TNFA deletion alters apoptosis as well as caspase 3 and 4 expression during otitis media
Abstract Background Tumor necrosis factor (TNFA) is the canonical member of the TNF superfamily, which plays a major role in both inflammation and apoptosis. To evaluate the role of TNFs in otitis media (OM), the most common disease of childhood, we evaluated middle ear (ME) expression of genes encoding the TNF and TNF receptor superfamilies during bacterial OM in the mouse, characterized OM in TNFA-deficient mice, and assessed apoptosis during OM in normal versus TNF-deficient MEs. Results TNFs and TNF receptors were broadly regulated during OM, with TNFA showing the highest level of up-regulation. TNF deficient mice exhibited mucosal hyperplasia even in the absence of infection and exuberant growth of the mucosa during OM, including the formation of mucosal polyps. Mucosal recovery during OM was also delayed, in parallel with a delay in mucosal apoptosis and reduced caspase gene expression. Conclusions The TNF and TNF receptor superfamilies mediate both inflammation and apoptosis during OM. TNF appears to be critical for the maintenance of mucosal architecture in both the normal and infected ME, since excessive accumulation of mucosal tissue is seen in TNFA-/- MEs both before and after bacterial inoculation of the ME. TNFA is also required for appropriate regulation of caspase genes.</p
Risky Play Approaches of Educators in German and Turkish Nature-Based Preschools
This cross-cultural multiple case study investigates the approaches of educators in nature-based preschools in Turkey and Germany towards risky play, exploring factors influencing play behaviours and educator roles. Four educators from Turkey and three educators from Germany participated in the study. Through qualitative analysis of interviews and observations, three themes emerged: factors affecting play (including age, gender, environment, and cultural norms), educator roles (pre-play, during play, post-play), and risky play requirements (rules, risk management). Findings reveal cultural differences in attitudes towards risk, the impact of policy frameworks on practice, and the importance of professional development for educators. Policy implications include the need for supportive regulations promoting outdoor play, while future research should focus on longitudinal studies to assess long-term outcomes of risky play and explore innovative approaches to risk management. This study underscores the importance of creating environments that empower children to engage in enriching, developmentally appropriate play experiences while ensuring their safety and well-being.Pamukkale University Scientific Research Projects Coordination UnitThis study is derived from the doctoral dissertation titled 'A Comparative Study of Risky Play in Nature-Based Preschools in Turkey and Germany,' completed by the first author at Pamukkale University. TNFA deletion alters apoptosis as well as caspase 3 and 4 expression during otitis media
Abstract
Background
Tumor necrosis factor (TNFA) is the canonical member of the TNF superfamily, which plays a major role in both inflammation and apoptosis.
Results
TNFs and TNF receptors were broadly regulated during OM, with TNFA showing the highest level of up-regulation.
Conclusions
The TNF and TNF receptor superfamilies mediate both inflammation and apoptosis during OM. TNF appears to be critical for the maintenance of mucosal architecture in both the normal and infected ME, since excessive accumulation of mucosal tissue is seen in TNFA-/- MEs both before and after bacterial inoculation of the ME. TNFA is also required for appropriate regulation of caspase genes.

