    Going Beyond Counting First Authors in Author Co-citation Analysis

    The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed.     “Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship

    Appropriate Similarity Measures for Author Cocitation Analysis

    We provide a number of new insights into the methodological discussion about author cocitation analysis. We first argue that the use of the Pearson correlation for measuring the similarity between authors’ cocitation profiles is not very satisfactory. We then discuss what kind of similarity measures may be used as an alternative to the Pearson correlation. We consider three similarity measures in particular. One is the well-known cosine. The other two similarity measures have not been used before in the bibliometric literature.     Dispelling the Myths Behind First-author Citation Counts

    We conducted a full-scale evaluative citation analysis study of scholars in the XML research field to explore just how different from each other author rankings resulting from different citation counting methods actually are, and to demonstrate the capability of emerging data and tools on the Web in supporting more realistic citation counting methods. Our results contest some common arguments for the continued use of first-author citation counts in the evaluation of scholars, such as high correlations between author rankings by first-author citation counts and other citation counting methods, and high costs of using more realistic citation counting methods that are not well-supported by the ISI databases. It is argued that increasingly available digital full text research papers make it possible for citation analysis studies to go beyond what the ISI databases have directly supported and to employ more sophisticated methods

    Les radiosensibilisants sont des agents qui rendent les cellules cancéreuses plus sensibles à l'irradiation en ciblant leurs voies de réparation de l'ADN, la régulation du cycle cellulaire, la production d’espèces réactives de l’oxygène (ROS), etc. Ils sont utilisés dans quelques types de radiothérapie. Certains de ces radiosensibilisants sont des ligands de l’ADN produisant des dommages à l’ADN tels que des complexes métalliques ou des ligands de structure G-quadruplex (G4). Pt-ctpy est un complexe de platine ligand de G4 qui a montré des propriétés de radiosensibilisation (RS) via un ciblage proposé des télomères. Par conséquent, nous avons essayé d’identifier les déterminants structuraux pour une RS (ligand G4 et/ou métal). Un criblage cellulaire de RS a donc été effectué, avec des complexes métalliques ligands ou non de G4 de trois séries, terpyridine, salphène et des complexes de platine N-hétérocycliques-Carbene (NHC-Pt) conjugués au ligand Pyridodicarboxamide PDC, ainsi que d’autres complexes de platine comprenant le cisplatine et des complexes NHC-Pt.Le potentiel RS de ces complexes a été testé à leurs concentrations subtoxiques combinées au rayonnement γ (IR) dans trois lignées de cellule différentes (ovaire, poumon et glioblastome). Seuls Pt-ttpy de la série des métal-terpyridines, 2 métal-salphènes (Ni et Cu) et 6 complexes NHC-Pt sont capables d'induire la RS dans les lignées cellulaires. La RS ne suit pas une relation d'activité de structure puisque, 1) tous les ligands métalliques de G4 et 2) tous les métaux n’induisent pas de RS. Les complexes Pt-NHC font l'objet d'un brevet pour leur structure et leur potentiel de radiosensibilisation.Des études mécanistiques ont été poursuivies pour le complexe ligand de G4, Pt-ttpy qui cible les télomères, se lie irréversiblement à l'ADN et induit également un dysfonctionnement mitochondrial. Les pré-requis pour son activité RS sont très différents de ceux des NHC-Pt et du cisplatine. Sa RS dépend du temps de pré-incubation qui a pu être corrélée à la quantité de Pt liée à l'ADN. La présence de Pt-ttpy post-irradiation est également indispensable suggérant que la réparation des dommages à l'ADN induits par l'IR peut être perturbée par la présence d’un adduit de Pt-ttpy. Cela a été corrélé à 1) un retard dans la réparation précoce et une augmentation des dommages à l'ADN jusqu'à 6 heures après irradiation 2) une faible augmentation des dommages aux télomères et 3) une diminution des cellules en phase G2-M aux premiers stades post-irradiation, suggérant une mauvaise réparation de l’ADN génomique et télomérique. La RS est également indépendante de la production de ROS et de la présence de dommages télomériques pré-irradiation.Tous les résultats nous amènent à émettre l'hypothèse d'un mécanisme d'action