas en la Empresa Carylu - Chiclayo
El objetivo es diseñar un plan de comercialización de telas plásticas para mejorar
las ventas en la empresa Carylu. Para esto, se realizó una investigación con
enfoque cuantitativo y de diseño no experimental descriptivo propositivo. La
muestra fueron 303 personas entre colaboradores y clientes, usándose un
cuestionario para recopilar información sobre la situación actual en la que se
encuentra la empresa Carylu. Se concluye en primer lugar, la necesidad de
implementar un plan operativo de comercialización que contribuya a incrementar
las ventas. Por consecuencia, es importante acelerar el cumplimiento de los
objetivos de los planes de comercialización y, por último, implementar estrategias
de mejora continua para la venta de telas plásticas que sirva de apoyo para la
gerencia y la toma de decisiones de los trabajadores
Degradación de metano en reactores de partición de dos fases líquidas y acumulación de PHB (poli-β-hidroxibutirato)
Las emisiones de gases que contribuyen al calentamiento global son un grave problema para los ecosistemas y la humanidad debido a sus implicaciones en el cambio climático. Durante las últimas décadas, el crecimiento demográfico y el desarrollo industrial han dado lugar a la acumulación de gases como el CO2, CH4 y N2O que contribuyen al efecto de invernadero. El metano tiene un potencial de calentamiento global (PCG), que es 21-25 veces mayor que el CO2. El metano es un gas de efecto invernadero, cuya concentración promedio en la atmósfera es de alrededor de 1760 ppbv. La degradación de metano ha sido estudiado principalmente en reactores de tanque agitado, biofiltros, biocubiertas y, recientemente, en reactores de partición de dos fases líquidas (TPPB), los cuales tienen como característica principal la adición de una fase no acuosa, como el aceite de silicón, la que aumenta la biodisponibilidad de CH4 mediante la mejora en los procesos de transferencia de masa del sustrato de la fase gaseosa hacia los microorganismos. En esta tesis, se estudió la degradación de CH4 por un consorcio y una cepa aislada del mismo empleando un STR (reactor de tanque agitado) y un TPPB. Al mismo tiempo, se estudió la capacidad de los microorganismos para acumular PHB (poli-β-hidroxibutirato), biopolímero intracelular, biodegradable y con propiedades plásticas similares a las del polipropileno. El PHB se acumula cuando hay limitación de algunos elementos como fósforo y nitrógeno. La primera etapa de la estrategia experimental consistió en aislar las cepas de un consorcio previamente adaptado al consumo de metano. 17 cepas con capacidad metanótrofa fueron aisladas y se mantuvieron en una atmósfera de CH4. El 75% de los aislados fueron Gram negativos. Las 10 cepas con mayor crecimiento sobre metano y metanol fueron elegidas. Los aislados seleccionados fueron sometidos a una prueba cualitativa de acumulación de PHB. La tinción de fluorescencia mostró que cuatro cepas fueron capaces de acumular PHB, se calcularon las tasas específicas de consumo de CH4 cuando las cepas no estaban limitadas por nitrógeno. Posteriormente, se hizo una comparación entre las cepas aisladas y una cepa de colección, Methylosinus trichosporium OB3b que tiene una alta capacidad de la degradación de CH4 (50 mg gX -1 h-1 ) y una acumulación de PHB de hasta 50% p p-1 . Bajo las mismas condiciones experimentales, utilizando una concentración del metano en el aire 1%, uno de los aislados (C2) mostraron una acumulación de PHB de 56% p p-1 en comparación con 24% p p-1 de Methylosinus trichosporium OB3b. En cuanto a las tasas de degradación específicas de metano, el C2 alcanzó una tasa específica de consumo de metano de 60 mg gX -1 h-1 y Methylosinus trichosporium OB3b 77 mg gX -1 h-1 , otra cepa estudiada, C147 presentó una acumulación de PHB 21% p p-1 y una tasa específica de consumo de 139 mg de CH4 gX -1 h-1 . La conclusión de este experimento fue que algunos aislados, tales como C2, fueron capaces de acumular alto contenido de PHB con valores incluso más altos que los reportados para otros metanótrofos y que algunos microorganismos como el C147 tienen la capacidad de alcanzar altas tasas de degradación de CH4. Los estudios continuaron a nivel reactor (STR y TPPB) utilizando el aislado C2 y el consorcio. Los resultados obtenidos mostraron un incremento de entre 33 a 45% en la capacidad de eliminación para el reactor operado con el consorcio y la cepa, respectivamente, debido a la presencia de una fase orgánica en el TPPB. Las tasas específicas para el consumo de metano fueron de 100 y 16.7 mg gX -1 h-1 para el aislado y el consorcio, respectivamente. Los resultados anteriores mostraron que la cepa C2 es más eficaz para el tratamiento de corrientes de gaseosas contaminadas con bajas concentraciones de CH4. El consorcio acumuló el 36% p p-1 de PHB, al principio de la acumulación de PHB hubo una disminución significativa en la producción de CO2, este fenómeno se observó en todos los experimentos con la limitación de nitrógeno y la acumulación de PHB. En el caso de la cepa C2, el efecto del aceite de silicón sobre la cantidad de PHB acumulado osciló entre 34 y 37% p p-1 para el experimento de STR y TPPB, respectivamente lo que indico que no