No full textThis article describes the development of a service portfolio for a research
data centre in the humanities; in particular, it explores which services are
necessary to support the long-term preservation and re-use of the results of
humanities research. First, the fundamental characteristics of digital research
data in the humanities are examined. In addition to file-based research data –
such as texts, images, videos, and so on – complex data structures become more
prominent and pose new challenges for institutions working with research (data)
infrastructures. Next, this paper describes the results of the test ingests
using three types of research data – database, digital edition, and interactive
visualisation – that were carried out during the design phase of the Humanities
Data Centre (HDC), followed by a discussion of their implications for the HDC’s
initial modular service portfolio, which could be expanded in the future in
coordination with other research data centres. This article reflects the
developments in the HDC project as of January 2016. Information about new

4Pi microscopy with linear fluorescence excitation
No full textPractical 4Pi microscopy has so far exclusively relied on multiphoton excitation of fluorescence, because the nonlinear suppression of contributions from higher-order sidelobes was mandatory for unambiguous axial superresolution. We show that novel lenses of 74 degrees semiaperture angle enable biological 4Pi microscopy with regular one-photon fluorescence excitation, thus increasing the signal and reducing system complexity and cost. An axial resolution of 95 nm, corresponding to a more than fourfold improvement over confocal microscopy, is verified in the imaging of microtubules in mammalian cells
