    MPTP Neurotoxicity: Actions, Mechanisms, and Animal Modeling of Parkinson’s Disease

    The study of neurotoxicity induced by MPTP led to drastically change the perspective on Parkinson’s disease. In fact, the selective neurotoxicity induced by MPTP rejuvenated PD research and generated a number of studies aimed at elucidating the mechanisms of action of MPTP. Remarkably, these molecular mechanisms turned out to be critical also for the survival of DA neurons in idiopathic PD. In this chapter, the main concepts developed over the last three decades to understand key molecular steps which are pivotal in MPTP toxicity are reported. This is the case of the role played by DAT and VMAT-2 in conditioning the sensitivity to MPTP neurotoxicity. Similarly, the mitochondria as targets of MPTP toxicity appear similarly affected by selective mutation of genes leading to PD. Again, the fate of mitochondria and the ability to clear these organelles when being dysfunctional are key in the modulation of MPTP toxicity. This also applies for misfolded proteins such as alpha-synuclein. Again, multiple brain areas as well as peripheral sites are increasingly recognized to be affected both during MPTP toxicity and sporadic PD patients. Nowadays, it seems that MPTP per se did not lead to the discovery of the environmental compound which causes PD; nonetheless, the study of MPTP did disclose several molecular and cellular pathways which are critical in the genesis of PD. This latter point fairly corresponds to what enthusiastically is expected from MPTP when it was identified as a causal agent of what it remains, a toxic form of environmental parkinsonism

    Caterina Franceschi Ferrucci è nota anzitutto per l’impegno pedagogico, svolto a favore dell’educazione femminile, lungo tutto il corso del XIX secolo. Dai rapporti giovanili con Leopardi, forse la pagina più indagata della sua vita, fino agli anni Settanta (morì nel 1887), Caterina Franceschi Ferrucci esaltò sempre il classicismo come fondamento di un sistema educativo, improntato a Gioberti, che includesse anche le donne. Tra il 1856 e il 1858 pubblicò per la prima volta le sue lezioni di Letteratura Italiana (furono riedite nel 1873), con il titolo I primi quattro secoli della Letteratura italiana, dal secolo XIII al secolo XVI, ribadendo anche in questo campo la centralità dei classici. La sua idea di letteratura, illuminata dallo storicismo giobertiano, propone una ricostruzione dei primi secoli della storia letteraria italiana che supera i modelli eruditi e si piega, ancora una volta, a dare supporto ad un progetto pedagogico che si offre ora al processo di unificazione nazionale. L’intervento cercherà di ricostruire i supporti teorici della storia letteraria della Franceschi Ferrucci e il ruolo assegnato in questo contesto alle figure femminili

    TURNING EMPTY OFFICE BUILDINGS INTO SUSTAINABLE SOCIAL HOUSING

    This paper deals with the issue of the increasing presence of empty office buildings. Indeed, compared to the industrial buildings, that have been turned into public services (libraries, museums, etc.), into luxurious homes, etc. (thanks to their architectural character and / or their identity value for local communities), the reuse of offices seems to be quite a different and less attractive issue for designers, and of little interest to developers. This may depend on their more recent age and lack of any architectural or identity significance, as well as on their out of date systems and low energy performance. Nevertheless, they are preserved, even if vacant, because of their value as part of the property assets of the owners, as well as the high demolition costs. Furthermore, as most of empty buildings, they are easily occupied by squatters, who cause degradation and social problems, with companies forced to prevent these situations by hiring expensive security services. ENPAM R.E., the real estate management company of the national pension and social security institute for doctors and dentists, has a number of these buildings, which are no longer required for rent. This led them to promote a collaboration with Politecnico di Milano, aimed at verifying the possibility that different office building types be easily and properly adapted for residential purposes (like dwellings, hostels, temporary residences, etc.), by the development of educational projects. The idea of turning empty offices into homes to rent comes from a comparison between the large availability of the former and the lack of the latter. Thus, this strategy could become a way for meeting the great need for social housing (even for emergencies) without using any more land, but rather preserving a part of energy and row materials already embodied in the existing building structures. In addition, the large number and rather small size of such situations seems to guarantee limited local problems and a large spread of urban rehabilitation opportunities in the future. By the empirical activity on selected case studies, typological models and technological solutions for the adaptive reuse and refurbishment have been outlined. These models and solutions are expected to help the evaluation of the opportunities for reusing different types of buildings, as well as the promotion of high environmental sustainability goals, through works of transformation of building systems and equipment

    Glioblastoma stem cells differentiation through epigenetic modulation is driven by miR-296-5p/HMGA1/Sox2 axis

    Glioma Stem Cells (GSCs) biology is finely regulated by a cross-talk of epigenetic mechanisms of regulation and the action of Oct4/Sox2 stemness gene drivers. Recently, a novel mechanism of regulation of miRNA-296-5p/Sox2 axis together with a new gene target (HMGA1) was described. The results raised questions of great relevance in glioma therapy, especially for the failure of the current radio/chemotherapies. Can miR-296-5p/HMGA1/Sox2 axis trigger the differentiation of GSCs subpopulation? Can these findings be used in a short term to develop novel therapeutics to fight glioma? We thus believe that targeting HMGA1 in GSCs, via nano-liposomes miRNA delivery together with epigenetic drugs, could overcome the failure of the current therapies. Additionally, in silico analyses to identify other gene targets of miR-296-5p with data available in literature, found a remarkable association with new gene targets whose functions are related to cell differentiation processes. Taken altogether these findings make miR-296-5p an attractive target for human brain cancer treatment

    Improved management of water resources in process industry by accounting for fluctuations of water content in feed streams and products

    In this paper, a general strategy is presented for optimizing water resources in industrial activities taking into account fluctuations of water flow-streams and water contents in feed and products as well as in the utility system. Indeed, considering fluctuations can be of primary importance for several water intensive industries such as the food or the paper&amp;pulp industry. Furthermore, whenever the water balance includes the water content of products (as is typically the case for the food industry) fluctuations in the composition of products can considerably affect the overall balance due to water being a major component of many foods. Therefore, the currently used optimization strategies that disregard water fluctuations, can lead to a severe bias in the water balance and thus overlook the potential of water recovery and reuse. Including this aspect in widely used algorithms for process monitoring (data reconciliation) and optimization might lead to a possibly prohibitive increase in the computational burden. In particular, it is shown in this paper that the resulting optimization algorithm should solve a dynamic non-convex stochastic Mixed Integer Nonlinear Programming problem. To tackle this (typically NP-hard) problem, a strategy has been developed that combines ordinal optimization for dealing with statistical information and nonlinear, non-convex deterministic algorithms. The outlined strategy has been applied to a complex process in the food industry (production of starch and starch-based products).
