    Potential bioaccessibility and functionality of polyphenols and cynaropicrin from breads enriched with artichoke stem

    In this study, an artichoke stem powder (ASP) was used at three concentrations (3%, 6% and 9%) in the formulation of new breads. The bioaccessibility of polyphenols and cynaropicrin from the ASP-enriched breads was evaluated in vitro by using a digestion model combined to high resolution mass spectrometry analysis. The overall total antioxidant capacity of the bioaccessible and unsolubilized fractions obtained during the intestinal steps and the potential ability to modulate alpha-glucosidase activity were tested. Data showed that 82% of totally bioaccessible polyphenols and 74% of cynaropicrin were released during the duodenal digestion whereas 88% of caffeic acid was released in the colon step. The antioxidant capacity and the alpha-glucosidase inhibitory activity of the duodenal extract correlated with the amount of ASP in the bread. Data demonstrated that ASP might be a valuable functional ingredient to create a reducing environment in the intestine and to partially modulate glucose metabolism

    In vitro bioaccessibility and functional properties of polyphenols from pomegranate peels and pomegranate peels-enriched cookies

    Obesity is an urgent social problem and new functional foods providing polyphenols and dietary fibers (DF) may be promising tools to modulate oxidative stress, inflammation and energy homeostasis. Pomegranate peels (PPe) are an agro-industrial by-product containing polyphenols such as ellagitannins (ETs), gallic acid (GA), ellagic acid (EA) and its derivatives (EAs), as well as DF. In this study, PPe enriched cookies (PPeC) were developed, and the bioaccessibility as well as the ability of their polyphenols to exert antioxidant activity along the Gastro-intestinal Tract (GiT) and to modulate digestive enzymes were evaluated in vitro. Data showed that the potential bioaccessibility of ETs was 40% lower from PPeC than PPe whereas EAs' and GA bioaccessibility increased by 93% and 52% for PPeC compared to PPe. The concentration of the polyphenols at each digestion step was associated with the total antioxidant capacity of the potentially bioaccessible material. Moreover the polyphenols released in the simulated duodenal phase upon PPeC digestion exhibited inhibitory activity towards α-glucosidase, α-amylase and lipase, being α-glucosidase > α-amylase > lipase. In conclusion, the data demonstrated that the inclusion of PPe at 7.5% in a bakery product potentially led to a high bioaccessibility of ETs' degradation products (mainly EA and EAs) in the duodenum, with a consequent antioxidant protection along the GiT and modulation of glucose metabolism. Further human studies are warranted to evaluate whether these effects also occur in vivo

    : During food processing most of the thermally-driven chemical reactions start off on the side chain amino group of lysine generating structurally modified compounds with specific metabolic routes. Upon human digestion, dietary Nε-carboxymethyllysine (CML) may enter the colon and undergo gut microbial metabolism. However, little is known about the in vivo metabolic fate of dietary CML and its relationship with the habitual diet. We explored by hydrophilic interaction liquid chromatography tandem mass spectrometry the metabolites of CML in urine samples collected from 46 healthy subjects and studied the associations with diet. Mean concentration of N-carboxymethylcadaverine (CM-CAD), N-carboxymethylaminopentanoic acid (CM-APA), N-carboxymethylaminopentanol (CM-APO), and the N-carboxymethyl-Δ1-piperideinium ion were 0.49 nmol mg-1 creatinine, 1.45 nmol mg-1 creatinine, 4.43 nmol mg-1 creatinine and 4.79 nmol mg-1 creatinine, respectively. The urinary concentration of CML, its metabolites and lysine were positively correlated. Dietary intake of meat products negatively correlated with urinary excretion of CML and CM-APA; conversely dietary plant-to-animal proteins ratio positively correlated with urinary CML and its metabolites. The identification and quantification of CML metabolites in urine and the associations with diet corroborate the hypothesis that CML, an advanced glycation end-product, can undergo further biochemical transformations in vivo. The gut microbiome may have a major role in human metabolism of dietary CML

