    Le déclin des abeilles est le syndrome d’effondrement de leurs colonies, est inquiétant. Les pesticides et la susceptibilité des abeilles à certaines infections contribuent à cette perte. Il est crucial de comprendre les mécanismes d’action des pesticides et parasites responsables de cette mortalité. J’ai étudié les effets de l’infection par la microsporidie T. ratisbonensis et/ou de l’insecticide fipronil chez la drosophile. Les microsporidies prolifèrent en subtilisant les lipides de l’hôte. La phagocytose est la seule réponse de l’hôte conférant une protection contre le parasite. Le fipronil induit un mécanisme de pertes lipidiques lequel repose sur la lipophagie et génère du tréhalose qui alimente un état d’hyperactivité. Le fipronil induit une production d’Akh mobilisant les réserves lipidiques et stimulant un réseau neuronal d’hyperactivité normalement induit en cas de jeûne. Ce mécanisme contribue à la sensibilité des drosophiles exposées à des doses faibles du pesticide.The decline of honeybees, colony collapse disorder, is alarming. Pesticides and the susceptibility of honeybees to some infections contribute to this loss. Understanding the mechanisms of action of pesticides and parasites responsible for this lethality is crucial. I have studied the effects of the infection by the microsporidium T. ratisbonensis and/or the fipronil insecticide in Drosophila. Microsporidia proliferate by hijacking host lipid reserves. Phagocytosis is the sole host defense conferring some protection against the parasite. Also, we have discovered a mechanism of lipidic loss upon ingestion of fipronil, which is mediated by lipophagy and produces trehalose that sustains a hyperactivity state. Fipronil indirectly induces the production of Akh, which partially mediates the mobilization of lipidic reserves and stimulates a hyperactivity neural network normally triggered by starvation. This mechanism contributes to the susceptibility of flies exposed to low doses of fipronil

    Contributions à l’étude des interactions hôte-pathogène entre Drosophila melanogaster et Serratia marcescens

    L’étude des interactions hôte-pathogène permettra de mieux comprendre les bases des maladies infectieuses. Durant ma thèse, j’ai étudié les interactions entre l’organisme modèle Drosophila melanogaster et la bactérie pathogène à Gram négatif Serratia marcescens (S.m). Cette bactérie est capable de tuer les mouches en moins de 24 heures une fois introduite directement dans l’hémolymph. Au contraire, les mouches peuvent survivre plusieurs jours après avoir ingéré du Serratia malgré les dommages à l’épithélium intestinal qui en résulte. Le travail de ma thèse a mené à comprendre la virulence des vésicules de la membrane externe purifiés de S.m et injectés dans la mouche. En plus, nous avons mis en évidence deux gènes qui jouent un rôle dans la virulence de la bactérie en infection intestinale, notamment dans la capacité de S.m à endommager les cellules intestinales. Enfin, nous avons identifié plusieurs gènes impliqués dans un mécanisme de résilience de la drosophile aux infections intestinales par S.m.The study of host-pathogen interactions will provide a better understanding of the basics of infectious diseases. During my thesis, I studied the interactions between the model organism Drosophila melanogaster and the Gram-negative pathogen Serratia marcescens (S.m). This bacterium is capable of killing flies in less than 24 hours once introduced directly into the hemolymph. On the contrary, flies can survive several days after ingesting Serratia despite he resulting damages to the intestinal epithelium. The work of my thesis led to an understanding of the virulence of the outer membrane vesicles purified from S.m and injected into the fly. In addition, we have identified two genes that play a role in the virulence of the bacterium in the intestinal infection model, particularly in the ability of S.m to damage the intestinal cells. Finally, we have identified several Drosophila genes involved in a resilience mechanism to intestinal infections by S.m

