Age-dependent decrease in 11beta-hydroxysteroid dehydrogenase type 2 (11beta-HSD2) activity in hypertensive patients
BACKGROUND: The prevalence of arterial hypertension lacking a defined underlying cause increases with age. Age-related arterial hypertension is insufficiently understood, yet known characteristics suggest an aldosterone-independent activation of the mineralocorticoid receptor. Therefore, we hypothesized that 11beta-HSD2 activity is age-dependently impaired, resulting in a compromised intracellular inactivation of cortisol (F) with F-mediated mineralocorticoid hypertension. METHODS: Steroid hormone metabolites in 24-h urine samples of 165 consecutive hypertensive patients were analyzed for F and cortisone (E), and their TH-metabolites tetrahydro-F (THF), 5alphaTHF, TH-deoxycortisol (THS), and THE by gas chromatography-mass spectroscopy. Apparent 11beta-HSD2 and 11beta-hydroxylase activity and excretion of F metabolites were assessed. RESULTS: In 72 female and 93 male patients aged 18-84 years, age correlated positively with the ratios of (THF + 5alphaTHF)/THE (P = 0.065) and F/E (P < 0.002) suggesting an age-dependent reduction in the apparent 11beta-HSD2 activity, which persisted (F/E; P = 0.020) after excluding impaired renal function. Excretion of F metabolites remained age-independent most likely as a consequence of an age-dependent diminished apparent 11beta-hydroxylase activity (P = 0.038). CONCLUSION: Reduced 11beta-HSD2 activity emerges as a previously unrecognized risk factor contributing to the rising prevalence of arterial hypertension in elderly. This opens new perspectives for targeted treatment of age-related hypertension
18F-RB390: innovative ligand for imaging the T877A androgen receptor mutant in prostate cancer via positron emission tomography (PET).
BACKGROUND
Detecting prostate cancer before spreading or predicting a favorable therapy are challenging issues for impacting patient's survival. Presently, 2-[(18) F]-fluoro-2-deoxy-D-glucose ((18) F-FDG) and/or (18) F-fluorocholine ((18) F-FCH) are the generally used PET-tracers in oncology yet do not emphasize the T877A androgen receptor (AR) mutation being exclusively present in cancerous tissue and escaping androgen deprivation treatment.
METHODS
We designed and synthesized fluorinated 5α-dihydrotestosterone (DHT) derivatives to target T877A-AR. We performed binding assays to select suitable candidates using COS-7 cells transfected with wild-type or T877A AR (WT-AR, T877A-AR) expressing plasmids and investigated cellular uptake of candidate (18) F-RB390. Stability, biodistribution analyses and PET-Imaging were assessed by injecting (18) F-RB390 (10MBq), with and without co-injection of an excess of unlabeled DHT in C4-2 and PC-3 tumor bearing male SCID mice (n = 12).
RESULTS
RB390 presented a higher relative binding affinity (RBA) (28.1%, IC50 = 32 nM) for T877A-AR than for WT-AR (1.7%, IC50 = 357 nM) related to DHT (RBA = 100%). A small fraction of (18) F-RB390 was metabolized when incubated with murine liver homogenate or human blood for 3 hr. The metabolite of RB390, 3-hydroxysteroid RB448, presented similar binding characteristics as RB390. (18) F-RB390 but not (18) F-FDG or (18) F-FCH accumulated 2.5× more in COS-7 cells transfected with pSG5AR-T877A than with control plasmid. Accumulation was reduced with an excess of DHT. PET/CT imaging and biodistribution studies revealed a significantly higher uptake of (18) F-RB390 in T877A mutation positive xenografts compared to PC-3 control tumors. This effect was blunted with DHT.