había efecto en la acumulación de PHB. Durante estos experimentos la velocidad específica de consumo de metano aumentó el 12,5% debido al aceite de silicón, las tasas fueron de 30 mg gX -1 h-1 en el STR y 45 mg gX -1 h-1 en TPPB. Estos resultados son comparables y en algunos casos, superiores a los reportados en la literatura. La cepa C2 fue preliminarmente identificada por técnicas de biología molecular, como Methylobacterium organophilum. Estos experimentos demostraron que durante el tratamiento de CH4 es posible generar un producto con alto valor agregado, si el metabolismo celular se redirige a la producción de PHB y biomasa reduciendo de esta manera las emisiones de CO2.The releases of gases that contribute to global warming are a serious problem to ecosystems and humanity for its implications for on the climate change. During the past decades, population growth and industrial development have resulted in accumulation of gases like CO2, CH4 and N2O emissions which contribute mainly to the greenhouse effect. The methane has a global warming potential (GWP) that is 21-25 times greater than CO2. Methane is a greenhouse gas, whose average concentration in the atmosphere is about 1760 ppbv. The methane degradation has been studied mainly in stirred tank reactors, biofilters biocovers and just recently in the two phases partitioning bioreactors (TPPB); its main characteristic is the addition of a non-aqueous phase, such as silicone oil, which increases the CH4 bioavailability to the microorganisms by improving the processes of mass transfer of substrate from the gas phase in to the microorganisms. In this thesis, the degradation of CH4, by a consortium and a strain isolated from it, was studied using a STR (stirred tank reactor) and a TPPB. At the same time, we studied the ability of microorganisms to accumulate PHB (poly-β-hydroxybutyrate), intracellular biopolymer, biodegradable and plastic properties similar to polypropylene. The PHB accumulates mainly when there is limitation of some elements like nitrogen o phosphorus.. The first stage of the experimental strategy was to isolate the strains from a consortium previously adapted to methane consumption. 17 strains with methanotrophic capacity were isolated and they were maintained in an atmosphere of CH4. 75% of the isolates were Gram negative and 10 strains with higher cell growth on methane or methanol were chosen. The isolates were qualitatively tested for the capability to accumulate PHB. The fluorescence staining showed that four strains were capable of accumulating PHB, also the specific consumption rates of CH4 were calculated for the isolates grown under non- nitrogen restricted mineral medium. Afterwards, a comparison was made between the isolates and a collection (ATCC) strain, Methylosinus trichosporium OB3b. It has a high capacity for CH4 degradation of (50 mg gx-1 h-1 ) and a PHB accumulation of up 50% w w-1 . At the same experimental conditions with a methane concentration of 1% in air, one of the isolates (C2) showed a PHB accumulation of 56% w w-1 compared to 24% w w-1 of Methylosinus trichosporium OB3b. Regarding to the specific degradation rates of methane, the C2 reached a specific rate of methane consumption 60 mg gx-1 h-1 and Methylosinus trichosporium OB3b 77 mg gx-1 h-1 , another studied strain, C147, has a PHB accumulation of 21% w w-1 and a specific consumption rate of 139 CH4 mg gx-1 h-1 . The conclusion of this experiment was that some isolated, such as C2, were able to accumulate high PHB content with values even higher than reported methanotrophs and some other have the ability to reach high CH4 degradation rates, such as C147. The studies continued at the 3-L reactor (STR and TPPB) using the C2 isolate and the consortium. The obtained results showed an increase between 33 - 45% on the elimination capacity for the reactor operated with the consortium and the strain, respectively due to the presence of an organic phase in the TPPB. Specific rates for methane consumption were 100 and 16.7 mg gx -1 h-1 for the isolate and the consortium, respectively. The above results showed that the isolate C2 is more efficient for the treatment of contaminated gas streams with low CH4 concentrations. The consortium accumulated 36% w w-1 of PHB and, at the beginning of PHB accumulation, there was a significant decrease on CO2 production, this phenomenon was observed in all experiments with nitrogen limitation and accumulation of PHB. In the case of the tested strain, the effect of silicone oil on the amount of accumulated PHB ranged between 34 and 37% w w1 for the experiment in STR and TPPB, respectively. During those experiments the specific methane consumption rate increased 12.5% due to silicone oil in which the result was 30 mg gx -1 h-1 in the STR and 45 mg gx -1 h-1 in TPPB. These results are comparable and, in some cases, higher than those reported in the literature. The isolate C2 was preliminarily identified by molecular biology techniques such as Methylobacterium organophilum. These experiments showed that during treatment of CH4 is possible to generate a product with high value, if cell metabolism is redirected to the production of PHB and biomass to reduce CO2 emissions
