Realizace auditů je v práci popsána komplexně, všechny druhy auditů jsou charakterizovány z hlediska svého průběhu. Improving pancreatic lineage commitment of human pluripotent stem cells in terms of pancreas development and disease modelling
No full textPancreatic lineage differentiation has predominantly focused on driving the generation of functional β-cells, given their significance in diabetes research and therapy. However, for comprehensive disease modeling and a better understanding of the entire pancreas, including its interactions and functions, it is likewise crucial to generate cells of the exocrine compartment. The objective of the first part of this doctoral thesis was to optimize the differentiation of hESCs toward the pancreatic lineage. This included a detailed characterization of pancreatic progenitors to understand their marker profiles and cellular heterogeneity, and to direct their further specification into endocrine and exocrine lineages. Additionally, this work aimed to implement an ex vivo culture system to achieve robust, long-term maturation of pancreatic cells.
The second part of the thesis aimed to utilize the optimized differentiation system for modelling monogenic neonatal diabetes (Figure 2). In this specific case, a child diagnosed with severe neonatal diabetes, and his family history of T2D, served as basis for this study. This part of the doctoral thesis involved integrating gene engineering with pancreatic differentiation, various sequencing approaches and functionality assays to decipher the underlying disease mechanisms. A key focus was to elucidate the regulatory role of a non-coding region on ONECUT1 expression and its impact on islet development

No full textDie in diesem Band vorgelegten Arbeiten zielen auf eine historisch-soziologische Analyse der Geschichte der Schweiz der Vor- und Zwischenkriegszeit. In diesem Zeitraum durchläuft die Schweiz eine Entwicklung, die vom brüchigen Burgfrieden 1914 über die klassenantagonistische Krise ausgangs des Ersten Weltkrieges in die «Hochzeit» des Bürgerblocks der 20er Jahre führt und schliesslich über die Krise der 30er Jahre - konkordanzpolitisch befriedet - in das sozialmarktwirtschaftliche Zeitalter einmündet. Dem Zweiten Weltkrieg stellt sich im Gegensatz zum Ersten eine weitgehend geeinte Schweiz entgegen. Diesen spannungsreichen, von Konflikt wie Konkordanz geprägten sozialen Wandel nachzuzeichnen und zu periodisieren, ist Thema der Arbeiten.
Die einzelnen Beiträge, deren gemeinsames Erkenntnisinteresse sich in den Begriffen «diskontinuierlicher sozialer Wandel» und «Öffentlichkeit» konkretisiert, nehmen sich diesem Thema auf besondere Weise an: Allesamt nähern sie sich der Vor- und Zwischenkriegszeit über eine Rekonstruktion des öffentlichen Diskurses - die Zeit wird über Zeitungen erschlossen, um die Einschätzung vergangener Gegenwarten und Zukünfte, die Eigen- und Fremdbilder, ihre Brüche und Kontinuitäten im öffentlichen politischen Raisonnement zu erfassen.
Der vorliegende Band ist der erste der Reihe «Krise und sozialer Wandel». Dementsprechend befasst sich ein Beitrag mit den theoretischen und methodischen Grundlagen dieser Reihe, die sich mit dem öffentlichen Diskurs auseinandersetzt. Der nächste Band beschäftigt sich mit der Nachkriegszeit und der Periode des Kalten Krieges, der dritte Band mit den 60er und 70er Jahren
