Abstract 2177: The olive biophenol oleuropein selectively induces apoptosis in pancreatic cancer cells <i>in vitro</i>
Abstract
Pancreatic cancer is a devastating disease with a 5-year survival rate of less than 5%. Resistance to conventional treatment options and toxicity of current chemotherapy agents (gemcitabine) makes pancreatic cancer a target for the development of novel therapeutic agents. Oleuropein is the most abundant biophenol found in olive products; it has anti-atherogenic and anti-inflammatory properties as well as activity against cancers of the breast, colon and prostate. However, there has yet to be any investigation into the effects of oleuropein on pancreatic cancer cells. Consequently, this study aimed to assess the anti-pancreatic cancer activity of oleuropein in vitro. Two cell lines were investigated; a pancreatic cancer cell line (MiaPaCa-2) and a normal pancreas cell line (HPDE). The viability of cells after treatment with 0-200μM oleuropein was assessed using the Dojindo CCK-8 viability assay and compared to gemcitabine. The induction of apoptosis was measured by way of caspase 3/7 activation, using a MUSE flow cell analyser, and expression of Bcl-2, Bax and Bac using Western blot. Cell cycle analysis was conducted using the MUSE flow cell analyser. RNA expression was assessed using Affymetrix GeneChip® Whole Transcript (WT) expression arrays. The IC50 values for oleuropein against MiaPaCa-2 cells was148μM. However, importantly, oleuropein did not decrease the viability of HPDE cells within the treatment range. In comparision, 20nM of gemcitabine did not show selectivity; it reduced the viability of MiaPaCa-2 cells to 21% and of HPDE cells to 2%. An increase in the expression of caspase 3/7 was seen in MiaPaCa-2 cells when treated with oleuropein but it had no effect on the HPDE cells. Furthermore, when treated with oleuropein, an increase in the expression of genes involved in the NRF-2 (oxidative stress) pathway was observed in MiaPaCa-2 cells, an effect not observed in HPDE cells.
Conclusion Oleuropein selectively induced apoptosis in the pancreatic cancer cells (MiaPaCa-2), appearing non-toxic to normal pancreas cells (HPDE) within the treatment ranges; this is significant, since gemcitabine was comparatively more toxic to HPDE cells. Furthermore, the link between oleuropein and the NRF-2 pathway in MiaPaCa-2 cells justifies further study into the mechanisms of action of oleuropein and its potential as a novel therapeutic approach for pancreatic cancer.

Longus’s influential novel, Daphnis and Chloe, is barely present in Hungarian literary education. This paper sets out to provide a methodological example for the understanding of the novel by applying some basic poetic-narratological aspects and by doing this the distance between ancient culture and today’s reader can be reduced. The interpretation of the novel starts with the most important questions expressed in the prologue (prooimium): the unanimity and difference between the real and the fictional author/narrator; the role of intertextuality; the interconnection of word and image, the novel as ekphrasis; the understanding of the relation between nature (physis) and human creation (technē). Then it continues to explore the characters of the two protagonists, Daphnis and Chloe, the gods who have a role in their love and experiences (Eros, Pan, and Dionysus), the threatening external forces, and the most crucial questions of space, time and bucolic idyll. All these are subordinated to the most significant interpretational possibilities (the novel as the allegory of Love), which provides the opportunity to show the author’s, Longus’s, ironic perspective.Longus’s influential novel, Daphnis and Chloe, is barely present in Hungarian literary education. Discussion of wildlife trade before and during the COVID-19 pandemic in professional opinion pieces and scientific articles
Wildlife trade is a multi-billion-dollar sector that impacts a wide range of species, and thus is of significant research and conservation interest. Wildlife trade has also become a prominent topic in the public-facing media, where coverage has intensified following the outbreak of the global COVID-19 pandemic due to the potential connection between wildlife trade and the origin of the SARS Cov2 virus. Given the importance of the media in shaping public understanding and discourse of complex topics such as wildlife trade, this could impact the implementation of and public support for policy decisions. In this study, we followed a standardised protocol to extract wildlife trade-related discussion from 285 professional opinion pieces (NGO reports or articles in conservation-themed forums) and 107 scientific articles published in two time periods: “pre-COVID” (June 1–December 31, 2019) and “during-COVID” (January 1–May 31, 2020). We compared opinion pieces and scientific articles across the two time periods and to each other to investigate potential differences in the presentation of wildlife trade and associated speakers. We found a shift in the way that wildlife trade was discussed in professional opinion pieces between the periods, in that the discussion became less specific in terms of defining the legality and purpose of trade, and the animal groups involved in the “during-COVID” period. The generalised framing of wildlife trade in our dataset also coincided with an increased discussion of highly generalised management strategies, such as blanket bans on wildlife trade. We also found that publications included more quotes from researchers in the “during-COVID” period. In both professional opinion pieces and scientific articles, we found that quotations or research were often from speakers whose affiliation region was different to the geographic range of the trade they were speaking about. This highlights the importance of incorporating local knowledge and considering the diversity of speakers and interviewees in both research and the public-facing media about the wildlife trade
