    Transcription factor oscillations in neural stem cells: implications for accurate control of gene expression

    Naturally occurring oscillations in glucocorticoids induce a cyclic activation of the glucocorticoid receptor (GR), a well-characterized ligand-activated transcription factor. These cycles of GR activation/deactivation result in rapid GR exchange at genomic response elements and GR recycling through the chaperone machinery, ultimately generating pulses of GR-mediated transcriptional activity of target genes. In a recent article we have discussed the implications of circadian and high-frequency (ultradian) glucocorticoid oscillations for the dynamic control of gene expression in hippocampal neural stem/progenitor cells (NSPCs) (Fitzsimons et al., Front. Neuroendocrinol., 2016). Interestingly, this oscillatory transcriptional activity is common to other transcription factors, many of which regulate key biological functions in NSPCs, such as NF-kB, p53, Wnt and Notch. Here, we discuss the oscillatory behavior of these transcription factors, their role in a biologically accurate target regulation and the potential importance for a dynamic control of transcription activity and gene expression in NSPCs

    Nella prima parte della nota, l’Autore analizza la fattispecie di «denuncia di un sinistro non accaduto» di cui all’art. 642, comma 2, c.p., introdotta nel 2002 nell’ambito della riforma delle assicurazioni R.C. Auto, per, poi, affrontare la questione su cui si è pronunciata la Corte di cassazione, relativa al significato da attribuire al concetto di “sinistro”. Infine, l’Autore offre le proprie riflessioni sui rapporti intercorrenti tra la fattispecie esaminata e le altre ipotesi delittuose previste dall’art. 642 c.p. e sulla compatibilità di tali reati con il principio di offensività.In the first part of the note, the Author analyzes the crime of “denunciation of a false accident” that was introduced into the art. 642 c.p. in 2002, and, then, the question about the meaning of “sinistro” (accident) on which the Supreme Court stated.     Antihistaminergics and inverse agonism. Potential therapeutic applications

    The accurate characterization of the molecular mechanisms involved in the action of receptor ligands is important for their appropriate therapeutic use and safety. It is well established that ligands acting at the histamine system currently used in the clinic exert their actions by specifically antagonizing G-protein coupled H1 and H2 receptors. However, most of these ligands, assumed to be neutral antagonists, behave as inverse agonists displaying negative efficacy in experimental systems. This suggests that their therapeutic actions may involve not only receptor blockade, but also the decrease of spontaneous receptor activity. The mechanisms whereby inverse agonists achieve negative efficacy are diverse. Theoretical models predict at least three possible mechanisms, all of which are supported by experimental observations. Depending on the mechanism of action engaged, the inverse agonist could interfere specifically with signaling events triggered by unrelated receptors. This possibility opens up new venues to explain the therapeutic actions of inverse agonists of the histamine receptor and perhaps new therapeutic applications.Fil: Monczor, Federico.     Suspended graphene beams with tunable gap for squeeze-film pressure sensing

    We present suspended graphene pressure sensors fabricated using an innovative surface micro-machining process. The great advantage of this process is that the molybdenum (Mo) catalyst layer for multi-layer graphene chemical vapor deposition (CVD) is also used as a sacrificial layer to suspend the graphene. This method allows for accurate control of the gap size under the beam by simply varying the catalyst thickness. Furthermore, the need for transfer of the graphene layer is eliminated.     2021 butterfly inventories within Boulder County open spaces, Boulder, Colorado

    April 4, 2022.Includes bibliographical references.The butterfly inventories continued for the 18th year in a few of the Boulder County Open Space properties. However, because of the Calwood Fire during October 17, 2020, two of the survey areas were inaccessible; Plumely Canyon and the work road through the streamlet near the Lichen Trail. Geer Watershed had been burned, but the meadows above the Resident Ranger's Cabin and associated streamlets were made accessible with our 2021 permit. Cabbage Whites (Pieris rapae), Orange Sulphurs (Colias eurytheme) and Common Wood-nymphs (Cercyonis pegala) were more numerous when compared to 2015-2020 populations before the burn. In 2021 within other Open Spaces, Anne U. White and Caribou, the resident species produced noticeably large populations: Julia Orangetips (Anthocharis julia), Pine Whites (Neophasia menapia), Northern Checkerspots (Chlosyne palla), Common Wood-nymphs, Small Wood-nymphs (C. oetus), Common Checkered-skippers (Burnsius communis). On the other hand, the well adapted non- native Cabbage Whites were again the most common during the long season; Variegated Fritillaries (Euptoieta claudia) and Dainty Sulphurs (Nathalis iole) were numerous flying in from colonies in the eastern plains and low foothills where their host plants were plentiful. 95 species were seen in 2021: 129 species between 2004 and 2021 in the county Open Spaces. 203 species are on the record for Boulder County (Butterflies and Moths of America 2021) website. The average number of species per year observed within the Open Spaces by this team is 88

    Doublecortin-like kinase : a potential therapeutic target for neuroblastoma

    There is an urgent need for a better and more effective therapy for neuroblastoma, a pediatric tumor of the sympathetic nervous system. Despite intensive treatment, less than 50 % of the patients with high-risk neuroblastoma survive. Therefore we have investigated if doublecortin-like kinase is a potential therapeutic target for neuroblastoma. Doublecortin-like kinase (DCLK1) gene encodes doublecortin-like (DCL) and DCLK-long that are crucial for correct proliferation and differentiation of neuroblasts. We found that these proteins are overexpressed in human neuroblastomas and gliomas but not in other tumors or normal tissue. DCL/DCLK-long knockdown in mouse and human neuroblastoma cells resulted in apoptotic cell death. We also found that DCL is involved in the control of mitochondrial activity, energy production and neuroblastoma cell proliferation. In vivo, we observed delayed tumor growth in neuroblastoma tumor xenografts with inducible DCL knockdown. Moreover, we showed that combining DCL/DCLK-long knockdown with low doses of vinca alkaloids results in a synergistic apoptotic effect, confirming that this is a potential approach to consider for neuroblastoma therapy. In conclusion, the data presented in this thesis provide new insights in the biological function of the DCLK1 gene and indicate that DCL and DCLK-long are potential therapeutic targets for neuroblastoma.UBL - phd migration 201
