    The External Dimension of EU Justice and Home Affairs: Tools, Processes, Outcomes

    This working document offers a conceptual framework for understanding the processes underpinning the external dimension of EU Justice and Home Affairs (ED-JHA). Practically, it defines how the export of JHA principles and norms inform the geopolitical ambitions of the EU, i.e. the use of space for political purposes, or the control and management of people, objects and movement. The author begins by investigating how the ENP reconfigures the ED-JHA, and then goes on to discuss various conceptual stances on governance, specifically institutionalism, constructivism, and policy instruments. To conclude he traces the evolution of this external dimension, emphasising, whenever possible, its continuities and bifurcations.     Guglielmo di Saint-Thierry: teologo dell’Eucaristia

    The study  is about the Eucharistic theology in the work of the medieval Cistercian theologian,  William of Saint-Thierry Abbey. First, his general concept of the spirituality of monastic  life is presented, which William based on the paradigm of the liturgical celebration  which by sign (signum) refers to reality (res). Then, his concept of spiritual communion  as inseparably connected with the sacramental communion is discussed. In this part of  the study, William’s images appear as important factors for understanding his theology:  a spiritual kiss as a way of touching the sacrament mystically, as well as an episode  from the monastic life, showing the necessity of receiving the sacrament in a spiritual  way so that it can be properly received in a physical one. In the third part of the study,  the author attempts to summarize the Eucharistic theology of William of Saint-Thierry, which seems to contrast with the colloquial vision of medieval intellectualism, deprived  of mysterious and spiritual elements. Wilhelm z Saint-Thierry: teolog EucharystiiOpracowanie dotyczy teologii eucharystycznej w twórczości średniowiecznego  teologa cysterskiego, Wilhelma z opactwa Saint-Thierry. W tej części studium pojawiają się ważne dla zrozumienia teologii  Wilhelma obrazy: pocałunek duchowy jako sposób dotykania sakramentu w sposób  mistyczny, a także epizod z życia klasztornego, ukazujący konieczność przyjęcia sakramentu  w sposób duchowy, aby móc właściwie przyjąć go na sposób fizyczny. W trzeciej  części opracowania autor podejmuje próbę podsumowania teologii eucharystycznej Wilhelma  z Saint-Thierry, która okazje się kontrastować z potoczną wizją średniowiecznego  intelektualizmu sakramentalnego, wyzbytego pierwiastka misteryjnego i duchowego.

    The authors analyze the relationship between the development of contractual savings institutions and banks'efficiency, credit, and liquidity risks. They discuss the potential mechanisms through which the development of contractual savings institutions may affect the banking sector. They show that the development of contractual savings institutions has a significant impact on bank spreads and loan maturity.     Microbiological and molecular approaches to fight against the vibriosis of sea bass (Dicentrarchus labrax)

    Les bactéries du genre Vibrio et plus particulièrement celles du clade Harveyi, sont responsables de la vibriose, entraînant des mortalités de masse associées à des pertes économiques considérables pour le secteur aquacole. Vibrio Harveyi est un agent pathogène émergent pour le bar commun d'élevage (Dicentrarchus labrax), capable de former des biofilms. Etant donné qu'aucune méthode prophylactique n'est aujourd'hui commercialisée, la lutte contre la vibriose réside dans la prévention et le développement de méthodes de surveillance et de diagnostic. L'objectif de cette thèse consiste à apporter des éléments de compréhension, via la combinaison d'approches microbiologiques et moléculaires, visant à optimiser la gestion du risque Vibrio pour le bar commun d'élevage. Dans un premier temps, des méthodes de quantification et d'identification ont été mises au point. Une méthode de PCR en temps réel, ciblant le gène mreB, a été développée et optimisée pour quantifier V. harveyi à la fois dans l'eau et les biofilms. La "Luvibase", une base de données composée de 23 profils de spectres de masse de souches de Vibrio de collection a ensuite été élaborée, permettant d'identifier les espèces du clade Harveyi par MALDI-TOF MS. Dans un second temps, ces méthodes ont été appliquées lors d'une campagne hebdomadaire d'échantillonnage d'avril à octobre 2018 dans une ferme d'élevage de bars communs. Des échantillons d'eau et de biofilms ont été collectés à partir des bassins d'élevage et des arrivées d'eau chaude et froide, alimentant la ferme. L'abondance de V.harveyi et des Vibrionaceae cultivables à 37°C, mais aussi leur diversité relative ont ensuite été déterminées. Plusieurs espèces potentiellement pathogènes ont régulièrement été identifiées, dont V.harveyi, tout au long de la campagne. L'étude de l'influence des paramètres environnementaux a mis en évidence une corrélation positive entre la température de l'eau et l'abondance de ces bactéries à la fois dans l'eau et les biofilms. Sachant que ces biofilms peuvent induire une persistance de V.harveyi au sein de la ferme, et que leur abondance pourrait prochainement augmenter du fait du réchauffement climatique, il est urgent de développer des solutions anti-biofilms, afin de limiter le risque Vibrio.Vibrio bacteria, and particularly members of the Harveyi clade, are the causative agents of vibriosis, responsible for mass mortality events and importatn economic losses on aquaculture farms. Vibrio harveyi, which can form biofilms on aquaculture facility surfaces, has recently been identified as an emerging threat for seabass (Dicentrarchus labrax) aquaculture. As no prophylactic method is available on the market, it is urgent to improve surveillance and diagnosis tools to successfully manage vibriosis outbreaks. This thesis focuses on the optimisation of vibriosis risk reduction strategies in seabass farms, using a combination of microbiological and molecular approaches. First, we develop quantification and identification methods. A mreB-targeted SYBR Green I real-time PCR method was developed to quantify V. harveyi directly from environmental samples, including seawater and biofilms. A new in-house database named Luvibase, composed of 23 reference MALDI-TOF mass spectra profiles from vibrio collections strains, was elaborated in order to identify Vibrio species belonging to the Harveyi clade, by MALDI-TOF MS. Second, those methods were applied during a sampling campaign from April to October 2018 in a seabass aquaculture facility. Seawater and biofilm samples were collected from rearing tanks, and from the cold- and hot-water inlets, supplying the farm. The abundance of V. harveyi and Vibrionaceae cultivable at 37°C and their relative diversity were then determined. Several pathogens for seabass were constantly isolated during the sampling campaign. A positive correlation was highlighted between seawater temperature and their abundance in both types of samples. As these biofilms may act as a reservoir of V. harveyi in the farm, and as its abundance is likely to increase due to global warming, anti-biofilm solutions are urgently needed, in order to manage the risk of Vibrio outbreak

