    The aim of the present study was to evaluate the ability of the calcium channel blocker (CCE), nimodipine (NIM), to interact with (+/-)-amphetamine (AMPH) in modifying ingestive behavior. Rats performed in a water-reinforced runway paradigm with multiple trials. Water was available in sufficient quantity to produce satiety under control conditions as measured by a decline in response rate over the session. NIM and AMPH, given alone, did not produce significant effects on performance but produced behavioral changes when administered in combination. In particular, the combination of the highest doses (13 mg/kg i.p. NIM plus 0.56 mg/kg i.p. AMPH) initially depressed both running and drinking, whereas in later trials it increased running rate, without producing a parallel increase in water intake. These results suggest that NIM enhances AMPH-produced inhibition of drinking, whereas it first depresses and then enhances the AMPH-mediated runway performance, suggesting the rate dependency of this latter effect

    The aim of this thesis concerning the setting of new methods to study methabolic pathaways in vivo using both stable isotope tracer (113C-leucine and D2O) both stable isotope at natural abundance. First chapter reports a new method to measure absolute and percentage of AA and DHA synthesis. Preterm infants were fed with a formula enriched in AA and DHA with a higher content of 13C than did the endogeusly sinthesized long chain fatty acids (LCP). The second chapter reports a new menthod to measure SP-B synthesis in vivo in humans by means of stable isotopes. We administered a 24 h infusion of 13C-Leucine as metabolic precursor of SP-B and 2H2O as precursor of dipalmitoyl phosphatidylcholine (DSPC).Obiettivo di questa tesi è stato l’applicazione sia di isotopi stabili ad abbondanze naturali sia di traccianti marcati con isotopi stabili (113C-leucina e D2O) per lo studio di diverse vie metaboliche, quali la sintesi, di acidi grassi, nel bambino pretermine e la sintesi proteica e lipidica nell’adulto. Il primo studio ha visto l’utilizzo di una dieta arricchita con due acidi grassi che presentavano un’abbondanza naturalmente diversa di 13C rispetto a quella degli altri acidi grassi per determinare, per un lungo periodo di tempo, la sintesi assoluta e in percentuale di questi due acidi grassi nei bambini pretermine. Nel secondo studio abbiamo utilizzato leucina marcata con 13C e acqua marcata con deuterio per determinare il metabolismo della fosfatidilcolina disatura (DSPC) e della proteina specifica B del surfattante (SP-B)

    Profiling of Mycobacterium tuberculosis gene expression during human macrophage infection: Upregulation of the alternative sigma factor G, a group of transcriptional regulators, and proteins with unknown function