    Une partie conséquente de mon travail a été d'effectuer un crible génétique en utilisant une bibliothèque de mutants générés par insertion aléatoire de Tn5-Sm, un minitransposon bactérien. Le crible a été réalisé dans un contexte défini: celui de mouches-hôtes auxquelles manquait le gène Eater, lequel code un récepteur de phagocytose (Kocks et al., 2005). Dans ces mouches, l'infection n'est plus contrôlée dans l'hémocoele par les hémocytes et les drosophiles mutantes succombent rapidement à une bactériémie. Plusieurs phénotypes bactériens étaient attendus à l'issue de ce crible. Une première catégorie de phénotype prévisible était une virulence accrue, par exemple si les bactéries mutantes devenaientcapables de traverser plus rapidement ou efficacement la barrière intestinale conséquemment à la perte d'un régulateur négatif. Un deuxième type de phénotype attendu était une virulence atténuée pouvant s'expliquer de plusieurs manières: 1- perte de résistance à l'environnement existant dans le lumen intestinal (enzymes digestives et lysozyme, radicaux libres et peptides antimicrobiens induits au niveau de l'épithélium intestinal dans le cadre d'une réponse immunitaire locale de l'hôte); 2- incapacité à traverser la matrice péritrophique; 3-incapacité à envahir les cellules épithéliales (adhésion, pénétration); 4- incapacité à résister aux défenses intracellulaires potentielles; 5- incapacité à sortir du côté basal des entérocytes 6- incapacité à proliférer dans l'hémolymphe ou perte de la résistance à l'action de la réponse immunitairesystémique qui est, quant à elle, fortement induite en l'absence de phagocytose, laquelle empêche chez les mouches sauvages la prolifération des bactéries ayant traversé la paroi intestinale. [...] Dans le cadre d'une infection intestinale, les mouches sauvages (et imd) succombaient en six jours alors que, de manière surprenante, les mouches mutantes de la voie Toll périssaient plus lentement, une situation opposée à celle du modèle de la piqûre septique. Quelques bactéries sont capables de traverser la paroi intestinale mais sont incapables de proliférer à moins que la réponse cellulaire ait été préalablement bloquée. L'épithélium intestinal apparaissait normal à la dissection et la presque totalité des bactéries ingérées étaient tuées dans l'intestin. Après avoir exclu l'hypothèse d'une toxine sécrétée dans le surnageant des bactéries adsorbées sur le filtre sur lequel viennent se nourrir les mouches, nous avons testé l'hypothèse qu'une suractivation de la réponse immunitaire était à l'origine du décès des mouches. La génétique mettant hors de cause les peptides antimicrobiens, la voie Toll n'étant apparemment pas activée dans l'épithélium intestinal, nous avons alors étudié laréponse oxydative induite par l'ingestion de bactéries (Ha et al., 2009), laquelle est capable de tuer les mouches lorsqu'elle n'est pas régulée correctement. Là-aussi, le résultat s'est avéré négatif. En fin de compte, j'ai pu établir que la mort des mouches était due à un état de famine, confirmé par des mesures des réserves métaboliques. Mes travaux ont permis d'établir un nouveau rôle de la voie Toll dans la résistance à la famine, en présence ou absence d'infection, qui sera peut-être à mettre en relation avec un rôle métabolique de la voie Toll consistant à bloquer la voie de réponse à l'insuline lors d'une infection. En conclusion, mes travaux permettent de mieux comprendre les relations hôte-pathogènequi s'établissent lors d'une infection intestinale.For the systematic study of bacteri al virulence factors, we initially planned to screen the 12,000 mutant strains of the miniTn5-Sm tranposon-induced mutant bank in a wild-type S. marcescens strain Db10. Phagocytosis-deficient eater mutant flies (Kocks et al., 2005) were used in this screen to isolate the bacterial strains mutated for virulence factors and the genes responsible for crossing the gut barrier. In the eater mutant background, flies succumb to septicemia caused by the rapid proliferation of the bacteria in the hemolymph. Out of 1348 mutant strains screened, 58 candidate mutants have been isolated. Only 20% percent of the potential mutant strains displayed an increased virulence indicating that there are very few factor(s) that negatively control the virulence program of the bacterium. The fly survival phenotypes induced by the candidate mutants isolated in the first round of screen were retested. Only those bacterial strains that were consistent with the phenotype were chosen for the molecular identification of the transposon insertion sites using one primer PCR (Karlyshev et al., 2000). Once the genes impaired in each case had been identified, they were knocked off in the S. marcescens Db10 by site specific plasmid insertion mutagenesis. A mutant strain with the transposon inserted into the fliR gene, a component of the type III flagellar protein export system, exhibited attenuation of virulence. The plasmid insertionmutant strain generated to interrupt the gene fliR reproduced the fly survival phenotyp, indicating that the fliR gene is important for the virulence of S. marcescens. The fliR mutants are able to cross the peritrophic matrix, functionally similar to the human mucus. The bacteria were found in the vicinity of the epithelial cells but were not able to efficiently invade the intestinal epithelium as compared to the wild-type strain. Consequently lower titer of FliR mutants was found in the hemolymph. The inefficiency of the FliR mutants to invade cells was also confirmed in ex-vivo assay using insect cells.I thus demonstrated that the fliR gene which is important in the motility apparatus is also required by S. marcescens for the crossing of the epithelial barrier of D. melanogaster.[...]A strong oxidative response is triggered by D. melanogaster in the midgut against commensals and pathogens (Ha et al., 2009). In order to check whether the strong oxidative immune response is eventually killing the flies themselves, hydrogen peroxide was chemically neutralized in the midgut during the S. xylosus A. oral infection. No difference inthe fly survivals was observed with or without neutralization of the oxidative response indicating that over-production of reactive oxygen species (ROS) does not seem to be responsible for the fly death caused by a very low number of bacteria. Flies could efficiently survive to killed bacteria and filtered supernatant solution from overnight bacterial culture indicating that they do not die to the toxins released by the bacteria. Most surprisingly MyD88-, the Toll pathway-, mutant flies were surviving better to S. xylosus A. oral infection. A series of experiments lead us to the finding that the flies actually succumbed to starvation when orally infected with S. xylosus and that the MyD88 is required for the starvation susceptibility in microbiota-mediated manner. In conclusion my work has lead us to the better understanding of the host-bacterial interactions in the intestine