Open access to scholarly communications: advantages, policy and advocacy
The Open Access (OA) movement regards OA modes of disseminating research as the unequivocal future of scholarly communication. Proponents of the open access movement itself have, over the last ten years, carried out systematic research to show how OA can tangibly benefit researchers, institutions and society at large. Even so, the number of research papers being uploaded to OA institutional repositories remains relatively low, frequently based on concerns which often contradict the facts. Policies for OA have been introduced to encourage author uptake, and these are also discussed here. Briefly delineating aspects of these phenomena, this paper will then move on to outline and discuss advocacy for OA in organisations, and whether this should be “downstream”, in the form of informational campaigns, or “upstream”, in the form of top-down change management. This paper seeks to make a contribution to these issues in the OA sphere, by brining into the debate strands from the literature of the sociology of science and management science that will hopefully elucidate aspects of author reactions to OA, and the perceived changes that its adoption gives rise to
Ultrasound increases the aqueous extraction of phenolic compounds with high antioxidant activity from olive pomace
Olive pomace is a waste produced by the olive oil industry in massive quantities each year. Disposal of olive pomace is difficult due to high concentrations of phenolic compounds, which is an environmental concern. However, phenolic compounds have applications in the health industry. Therefore, extraction of phenolic compounds from olive pomace has the potential to remove an environmentally hazardous portion of pomace while creating an additional source of income for farmers and producers. Using advanced technologies including Ultrasound Assisted Extraction (UAE), combined with water as an extraction solvent, has recently gained popularity. The present study outlines the optimal UAE conditions for the extraction of phenolic compounds with high antioxidant activity from olive pomace. Optimal conditions were developed using RSM for parameters power, time and sample-to-solvent ratio. Total phenolic compounds determined by Folin Ciocalteu method and total major bioactive compounds determined by HPLC as well as antioxidant capacity (DPPH and CUPRAC) were investigated. The optimal conditions for the extraction of phenolic compounds with high antioxidant activity were 2 g of dried pomace/100 mL of water at 250 W power for 75 min. UAE improved the extraction efficiency of water and yielded extracts with high levels of phenolic compounds and strong antioxidant activity

Although the plot is somewhat predictable and at times leaves the reader questioning the characters’ motivations, it engages the reader through the anticipation of Chloe’s gradually-revealed memories. PLASTICITY: Shaping the transitions to a resilient and circular plastic manufacturing system
Different government bodies, from the European Union down to the Port of Rotterdam Authority have implemented the policy goal to become circular by 2050 to minimize demand and dependence on finite resources. However, a circular economy - an economic model in which materials circulate in closed loops and waste is viewed as a resource - is vastly different from the current linear system of extraction, consumption and waste. The plastics industry that is currently present in the port of Rotterdam is one such industry where large amounts of raw material is being used and large amounts of waste generated. It is still far from circular. Thus, transitions in the industry itself and in our interaction with products are necessary. The question that arises is: how can the transition to circular plastic manufacturing in the Rotterdam maritime region contribute to a more sustainable, resilient and just economy?Theory on transition management and socio-technical systems, analyses of spatial use and networks of the plastics industry in South Holland, and research on the developments in plastic manufacturing, has led to the understanding of the current networks and flows in the plastics industry. A subsequent analysis of stakeholders, policies and design options has led to a vision and strategy for the South Holland region, on how to shape a new circular plastics economy.In the proposed strategy of Plasticity, (1) the strength of a strategic location in the port of Rotterdam is used to expand the renewable cycle of the bioplastics industry, and (2) by actively engaging citizens in reusing and recycling plastic products on a local level in the whole region, a technical circular cycle is enhanced in the whole province. Plastic is used as an example to demonstrate the contemporary issues around dependency on fossil material in the South Holland context, but similar principles regarding integrating the biological and technical cycle, facilitating space for innovation and growth of circular models, and engaging the whole socio-technical system in the transition process can be applied to other sectors and places. Explanation of raw data: We measured development time (measured in h), egg-to-adult survival (proportion viability), chill coma recovery time (measured in h), oxidative stress resistance (age at death, measured in h), triglyceride content (measured in µg), wing size (measured in mm2), femur and tibia length (measured in mm), allometry differences (ratios of femur length to tibia length, (2) femur length versus wing area, and (3) tibia length versus wing area), Jacobian determinant values of wing shape differences (for males and females separately) for isochromosomal lines of D. melanogaster from Florida and Maine that were either homokaryotypic for the In(3R)Payne inverted arrangement or for the standard chromosomal arrangement. Our main goal was to analyze how traits covary with inversion karyotype (inverted versus standard arrangement) and geography (Florida versus Maine).
