    Going Beyond Counting First Authors in Author Co-citation Analysis

    The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed.     “Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship

    Appropriate Similarity Measures for Author Cocitation Analysis

    We provide a number of new insights into the methodological discussion about author cocitation analysis. We first argue that the use of the Pearson correlation for measuring the similarity between authors’ cocitation profiles is not very satisfactory. We then discuss what kind of similarity measures may be used as an alternative to the Pearson correlation. We consider three similarity measures in particular. One is the well-known cosine. The other two similarity measures have not been used before in the bibliometric literature.     Dispelling the Myths Behind First-author Citation Counts

    We conducted a full-scale evaluative citation analysis study of scholars in the XML research field to explore just how different from each other author rankings resulting from different citation counting methods actually are, and to demonstrate the capability of emerging data and tools on the Web in supporting more realistic citation counting methods. Our results contest some common arguments for the continued use of first-author citation counts in the evaluation of scholars, such as high correlations between author rankings by first-author citation counts and other citation counting methods, and high costs of using more realistic citation counting methods that are not well-supported by the ISI databases. It is argued that increasingly available digital full text research papers make it possible for citation analysis studies to go beyond what the ISI databases have directly supported and to employ more sophisticated methods

    Die Rolle von T-Zellen in einem akuten und subchronischen Tiermodell der Zigarettenrauch-induzierten pulmonalen Entzündung

