Large datasets, bias and model‐oriented optimal design of experiments
We review recent literature that proposes to adapt ideas from classical model based optimal design of experiments to problems of data selection of large datasets. Special attention is given to bias reduction and to protection against confounders. Some new results are presented. Theoretical and computational comparisons are made
Model-based Design of Experiments for Large Dataset
The first part of this thesis presents the motivation for adapting ideas and methods from the theory of the model-based optimal design of experiments in the context of Big Data while guarding against different sources of bias. In particular, the key focus is on the issue of guarding against bias from confounders and how to use the theory of the design of experiment and randomization to remove bias depending on the constraints in the design. Starting with A/B experiments, largely used by major Tech Companies in online marketing, the theory of circuits is introduced and an algebraic methods which gives a wide choice of randomization schemes is presented. Furthermore, a robust exchange algorithm to deal with the problem of outliers in a Big Dataset is proposed.
The second part is based on a marine insurance use case sponsored by Swiss Re Corporate Solutions, commercial insurance division of the Swiss Re Group. Several temporal disaggregation methods for dealing with time series collected at different time frequencies are reviewed and applied to real data in order to obtain a curated dataset for predicting future losses
Generation of all randomizations using circuits
After a rich history in medicine, randomized control trials (RCTs), both simple and complex, are in increasing use in other areas, such as web-based A/B testing and planning and design of decisions. A main objective of RCTs is to be able to measure parameters, and contrasts in particular, while guarding against biases from hidden confounders. After careful definitions of classical entities such as contrasts, an algebraic method based on circuits is introduced which gives a wide choice of randomization schemes

Surface charge and hydrodynamic coefficient measurements of Bacillus subtilis spore by optical tweezers
In this work we report on the simultaneous measurement of the hydrodynamic coefficient and the electric charge of single Bacillus subtilis spores. The latter has great importance in protein binding to spores and in the adhesion of spores onto surfaces. The charge and the hydrodynamic coefficient were measured by an accurate procedure based on the analysis of the motion of single spores confined by an optical trap. The technique has been validated using charged spherical polystyrene beads. The excellent agreement of our results with the expected values demonstrates the quality of our procedure. We measured the charge of spores of B. subtilis purified from a wild type strain and from two isogenic mutants characterized by an altered spore surface. Our technique is able to discriminate the three spore types used, by their charge and by their hydrodynamic coefficient which is related to the hydrophobic properties of the spore surface
