    In the vertebrate central nervous system, exploratory filopodia transiently form on dendritic branches to sample the neuronal environment and initiate new trans-neuronal contacts. While much is known about the molecules that control filopodia extension and subsequent maturation into functional synapses, the mechanisms that regulate initiation of these dynamic, actin-rich structures have remained elusive. Here, we find that filopodia initiation is suppressed by recruitment of ArhGAP44 to actin-patches that seed filopodia. Recruitment is mediated by binding of a membrane curvature-sensing ArhGAP44 N-BAR domain to plasma membrane sections that were deformed inward by acto-myosin mediated contractile forces. A GAP domain in ArhGAP44 triggers local Rac-GTP hydrolysis, thus reducing actin polymerization required for filopodia formation. Additionally, ArhGAP44 expression increases during neuronal development, concurrent with a decrease in the rate of filopodia formation. Together, our data reveals a local auto-regulatory mechanism that limits initiation of filopodia via protein recruitment to nanoscale membrane deformations

The OAI (Open Archives Initiatives)and ACIS (Academic Contributor Information System)are progressive in the direction ,which may lead to realise the "Collective Genius" at global level. Focus of present study is on Author-Self-Archiving (A-S-A)Metadata of the 983 Eprints in the Live Archives of the E-LIS (EPrints of Library and Information Science),which were approved till 7th July 2004.The A-S-A Metadata was used for librametric analysis.     The evolution of Aristotle's ideal state

    Aristotle is explaining the entire reality through two most abstract principles, form and matter. The matter that is in the state of motion is taking a concrete form, becoming an object, or in other words an idea with a purpose. The evolution of all ideas in the world is permanent, and since the beginning of history the matter is constantly falling, and the form is rising, making the entire world more perfect. The state is an idea in which the population and the territory are representing the matter, and the authority is representing the form, with a purpose of achieving pleasant life of citizens. Superior forms of authority are those aimed at achieving general interest, which by Aristotle are: kingdom, aristocracy and republic. Poor forms of authority are those aimed at achieving personal interest of those who rule, to the detriement of the rest of the population which are: tiranny, oligarchy and democracy. The ideal state can only arise where there are conditions for its creation, in other words where there is an ideal territory and population, taking ideal form of authority. The types of ideal state are: pambasileia, politeia ariste and polity. Aristotle states that polity existed in Athens during the reign of Solon. There is no information in his works about the existence of pambasileia and politeia ariste. Considering the actual nature of Arisotle's philosophy and the general teaching about the evolution of all ideas in the world, the author came to the conclusion that Aristotle created those types of states like pambasileia and politeia ariste based on the fact that the conditions for their existence will develop later on in the future. As the world is becoming more arranged, first there will emerge conditions for politeia ariste coming into existence and in the end of history pambasileia too

    Sea buckthorn (Hippophae rhamnoides L.) is used in beverages, pharmaceuticals, cosmetic products and animal feeds. Although Sea buckthorn has been shown to be easy to propagate vegetatively, currently, there is little information on reliable techniques to vegetatively propagate the plant repetitively within a single year.. To address this, three experiments were conducted to study whether season and chilling affected the successful rooting of cuttings. Four cultivars, Chuskaya, Golden Rain, Lord, and Sunny were used in the season - and chilling-effect experiments. Hardwood and softwood cuttings from field-grown plants did not root from October to December. The percent of rooted cuttings in January was cultivar dependent. The number and percent of rooted greenhouse produced cuttings were significantly affected by length of chilling. Most cuttings were produced, and had the highest rooted when the plants chilled for at least 6 weeks. Lord had the most and Golden Rain the least number of rooted cuttings. This study indicated that greenhouse grown stock plants were a viable source of sea buckthorn cuttings for vegetative propagation. When combined with field-grown sources it is possible to produce sea buckthorn planting material year round.The accepted manuscript in pdf format is listed with the files at the bottom of this page.     The Highlander