pour la RS de Pt-ttpy, principalement axé sur le retard de la réparation des dommages à l’ADN dus à l’IR. Nous proposons que lorsque l’adduit encombrant à l’ADN de Pt-ttpy, présent avant irradiation et formé après irradiation, est situé près des dommages à l'ADN induits par l’IR, il se forme un dommage à l’ADN complexe plus difficile à réparer et qui conduirait à la mort cellulaire.De plus, Pt-ttpy est également capable d'induire une chimio sensibilisation aux inhibiteurs de la signalisation « DNA Damage Response » (ATM, ATR, DNA-PK) et à un inhibiteur de PARP1, protéine impliquée dans plusieurs voies de réparation d’ADN, mettant en évidence sa valeur dans une approche chimio-thérapeutique combinatoire et l’implication des adduits de Pt-ttpy à l’ADN dans sa cytotoxicité.En conclusion, ce projet met en évidence de nouvelles propriétés pour le complexe métallique ligand de G4, Pt-ttpy, qui sont différentes de celles du cisplatine et de Pt-ctpy, ce qui ouvre la voie à une sélection de nouveaux complexes métalliques dotés de propriétés de ciblage radiosensibilisant, chimiosensibilisant et mitochondrial.Radiosensitizers are drugs that can sensitize cancer cells to irradiation by targeting their DNA repair pathways, cell cycle regulation, reactive oxygen species (ROS) production, etc. Some of these radiosensitizers are DNA binding and damaging agents such as metal complexes or G-quadruplex (G4) binding ligands. The results show that only Pt-ttpy in the metal-terpyridine series, 2 metal-salphen (Ni and Cu) and 6 NHC-Pt complexes non G4-ligands are able to induce RS in different cell lines. This indicates that the RS effect does not follow a structure activity relationship since, 1) not all G4 ligands induces RS and 2) Not any specific metal induces RS. The Pt-NHC complexes are currently undergoing a patent for their structure and their radiosensitization potential.The drug chosen for in depth mechanistic studies was the G4-binding complex Pt-ttpy that targets telomeres and irreversibly binds to DNA by coordination, leading to damaged bases and DNA double stranded breaks. Interestingly, Pt-ttpy also induces mitochondrial dysfunction without ROS formation, in a complete contrast from cisplatin activity. The requirements for its RS activity is quite different from the ones of NHC-Pt and cisplatin. RS depends on pre-incubation time that could be correlated to the amount of Pt bound to DNA which has been seen in the case of cisplatin as well. Pt-ttpy needs to be present post irradiation, hinting that the IR-induced DNA damage repair may be perturbed by the presence of Pt-ttpy DNA-adduct, in contrast to cisplatin. This has been correlated to 1) a delay in early DNA repair and an increase in DNA damage up to 6hrs post irradiation when treated with Pt-ttpy, and 2) a weak telomeric DNA damage and 3) a decrease of cells in G2-M phase in early stages post irradiation when treated with Pt-ttpy suggesting improper repair of the cellular genomic DNA. In contrast, cisplatin and other non RS inducing complexes do not show any of these features, suggesting a unique RS mechanism for Pt-ttpy. It is also devoid of ROS production (unlike cisplatin) and independent of telomere targeting pre-irradiation (unlike RHPS4 and Pt-ctpy).All the results lead us to hypothesize a mechanism of action for Pt-ttpy induced RS, mostly focused on the delay of IR-induced-DNA damage repair. We propose that the bulky Pt-ttpy-DNA adduct, when present before and formed after irradiation, is located near the IR-induced DNA damage inducing the formation of a complex DNA damage that is more difficult to repair and can lead to cell death.In addition, Pt-ttpy is also able to induce chemo sensitization for DNA Damage Response (DDR) signaling (ATM, ATR, DNA-PK) inhibitors and DNA damage sensor (PARP1) indicating its value in a combinatorial chemotherapeutic approach and also highlighting the involvement of Pt-ttpy DNA-adducts in its cytotoxicity.     We have done our best to complete the author checklist relating to the use of animals in the hut study. Note that the objective for the hut study was to evaluate the IRS treatment applications for residual efficacy against Anopheles mosquitoes, including the local An. coluzzii mosquito population. Cows were only used to attract mosquitoes into the huts and no tests were carried out directly on the cows. The author checklist is intended for use with studies where experiments are carried out on animals, which is why we have had such difficulty in completing this for the hut study, as many of the questions do not relate to how the cows were used