    In the present study, a mouse model of cigarette smoke-induced lung injury was thoroughly characterized and shown to result in a distinct and reproducible pulmonary inflammation in the short-term and mid-term setup. Special emphasis was placed on the role of T-cells and the interplay between the innate immune system and the adaptive immune system within the model. To unravel the role of different T-cell subtypes, different genetically modified mice were used. CD4-/- or CD8-/- mice lack either T helper cell acitivity or cytotoxic T-cell activity and therefore share an increased susceptibility to infections. A common feature of CD4-/- and CD8-/- mice is the compensation of the the lack of one T-cell subtype by the expansion of the other T-cell subtype. A major finding using these mice was that neither CD4+ T-cells nor CD8+ T-cells are required to cause pulmonary inflammation induced by mid-term cigarette smoke exposure. This was consistently seen in several parameters such as cell counts, mediator concentrations in the BAL fluid and gene expression in lung tissue. Using T-cell deficient nude mice, which appear hairless and athymic and are characterized by a complete lack of cell-mediated immunity, it was confirmed that T-cells do not have a detrimental function, neither in the short-term nor in the mid-term setup of the model. However, some compensation mechanisms were observed in nude mice, leading to increased inflammatory response of the innate immune system. Furthermore, the present data suggest that T-cells are not involved in resolving cigarette smoke-induced airway inflammation, even though the resolution may be attenuated by T-cells in a specific setup of the model. More studies are needed to unravel this putative harmful function of T-cells. In contrast to the suggestions from studies using other stimuli, T-cells do not seem to protect from inflammation induced by a recurrent smoke stimulus following the resolution of the primary inflammation. Th1 and Tc1 cells were shown to be involved in cigarette smoke-induced pulmonary inflammation by using IFN-γ reporter mice, which enable tracking of IFN-γ in viable cells due to the co-expression of IFN-γ and the reporter protein YFP. Th1 and Tc1 cells are regulated by IL-10, a cytokine with distinct immune-modulatory function. The role of IL-10 was investigated using IL-10 deficient mice which share an increased susceptibilitiy to inflammatory stimuli and are prone to develop intestinal inflammation. Accordingly, IL-10 controls pulmonary inflammation by preventing overwhelming accumulation of neutrophils, macrophages and T-cells in the model presented. Thereby, IL-10 modulates a complex immune network linking the innate and adaptive immune system in cigarette smoke-induced lung injury. Even if analyses of biopsies from COPD patients indicate that the adaptive immune system may be involved in the pathophysiology of the disease, the specific role of lymphocytes is still unclear. However, observations from animal models remain conflicting to date. There are some publications supporting the findings of this study that T-cells may not be crucial in cigarette smoke-induced airway inflammation in mice. Others are in contrast, demonstrating a detrimental function of T-cells especially in later stages of disease models. More studies are needed to investigate the linkage between the early phase and the late phase of cigarette smoke models and the role of T-cells therein. Moreover, there is still a lack of knowledge about the antigens which activate T-cells and promote their putative detrimental function.In der vorliegenden Studie wurde ein Mausmodell der Zigarettenrauch-induzierten Lungenschädigung eingehend charakterisiert. In einem akuten und subchronischen Setup zeigte sich eine deutliche und reproduzierbare pulmonale Entzündung. Ein besonderes Augenmerk lag auf der Untersuchung der Rolle der T-Zellen und dem Zusammenspiel zwischen dem angeborenen