cexA and its regulatory processes – a closer look into the citric acid production mechanisms of Aspergillus niger Authors: Aline Reinfurt, Valeria Ellena, Matthias G. Steiger Aspergillus niger is an important filamentous fungus used for the industrial production of citric acid. One of the main contributors to high citric acid accumulation by the fungus is the citrate transporter CexA. It belongs to the major faciliatory superfamily subclass DHA1 which act as drug-H+ antiporters1. Since cexA and its regulators are essential within the citric acid production process, it is important to study their regulatory mechanism, which is the focus of this work. LaeA for one is known to be a major regulator of the cexA gene. It regulates the expression via methylation levels of the histones H3K4 and H3K92. There are indications that other transcriptional regulators such as AmyR and XlnR are also involved in the regulation of cexA on a transcriptional level. Another factor that affects citric acid production is the amount of manganese that is present during the production process. Observations showed that the fungus develops a certain pellet-like morphology under manganese limitation conditions and that this limitation is decisive for high citric acid accumulation by A. niger3,4. However, the exact mode of action of manganese in the cell is not clear. The transcriptional influence of manganese on cexA and laeA is investigated in order to find out more about the connection between manganese limitation and citric acid production

Sexual reproduction is an important proliferation mechanism which provides advantages to a species by promoting genetic variation. Interestingly, for a large group of fungal species, including the industrially relevant Aspergillus niger, this mode of reproduction has not been described yet. In recent years, strong indications of the sexual potential of A. niger have accumulated 1. These include the ability of certain strains to produce pre-mature sexual structures (sclerotia) and the presence of a mating type MAT1-1 locus in the commonly used A. niger strains, such as ATCC 1015 and CBS 513.88. Here we show additional evidence of the sexual potential of A. niger. Formation of sclerotia could be induced in the progenitor of the industrial citric acid producing strains of A. niger, ATCC 1015, and in pyrG mutants derived from it 2. The capability of ATCC 1015 to form sclerotia, known to act as sexual structures in related organisms, represents a step forward towards the discovery of a sexual cycle in this important industrial species. To find the second mating type locus, we performed genome sequencing of the A. niger neotype strain CBS 554.65 and identified the missing MAT1-2 locus 3. A nucleotide alignment showed a different orientation of the MAT1-1 locus of ATCC 1015 compared to the MAT1-2 locus of CBS 554.65. While the genomic context of the MAT1-2 locus in CBS 554.65 is similar to the one of other MAT1-2 A. niger strains and other Aspergillus species, the region comprising the MAT1-1 locus is inverted in all sequenced strains of A. niger. These observations suggest the occurrence of genetic flipping or switching events at the MAT1-1 locus of A. niger, which might have a direct impact on its sexuality. These results provide new insights in the mating system of A. niger and pave the way for the discovery of a sexual cycle in a species long thought to be asexual. 1. Tempi strambi.

Can we consider queer theory as a successful example of recent anglo-american cultural imperialism? The term "queer" itself has proved so culture-bound to make it almost impossible to translate into other languages, so that despite the availability of a number of synonims, in Italy (and in most European languages)the term has remained unchanged. But are the theoretical assumptions behind the uses of "queer theory" really new? This essay explores the possibility to read "queer theory" in the international tradition of "feminist studies"

Valeria Della Valle, The language of Luigi Einaudi between classicism and pathos, in Luigi Einaudi: economic freedom and social cohesion, by Alfredo Gigliobianco, Rome-Bari, 2010, p. 138-154. The author makes a linguistic reading of texts by Luigi Einaudi, to identify the lexical, stylistic and rhetorical characters of the writings of the economist, the popularizer, the journalist, the President of the Italian Republic. On them has long weighed the label of a crisp, neat, icy writer. In fact, subjecting its vast production on close examination, it reveals the extraordinary ability in the use of various language modes: solemnity, friendly conversational syntactic, lexical inventions and popular modes, descriptions with bright colors and extreme simplicity

Towards a sexual cycle in Aspergillus niger

Sexual reproduction is an important mechanism which can provide advantages to a species by promoting genetic variation. Interestingly, however, for a large group of fungal species this mode of reproduction has not been described yet. In recent years evidence of the sexual potential of presumably asexual filamentous fungi has been gathered. Aspergillus niger, a relevant industrial microorganism, belongs to the fungal species so far considered asexual. Genome analyses provided strong indications that this fungus might be able to undergo sexual development. One of these indications is the presence of the MAT1-1 gene in the sequenced A. niger strains, suggesting the existence of a heterothallic lifestyle. The discovery of a sexual cycle in this important industrial microorganism would not only broaden its current biological knowledge but also allow to develop new strategies for strain improvement. Therefore, this work was conceived to expand the current knowledge of the sexual potential of A. niger. In this study an A. niger strain containing the opposite mating-type gene, MAT1-2, was identified and characterized. Experiments were set up to study the reciprocal influence of the opposite mating-type strains and, ultimately, to induce a sexual cycle. Although complete sexual development was not observed in the performed crossing experiments, the formation of sclerotia, known to act as sexual structures in related species, was induced. The nature and the morphology of the sclerotia were determined by macroscopical and microscopical methods. Moreover, genetic engineering strategies were used in order to inactivate or overexpress the MAT genes and get an insight into their function. The results obtained here set the basis for further analysis and understanding of the mating system in A. niger. Although strain improvement in A. niger cannot be yet performed through breeding, the alternative parasexual cycle was shown to allow the successful generation of novel strains.submitted by Valeria EllenaDissertation Universität für Bodenkultur Wien 2020Mit deutscher Zusammenfassun

White middle-class men in Rio de Janeiro. The making of a dominant subject

    No full text
Based on the biographical accounts of upper-middle-class white men living in wealthy parts of Rio di Janeiro, Valeria Ribeiro Corossacz analyzes specific experiences of whiteness as they are produced at the intersection of multiple categories, in particular gender, class, and sexuality. White middle class men in Rio de Janeiro investigates what it means to be classified as a white person and a man in a society known for its valorization of racial mixing and yet deeply structured by racism, class and gender inequalities. Ribeiro Corossacz focuses on certain experiences in the men’s biographical trajectories representing moments of apprenticeship in a specific model of white, heterosexual and heteronormative middle-class masculinity and describes how these experiences are constructed as normative. This book explores how class, gender and race privilege are mutually produced and perceived by these men as “normal”. Examining instances of silence and what is left unsaid but also these men’s ability to provide precise descriptions of power relations and violent episodes, the author encourages us to observe the condition of dominant subjects as a keystone of the reproduction social discrimination