CONCLUSION
Given the differential binding capacity and the favorable radioactivity pattern, (18) F-RB390 represents the portrayal of the first imaging ligand with predictive potential for mutant T877A-AR in prostate cancer for guiding therapy. Prostate 75:348-359, 2015. © 2014 Wiley Periodicals, Inc
'Pergularain e I'--a plant cysteine protease with thrombin-like activity from Pergularia extensa latex
Pergularain e I, a cysteine protease with thrombin-like activity, was purified by ion exchange chromatography from the latex of Pergularia extensa. Its homogeneity was characterized by sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE), native PAGE and reverse-phase high-performance liquid chromatography (RP-HPLC). The molecular mass of pergularain e I by matrix-assisted laser desorption ionization-time of flight (MALDI-TOF) was found to be 23.356 kDa and the N-terminal sequence is L-P-H-D-V-E. Pergularain e I is a glycoprotein containing approximately 20% of carbohydrate. Pergularain e I constituted 6.7% of the total protein with a specific activity of 9.5 units/mg/min with a 2.11-fold increased purity. Proteolytic activity of the pergularain e I was completely inhibited by iodoacetic acid (IAA). Pergularain e I exhibited procoagulant activity with citrated plasma and fibrinogen similar to thrombin. Pergularain e I increases the absorbance of fibrinogen solution in concentration-dependent and time-dependent manner. At 10 microg concentration, an absorbance of 0.48 was reached within 10 min of incubation time. Similar absorbance was observed when 0.2 NIH units of thrombin were used. Thrombin-like activity of pergularain e I is because of the selective hydrolysis of A alpha and B beta chains of fibrinogen and gamma-chain was observed to be insusceptible to hydrolysis. Molecular masses of the two peptide fragments released from fibrinogen due to the hydrolysis by pergularain e I at 5-min incubation time were found to be 1537.21 and 1553.29 and were in close agreement with the molecular masses of 16 amino acid sequence of fibrinopeptide A and 14 amino acid sequence of fibrinopeptide B, respectively. Prolonged fibrinogen-pergularain e I incubation releases additional peptides and their sequence comparison of molecular masses of the released peptides suggested that pergularain e I hydrolyzes specifically after arginine residues

In amyotrophic lateral sclerosis (ALS) the large motoneurons that innervate the fast-contracting muscle fibers (F-type motoneurons) are vulnerable and degenerate in adulthood. In contrast, the small motoneurons that innervate the slow-contracting fibers (S-type motoneurons) are resistant and do not degenerate. Intrinsic hyperexcitability of F-type motoneurons during early postnatal development has long been hypothesized to contribute to neural degeneration in the adult. Here, we performed a critical test of this hypothesis by recording from identified F- and S-type motoneurons in the superoxide dismutase-1 mutant G93A (mSOD1), a mouse model of ALS at a neonatal age when early pathophysiological changes are observed. Contrary to the standard hypothesis, excitability of F-type motoneurons was unchanged in the mutant mice. Surprisingly, the S-type motoneurons of mSDO1 mice did display intrinsic hyperexcitability (lower rheobase, hyperpolarized spiking threshold). Reduced 11β‐hydroxysteroid dehydrogenase activity in experimental nephrotic syndrome
BACKGROUND: The disease state of the nephrotic syndrome is characterized by abnormal renal sodium retention that cannot be completely explained by a secondary hyperaldosteronism for the following reasons. Firstly, in rats an enhanced sodium retention is observed before proteinuria with intravascular volume depletion occurs. Secondly, in patients with the nephrotic syndrome, volume expansion with hypertension has been reported despite suppression of the renin-aldosterone system. Therefore, another mechanism for sodium retention must be postulated for this disease state. We hypothesize that this mechanism is a reduced 11beta-hydroxysteroid dehydrogenase 2 (11beta-HSD2) activity, a phenomenon known to cause enhanced access of cortisol or corticosterone to the mineralocorticoid receptor. METHODS: We assessed the 11beta-HSD activity by measuring the urinary ratio of tetrahydrocorticosterone (THB) plus 5alpha-tetrahydrocorticosterone (5alpha-THB) to 11-dehydro-tetrahydrocorticosterone (THA) by gas chromatography-mass spectrometry in rats with puromycin aminonucleoside (PAN)-induced proteinuria and with adriamycin nephrosis. Furthermore, the plasma ratios of corticosterone to 11-dehydrocorticosterone were measured. RESULTS: The urinary ratio of (THB+5alpha-THB)/THA increased in all animals following injection of PAN or adriamycin, indicating a reduced activity of 11beta-HSD. The reduced activity of 11beta-HSD was confirmed by an increased plasma ratio of corticosterone to 11-dehydrocorticosterone. The changes in the glucocorticoid metabolite ratios were already present before significant proteinuria appeared. CONCLUSION: PAN- or adriamycin-treated rats develop proteinuria with a reduced activity of 11beta-HSD, a mechanism contributing to the abnormal sodium retention in nephrotic syndrome
Taxonomy and phylogeny of the Central European bug genus "Psallus" (Hemiptera, Miridae) and faunistics of the terrestrial Heteroptera of Basel and surroundings (Hemiptera)
True bugs or Heteroptera are a biologically diverse, species-rich group containing some
37,000 species described worldwide (Schaefer & Panizzi, 2000). It is the largest taxon of
endopterygotan insects and constitutes a well-supported monophylum within the order
Hemiptera. Species are usually trophically specialised; they are phytophagous, zoophagous or
mycophagous (Schuh & Slater, 1995). For their often specialised feeding habits, many species
are economically important as crop pests, biological control agents of phytophagous insect
pests (Schaefer & Panizzi, 2000) or as vectors of human diseases (Schofield & Dolling, 1993;
Schaefer, 2000; Garcia et al., 2000). The last one is, however, without significance in Central
Europe. Some bugs constitute an important protein source in human diet (Fritsche Gitsaga,
2000).