und adaptiven Immunsystem in diesem Tiermodell. Um die Rolle der verschiedenen T-Zell Subtypen darin zu entschlüsseln, kamen verschiedene genetisch veränderte Mäuse zum Einsatz. CD4-/- oder CD8-/- Mäusen fehlen entweder die T-Helfer-Zell Funktion oder die zytotoxische T-Zell Funktion. Beide Mausstämme zeigen daher eine erhöhte Infektionsanfälligkeit. CD4-/- und CD8-/- Mäusen ist gemeinsam, dass sie das Fehlen der einen T-Zell Subpopulation durch die Erweiterung der jeweils anderen T-Zell Subpopulation kompensieren. Eine wichtige Erkenntnis war, dass weder CD4+ T-Zellen noch CD8+ T-Zellen dafür notwendig sind, die Lungenentzündung durch subchronische Zigarettenrauchexposition auszulösen. Dies wurde übereinstimmend in mehreren Parametern wie Zellzahl, Mediator-Konzentrationen in der BAL und Genexpression im Lungengewebe detektiert. Durch die Verwendung von T-Zell defizienten Nacktmäusen, welche haarlos, ohne Thymus und durch ein komplettes Fehlen der zellvermittelten Immunität charakterisiert sind, wurde der Befund bestätigt, dass T-Zellen weder nach akuter noch nach subchronischer Zigarettenrauchexposition eine schädliche Funktion haben. Es wurden jedoch einige Kompensationsmechanismen in Nacktmäusen beobachtet, welche zu einer verstärkten entzündlichen Reaktion des angeborenen Immunsystems führten. Darüber hinaus deuten die vorliegenden Daten darauf hin, dass T-Zellen nicht an der Beseitigung der Zigarettenrauch-induzierten Entzündung der Atemwege beteiligt sind, auch wenn es so scheint als ob T-Zellen die Auflösung der Entzündung in einer bestimmten Konfiguration des Modells verzögern würden. Weitere Untersuchungen sind notwendig, um diese vermeintlich schädliche Funktion von T-Zellen aufzudecken. Im Gegensatz zu den Hinweisen aus Studien die andere Stimuli verwendet haben, scheinen T-Zellen auch nicht von einer Entzündung durch wiederkehrende Rauchexposition zu schützen, nachdem die initiale Entzündung beseitigt wurde. Durch die Verwendung von IFN-γ Reporter Mäusen, welche die Nachverfolgung von IFN-γ in lebenden Zellen durch die Co-Expression von IFN-γ und dem Reporterprotein YFP erlauben, wurde gezeigt, dass Th1- und Tc1 Zellen an der Entstehung der Zigarettenrauch-induzierten Lungenentzündung beteiligt sind. Beide Zelltypen werden durch IL-10, einem Zytokin mit ausgeprägter immunmodulatorischer Funktion, reguliert. Die Rolle von IL-10 wurde mittels IL-10-/- Mäusen untersucht, die durch ein verstärktes Ansprechen auf entzündliche Stimuli und durch eine erhöhte Anfälligkeit für Darmentzündungen charakterisiert sind. Dementsprechend kontrolliert IL-10 die Lungenentzündung im hier beschriebenen Modell durch die Verhinderung einer überschießenden Akkumulation von neutrophilen Granulozyten, Makrophagen und T-Zellen. Demnach moduliert IL-10 ein komplex vernetztes Immunsystem und stellt so eine Verknüpfung der angeborenen und adaptiven Immunantwort im Modell der Zigarettenrauch-induzierten Lungenschädigung dar. Auch wenn Analysen von Biopsien von COPD-Patienten nahelegen, dass das adaptive Immunsystem in der Pathophysiologie der Krankheit beteiligt ist, bleibt die spezifische Funktion von Lymphozyten weiterhin unklar. So sind Befunde aus Untersuchungen von Tiermodellen für COPD bislang widersprüchlich. Einige Publikationen unterstützen die Ergebnisse der hier vorgelegten Studie, dass T-Zellen möglicherweise nicht entscheidend an der Zigarettenrauch-induzierten Entzündung der Atemwege bei Mäusen beteiligt sind. Andere Publikationen hingegen deuten auf eine zerstörerische Funktion der T-Zellen vor allem in späteren Stadien der Krankheitsmodelle hin. Weitere Untersuchungen sind notwendig, um die Verbindung zwischen der frühen Phase und der späten Phase der Zigarettenrauch Modelle und die spezifische Rolle der T-Zellen darin zu untersuchen. Es ist auch noch sehr wenig über die Antigene bekannt, die die T-Zellen aktivieren und zur Förderung ihrer vermeintlich schädlichen Funktion beitragen

