    Proses pembentukan pembuluh darah baru (angiogenesis) merupakan tahapan yang penting dalam pertumbuhan tumor dan kemampuan untuk bermetastase ke tempat yang jauh. Endothelial cell-Selective Adhesion Molecule (ESAM) adalah protein yang ditemukan pada sel-sel endotel pembuluh darahVon Willebrand Factor (vWF) adalah glikoprotein plasma yang diproduksi oleh sel endotel dan megakariosit. Ki-67 adalah protein sel yang dikode oleh gen MKi-67 pada sel manusia dan berhubungan erat dengan proliferasi sel. Telah dilakukan pemeriksaan pada 79 sampel jaringan karsinoma mamma dan dilakukan perwarnaan secara immunohistokimia dengan antibodi ESAM, vWF dan Ki-67 untuk mengevaluasi jumlah pembuluh darah dan proliferasi sel. Dari 79 sampel yang diperiksa, didapatkan 31 (39%) tumor yang memiliki vaskularisasi kurang dan 48 (61%) tumor yang memiliki vaskularisasi banyak menurut ekspresi dari ESAM. Sementara berdasarkan ekspresi vWF didapatkan 34 (43%) tumor yang memiliki vaskularisasi kurang dan 45 (54%) tumor yang memiliki vaskularisasi yang banyak. Terdapat hubungan yang signifikant antara jumlah vaskularisasi menurut ekspresi ESAM dan vWF dalam tumor dan peningkatan jumlah sel yang terekspresi Ki-67 dengan ukuran/volume tumor. Hanya jumlah vaskularisasi menurut ekspresi ESAM yang memiliki hubungan signifikant dengan ada tidaknya metastasis ke pembuluh limfe. Terdapat pula hubungan yang signifikant antara peningkatan jumlah vaskularisasi dengan peningkatan jumlah sel yang mengekspresikan Ki-67. Dapat disimpulkan bahwa proses angiogenesis sangat penting dalam tahap pertumbuhan dan metastasis sel tumor dan ESAM maupun vWF juga berperan penting dalam proses angiogenesis pada karsinoma payudara. Namun, ESAM jauh lebih sensitif dibanding vWF dalam memprediksi ada tidaknya metastasis dari karsinoma payudara

    Efficient Summation of Arbitrary Masks -- ESAM

    Searches for impulsive, astrophysical transients are often highly computationally demanding. A notable example is the dedispersion process required for performing blind searches for Fast Radio Bursts (FRBs) in radio telescope data. We introduce a novel approach - Efficient Summation of Arbitrary Masks (ESAM) - which efficiently computes 1-D convolution of many arbitrary 2-D masks, and can be used to carry out dedispersion over thousands of dispersion trials efficiently. Our method matches the accuracy of the traditional brute force technique in recovering the desired Signal-to-Noise ratio (S/N) while reducing computational cost by around a factor of 10. We compare its performance with existing dedispersion algorithms, such as the Fast Dispersion Measure Transform (FDMT) algorithm, and demonstrate how ESAM provides freedom to choose arbitrary masks and further optimise computational cost versus accuracy. We explore the potential applications of ESAM beyond FRB searches

    Endothelial cell-selective adhesion molecule (ESAM) is specifically expressed at endothelial tight junctions and on platelets. To test whether ESAM is involved in leukocyte extravasation, we have generated mice carrying a disrupted ESAM gene and analyzed them in three different inflammation models. We found that recruitment of lymphocytes into inflamed skin was unaffected by the gene disruption. However, the migration of neutrophils into chemically inflamed peritoneum was inhibited by 70% at 2 h after stimulation, recovering at later time points. Analyzing neutrophil extravasation directly by intravital microscopy in the cremaster muscle revealed that leukocyte extravasation was reduced (50%) in ESAM(-/-) mice without affecting leukocyte rolling and adhesion. Depletion of >98% of circulating platelets did not abolish the ESAM deficiency-related inhibitory effect on neutrophil extravasation, indicating that it is only ESAM at endothelial tight junctions that is relevant for the extravasation process. Knocking down ESAM expression in endothelial cells resulted in reduced levels of activated Rho, a GTPase implicated in the destabilization of tight junctions. Indeed, vascular permeability stimulated by vascular endothelial growth factor was reduced in ESAM(-/-) mice. Collectively, ESAM at endothelial tight junctions participates in the migration of neutrophils through the vessel wall, possibly by influencing endothelial cell contacts