    Many philosophers have propounded a vision of an improved society, what distinguishes Antonio Gramsci is his continuous effort to make it happen by understanding the process in order to put into practice. Gramsci's conviction about the importance of educative development came from both theory and experience. While there has been considerable examination of Gramsci's work in relation to the Prison Notebooks, this study will seek to address a lacuna in Gramsci scholarship. Using Gramsci's philological method, I analyse Gramsci's pre-prison activity; his pre-prison articles and letters, which, together with his letters from prison, formed part of his educative mission. This educative process was necessary, in order to construct a new party which would develop a collective will, collaboratively, with the masses.In this study therefore, I explore the contexts and formative experiences of the first part of his life together with the intellectual sources from which Gramsci developed his later theories, making central hitherto underemphasised connections between them which informed his writing and ideas. I intend to illustrate that Gramsci's underlying purpose in his writing, and political activity, was not only practical, on how to create a new socialist ruling class, but also educative in forming the mindset and values of his comrades. So that in addition to outlining his vision of a new order, he implicitly guided or explicitly explained the processes by which the necessary changes in social relations and moral climate could be made in order to achieve it. Each person had to engage with the values of the new order so that each could contribute to the construction of a new robust state. It was essential to build a hegemony at the most profound level, one which was dependent on collective understandings and a collective will

    Development and functional characterization of new antioxidant dietary fibers from pomegranate, olive and artichoke by-products

    A novel ingredient acting as a slow digestible dietary fiber (DF) was developed by including native corn starch in calcium alginate microspheres (MS). In this study three types of antioxidant DF-rich ingredients were designed and developed by including in the MS, polyphenol-rich vegetable by-product extracts (obtained from pomegranate peels, olive leaves and artichoke leaves) and their potential functionality was assessed in vitro. Specifically, the physico-chemical properties of the new MS were compared with those of six commercially available DF concentrates and with wheat and oat brans. To evaluate the potential efficacy to release PPs along the gastrointestinal tract (GiT), pomegranate peels-microspheres (PPe-MS) were subjected to in vitro simulated gastrointestinal digestion. Results showed that the newly developed MS had higher free antioxidant capacity (free-TAC) than commercial DF rich products, and the bound antioxidant capacity (bound-TAC) of PPe-MS was comparable to that of wheat bran and 4.4 folds higher than that of oat-bran. Furthermore, it was shown that the release of ellagitannins from cooked PPe-MS along in vitro simulated gastro-intestinal digestion decreased from the salivary to the small intestine phase whereas gallic acid, ellagic acid and its derivatives had an opposite trend. A certain amount of PPs was found in the spent pellet obtained from the in vitro digestion, which was mimicking the residue reaching the colon in vivo. In conclusion data showed that the new antioxidant MS have physical-chemical properties like those of wheat and oat brans, mainly including the bound antioxidant capacity. This open to new possibilities of functional utilization of vegetable by-products for obtaining valuable and healthy food ingredients

Primeiramente, um estudo introdutório sobre o autor, Antonio Rosmini, sobre sua obra filosófica e, mais especificamente, sobre o livro Principi della scienza morale, que, na segunda parte, traduzimos para o português. Na introdução, expomos brevemente o sistema filosófico rosminiano e o contextualizamos no âmbito histórico-filosófico. Além disso, apresentamos o percurso por nós desenvolvido para obter uma tradução filologicamente correta do texto original, incluindo um breve glossário dos termos filosóficos empregados pelo autor.This work is composed of two parts. Firstly, there is an introductoty study on the author, Antonio Rosmini, on his phylosophical system and, more specifically, on the book Principi della scienza morale, which we have translated to Portuguese, in the second part. In the introduction, we expose briefly the rosminian phylosofical system and we place it in its historical-phylosofical context. Furthermore, we show the path weve followed to reach a phylologically correct translation from the original text, including a brief glossary of phylosophical terms employed by the author
