Espírito Santo: Santa Teresa, Valsugana Velha, Santa Lúcia Biological Station, 750 m, 16 Jun 2001 (fl), <i>Fraga 780</i> (holotype: MBML!; isotype: RB).</p> <p> <b>Distribution:</b> —Southeastern Brazil.</p> <p> <b>Notes:—</b> Like <i>M. conceicionensis</i>, <i>M. lonchophyllus</i> and <i>M. punctatus</i>, this species is endemic to southeastern Brazil and has a dark red to purple-spotted perianth, lip base with a small auricle on both sides and margins revolute to the apex and the characteristic fimbriate anther cap. No full textEste dataset está compuesto por 2 archivos, de los cuales el primero es el conjunto de datos con 1865 análisis de muestras de agua de temperatura, salinidad, oxígeno, nutrientes, pH, alcalinidad y clorofila, y el otro (Readme.txt) incluye una pequeña descripción de las variables calculadasLa campaña se realizó del 11 de Julio al 8 de agosto de 1984 a bordo del B/O "García del Cid" abarcando la zona costera del NW de la península ibérica. El tiempo, en general fue bueno, predominando el viento norte como es general en esta época del año, con una velocidad media de 19 nudos, llegando a 32 nudos. El final de la campaña coincidió con una purga de mar intensa localizada principalmente en las bocas de las Rías Bajas al oeste de la Rías Bajas
Objetivos.- El principal objetivo fue el estudio del afloramiento costero y en especial el afloramiento producido en el frente de contacto entre los dos cuerpos de Agua Central frente al Cabo Finisterre. Otro de los objetivos era la variación de nivel de la capa de separación entre la parte superior del Agua Central y el agua superficial, durante un ciclo de marea y durante un ciclo mensual y la penetración del Agua Central en la Rías. Se presentó frente al Cabo Finisterre un núcleo de afloramiento muy rápido que no da tiempo a la proliferación de fitoplancton por lo que es detectado no solo por su temperatura baja, 13,6ºC, sino también por su alto contenido de nutrientes (ver figura 9 del informe). En la costa NW de Galicia aparece un afloramiento
muy costero bien visible por la distribución de la isoterma de 15,5 V (Figura 7 del informe) pero la velocidad de fotosíntesis es superior a la velocidad de aporte de nutrientes por lo que la concentración de éstos es próxima a cero y la cantidad de fitoplancton es alta. En la parte sur frente a la desembocadura del rio Miño, hay un afloramiento muy costero que rompe la capa superior de agua cálida y poco salada procedente de las rías.
Se usaron botellas Niskin "General Oceanics" de 1,7 L.Temperatura. - Todas las botellas hidrográficas llevaban dos termómetros "Richter y Wiese" protegidos y uno no protegido. La corrección de temperaturas se hizo, según Andersson (l974) con la ecuación no simplificada. La profundidad se calculó a partir de las lecturas termométricas
redondeando el resultado de la decena más próxima.
Salinidad. - Se determinó con un salinómetro de inducción AUTOSAL modelo 8400A calibrado con “agua normal” IAPSO, La salinidad se calculó utilizando las ecuaciones dadas en UNESCO (1981).
Nitrato. - Se determinaron en un auto-analizador por reducción a nitritos en columna de Cd-Cu según la modificación de C. Mouriño y F. Fraga (1985). Las muestras se analizaron inmediatamente después de recogidas
Nitrito. - Por diazotación de sulfanilamida y copulación con naftiletilendiamina, según la modificación de la cita indicada anteriormente en nitratos.
Fosfato y silicato. - Se determinaron por el método descrito en K. Grasshoff (1976).
Oxígeno. - Se determinó por el método de Winkler. La valoración del iodo con tiosulfato se hizo con el titulador automático Metrohm E-425, E-473, con un electrodo de platino.
pH. - Se midió inmediatamente con un pHmetro "Metrohm E-510 con electrodo de vidrio y como referencia uno de Ag-ClAg calibrado con tampón 7,413 NBS. La temperatura en el momento de la medida se toma con un termómetro graduado en décimas y se hizo la corrección de temperatura para referir todos los valores de pH a 15 ºC (Pérez & Fraga, 1987).
Alcalinidad. - Se midió por valoración con HCl 0,1 N a pH final 4,45 siguiendo Pérez & Fraga (1987)Clorofila. - La muestra se filtró por filtros "Whatman GF/C” de 2,5 cm de diámetro. La clorofila se midió fluorométricamente siguiendo la técnica descrita en Strickland y Parsons (1972). Integration of multi-omics layers empowers precision diagnosis through unveiling pathogenic mechanisms on maple syrup urine disease
No full textMaple syrup urine disease (MSUD) is a rare inherited metabolic disorder characterized by deficient activity of the branched-chain alpha-ketoacid dehydrogenase (BCKDH) complex, required to metabolize the amino acids leucine, isoleucine, and valine. Despite its profound metabolic implications, the molecular alterations underlying this metabolic impairment had not yet been completely elucidated. We performed a comprehensive multi-omics integration analysis, including genomic, epigenomic, and transcriptomic data from fibroblasts derived from a cohort of MSUD patients and unaffected controls to genetically characterize an MSUD case and to unravel the MSUD pathophysiology. MSUD patients exhibit a defined episignature that reshapes the global DNA methylation landscape, resulting in the stimulation of HOX cluster genes and the restriction of cell cycle gene-related signatures. Subsequent data integration revealed the impact of AP1-related and CEBPB transcription factors on the observed molecular reorganization, with MEIS1 emerging as a potential downstream candidate affected by robust epigenetic repression in MSUD patients. Furthermore, the integration of multi-omics layers facilitated the identification of a strong epigenetic repression in the DBT promoter in a patient wherein no BCKDH pathogenic variants had been detected. A Circular Chromatin Conformation Capture assay indicated a disturbance of the interactions of DBT promoter, thereby unveiling alternative modes of disease inheritance. Integration of multi-omics data unveiled underlying molecular networks rewired in MSUD patients and represents a powerful approach with diagnostic potential for rare genetic disorders with unknown genetic bases.This work was supported by the Health Research Institute Carlos III (ISCIII) (grant numbers PI22/00699 to Belén Pérez and PI21/01067 to Mario F.