Within the Heteroptera the plant bugs or Miridae constitute the largest family with more
than 10,000 described species (Schuh, 1995). It is expected that the family may contain twice
as many species (Wheeler, 2001). Even though the mirids contain many economically
important species, its taxonomy and systematics contains many unsolved problems. This is a
result of the large number of included taxa and the uniformity of external morphology of
many genera. Current problems exist at species level where some taxa are ill-defined or where
many species can be identified only by male genitalic characters, and at genus level where
phylogenetic considerations are missing for many taxa.
The first list of Swiss Heteroptera containing 50 species is part of Füessly’s (1775)
catalogue of insects from Switzerland. Several additional records were inlcuded in general
works of 18th and 19th century entomologists (Sulzer, 1771, 1776; Razoumowsky, 1789;
Roemer, 1789; Schellenberg, 1800; Meyer-Dür, 1843). The only catalogue dealing with Swiss
bugs was provided by Frey-Gessner (1864a, b, 1865, 1866a, b, 1871a). Basel and the
surrounding region have, compared to the rest of Switzerland, a distinct fauna and flora due to
the geografical position and the relatively dry climate. The region is open to the Rhône valley
via the "Burgunderpforte" from where mediterranean floristic and faunistic elements may
immigrate. The first part of the present thesis deals with the ill-defined mirid genus Psallus
(Phylinae, Phylini). One aim was to test the monophyly of Psallus and to propose, based on
the study of the Central European species, a classification applicable to the world fauna. The
second goal was to investigate the female genitalia in view of taxonomic and phylogenetic
significance. Female genitalia are so far rarely used for species definition within mirids
(exeptions e. g. Calocoris, Rosenzweig, 1997) in contrast to other families (e. g. Nabidae,
Péricart, 1987). As for Psallus the few species of which the female genitalia have been
described so far (Kullenberg, 1947; Seidenstücker, 1972; Matocq, 1989, 1997) suggest this
may be an useful set of characters. A third aim of the present study was to provide keys for
the identification of the Central European species based exclusively on male and female
genitalic characters.
The second part of the present study deals with the terrestrial bug fauna of the region of
Basel. Currently most of our knowledge goes back to Frey-Gessner's catalogue (1864a, b,
1865, 1866a, b, 1871) – obsolete for a long time. A few scattered papers have been published
since (e. g. Voellmy & Egli, 1981; Meduna et al., 2001) but a thorough inventory lacks to
date. In Switzerland, in general, faunistic surveys on true bugs are scarce in contrast to other
European countries. The relatively few papers recently published on Swiss Heteroptera are
based, to a large extent, on material from traps run for ecological investigations (e. g. Otto,
1996; Di Giulio et al., 2000).
The combination of systematic revisions and faunistic investigations is necessary if we
want to know and conserve our fauna. Turnball (1979) brought this to the point: “How can we
detect change in the future if we cannot define the fauna we now have?
On Geography and Institutions as Determinants of Foreign Direct Investment. A cross country comparative analysis of sub-Saharan African relative to developing countries
This work explores what factors determine foreign direct investment (FDI) in sub-Sahara Africa (SSA) relative to non-sub-Saharan African countries, using a panel data set which encompasses most of the world´s developing countries between 1997 and 2006. The results indicate that institutions and infrastructure development promoted FDI to non-SSA but did not induce FDI to sub-Saharan Africa. Geography played a modest and indirect role. The marginal benefit from openness to trade was higher for SSA, which is closely related to resource-seeking FDI that did not translate into sustained economic growth, neither institutional change, but consequently crowded out the second FDI wave of manufacturing. At the same time, FDI into value-added manufacturing largely located in non-SSA countries acted as engine for scaling the economic development ladder through institutional improvement for a number of non-SSA countries. Hence, FDI has the potential to act as a reliable and equitable driver of sustained economic development and poverty alleviation.