    Nonlinear ultrasonic spectroscopy using ESAM and DORT symbiosis

    National audienceCombination of DORT (Décomposition de lOpérateur du Retournement Temporel) and ESAM (Excitation Symmetry Analysis Method) presented as a powerful tool for detection and localization of defects by analysis of their nonlinear signature[1,2]. The third order nonlinear responses are extracted from ESAM eigen-excitations and constitute the multistatic data matrix associated with the singular value decomposition. The symbiosis of ESAM-DORT provides a normalization process of singular values associated with scatterers, validated with simulations and experiments of acoustic propagation[3]. Piezoelectric transducers are used for both excitation and data acquisition. Transmitters consequently emit the excitation signals and corresponding responses are measured by the array of receivers. The amplitude of excitation signals is variable as to separate the nonlinear parts of the measured signal. ESAM signal pre-processing is used for nonlinear parts extraction. Separated signal records form linear and nonlinear multistatic data matrices. DORT method is applied on data matrices to separate echoes of defects in the tested medium. Data obtained from DORT method are used for evaluation of nonlinear parameters corresponding to separated defects and also for their localization. The procedure is completed by visualization of nonlinear signatures of detected defects which is referred to as pseudotomographic imaging.     Meanings and Visions of Healthy and Unhealthy Food in Flevoland, the Netherlands

    This chapter turns attention to the Netherlands, precisely the province of Flevoland, to show the meanings of healthy and unhealthy food. The choice of Flevoland was because the province is on an agricultural polder in the Netherlands at the forefront of agricultural transformations. By combining photovoice and online surveys, findings show that meanings of healthy food are predominantly nutrition-based. This is determined by the government recommendation (Disc of five) promoting a nutrition-based understanding of healthy and unhealthy food. This chapter highlights the overemphasis on nutrition in meanings of healthy and unhealthy food. The chapter argues that although the nutrition-based discourse on healthy and unhealthy food is dominant amongst Dutch consumers, there also exist socio-culturally informed meanings of un/healthy which include the pleasure derived from the acts of sharing food experiences that positively contribute to consumers’ overall pleasure and satisfaction with consumer’s food consumption. These socio-culturally informed meanings of un/healthy food are equally important to consumers. Therefore, the author argues that the key to the future of healthy food is a middle ground where nutritionally rich food meets socially informed meanings of healthy food

    Development of three mango cultivars on four rootstocks

    The experiment was conducted at the physics structure of the Rio Grande do Norte State Farming Research Company (EMPARN) in Açu-RN, with the objective of evaluate the behavior of grafts-holder cultivates in the plants formation of cultivates used as tops. The experimental design was randonizated blocks in the factorial outline 3 x 4, with 3 repetitions. The analyzed factors were four cultivates of grafts-holder and three cultivates of grafts. As grafts-holders were used the cultivates Carabao and Manga d'agua, originated at the Campinas Agrarian Institute, and the cultivates Rosinha and Espada, obtained from the EMPARN collection. The cultivates used as tops were Tommy Atkins, Van Dyke and Keitt, with the fork that were also obtained at EMPARN. In a period of six months was observed the diameters of the grafts-holder, grafts and hight of the plants.     Endothelial cell-selective adhesion molecule (ESAM) is an immunoglobulin-like transmembrane protein associated with endothelial tight junctions (TJ). Based on a yeast two-hybrid screen, we have identified the membrane-associated guanylate kinase protein MAGI-1 as an intracellular binding partner of ESAM. MAGI-1 is a multidomain adaptor protein, which binds to transmembrane, cytoskeletal, and signaling molecules, and has been localized to tight junctions in epithelial cells. MAGI-1 associates with the very C-terminal sequence of ESAM most likely through a PDZ domain-mediated interaction. The direct interaction between ESAM and MAGI-1 was confirmed by pull-down experiments. The two proteins formed stable complexes in transfected Chinese hamster ovary (CHO) cells, which could be immunoisolated. We found MAGI-1 to be associated with cell-cell contacts in human umbilical vein endothelial cells (HUVECs) and in mouse endothelium, where it colocalizes with ESAM. In CHO cells, recruitment of MAGI-1 to cell contacts required the presence of ESAM. Hence, ESAM may be involved in anchoring MAGI-1 at endothelial tight junctions
