Investigation of Taxonomic Relationship and Effect of Seasonal Temperature Changes Based on Protein Profiles of Fishes From Beyşehir, Suğla Lakes and Dam Apa
Marine Science and Technology BulletinBesides traditional methods based on morphological characters, electrophoretic methods such as SDS-PAGE are preferred by taxonomists to make the right decision in the species identification process. In addition, the effect of environmental factors, such as pH, salinity, heat, and temperature on protein profiles are essential in various studies. In this study, we aimed to determine the degree of relationship in some fish species, such as Squalius lepidus, Cyprinus carpio, Carassius gibelio, Pseudophoxinus anatolicus, Tinca tinca, Alburnus orontis, Scardinius erythrophthalmus, Capoeta capoeta, Vimba vimba, Sander lucioperca living in Beyşehir, Suğla lakes and Apa Dam by SDS-PAGE method, and to examine seasonal differences by evaluating the effect of hot/cold water on protein profiles in fish. Although there were common major protein bands in all fish species studied, the presence of species-specific minor protein bands led to the separation of the species. The same fish species distributed in different lakes and dams were different both in minor bands, and changes in protein profiles were observed consequently on the same fish species synthesizing different proteins in different seasons. Molecular Typing with BOX-PCR of Staphylococcus aureus Isolated from Milk Samples

A total of 28 band profiles ranging from 950-300 bp were generated by the BOX primer. This band profile was evaluated according to Nei homology and a dendogram (UPGMA) was obtained. According to the data obtained, 95 isolates with genetic similarity varying between 50-100 were in the same group. This study demonstrates that the BOX-PCR technique provides a fast and reliable tool to investigate epidemiology in S. aureus.Bu çalışmanın amacı Konya bölgesindeki sığır subklinik mastitinden izole edilen 98 Staphylococcus aureus'u REP ve BOX primerleri ile karakterize etmektir. İlk olarak REP primeri tarafından oluşturulan bant profili ve BOX primeri tarafından oluşturulan bant profili ayrı ayrı değerlendirildi. Daha sonra her iki primerin oluşturduğu bant profili birlikte değerlendirildi. Bant profillerinin değerlendirilmesi Nei homolojisine göre yapıldı ve bir dendogram (UPGMA) elde edildi. 4000-300 bp arasında değişen toplam 44 bant profili değerlendirildi. Elde edilen verilere göre, hemen hemen aynı izolatlar aynı gruptaydı. Her iki primerin kombinasyonuna dayanan dendrogram, S. aureus suşlarının çoğunun 62'den fazla benzer olduğunu gösterdi. Bu çalışma, kombine REP ve BOX-PCR tekniğinin S. aureus'ta epidemiyolojiyi araştırmak için hızlı ve güvenilir bir araç sağladığını göstermektedir
Effect of grape seed extract on β-catenin gene expression and hyperglycemia in rats induced by streptozotocin
Eurasian Journal of Veterinary SciencesAim: Type 2 Diabetes Mellitus is a metabolic disorder characterized by high blood sugar in case of insulin resistance and/or insulin deficiency or insufficiency. The Wnt/β-catenin signaling pathway plays a role in in all physiological processes. Any defect in the pathway causes diabetes to develop. The aim of this study was to determine the effect of grape seed extract (GSE) on body weight, blood glucose and expression of β-catenin gene in experimental Type 2 diabetic rats. Materials and Methods: Rats were divided into five groups as Control, Diabetic Control and Treatment groups (100, 200, 400 mg/kg GSE). The rats in the diabetes and treatment groups were fed with high-fat diet and were administered low dose of streptozotocin (35 mg/kg). Body weights of the rats were recorded weekly during the experiment. The water-soluble extracts were administered by gavage for four weeks and liver, pancreas and blood samples taken from animals at the end of treatment were used in the study. Results: It was observed that the effect of GSE treatment on overall weight loss due to diabetes was minimal, and 100 and 400 mg/kg GSE doses lowered blood sugar (p0.05) and pancreatic tissue (p<0.05). Conclusion: According to the findings, grape seed may be promising in the treatment of diabetes
Production and biological properties of nano porous glass microparticles for anticancer drug carrier
Nanoporous glass (NPG) microparticles were produced by conventional melt-quenching followed by acid-alkali leaching to get material for anticancer drug carriers. NPG exhibited a positive zeta potential of 34 mV after [3-(2-aminoethylamino) propyl] trimethoxysilane treatment. The specific surface area and the total pore volume of NPG were 47.3 m2/g and 0.692 cm3/g, respectively. The 5-Fluorouracil (5FU) loading capacity of NPG was measured as 18.2 ± 0.2 mg5FU/gNPG. The drug release rate was monitored for 120 hours. To evaluate the cytotoxic effects of NPG on both MCF-7 breast cancer cells and MCF-12A, an immortalized cell line, the study employed the 2, 3-bis [2-methoxy-4-nitro-5-sulfophenyl]-2H-tetrazolium5-carboxanilide inner salt (XTT) assay. The XTT results revealed that NPG showed a time and concentration-dependent cytotoxic effect. It is anticipated that NPG is a safe and effective material for drug delivery systems for in vitro and a promising alternative material for in vivo applications