    Nous étudions les interactions hôte/pathogène entre la drosophile et un parasite intracellulaire, la microsporidie Tubulinosema ratisbonensis.Les microsporidies dérivent du groupe des champignons possédant un génome réduit et pas de mitochondrie. Ces parasites dépendent fortement du métabolisme de leur hôte pour proliférer. Les interactions entre les microsporidies et leurs hôtes reste peu étudiées au niveau métabolique.In vivo, les spores ciblent beaucoup de tissues, en particulier le corps gras qui perd ces gouttelettes lipidiques. Les réserves métaboliques sont épuisées au cours de l’infection et les mouches présentent les caractéristiques de famine. La supplémentation en acide gras dans la nourriture des mouches infectées augmente la prolifération du parasite. Cet effet est bloqué lorsque l’on perturbe l’assimilation ainsi que le transport des lipides provenant de l’intestin. En conséquence, les mouches résistent mieux à l’infection lorsque la synthèse des lipides est bloquée.We study the host/pathogen interactions between Drosophila and a natural intracellular parasite : the microsporidium Tubulinosema ratisbonensis. Microsporidia are highly derived group of fungi with a compact genome and no mitochondria. These obligate parasites thus rely intensively on host metabolism for growth and proliferation. The interactions between microsporidia and their hosts remain poorly understood at the metabolic level.In vivo, spores target many tissues especially the fat body that loses its lipid droplets. Metabolic reserves become depleted during the course of the infection and flies display the hallmarks of severe starvation. Fatty acids supplementation of the infected flies diet increases parasite proliferation and host susceptibility to the parasite. This effect is blocked when perturbing lipid assimilation and transport originating from the gut. Accordingly, flies resist better infection when lipid synthesis is blocked, as the parasite proliferation is hampered

    Plus de 150 années de recherches sur les Microsporidies ont conduit à une connaissance relativement basique de divers aspect de leur biologie. Malgré cela, peut d’informations existent concernant la génétique et les mécanismes moléculaire des interactions hôte-pathogène qui gouvernent les infections aux Microsporidies.Dans un premier temps, je décris comment détecter, traiter et éradiquer les infections microsporidiales avec Tubulinosema ratisbonensis dans des lignées de Drosophila melanogaster. Jusqu’à présent, les connaissances concernant les défenses de l’hôte chez la drosophile contre les parasites intracellulaire obligatoires restent incomplète due au manque d’un bon modèle d’infection. De ce fait, j’ai développé des modèles d’infection de D. melanogaster par la microsporidie T.ratisbonensis, à la fois en culture cellulaire et drosophiles adultes. Mes travaux sur le modèle d’infection cellulaire englobent des approchent en transcriptomique et métabolomique qui analysent les deux cotées de cette relation hôte-pathogène. En fin, je présente les fonctions biologiques des glycosylphosphatidyl inositoles de Toxoplasma gondii.More than 150 years of Microsporidia research led to a basic understanding of many aspects of microsporidial biology, yet little is known about the genetic basis and molecular mechanisms of the intimate host-parasite relationship that govern Microsporidia infections.Here, I first report on the detection, prophylaxis, and eradication measures against microsporidial infestations with Tubulinosema ratisbonensis, infecting cultures of Drosophila melanogaster. To date,knowledge about Drosophila host defense against obligate intracellular parasites remained incomplete for lack of good infection models.To this end, I have developed infection models of Drosophila by the microsporidian T. ratisbonensis,both in cell lines and in adults. The work on the cellular infection model encompasses transcriptomics and metabolomics approaches, which aim to attempt both sides of the host-pathogen equation. Finally, I report on the biological roles of glycosylphosphatidyl inositols of Toxoplasma gondii