    La fraîcheur est un paramètre clé de la qualité du poisson. Les méthodes actuelles appliquées en routine pour déterminer la fraîcheur du poisson ne sont pas applicables à toutes les espèces et reflètent davantage un début d'altération du produit. Ainsi, la recherche d'indicateurs précoces de fraîcheur du poisson représente encore un défi majeur et d'actualité dans l'industrie de la pêche. Le but de ces travaux de thèse était de démontrer que les fonctions et l'intégrité mitochondriales étaient susceptibles de constituer des indicateurs précoces de la fraîcheur de filets de poisson. En effet, la mitochondrie est la "centrale" énergétique de la cellule eucaryote et joue un rôle clef dans les mécanismes de mort cellulaire tels que l'apoptose et la nécrose. Les fonctions et l'intégrité mitochondriales de cellules musculaires de filets de poisson ont été étudiées à différents temps de conservation post mortem à 4°C. Le modèle d'étude était la daurade royale (Sparus aurata) (lignée cellulaire de fibroblastes (SAF-1) et muscles de poisson). Dans un premier temps, la structure des mitochondries de poisson a été étudiée par microscopie électronique à transmission. De nombreuses dégradations de la structure des mitochondries ont été observées dans les filets à partir de 72 heures (J3) de conservation à 4°C. Ces altérations se sont accentuées à J4 et J6. La fonctionnalité des mitochondries a ensuite été évaluée selon deux approches : la respiration mitochondriale (oxygraphie) et le potentiel membranaire mitochondrial (ΔΨₘ) estimé avec la sonde fluorescente Rhodamine 123. A partir de 96 heures de conservation à 4°C (J4), ces deux paramètres ont été significativement impactés témoignant d'une altération des fonctions et de l'intégrité mitochondriales.Ces résultats sont ainsi en corrélation avec l'altération structurale observée par microscopie. En parallèle, une méthode d'évaluation du potentiel membranaire a été développée avec un fluorimètre à microvolume à partir d'un modèle bactérien puis de mitochondries isolées. Ces travaux de thèse ont démontré que l'étude des fonctionnalités mitochondriales constitue un marqueur fiable et précoce de la fraîcheur des filets de poisson. Des connaissances supplémentaires sur les mécanismes cellulaires post mortem ont également été apportées. Ces résultats constituent ainsi le point de départ pour le développement d'un kit d'évaluation de la fraîcheur et ouvrent la voie pour la recherche de marqueurs de fraîcheur et de congélation/décongélation basés sur les fonctionnalités et intégrité mitochondriales.Freshness is a key parameter of fish quality. Current routine techniques to determine fish freshness are not applicable to all species and reflect a late stage of alteration. Thus, research on early indicators of fish freshness still represents a major and topical challenge in fishing industry. This PhD research project aimed to demonstrate that mitochondrial functions and integrity constitute early indicators of fish fillet freshness. Mitochondria are the powerhouse of the cell and play a central role in cell death mecanisms such as apoptosis and necrosis. Mitochondrial function and integrity in fish filet muscle cells were studied at different times of storage post mortem at 4°C. The species studied as a model was the gilthead seabream (Sparus aurata) (gilthead seabream fibroblast cell line (SAF-1) and fish fillets). Firstly, the structure of fish mitochondria was studied by transmission electron microscopy. Numerous mitochondrial structural alterations have been observed in fish fillet from 72 hours (D3) of storage at 4°C. These alterations were more pronounced at D4 and D6. Then, mitochondrial functionality was assessed with two approaches: mitochondrial respiration (oxygraphy) and mitochondrial membrane potential (ΔΨₘ) estimated with the fluorescent probe Rh123. From 96 hours of storage at 4°C (D4), these two parameters were significantly disrupted demonstrating the alteration of mitochondrial function and integrity. The results are in correlation with the mitochondrial structural alterations described by microscopy. In parallel, a method of mitochondrial membrane potential evaluation has been developed with a micro-volume fluorimeter, first using bacteria and then isolated mitochondria. This work demonstrated that the mitochondrial functionality study constitutes a reliable and early fish filet freshness indicator. Additional knowledge on cell mechanisms in post mortem condition has been brought. These results constitute the starting point for the development of a fish freshness assay kit and pave the way to research on others freshness and freeze-thawing indicators based on mitochondrial integrity and functionality
