    Economic factors affecting obesity: an application in Italy

    The World Health Organization has stated that obesity is spreading around the world like a “global epidemic”. In 2004 the percentage of obese people in the Italian population was 9%, but the trend s increasing in recent years. Focusing on this country, the purpose of the paper is to analyze the socio-economic variables affecting obesity by means of a survey conducted in a consumer sample. Our analysis is based on a survey conducted in Italy, and the sample was composed of 999 consumers. We used a binary logit model and the dependent variable is body mass index (BMI), expressed in a dichotomic way (seriously overweight and obese, value 1, and normal weight, value 0). The results show that the condition of the seriously overweight and obese increases with age, especially in people over 65 of age. Also gender is correlated with the pathology: being seriously overweight and obese is far more likely for men than for women. An inverse relation was shown between obesity and education, and between obesity and the level of food knowledge. The results highlight that disadvantaged social categories are more susceptible to the problem of overweight and obesity. A policy implication of the analysis, to limit the spread of obesity, could lie in programs aimed at improving health and food awareness and focused on these minority groups.economics of obesity, BMI and consumer, logit model, Food Consumption/Nutrition/Food Safety, Health Economics and Policy,

    Recent observation identifies subendothelial (mural) cells expressing MCAM, a specific system of clonogenic, self-renewing, osteoprogenitors (a.k.a, "mesenchymal stem cells") in the microvascular compartment of post-natal human bone marrow (BM). In this study, we used MCAM/CD146, as a marker to localize, isolate and assay subendothelial clonogenic cells from the microvasculature of postnatal human skeletal muscle. We show here that these cells share with their BM counterpart, anatomic position (subendothelial/adventitial) and ex vivo clonogenicity (CFU-Fs). When assayed under the stringent conditions, these cells display a high spontaneous myogenic potential (independent of co-culture with myoblasts or of in vivo fusion with local myoblasts), which is otherwise only attained in cultures of satellite cells. These muscle-derived mural cells activated a myogenic program in culture. Cultured CD146+ cells expressed the myogenic factors (Pax7, Pax3 and Myf5), NCAM/CD56, desmin as well as proteins characteristic of more advanced myogenic differentiation, such as myosin heavy chain. In vivo, these cells spontaneously generate myotubes and myofibrils. These data identify the anatomy and phenotype of a novel class of committed myogenic progenitor in human post-natal skeletal muscle of subendothelial cells associated with the abluminal surface of microvascular compartment distinct from satellite cells

    Detecting pathogens and mounting immune responses upon infection is crucial for animal health. However, these responses come at a high metabolic price (McKean and Lazzaro, 2011, Kominsky et al., 2010), and avoiding pathogens before infection may be advantageous. The bacterial endotoxins lipopolysaccharides (LPS) are important immune system infection cues (Abbas et al., 2014), but it remains unknown whether animals possess sensory mechanisms to detect them prior to infection. Here we show that Drosophila melanogaster display strong aversive responses to LPS and that gustatory neurons expressing Gr66a bitter receptors mediate avoidance of LPS in feeding and egg laying assays. We found the expression of the chemosensory cation channel dTRPA1 in these cells to be necessary and sufficient for LPS avoidance. Furthermore, LPS stimulates Drosophila neurons in a TRPA1-dependent manner and activates exogenous dTRPA1 channels in human cells.     Evaluation of the osteoconductive potential of bone substitutes embedded with schneiderian membrane- or maxillary bone marrow-derived osteoprogenitor cells

