    AC electrokinetic analysis of chemically modified microparticles

    Electrokinetic manipulation offers a flexible and versatile tool to analyse and precisely handle microparticles, it is non invasive and can be easily implemented in microfluidic devices. Those characteristics make the electrokinetic methods appealing in the field of medical diagnostics because of their ability to perform analysis on a single particle basis, for example to detect binding events of biomolecules such as antibodies and nucleic acids. The detection is performed without fluorescent tags or chemical modifications, because it uses the electrical properties of the surface of the particles as label. The aim of this project is to study the electrical properties of microparticles according to the chemical properties of their surface using electrokinetic on a single particle basis.A new method to analyse single particles that relies on electrorotation and electroorientation has been introduced. Using quadrupolar electrodes it has been possible to obtain both spectra for the same particle allowing a combined fitting of the data. Rod shaped particles have been analysed after each step of the biochemical modifications needed to obtain a surface functionalised with antibodies. We found that the chemical modifications happen mostly in the bulk of the particles, therefore not many charges were available onto the surface. Anyway, it has been possible to distinguish different types of particles using the surface conductance of the particles and their permittivity.Along with biochemically modified particles, also metal coated particles have been analysed to study the electrical double layer under different conditions, like different suspending medium conductivities and surface functionalisations. Particles of different shapes were metal coated with gold after a suitable chemical modification to sensitize their surface. The gold coating is an autocatalytic electroless process; therefore it has been used to plate particles in suspension without the need of an external current source. Metal coated particles’ behaviour is dominated by the double layer at low frequency, but the results are only in qualitative accord with the theory due to the roughness of the metal surface

    Topographical orientation in Parkinson’s disease: the role of embodiment in performing the VR Road Map Test

    Recent findings in neuroscience allow increasing insights in the interplay of perception and action determining knowledge acquisition. One of the findings for spatial cognition consists on the adoption of the embodied perspective, that considers knowledge as unfolded through action: stored in the form of motor responses and acquired by the act of "doing". In order to understand the impairment of spatial orientation in Parkinson’s disease, we tested 49 Parkinson’s subjects (23 female, 26 male; age 50–80 years) in a virtual version of the Money’s Road Map test (Morganti et al. 2009). Thirty-two patients are classified as Parkinson’s without dementia (PD) (MMSE: M = 28.94; sd = 1.7); 17 patients as Parkinson’s with dementia (PDD) (MMSE: M = 25.59; sd = 1.8). They were compared with a control group of healthy subjects (n = 68). We found significant differences in performing the VR Road Map Test between Parkinson’s patients (PD: M = 5.5; sd = 4.1; PDD: M = 2.18; sd = 1.8) and controls subjects (M = 15.53 SD 11.35). Moreover a 3(group) 9 4(age cohort) ANOVA revealed a significant difference between groups in the different age cohorts (Group*Age Cohort F (12.00) p \ 0.005). PD-patients showed reduced ability to navigate in comparison to healthy controls of same age (p \ 0.001), PDD- patients showed reduced topographical orientation compared to PD- patients (p \ 0.001). Results will be discussed in the light of the assumption that the allocentric/egocentric (embodied) perspective, requested by the virtual reality version of the Road Map Test, appears to be relevant for topographical orientation

    La sfida della causalità alla pratica medica: il modello eziologico galenico e il dibattito medico-tardo-seicentesco

    Il problema della causalità ha rappresentato e continua a rappresentare una sfida per la pratica medica: diversi e articolati sono stati i modelli eziologici prodotti nel tempo allo scopo di adattare una dottrina di ordine filosofico ai problemi concreti del medico. In che misura la ricerca delle cause può rivelarsi utile alla pratica clinica? Fin dove il medico può spingere la sua indagine? Il successo terapeutico dipende anche dalla comprensione delle cause occulte? In questo contributo, in particolare, verranno analizzate le tesi di Thomas Sydenham e Giorgio Baglivi in merito al ruolo delle cause in ambito clinico e alla possibilità di conoscere le cosiddette ‘causae primae et proximae’. Dopo aver presentato la dottrina galenica e i suoi rapporti con la tradizione ippocratica, il saggio illustrerà la posizione di Sydenham. Quindi, approfondirà la posizione di Baglivi, i suoi punti di contatto con Sydenham e il suo diverso orientamento sul problema delle cause