Özet - Bu araştırmada okul öncesi çocukların, iyi ve kötü kavramına ilişkin algılarıyla, bu kavramlara yükledikleri duygu ve değerler çizdikleri resimler Müge Yukay Yüksel, Nilgün Canel, Nuray Mutlu, Süheyla Yılmaz, Elif Çap aracılığıyla incelenmiştir. Araştırmaya Milli Eğitim Bakanlığına bağlı ana- okullarına devam eden 4-6 yaş aralığındaki 180 öğrenci katılmıştır. Katılım- cılardan önce iyi bir şey çizmeleri, ardından kötü bir şey çizmeleri istenmiştir. Katılımcılar tarafından yapılan çizimler, analizler sonucunda kategoriler oluş- turularak kodlanmıştır. Veriler nitel araştırma yöntemleriyle analiz edilmiştir. Araştırma sonucuna göre, çocukların iyi kavramına karşılık olarak en fazla sevgi, neşe-eğlence ve mutluluk; kötü kavramına karşılık olarak da korku, üzüntü, kızgınlık öfke duygularına ve 5-6 yaşta acı ve şaşkınlık duygularına yer verdikleri görülmüştür. 4-5 yaşta herhangi bir değer ifadesine rastlanmaz- ken, 5-6da iyi ile ilişkili olarak sevgi, dostluk, sorumluluk ve yardımsever- lik değerlerine rastlanmıştır. İyi ve kötü temalı resimlerde doğa, gökyüzü ve doğa olayları ile hayal gücüne ilişkin şekiller en çok rastlanan temalarken, mavi, kırmızı, sarı, yeşil, mor ve turuncu en çok kullanılan renkler olmuştur

This thesis aims to assess the EU’s external energy policy and to what degree it has been embraced by small states during the energy crisis by posing the question, “How does European Union membership influence the external energy policy decisions of small states within the foreign policy dimension during energy security crises?”. To answer this, the research is taking Bulgaria and Serbia as case studies due to their similar population sizes, GDP, and energy consumptions. This research conceptualizes energy security and energy diplomacy to understand how diplomacy and energy dependencies are interrelated and how this relationship can be utilised as a tool in international relations. Through supporting the qualitative work on foreign policy analysis with the quantitative analysis of the energy industry structures and energy dependencies of Serbia and Bulgaria in a comparative manner, this research is conducting mixed research on the issue. The comparison of Bulgaria and Serbia enables the author to analyse and understand the role of EU membership
Effects of Exposure to Radiofrequency at 2.45 GHz on Structural Changes Associated with Lipid Peroxidation in Prepubertal Rat Testicular Tissue
Objective: The increasing use of electronic devices, accompanied by advancing technologies, has led to heightened exposure to non-ionizing electromagnetic radiation (EMR). This exposure instigates the accumulation of free radicals and oxidative damage in tissues, consequently impacting biological systems. Notably, the testis is among the tissues adversely affected by EMR. Numerous studies have highlighted the pivotal role of the testis in sperm production, emphasizing the potential implications of any damage on the reproductive system. This study aims to assess the levels of lipid peroxidation through histological evaluation in the testicular tissue of prepubertal male rats exposed to electromagnetic radiation at varying electric field intensities within the 2.45 GHz radiofrequency (RF) range. Methods: The experimental group comprises six subdivisions, including a sham control group, as well as groups exposed to varying electric field strengths (EFS) of 0.6 V/m, 1.9 V/m, 5 V/m, 10 V/m, and 15 V/m, respectively. Excluding the sham control group, the remaining subgroups were subjected to a daily 2.45 GHz RF exposure for 1 hour starting immediately after fertilization. This exposure to different electric field intensities continued for 45 days post-birth. Results: The samples obtained from the RF radiation-exposed rats exhibited elevated malondialdehyde (MDA) values and decreased glutathione (GSH) values in the testicular tissue. Furthermore, a comparative analysis between the microwave radiation-exposed group and the control group revealed distinct histological alterations in the testicular tissue. Conclusion: In conclusion, our findings indicate that exposure to microwave radiation at an electric field intensity of 15 V/m can lead to significant histopathological and oxidative parameter changes in Wistar rats. These results underscore the potential effects of such exposure on human health

Determination of Antioxidant and Anticancer Activities of C. Sativa Leaf Extracts on MCF7 Human Breast Cancer Cell Line
Medical RecordsAim: In this study, it was aimed to determine the total phenolic content, flavonoid content, and antioxidant activity of Castanea sativa Mill. (chestnut) leaf extract and their anti-proliferative effect on MCF 7 cell line. Materials and Methods: It determined antioxidant capacities by using DPPH assay, total phenolic content by using Folin-Ciocaltaeu method and Total flavonoid content by aluminum chloride colorimetric method in the ethanolic extracts of Chesnut leaf. In addition, the anti-proliferative effect of chestnut leaf extract was determined by XTT method in MCF7 breast cell line. The leaf extract used was applied at different concentrations for 24 hours, 48 hours and 72 hours and the results were evaluated with the Graphpad Prism software program. Results: In this study, it was revealed that the total phenolic contents of ethanolic extracts of chestnut leaf 58,22 mg GAE/g. Total flavonoid content was 64,62 mg QE/g. The DPPH activity of the leaf extract of chestnut was 80.06%. Moreover, findings showed that Chestnut leaf extract had cytotoxic effect against breast cancer cells depends on concentration and time. The 24h, 48h and 72h most effective IC50 dose were 100.1 μL ,193 μL 15.23 μL, respectively. Conclusion: Chestnut has potential antiproliferative and antioxidant activities in conjunction with its high phenolic and flavonoid content on the MCF-7 cell line. However, more studies are needed
