Born in San Francisco on November 6, 1939, one of the twin sons of Elizabeth and Frank Erlin of Palo Alto, Peter Erlin passed away January 9, 2015 at his home in St. Louis, Missouri. He was 75. He is survived by Susana Erlin, his wife of 47 years, his three children, (Shawn Meisl of Jackson, WY, Matt Erlin of St. Louis, MO, and Tim Erlin of Minneapolis, MN) and six grandchildren. He also leaves his twin brother Michael Erlin of La Honda, CA, his nephews Michael Erlin Jr of Sydney, Australia and Christopher Erlin of San Rafael, CA, his Aunt Mary Folger Barrett of Techapi, CA, and many beloved cousins. Peter grew up in Palo Alto, where he attended Walter Hayes Elementary School, Jordan Junior High School and Palo Alto High School. He attended Stanford University on an ROTC scholarship and graduated in 1961 with a degree in Political Science. After college he spent six years in the Navy on the USS Providence. Upon completion of his service he moved to Newport Beach, where he met and married Susana. They moved to the East Coast to work and start a family but returned to California after five years. Peter spent much of his life in the San Francisco Bay Area, living in Los Altos and Orinda and working for Western Contract Furnishings before starting his own office furnishings business. Upon retirement he and Susana moved to South Portland, ME, where they enjoyed ten years of beach living. In 2014 they moved to St. Louis to be close to family. Peter was a man of principle, a dedicated husband, father, and son, an engaged member of all of the communities in which he lived, a witty conversationalist and talented writer, and a fantastic cook

Characterization of the <it>C. elegans </it>erlin homologue

Abstract Background Erlins are highly conserved proteins associated with lipid rafts within the endoplasmic reticulum (ER). Biochemical studies in mammalian cell lines have shown that erlins are required for ER associated protein degradation (ERAD) of activated inositol-1,4,5-trisphosphate receptors (IP3Rs), implying that erlin proteins might negatively regulate IP3R signalling. In humans, loss of erlin function appears to cause progressive intellectual disability, motor dysfunction and joint contractures. However, it is unknown if defects in IP3R ERAD are the underlying cause of this disease phenotype, whether ERAD of activated IP3Rs is the only function of erlin proteins, and what role ERAD plays in regulating IP3R-dependent processes in the context of an intact animal or embryo. In this study, we characterize the erlin homologue of the nematode Caenorhabditis elegans and examine erlin function in vivo. We specifically set out to test whether C. elegans erlin modulates IP3R-dependent processes, such as egg laying, embryonic development and defecation rates. We also explore the possibility that erlin might play a more general role in the ERAD pathway of C. elegans. Results We first show that the C. elegans erlin homologue, ERL-1, is highly similar to mammalian erlins with respect to amino acid sequence, domain structure, biochemical properties and subcellular location. ERL-1 is present throughout the C. elegans embryo; in adult worms, ERL-1 appears restricted to the germline. The expression pattern of ERL-1 thus only partially overlaps with that of ITR-1, eliminating the possibility of ERL-1 being a ubiquitous and necessary regulator of ITR-1. We show that loss of ERL-1 does not affect overall phenotype, or alter brood size, embryonic development or defecation cycle length in either wild type or sensitized itr-1 mutant animals. Moreover we show that ERL-1 deficient worms respond normally to ER stress conditions, suggesting that ERL-1 is not an essential component of the general ERAD pathway. Conclusions Although loss of erlin function apparently causes a strong phenotype in humans, no such effect is seen in C. elegans. C. elegans erlin does not appear to be a ubiquitous major modulator of IP3 receptor activity nor does erlin appear to play a major role in ERAD.</p

Analysis of phosphatidylinositol 3-phosphate binding to the erlin complex

The Endoplasmic reticulum (ER) membrane lipid raft-associated proteins erlin 1 (E1) and erlin2 (E2) are ~40 kDa proteins, and they are members of a superfamily of stomatin/prohibitin/ flotillin/HflK/C (SPFH) domain-containing proteins. E1 and E2 form a massive (~2 MDa) hetero-oligomeric complex that is an essential mediator of inositol 1,4,5-trisphosphate receptor (IP3R) ubiquitination and degradation. Mutations of E1 and E2 are involved in many pathological processes in neurological disorders, such as Hereditary Spastic Paraplegia (HSP), with unknown molecular mechanisms. The Wojcikiewicz laboratory has previously provided evidence that the erlin complex, immunopurified from mammalian cells binds to phosphatidylinositol 3-phosphate (PI(3)P), a key player in membrane dynamics and trafficking regulation in endocytosis and autophagy. In addition, the erlin complex may be involved in different cellular processes beyond IP3Rs degradation, but the exact nature of these roles has remained elusive. My research described in this thesis has uncovered intriguing new insights into the erlin complex and its role in previously unknown aspects of cellular biology. Through the application of diverse biochemical and molecular biology assays, I successfully identified specific regions on E2 that are essential for its binding to PI(3)P. Additionally, my research revealed that the erlin complex plays an important role in regulating cellular PI(3)P levels through its interaction and stabilization with this lipid. This binding and stabilization of PI(3)P are crucial for the regulation of autophagy and lysosome function. These findings contribute to our understanding of the erlin complex's importance in cellular biology and provide valuable knowledge about related processes that have implications for human health and disease.NAUpstate Medical UniversityPharmacologyPh