    This thesis explores James Macpherson’s The Highlander (1758) in relation to originality, Scottish identity and historiography. It also situates the Ossianic Collections in the context of Macpherson’s earlier poetical and later historical works. There are three parts to it: a biographical sketch of Macpherson’s early life, the annotated edition of The Highlander, and discursive commentary chapters. By examining The Highlander in detail this thesis questions the emphasis of other Macpherson criticism on the Ossianic Collections, and allows us to see him as a writer who is historically minded, very aware of sources, well versed in established forms of poetry and thoroughly, and positively, British. The Highlander stands out among the corpus of his works not because it can give us insights into the Ossianic Collections, which is its usual function in Macpherson criticism, but because it can help us understand what it is that connects Macpherson’s earlier and later works with the Ossianic Collections: history, Britishness, tradition. Macpherson’s poetical works are united by a desire to translate Scotland’s factual past into sentimental British poetry. In the Ossianic Collections he does so without particular faithfulness to his sources, but in The Highlander he converts historical sources directly into neo-classic verse. This is where Macpherson’s originality lies: his ability to adapt history. In different styles and genres, and based on different sources, Macpherson’s works are early examples of Scotland’s great literary achievement: historical fiction. Instead of accusing him of forgery or trying to trace his knowledge of Gaelic ballads, this thesis presents Macpherson as a genuine historian who happened to write in a variety of genres

    AMP-activated protein kinase (AMPK) is a known regulator of whole-body energy homeostasis, but the downstream AMPK substrates mediating these effects are not entirely clear. AMPK inhibits fatty acid synthesis and promotes fatty acid oxidation by phosphorylation of acetyl-CoA carboxylase (ACC) 1 at Ser79 and ACC2 at Ser212. Using mice with Ser79Ala/Ser212Ala knock-in mutations (ACC DKI) we find that inhibition of ACC phosphorylation leads to reduced appetite in response to fasting or cold exposure. At sub-thermoneutral temperatures, ACC DKI mice maintain normal energy expenditure and thermogenesis, but fail to increase appetite and lose weight. We demonstrate that the ACC DKI phenotype can be mimicked in wild type mice using a ghrelin receptor antagonist and that ACC DKI mice have impaired orexigenic responses to ghrelin, indicating ACC DKI mice have a ghrelin signaling defect. These data suggest that therapeutic strategies aimed at inhibiting ACC phosphorylation may suppress appetite following metabolic stress.</p

    Performance of hazelnut cultivars and selections in southern Ontario

    The global demand for hazelnuts is increasing steadily, driven by increasing use by chocolate companies, pharmaceuticals, health products, and others. North America only produces 5% of the world crop, of which 99% is produced in Oregon (OR, USA). Most available cultivars are adapted to areas with mild winters and thus do not perform well in Ontario (ON, Canada). Our objective was to identify genotypes capable of supporting the newly formed hazelnut industry in southern Ontario. In the last several decades, selections have been identified in ON, New York (NY, USA), and Michigan (MI, USA) that may be better adapted in ON than cultivars from Europe and OR. To test our hypothesis that these new selections would outperform cultivars from Europe and OR in southern Ontario, two trials were performed to evaluate yield, nut quality, and winter hardiness. As hypothesized, selections from ON and NY such as ‘Gene’, ‘Alex’, and ‘Slate’ were winter hardy and highest yielding, followed by ‘Butler’ and ‘Gamma’ from OR; however, these selections did not perform well in other assessed parameters such as nut quality and catkin survivability. Possible explanations for the poor performance of the European and OR cultivars include (i) longer time needed to acclimatize and enter production phase relative to selections from ON, NY, and MI, and (ii) cold susceptibility during the initial establishment phase. The need to allocate resources towards survival probably delays the ability of the European and OR cultivars to enter the production phase. In conclusion, hazelnut cultivars brought from regions with a different climate need to be tested for cold hardiness.The presentation of the authors' names and (or) special characters in the title of the pdf file of the accepted manuscript may differ slightly from what is displayed on the item page.