    Aim: Sinus augmentation procedures commonly employ osteoconductive scaffolding materials to stimulate and support bone formation. The aim of this study was to develop a simple screening methodology for the evaluation of the osteoconductive potential of various bone graft materials prior to clinical use. Materials and methods: Materials tested were Bio-Oss, Bi-Ostetic, OraGraft, and ProOsteon. These Simple and composite bone substitutes were embedded with osteoprogenitor cells derived from either the human maxillary sinus schneiderian membrane (hMSSM) or from maxillary tuberosity bone marrow and then monitored both in vitro and in vivo. Results: Cell adherence and proliferation was most pronounced in OraGraft, followed by ProOsteon. In vivo bone formation, within the bone graft, was also observed, with most marked results in OraGraft and ProOsteon grafts. Conclusions: The proposed osteoconductivity testing method proved simple, informative, and reliable for the purpose of screening candidate biomaterials for sinus lifting or sinus augmentation. © 2012 John Wiley & Sons A/S

Sorsero così diverse strutture: i monasteri di Santa Maria Maddalena al Corso e di San Giacomo alla Lungara per ex prostitute che volevano diventare monache di clausura; le case pie di Santa Marta, Santa Chiara e Santa Croce alle Scalette per le donne costrette a prostituirsi dai mariti e per le prostitute che non volevano professare i voti solenni; il conservatorio di Santa Caterina dei Funari per le figlie delle cortigiane; le comunità di San Filippo Neri e di Santa Maria del Rifugio per altre donne fuorviate. Variegate furono inoltre le procedure e i requisiti per essere ammesse in tali istituti, spesso sovraccarichi di debiti e turbati da disordini interni e conflitti.Between the 16th and 17th century, the Roman ecclesiastical hierarchies undertook to dam the wide-spreading female prostitution in the city by providing a series of sanctions but also trying to redeem these women by setting up some institutions to recover them.     Human sinusoidal subendothelial cells regulate homing and invasion of circulating metastatic prostate cancer cells to bone marrow

    : Subendothelial cells (pericytes) are the clonogenic, multipotent and self-renewing skeletal stem cells (SSCs) found in bone marrow (BM) stroma. They express genes maintaining hematopoietic stem cell (HMC) niche identity and, transplanted in immunocompromised mice, organize the hematopoietic microenvironment (HME) generating humanized bone/BM ossicles. To create a mouse model of hematogenous metastasis of human prostate cancer (PC) cells to human bone/BM, we injected PC cells in the blood circulatory system of Severe Combined Immunodeficiency (SCID)/beige mice bearing heterotopic ossicles. Results indicate that PC cells could efficiently home to mice-implanted extraskeletal BM ossicles, but were not able to colonize mice skeletal segments. In humanized bone/BM ossicles, early foci of PC cells occupied a perisinusoidal position, in close contact with perivascular stromal cells. These findings demonstrate the importance of the SSC compartment in recreating a suitable environment to metastatic PC cells. Our data support the hypothesis that BM SSCs committed to a pericyte fate can specify for homing niches of PC cells, suggesting an involvement of specific interactions with subendothelial stromal cells in extravasation of circulating metastatic PC cells to BM

    The paper aims to analyse the phenomenon of Violence against women in the Italian context during 2020. It proposes to study the relationship between ‘victim-author’ and ‘motivation’ in femicides committed in domestic environment. By means of the properties of the Social Network Analysis on bimodal data, the study detected main actors and motivations that generated the homicides with female victims. At the same time, the structural relationships allowed to investigate the existence of motivations that better characterized the action of the various actors. The bipartite graph visualization and centrality scores calculated have demonstrated the effectiveness of the methodology for the pursued objectives

    Human maxillary tuberosity and jaw periosteum as sources of osteoprogenitor cells for tissue engineering

    Objective. Bone tissue engineering is a promising approach for bone reconstruction in oral-maxillofacial surgery. This study investigates the suitability of oral skeletal tissues as convenient and accessible sources of osteogenic progenitors as an alternative to the iliac crest bone marrow. Study design. Samples of maxilla tuberosity (MT) and maxillary and mandibular periosteum (MP) were obtained during routine oral surgery, and donor site morbidity was assessed using a "split-mouth" approach. Cells isolated from MT (bone marrow stromal cells; MT-BMSCs) and from MP (periosteal cells; M-PCs), were analyzed for clonogenicity, phenotype, expression of osteogenic markers, and ability to form bone in vivo. Results. Both MT-BMSCs and M-PCs included clonogenic cells, showed comparable phenotypic profiles, and expressed early osteogenic markers. Most importantly, both cell populations formed bone upon ectopic in vivo transplantation. Conclusion. MT-BMSCs and M-PCs behaved as osteoprogenitor cells in vitro and in vivo. MT and MP may be considered as suitable sources of cells for bone tissue engineering in humans