    Die Bedeutung der Prostaglandin-E2-Rezeptoren in der murinen Lyme-Borreliose

    Background and purpose: Prostaglandines are important mediators of allergies, tumors and inflammations, and locally regulate cell homeostasis in many tissues. They are produced by cyclooxygenases (COX) and specific synthetases for example in the CNS, the vessel system and immune cells. Most prostaglandines bind to only one specific G protein-coupled receptor. PGE-2 plays an exceptional role among prostaglandines. PGE-2 not only binds to four different receptors (EP-1 to EP-4), whose expressions are cell- and tissue-specific, but depending on the receptor can act as a pro- or anti-inflammatory mediator. In previous experiments, Prof. Gessner’s workgroup identified prostaglandines to be important factors during inflammatory response due to Borrelia burgdorferi infection. Upon pharmacologic COX-2 inhibition, Borrelia-induced arthritis was greatly reduced in mice. The purpose of this work was to investigate the relevance of EP-3 and EP-4 receptors in inflammation during experimental Lyme arthritis. Hereto, C3H/HeJ mice were treated with a highly selective EP-3 receptor (ONO-AE-248) agonist and a highly selective EP-4 receptor (ONO-AE3-208) antagonist. Additionally, the possible influence of these substances on specific immune responses and on the elimination of pathogens was examined. Methods: Mice were injected subcutanously with the EP-4 antagonist on the day of infection with B. burgdorferi and with the EP-3 agonist on day 7 p. i.. COX-2 inhibitor Rofecoxib was administered using a gavage from day 1 of infection with B. burgdorferi. The swelling and the inflammatory process were measured repeatedly at tibiotarsal joints using an outside caliper. In order to examine the expression profile of relevant genes in the process of treated and non-treated borrelia infection, total RNA from joints of B. burgdorferi infected C3H/HeJ mice was isolated and analysed by quantitative RT PCR. Furthermore, histologic frozen sections of borrelia-infected tibiotarsal joints were prepared and stained with hematoxylin-eosin in order to evaluate the influence of pharmaceutical products on the extent of inflammation. Thioglycollat-elicited peritoneal macrophages as well as endothelial und fibroblast cell lines were stimulated in vitro with B. burgdorferi in the presence or absence of different test substances. Cytokines and PGE-2 secreted by those cells were detected using specific ELISAs. In order to assess genome-wide differences in gene expression resulting from treatment with certain substances, microarray analysis of the total RNA from entire tibiotarsal joints from C3H/HeJ mice on day 9 p. i. was carried out in cooperation with the HZI Braunschweig. Results and observations: The four EP-receptors were found to be expressed on PMNs, M, DCs, endothelial cells, PECs and in tibiotarsal joints of B. burgdorferi-infected mice. A significant reduction in joint swelling could be observed in C3H/HeJ mice treated with the EP 3-agonist from day 7 p.i. and with the EP-4 antagonist from day 1 of infection. The reduced inflammatory response was confirmed histologically where fewer cell infiltrates were found in joints of treated mice. Microarray analysis detected a significantly reduced expression of the chemokine Cxcl-5 in C3H/HeJ mice treated with the EP-4 antagonist and the COX-2 inhibitor Rofecoxib compared to control mice. The chemokine CXCL-5 is known to attract granulocytes. Regulation of this chemokine through PGE-2 was confirmed in experiments with COX-2-deficient mice. Peritoneal macrophages were identified as important cellular source for CXCL-5 production. Further experiments revealed that neither the specific immune response nor the elimination of borrelia were influenced by treatment with the EP-4 antagonist or COX-2 inhibitor. Practical conclusion: Severe Lyme arthritis could be assuaged by EP-3 agonists as well as EP 4 antagonists. Pro-inflammatory effects of PGE-2, produced upon Borrelia infection, were apparently mediated via the EP-4 receptor, anti-inflammatory effects via the EP-3 receptor. The dichotomous function of PGE-2 is defined by the receptor’s expression kinetics. The EP-3 agonist could be of therapeutical importance, as it displays anti-inflammatory effects even if treatment is initiated on day 7 p. i., when Lyme arthritis is clinically established.Hintergrund und Ziele: Prostaglandine sind wichtige Mediatoren u. a. von Allergien, Tumoren und Entzündungen und regulieren in vielen Geweben lokal die Zellhomöostase. Sie werden durch Cyclooxygenasen und spezifische Synthetasen z. B. im ZNS, dem Gefäßsystem und von Immunzellen hergestellt. Die meisten Prostaglandine binden an einen einzigen spezifischen G-Protein-gekoppelten Rezeptor. Unter den Prostaglandinen nimmt PGE-2 eine außergewöhnliche Rolle ein. PGE-2 bindet nicht nur an 4 verschiedene Rezeptoren (EP-1 bis EP-4), welche zell- und gewebespezifisch exprimiert