    Au cours de ma thèse je me suis intéressée aux relations hôte-pathogène entre Drosophila melanogaster et Pseudomonas aeruginosa PA14. RhlR, un facteur de transcription bactérien permet à la bactérie d’échapper à la phagocytose. Mon projet de thèse consistait à identifier comment RhlR exerce cette fonction. Mes résultats suggèrent que RhlR exercerait également une fonction indépendante du quorum sensing. Un crible de mutants PA14 nous a permis d’isoler trois gènes importants pour la virulence de la bactérie et possiblement reliés à RhlR: xcpR, vfR et sltB1. L’utilisation de mutants de drosophile tep4, m’a permis de montrer que le rôle d’échappement à la phagocytose se ferait au niveau de la détection de la bactérie. Par ailleurs, mes résultats suggèrent aussi l’intervention d’un composé volatil qui permettrait de synchroniser la virulence de la bactérie. Dans une dernière partie, j’ai étudié les effets d’une co-infection entre un virus entérique et PA14.During my PhD, I studied the host-pathogen interactions between Drosophila melanogaster and Pseudomonas aeruginosa PA14. We previously identified RhlR as a bacterial transcription factor that allows the bacteria to circumvent phagocytosis. My main PhD project was to study and identify how RhlR exerts this function. My first results suggested that RhlR plays also a role independently its the quorum sensing. A screen of PA14 mutants allowed me to identify three genes involved in PA14 virulence and possibility in RhlR function: xcpR, vfR and sltB1. By using tep4 fly mutants, I have shown that RhlR’s role against phagocytosis is most likely required at the level of PA14 detection. Beside this, my results indicated that possibly a volatile compound is involved to synchronize PA14 virulence.     Going Beyond Counting First Authors in Author Co-citation Analysis

    The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed. Reasons for these effects are discussed

    S. marcescens ingérée par la mouche la tue en six à huit jours. Afin d'identifier les facteurs de l'hôte impliqués dans le processus infectieux, nous avons mis en oeuvre un crible compréhensif à l'échelle du génome basé sur l'interférence ARN (ARNi) in vivo dans des mouches transgéniques. Ce crible a été réalisé à Vienne en collaboration avec le groupe dirigé par le Pr. J. Penninger (IMBA, Vienne), essentiellement par Nadine Nehme et Shane Cronin. Un crible secondaire concentré sur les gènes candidats possédant des homologues chez les vertébrés et présentant un phénotype lorsque leur expression était ciblée seulement dans l'intestin moyen a permis de restreindre la liste, laquelle contenait plusieurs gènes de la voie JAK-STAT. Nous avons établi que la voie JAK-STAT contrôle le taux de prolifération des cellules souches intestinales au cours de l'infection. Pseudomonas aeruginosa tue la mouche en huit à dix jours après ingestion. La forte prolifération des bactéries dans l'hémolymphe induit les deux voies de la réponse humorale systémique, Toll et Immune Deficiency (IMD). La réponse cellulaire est aussi impliquée. Des bactéries mutants pour le gène rhIR, lequel est requis dans un système de perception du quorum sont moins virulent. La virulence des bactéries rhIR est seulement rétablit dans des mouches dépourvues de réponse cellulaire sont infectées. Nos résultats établissent que le gène rhIR est nécessaire essentiellement pour contrebalancer la seule réponse cellulaire et non la réponse humorale.S. marcescens fed to the fly are killing in 6-8 days. To better understand the host factors implicated in the infection process we performed an ill vivo genome-wide RNAi (RNA interference) screen. This screen was realized in collaboration with the Penninger lab (Vienna). A secondary screen on the candidate genes that have vertebrate homolgs (mouse or human) was carried out using a midgut-specific driver to express the RNAi hairpin constructs. Amongst the genes that showed, in addition to the primary screen, a phenotype in the intestine specific screen, were several members of the JAK-STAT pathway. We were able to show that the JAK-STAT pathway is needed to control the level of compensatory proliferation of ISCs during the infectious process. P. aeruginosa kills the flies within 8-\0 days upon ingestion. A strong proliferation of the bacteria in the hemolymph induces the two pathways of the humoral systemic immune response, Toll and Immune Deficiency (IMD). Phagocytosis is also needed for host defense. Bacteria mutant for the gene rhIR are less virulent. rhIR mutant bacteria just regain their virulence in flies deficient for phagocytosis Our results show that RhIR is needed to counteract the cellular immune response, but not the humoral immune response

    “Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship
