    Jörg Fauser was the enfant terrible of the German literary scene. Born in the Taunus town of Bad Schwalbach in 1944, he worked a number of different jobs (temporary worker, airport worker, night guard, journalist, author and rock songwriter). The night after his forty-third birthday, he died tragically and under mysterious circumstances when walking on the A94 highway between Feldkirchen and Riehm near Munich, where he was hit by a truck. It was only after his death that he became an iconic figure. In 1988, Das Syndikat, a group of German crime writers, awarded him the prestigious Ehrenglauser prize for crime fiction, named after the Swiss author Friedrich Glauser

    Alla luce della definizione provocatoria di biblioteca "virtuale" dei fratelli Schlegel, l'articolo si occupa di discutere come l'attività di recensore di August Wilhelm Schlegel, fondatore - con il fratello Friedrich - della cultura ufficiale della Frühromantik, ci permetta oggi di ricostruire una biblioteca che non esiste più, in quanto fu dispersa già in occasione della morte del noto studioso tedesco. Le recensioni di August Wilhelm, dottissimo critico e traduttore, consentono altresì di considerare la gran messe di generi che la scrittura del '700 poteva offrire ai suoi lettori: almanacchi e plaisanterien, florilegi e libretti per le dame. Partendo da questo primo approccio alla cultura letteraria del '700, l'articolo esplicita come gli stessi Schlegel amassero sperimentarsi su generi nuovi, come, ad esempio, le "Charakteristiken": una forma mista di biografia, interpretazione testuale e profilo della personalità dell'autore. Dalle scelte tematiche e di gusto, che riflettono la tensione della cultura tedesca a un dialogo con le altre civiltà europee, si configurano anche i presupposti per discutere il tema della "socievolezza" nel primo romanticismo. Questo atteggiamento culturale, tipico dei saloni letterari di inizio '800, ma discendente dalla tradizione delle riunioni nelle case delle gran dame francesi, viene elevato nella prospettiva schlegeliana a una forma comunicativa e di scambio di idee che viene definita sinfilosofia. Nelle recensioni i critici jenensi non si limitano a argomentare intorno al tono, alla maniera e allo stile di un'opera, bensì affrontano anche la questione della Weltanschauung e lo stile di vita dello scrittore. Dalle suddette premesse, è molto più facile comprendere come la cerchia di Jena non fosse solo un ricettore di idee culturali, ma anche fucina e laboratorio di originali forme poetiche.     The possibility to predict the outcome of targeted DNA double-stranded break (DSB) repair would be desirable for genome editing. Furthermore the consequences of mis-repair of potentially cell-lethal DSBs and the underlying pathways are not yet fully understood. Here we study the clustered regularly interspaced short palindromic repeats (CRISPR)/Cas9-induced mutation spectra at three selected endogenous loci in Arabidopsis thaliana by deep sequencing of long amplicon libraries. Notably, we found sequence-dependent genomic features that affected the DNA repair outcome. Deletions of 1-bp to &lt;1000-bp size and/or very short insertions, deletions &gt;1 kbp (all due to NHEJ) and deletions combined with insertions between 5-bp to &gt;100 bp [caused by a synthesis-dependent strand annealing (SDSA)-like mechanism] occurred most frequently at all three loci. The appearance of single-stranded annealing events depends on the presence and distance between repeats flanking the DSB. The frequency and size of insertions is increased if a sequence with high similarity to the target site was available in cis. Most deletions were linked to pre-existing microhomology. Deletion and/or insertion mutations were blunt-end ligated or via de novo generated microhomology. While most mutation types and, to some degree, their predictability are comparable with animal systems, the broad range of deletion mutations seems to be a peculiar feature of the plant A. thaliana

    The two books reviewed here both contribute to our understanding of how transnational participation and negotiation are embedded in the classical music tradition of the United States. Hess shows us that the study of music in the Western classical tradition cannot escape its intrinsic cosmopolitanism and that transnational dialogues are actually central to its expressive and discursive universes. Fauser asks us to attend to musical practices beyond the concert hall and that may involve musical actors peripheral to grand narratives of music

    The hydrolysis of starch is a key step in plant germination, which also has relevance in the malting and brewing processes for beer and spirit production. Gaps in knowledge about this metabolic process exist that cannot easily be addressed using traditional genetic techniques, due to functional redundancy in many of the enzyme activities required for alpha-glucan metabolism in cereal crop species. Chemical inhibitors provide opportunities to probe the role of carbohydrate-active enzymes and the phenotypes associated with inhibition of specific enzymes. Iminosugars are the largest group of carbohydrate-active enzyme inhibitors and represent an underused resource for the dissection of plant carbohydrate metabolism. Herein we report a method for carrying out a reverse chemical genetic screen on α-glucosidase, the enzyme that catalyzes the final step in starch degradation during plant germination, namely the hydrolysis of maltose to release glucose. This chapter outlines the use of a high-throughput screen of small molecules for inhibition of α-glucosidase using a colorimetric assay which involves the substrate p-nitrophenyl α-D-glucopyranoside. Identified inhibitors can be further utilized in phenotypic screens to probe the roles played by amylolytic enzymes. Furthermore this 96-well plate-based method can be adapted to assay exo-glycosidase activities involved in other aspects of carbohydrate metabolism

    Novel micro-phenotyping approach to chemical genetic screening for increased plant tolerance to abiotic stress

    Studying the effects of small molecules on root system development in the context of a large-scale chemical genetic screen has previously been a technical challenge. The recent development of novel seedling growth devices (“Phytostrips”), used in combination with standard 96-well microtiter plates, has made it possible to perform detailed studies of changes in root morphology and root system architecture following the application of a library of chemical compounds. Phytostrips were originally designed to allow automated robotic capture of images of roots and shoots of the model species Arabidopsis thaliana, but can also be used for manual screens that are more laborious but do not require the investment in expensive robotics. Here we describe a protocol for the use of Phytostrips to perform chemical genetic screens that rely on clearly observable changes in root morphology or root system architecture. As an example, we describe the use of polyethylene glycol to impose an abiotic stress related to reduced water potential and the application of a chemical screen for small molecules that are able to rescue Arabidopsis root development from the disruptive effect of the polyethylene glycol treatment. The protocol we describe provides a template for the application of a multiplicity of other screens for compounds that can antagonize the effects of a range of abiotic stresses on root development

Epilepsia. 2021.OBJECTIVE: Rasmussen encephalitis (RE) is a progressive and destructive inflammatory disease of one hemisphere. Its cause is unknown. We investigated comorbidity and laterality factors that might predispose to RE.; METHODS: We retrospectively compared the histories of 160 RE patients to those with genetic generalized epilepsy (n=154) and those with focal cortical dysplasia Type II (FCD II; n=148).; RESULTS: The median/mean age at symptom onset in RE was 7/10years (range = 1-53years), and 58.1% of the patients were female. The female sex predominated in RE patients, with age > 7years at disease manifestation. The left hemisphere was affected in 65.6%. Perinatal complications (preterm birth, twin pregnancies, early acquired brain lesions) were more frequent in RE than in control patients. Ipsilateral facial autoimmune conditions (scleroderma en coup de sabre, uveitis, or chorioretinitis) were only observed in RE patients (6.9%). Onset of RE was more frequently associated with fever than that of FCD II. In 33.1% of RE patients, ≥1 potential risk factor was found. Interestingly, 11.9% of patients had one-sided early brain lesions or facial autoimmune lesions ipsilateral to subsequent RE; none had such a lesion contralaterally.; SIGNIFICANCE: Perinatal complications and facial autoimmune conditions may act as predisposing factors for RE. Fever might trigger RE manifestation. Further genetic or infectious contributors may be identified in the future. Single or combined hits may be required to elicit or facilitate the start of the disease. Ipsilateral early comorbid lesions or facial autoimmune processes might in part explain the enigmatic unilaterality of RE. © 2021 The Authors.