werden, sondern kann in Abhängigkeit vom Rezeptor sowohl pro- als auch anti-inflammatorisch wirken. In vorangegangenen Versuchen der Arbeitsgruppe von Prof. Gessner haben Prostaglandine sich als wichtige Faktoren bei der Entzündungsreaktion infolge einer Borrelia burgdorferi-Infektion erwiesen. Bei pharmakologischer COX-2-Inhibition war die Borrelien-induzierte Arthritis in Mäusen stark abgeschwächt. Ziel dieser Arbeit war es, die Relevanz des EP-3- und EP-4-Rezeptors für die Entzündungsgenese im Modell der murinen Borreliose zu untersuchen. Hierzu wurden C3H/HeJ-Mäuse mit einem hochselektiven Agonisten des EP-3-Rezeptors (ONO-AE-248) und einem hochselektiven Antagonisten des EP 4-Rezeptors (ONO-AE3-208) behandelt. Des Weiteren sollte ein etwaiger Einfluss der Substanzen auf die spezifische Immunantwort und die Erregerelimination geprüft werden. Methoden: Der EP-4-Antagonist wurde den Mäusen am Tag der Infektion mit B. burgdorferi und der EP-3-Agonist ab dem siebten Tag p. i. subkutan injiziert. Der COX 2 Inhibitor Rofecoxib wurde den Mäusen ab dem Tag der Infektion mit B. burgdorferi mittels Schlundsonde verabreicht. Durch wiederholtes Messen der Tibiotarsalgelenke mit Hilfe eines Außentasters wurde anschließend der Schwellungs- und Entzündungsverlauf registriert. Um das Expressionsprofil relevanter Gene im Verlauf einer Borrelieninfektion mit und ohne Behandlung zu untersuchen, wurde die Gesamt-RNA aus Gelenken B. burgdorferi-infizierter C3H/HeJ-Mäuse isoliert und nach reverser Transkription mittels quantitativer PCR analysiert. Des Weiteren wurden histologische Gefrier-Schnitte Borrelien-infizierter Tibiotarsalgelenke angefertigt und mit Hämatoxylin-Eosin gefärbt, um den Einfluss der Pharmaka auf das Ausmaß der Entzündung zu beurteilen. Thioglycolat-elizitierte Peritonealmakrophagen sowie Endothel- und Fibroblastenzelllinien wurden in vitro unter An- oder Abwesenheit verschiedener Test-Substanzen mit B. burgdorferi stimuliert. Von den Zellen sezernierte Zytokine und PGE-2 wurden mit spezifischen ELISAs detektiert. Um genomweit Genexpressions-Unterschiede feststellen zu können, die sich durch die Behandlung mit bestimmten Substanzen ergeben, wurde in Kooperation mit dem HZI Braunschweig eine Analyse der Gesamt-RNA aus kompletten Tibiotarsalgelenken von C3H/HeJ-Mäusen am Tag 9 p. i. mittels Microarray durchgeführt. Ergebnisse und Beobachtungen: Bei Untersuchungen des Expressionsprofils der 4 EP-Rezeptoren konnten diese in PMNs, Makrophagen, DCs, Endothelzellen, PECs und auch in Tibiotarsalgelenken B. burgdorferi-infizierter Mäuse nachgewiesen werden. Durch die Behandlung von C3H/HeJ-Mäusen mit dem EP-3-Agonisten ab Tag 7 p. i. und dem EP 4 Antagonisten ab dem Tag der Infektion konnte eine signifikante Reduktion der Gelenkschwellung erreicht werden. Histologisch konnte die abgeschwächte Entzündungsreaktion insofern bestätigt werden, als in den behandelten Gelenken weniger Zellinfiltrate beobachtet wurden. In der Microarray-Analyse konnte eine verglichen mit Kontrollmäusen signifikant verminderte Expression des Chemokins Cxcl-5, das neutrophile Granulozyten anlockt, infolge einer Behandlung von C3H/HeJ-Mäusen mit dem EP 4 Antagonisten und dem COX-2-Inhibitor Rofecoxib festgestellt werden. Die Regulation dieses Chemokins durch PGE-2 konnte in Experimenten mit COX 2 defizienten Mäusen bestätigt werden. Als eine wichtige zelluläre Quelle für die CXCL-5-Produktion wurden Peritonealmakrophagen identifiziert. In weiteren Versuchen konnte gezeigt werden, dass weder die spezifische Immunantwort noch die Elimination der Borrelien durch die Behandlung mit dem EP-4-Antagonisten oder dem COX-2-Inhibitor beeinflusst werden. Praktische Schlussfolgerung: Die Schwere der Lyme-Arthritis ließ sich sowohl durch den EP-3-Agonisten als auch durch den EP-4-Antagonisten reduzieren. Pro-inflammatorische Effekte des bei einer Borrelieninfektion produzierten PGE-2 werden daher offensichtlich über den EP-4-Rezeptor vermittelt, anti-inflammatorische über den EP-3-Rezeptor, wobei die unterschiedliche Expressionskinetik der Rezeptoren die dichotome Funktion von PGE-2 verursacht. Von therapeutischer Bedeutung könnte der EP-3-Agonist sein, da dieser eine anti-inflammatorische Wirkung auch noch bei Behandlungsbeginn am Tag 7 p. i., d. h. bei schon manifester Entzündungssymptomatik, entfaltet

    We have done our best to complete the author checklist relating to the use of animals in the hut study. Note that the objective for the hut study was to evaluate the IRS treatment applications for residual efficacy against Anopheles mosquitoes, including the local An. coluzzii mosquito population. Cows were only used to attract mosquitoes into the huts and no tests were carried out directly on the cows. The author checklist is intended for use with studies where experiments are carried out on animals, which is why we have had such difficulty in completing this for the hut study, as many of the questions do not relate to how the cows were used