Examining Kindergarten Children’s Numerosity Estimation Skills
© 2023, The Author(s), under exclusive licence to Springer Nature B.V.This study aimed to examine kindergarten children’s performance and strategies when answering different types of numerosity estimation questions. To this end, interviews were conducted with 44 children (aged 61–80 months) in an online setting, during which they were asked nine estimation questions of three types. These questions (open-ended, number referent, and visual referent estimation questions) differ in the size of the set to be estimated and in providing reference points for the estimation. The data analysis was based on the content analysis method. The verbal expressions, mimics, and gestures used by the children during the interviews were coded in two stages. In the first stage, the children’s estimation performances on different types of questions were evaluated. In the second stage, the strategies used by children for the estimation questions were identified. The estimation performance of children participating in the research was better in number referent estimation questions than in open-ended estimation questions. Moreover, the type of the questions affected the children’s strategies. Reportedly, although children tend to use counting strategy in open-ended estimation questions, when a reference point is presented, they move away from these tendencies and turn to more perceptual strategies such as eyeball and benchmark comparison
Buket Uzuner'in tabiat üçlemesinin (Su, toprak, hava) ekokritisizm açısından incelenmesi
Water, Earth, Air novels, which are included in the series of Misfits Defne Kaman's Adventures, are Buket Uzuner's work written in the environment-literary genre, which emerged as a new breath in Turkish literature. The author has fictionalized the events in the novel by being inspired by the ancient Kaman traditions and Central Asian and Siberian mythologies, which are still alive in today's Anatolia and exist in all cultures. Although the heroes in the novels seem to live in today's world and cope with the problems of this time, in fact, they have not broken away from traditional culture with their lifestyles and mentality. The author writes his novels, which she created by supporting mythological elements; It has been created by feeding it with an ecofeminist approach, which argues that the source of women and environmental problems is the male-dominated understanding and argues that the longed and desired life in this world built with relations based on domination can be achieved by dealing with environmental problems with a ecocritical approach. She made the selection of the subjects by considering the urban and cultural structures of the cities and the attitudes of the people living in the city. In the novel Water, the first book of the series, the subject is domestic violence and femicide within the framework of environmental problems. In the second book, Toprak, the subject is the smuggling of historical artifacts resulting in the plunder of our cultural heritage. In Air, which is the third book in the series of novels, which is stated to be a quartet in which the four elements, which are actually "Water, Earth, Air, and Fire", will be explained, the author, who focuses entirely on environmental problems, also deals with nuclear energy and veganism and vegetarianism as well as this subject. In all of the subjects in the three books of the series, there is damage to the nature and culture in which a person succumbs to his greed, as well as to himself and other living things. In the meantime, by not forgetting to show the way, she tried to show the ways of being a good person by putting Umay Nine, who is keeping the Kaman tradition alive, and Defne Kaman, her granddaughter, against these harmful people.</p
Our speech is filled with others' words: Understanding university student and instructor opinions towards paraphrasing through a Bakhtinian lens
At the center of academic writing sits the practice of paraphrasing. Writers are constantly paraphrasing the ideas, findings and language of others. Despite this commonplace presence, it continues to be a challenging activity for both students to engage in and for teachers to instruct. Bakhtinian theory on language use and learning could help address struggles regarding paraphrasing observed in academia. Through a Bakhtinian lens, this article presents the attitudes towards paraphrasing held by undergraduate students and writing instructors at a large US university. Using classroom ethnographic methods, the author relies on observations, interviews and textual analysis to discover how paraphrasing is understood by a specific group of individuals and how Bakhtinian theory can explain it, specifically the aspects of dialogism, response and language appropriation. Three related themes identified connect paraphrasing to work and critical thinking, maintenance of voice and textual ownership, and its value as a preferred form of source use despite its associations to plagiarism. Theoretical and pedagogical implications are also discussed. Keywords: Academic writing, Plagiarism, Paraphrasing, Bakhtin, Attitudes, Citatio

The Effect of Peer Instruction on Conceptual and Operational Approaches Preferred in Problem Solutions
The level of using conceptual and operational knowledge is of great importance in the problem-solving process. The previous studies on the measurement of conceptual and operational knowledge, have been, especially examining the problem solutions or in discussing the views. On the other hand, rich learning environments should be established to overcome such difficulties in teaching mathematics in general and analytical geometry in particular. In this study, the researchers provide useful information to the literature since this study aimed to examine quantitatively, whether the peer instruction in analytic geometry has any effect on the conceptual and operational approaches of secondary school mathematics teacher candidates in problem solving. The sample of the research is composed of (including 22 females, and 1 males) secondary school mathematics teacher candidates. Considering the advantages of technological resources on learning, the learning management system (LMS) designed by the second author, was used during the question-answer process. Before the application, the course content was integrated into the online learning management system (LMS). The teacher candidates were divided into groups before the application. In this research, the first group will be presented in the context of the research problem. In the first group, the scores of the prospective teachers who wanted to use the second right to answer were not reduced. Conceptual & Operational Approach towards Problem-Solving scale, developed by Özyıldırım Gümüş and Umay (2018) was used as the data collection tool. One of the two options is conceptual and the other one is the operational. Thus, it is possible to decide on approaches from the responses. Since the value of this approach could be related with the data set, the cut-off point was determined by the ROC curve method in SPSS program and the data were analyzed descriptively by the reference to this cut-off point. It has been determined that the secondary school mathematics teacher candidates display a conceptual knowledge-based approach through peer instruction.</jats:p