    Fibrous dysplasia is a skeletal disorder caused by gain-of-function mutations of Gs-alpha. Although the genetic event occurs in early development, pre-natal skeletal growth is unaffected in FD patients. This suggests that skeletal progenitors display a different sensitivity to aberrant Gs signaling at different developmental ages. To address this issue, we infected mouse embryonic stem cells (ES) and human post-natal skeletal stem cells (Bone Marrow Stromal Cells, BMSCs) with a lentivector expressing the R201C rat Gs-alpha cDNA and analyzed their ability to differentiate into skeletal phenotypes. The expression of the transgene was confirmed at both RNA and protein levels. Surprisingly, neither ES cells nor BMSCs showed excess intracellular cAMP upon infection. However, treatment with IBMX revealed a different response in the two cell types. In transduced BMSCs, IBMX significantly increased intracellular cAMP compared to wild-type cells. Accordingly, high expression of IBMX-sensitive PDE isozymes (i.e. PDE 3, 4 and 7) was detected by q-PCR in basal conditions. IBMX alone did not modify cAMP levels in transgenic ES cells in which increased expression of the IBMX-insensitive PDE 8 was observed. Following incubation in specific inductive media, the expression of cartilage and bone markers was, overall, unaffected in mutated ES cells. Furthermore, mature cartilage undergoing endochondral ossification and normal bone were observed in teratomas generated upon in vivo transplantation. In contrast, differentiation of BMSC was profoundly affected by the mutation. In vitro, abnormal expression of skeletal markers was observed in specific differentiation assays. In vivo, ectopic osteogenic activity was limited to the deposition of a scarce amount of woven bone. In conclusion, these data demonstrate the different behavior of Gs-alpha mutated embryonic vs adult stem cells during skeletal differentiation. Most important, they show that a PDE-mediated adaptive response to Gs-alpha activating mutations operates in both cell types. However, different, and differentially regulated, PDEs isozymes are activated in mutated ES cells compared to postnatal skeletal progenitors, and the resulting adaptive response seems more efficient in ES cells than in postnatal stem cells. This could contribute to explain the normal development and abnormal postnatal growth of FD bone

    Is Chronic Exposure to Raw Water a Possible Risk Factor for Amyotrophic Lateral Sclerosis? A Pilot Case-Control Study

    Background: The etiopathogenesis of amyotrophic lateral sclerosis (ALS) is still largely unknown. Methods: We performed a case-control study (33 cases and 35 controls) in Umbria, Italy. We investigated associations between common lifestyle, clinical factors, as well as environmental exposures potentially implicated with ALS onset. Face-to-face interviews were carried out. All cases were recruited and diagnosed according to El Escorial criteria. Case-control comparisons were made for educational and residential status, occupational exposures, and clinical and lifestyle factors prior to cases’ dates of diagnosis. Results: Our results showed an increased risk of ALS for subjects chronically exposed to raw water use (odds ratio (OR) = 6.55, 95% confidence interval (CI): 2.24–19.12). Garden activities showed a tight association with ALS as well, very likely as a consequence of chronic raw water exposure. Indeed, we could exclude an impact for pesticides, as no significant differences were observed in pesticide exposure in the two groups interviewed. However, cases were more often exposed to fertilizers. After adjustment for age, sex, and heavy physical activities, exposure to raw water was still associated with increased ALS risk (OR = 4.74, 95% CI: 1.33–16.85). Discussion: These findings suggest an association between ALS and exposure to raw water, which should be further investigated for the presence of chemicals interfering with nervous system functionality
