Analysis of maturation-specific genes in soybean seeds: Gene structure, desiccation induction and abscisic acid responsiveness

    No full text
This study describes two maturation (Mat) genes which share some structural similarities with a special group of genes, whose expression can be induced by water stress and also by exogenous abscisic acid (ABA) application. Previous studies have identified a 31-kDa maturation polypeptide (MAT) which accumulates during normal and precocious maturation of soybean (Glycine max L. Merr) seeds, and disappears rapidly during germination. The corresponding mRNA can also be induced in germinating seeds with ABA treatment, but not in normally developing seeds. By screening a 65 day-after-flower (DAF) seed cDNA library with a partial cDNA clone corresponding to the 31 kDa MAT protein, we were able to isolate and characterize two closely-related gene sequences represented by the pMat1 and pMat9 clones. Primer extension studies indicated that these two clones were full length cDNA clones. In the soybean genome, we identified at least two copies of the Mat1 genes and one Mat1-related gene, probably the Mat9 gene, in both the Williams'82 and T157 varieties. The nucleotide sequence comparison of cDNA and genomic clones showed these Mat genes do not contain any introns, which was confirmed by polymerase-chain-reaction (PCR) analysis of soybean seed mRNA and genomic DNA. The pMat 1 and pMat9 clones not only differ in size (51 nucleotides less in the coding region of pMat9 than in the coding region of pMat1), but the corresponding genes also differ in their expression patterns during normal and precocious maturation stages, and also by differential response to exogenous abscisic acid during germination. The protein sequence analysis showed that MAT proteins contain a highly conserved C-terminal region, which is also present in rice RAB (ABA-responsive) proteins and also in the cotton LEA and barley dehydrin proteins. The N-termini of MAT proteins also contains a short 7-amino acid stretch which might be related to the tissue-specificity expression or activity of these proteins. Possible roles of MAT proteins in seed maturation processes are discussed.Made available in DSpace on 2011-05-07T12:28:46Z (GMT). One preschool teacher's approach to play intervention: An interpretive study based on Vygotsky's concept of activity

    No full text
This study explores how the social interactions in a play intervention are socially constructed by the teacher and the children within the context of classroom organization and structure. Further, it explores how the organization and structure of the classroom are affected by the demands placed on the teacher by the preschool in which she works, by her beliefs about child development, and by the larger contemporary discourse on early childhood education. This study is based upon Vygotsky's concept of activity. Vygotsky's concept of activity asserts that the meaning of an action lies in the actor's intentions, that is, action is the means by which an actor achieves her intentions. Thus intention states must be interpreted within the social context within which the actions are embedded."Using ethnographic methods, I interpret the intentions of teacher and children in a play intervention episode by combining ""close analysis of fine details of behavior and meanings in (the) social interaction with analysis of the wider social context--the field of broader social influence--within which the face-to-face interaction takes place"" (Erickson, 1986, p.120)."The teacher's play intervention strategies were constituted by her practical-knowledge-in-use (Clandinin, 1986). Her practical-knowledge-in-use reflects her beliefs about early childhood education and the demands placed on her as a teacher, e.g., parents' expectations, school philosophy. The usefulness of Vygotsky's concept of activity to understand classroom process is discussed. First, Vygotsky's concept of activity provides a way to study teachers' intentions in their interactions with children. Teaching becomes, then, not a series of behaviors but of meaningful actions by which the teacher struggles to meet the many demands placed on her as a professional. Second, Vygotsky's concept of activity provides a most useful framework for studying face-to-face interactions within a classroom. Revealed are the lived experiences of children and teachers as they make sense out of the constraints placed upon them as they interact in classroom.Made available in DSpace on 2011-05-07T14:15:55Z (GMT). No full text
[[abstract]]practices of occupational health nurses and their deterrent factors. A questionnaire was mailed to all occupational health nurses in industrial manufacturing plants with more than 300 employees in Taiwan. The response rate was 50.51%. For the study, the duties of occupational health nurses were classified into five categories: medical service delivering, emergency planing, environmental health hazards identification, medical examination and case management, and health education. Among all of the duties, 'medical service delivering' was practiced best, and 'health education' was the worst. For the total working time, 29.8% was spent on 'medical service delivering', 23.39% on 'medical examination and case management', and 26.74% on 'other fairs'. The result different task had different deterrent factors was also showed. However, increasing some specific knowledge and skills, the sense of duty, and the support from the authorties were suggested for better occupational health nursing practice.