    Mycobacterium tuberculosis is one of the most prolific pathogens worldwide, and its virulence resides in its capacity to survive in human macrophages. In the present study, we analyzed the gene expression profile of M. tuberculosis H37Rv in macrophages and synthetic medium at the whole genome level. Out of 3875 spots tested, 970 genes passed the statistical significance filter (t scores >= +/- 2.5). A total of 22% of those assayed were found to be active genes (up- or downregulated), representing 5.5% of the whole MTB genome. Interestingly, 32.5% of the genes induced in our macrophage experiments are still classified as hypothetical proteins; 19.5% take part in the cell wall and processes (half of which are membrane proteins); 16% are involved in regulation and information pathways; and the PE family accounts for 3.6% of total induced genes. It is important to note that in the course of MTB replication in macrophages, we observed the upregulation of alternative sigma factor sigG and 13 MTB transcriptional regulators. The data for a selected group of upregulated genes were confirmed by real-time RT-PCR. The global MTB transcriptome described in this study suggests an intracellular MTB actively sensing its environment; it repairs and synthesizes its cell wall and DNA, so as to either repair oxidative and nitrosative damage and/or to augment its copy number and evade host cell killing.     This report is an attempt to analyze the narrative discourse order in the novel “La esposa del Dr. Thorne” (Dr. Thorne’s Wife) of the Venezuelan writer Denzil Romero, in the light of ¨Las cuatro estaciones de Manuela” (“The Four Seasons of Manuela”) of the American author Víctor W. von Hagen, and of “Manuela. Sus diarios perdidos y otros papeles”, (Manuela. Lost Diaries and Other Papers) of the Ecuadorian Carlos Álvarez Saá. This paper is a risky comparison between a book of fiction, presumably based on real facts, where Manuela’s eroticism is the central theme in Romero, the large literary chronicles issued by von Hagen and the compilation and discovery of alleged original and historic texts (letters and diaries) produced by Álvarez Saá. It is also a research exercise where, whimsically, what the official discourse says, as it may be viewed by the school of Venezuela or that of Quito, is mixed with the fictionalization of that discourse. It is, too, a possible vindication of Manuela Sáenz as a person and/or predominant character in the fights for the independence of America beyond her ¨circumstantial¨ relationship with Simón Bolívar. It is, to a certain extent, a look into how literary texts and the perceptions of history about a character (Manuela) tend to a “rearrangement” in present times and how there has been an attempt to consolidate the figure of Manuela Sáenz as a heroine, awarding a victory to what von Hagen calls “the school of Quito”.Si prese l’analisi dell’ordine discorsivo narrativo del romanzo dell’autore venezuelano Denzil Romero, La sposa del Dott.re Thorne, attraverso le comparazioni delle opere dell’autore statunitense Víctor W. Von Hagen, Le quattro stagioni di Manuela e dell’ autore ecuadoregno Carlos Álvarez Saá, Manuela, i suoi diari persi ed altre carte. È una comparazione azzardadata tra un libro di finzione, forse basato in fatti reali, dove l’erotismo della donna (Manuela) costituisce la tematica centrale, nell’opera di Romero, con la lunga cronaca letteraria che fa von Hagen e la compilazione e la scoperta di supposti testi originali e storici (lettere, diari) che fa Álvarez Saá. È un esercizio di ricerca dove si mescolano, in modo capriccioso, il campo d’attività del Venezuela o quello del Quito, e la finzione di quello ufficiale. È anche la rivendicazione possibile di Manuela Sáenz come persona e/o personaggio preponderante nelle lotte indipendentistiche dell’America che era aldilà della relazione congiunturale con Simón Bolívar. Também é um exercício de pesquisa em que, de maneira especial, se mistura o que diz o discurso histórico oficial, segundo seja visto pela escola da Venezuela ou a de Quito, com a visão dada pela ficção a esse discurso. É, ao mesmo tempo, um pouco a possível reivindicação de Manuela Sáenz como pessoa e/ou personagem preponderante nas lutas de independência da América que chegavam além do seu ¨controvertido¨ relacionamento com Simón Bolívar.     Molecular and functional diversity of culturable bacterial strains associated with Tuber borchii fruit bodies from different Italian sites

    Tuber borchii is an ectomycorrhizal fungus establishing important beneficial symbiosis with the roots of several tree species from different botanical families. Tuber borchii fruit bodies are associated with diverse bacterial communities, characterized by their plant growth promoting (PGP) abilities, such as the production of compounds capable of contrasting the growth of common soil-inhabiting phytopathogens. In this work we studied the molecular and functional diversity of bacterial strains, referable to the Pseudomonas fluorescens, selectively isolated from Tuber borchii samples, collected in three distinct Tuscany geographic areas with high truffle vocation. To this aim, 120 fluorescent colonies were randomly selected and isolated on King’s B agar, molecular characterized by Amplified Ribosomal DNA Restriction Analysis and then identified by amplicon sequencing of the 16S rRNA gene and two housekeeping genes, namely gyrase subunit B (gyrB) and RNA polymerase sigma factor (rpoD). Most of them were affiliated to the Pseudomonas fluorescens group and some isolates to Paenibacillus taiwanensis. Such isolates were functionally characterized assessing their ability to produce siderophores, using the universal Chrome Azurol Sulphonate (CAS) assay. The results obtained showed that siderophores producing isolates were 81,4% of the total, as revealed by a visible halo on CAS agar. Actually, 42,4% produced a discoloration halo from 0.3 to 0.8 cm and 27,9% from 0.9 to 1.4%. Finally, 11% accumulated the higher amounts, producing discoloration halos larger than 1,5 cm. In conclusion, our results allow us to identify functional bacterial strains to be used as specific bacterial inocula for both biocontrol, plant growth promotion and improvement of fungal performance and mycorrhizal establishment
