Progress and Distress on the Stratford Estate in Clare during the Eighteen Forties
In the late 1980s and early 1990s, the author acquired about 30,000 letters written mainly in the 1840s. These pertained to estates throughout Ireland managed by James Robert Stewart and Joseph Kincaid, hereafter denoted SK. Until the letters - called the SK correspondence in what follows - became the author’s property, they had not seen light of day since the 1840s. Addressed mainly to the SK office in Dublin, they were written mainly by landlords, tenants, the partners in SK, local agents, etc. After about 200 years in operation as a land agency, the firm in which members of the Stewart family were the principal partners - Messrs J. R. Stewart & Son(s) from the mid-1880s onwards -- ceased business in the mid-1980s. Since 1994 the author has been researching the SK correspondence of the 1840s. It gives many new insights into economic and social conditions in Ireland during the decade of the great famine, and into the operation of Ireland’s most important land agency during those years. It is intended ultimately to publish details on several of the estates managed by SK in book form. The proposed title is Landlords, Tenants, Famine: Business of an Irish Land Agency in the 1840s, a draft of which has now been completed. A majority of the letters in the larger study from which the present article is drawn are on themes some of which one might expect - rents, distraint (seizure of assets in lieu of rent) ; ‘voluntary’ surrender of land in return for ‘compensation’ upon peacefully quitting; formal ejectment (a matter of last resort on estates managed by SK); landlord-assisted emigration (on a scale much more extensive than most historians of Ireland in the 1840s appear to believe); petitions from tenants; complaints by tenants, both about other tenants and local agents; major works of improvement (on almost all of the estates managed by SK); applications by SK, on behalf of proprietors, for government loans to finance improvements; recommendations of agricultural advisers hired by SK, ete. Thus, most of the SK correspondence is about aspects of estate management. It seems, in the 1840s, that the only estate in Clare managed by SK was that of the elderly Col. Stratford. Although the files on the relatively small Stratford estate are much less extensive than those on some of the estates investigated in detail in the draft of Landlords, Tenants, Famine, they do refer to most of the core aspects of estate management mentioned above. But in the case of the Clare estate, the material on some of those themes is extremely thin.
NF1 loss disrupts Schwann cell-axonal interactions:a novel role for semaphorin 4F
Neurofibromatosis type 1 (NF1) patients develop neurofibromas, tumors of Schwann cell origin, as a result of loss of the Ras-GAP neurofibromin. In normal nerves, Schwann cells are found tightly associated with axons, while loss of axonal contact is a frequent and important early event in neurofibroma development. However, the molecular basis of this physical interaction or how it is disrupted in cancer remains unclear. Here we show that loss of neurofibromin in Schwann cells is sufficient to disrupt Schwann cell/axonal interactions via up-regulation of the Ras/Raf/ERK signaling pathway. Importantly, we identify down-regulation of semaphorin 4F (Sema4F) as the molecular mechanism responsible for the Ras-mediated loss of interactions. In heterotypic cocultures, Sema4F knockdown induced Schwann cell proliferation by relieving axonal contact-inhibitory signals, providing a mechanism through which loss of axonal contact contributes to tumorigenesis. Importantly, Sema4F levels were strongly reduced in a panel of human neurofibromas, confirming the relevance of these findings to the human disease. This work identifies a novel role for the guidance-molecules semaphorins in the mediation of Schwann cell/axonal interactions, and provides a molecular mechanism by which heterotypic cell-cell contacts control cell proliferation and suppress tumorigenesis. N-cadherin directs the collective Schwann cell migration required for nerve regeneration through Slit2/3-mediated contact inhibition of locomotion
Collective cell migration is fundamental for the development of organisms and in the adult for tissue regeneration and in pathological conditions such as cancer. Migration as a coherent group requires the maintenance of cell–cell interactions, while contact inhibition of locomotion (CIL), a local repulsive force, can propel the group forward. Here we show that the cell–cell interaction molecule, N-cadherin, regulates both adhesion and repulsion processes during Schwann cell (SC) collective migration, which is required for peripheral nerve regeneration. However, distinct from its role in cell–cell adhesion, the repulsion process is independent of N-cadherin trans-homodimerisation and the associated adherens junction complex. Rather, the extracellular domain of N-cadherin is required to present the repulsive Slit2/Slit3 signal at the cell surface. Inhibiting Slit2/Slit3 signalling inhibits CIL and subsequently collective SC migration, resulting in adherent, nonmigratory cell clusters. Moreover, analysis of ex vivo explants from mice following sciatic nerve injury showed that inhibition of Slit2 decreased SC collective migration and increased clustering of SCs within the nerve bridge. These findings provide insight into how opposing signals can mediate collective cell migration and how CIL pathways are promising targets for inhibiting pathological cell migration
EphB Signaling Directs Peripheral Nerve Regeneration through Sox2-Dependent Schwann Cell Sorting
SummaryThe peripheral nervous system has astonishing regenerative capabilities in that cut nerves are able to reconnect and re-establish their function. Schwann cells are important players in this process, during which they dedifferentiate to a progenitor/stem cell and promote axonal regrowth. Here, we report that fibroblasts also play a key role. Upon nerve cut, ephrin-B/EphB2 signaling between fibroblasts and Schwann cells results in cell sorting, followed by directional collective cell migration of Schwann cells out of the nerve stumps to guide regrowing axons across the wound. Mechanistically, we find that cell-sorting downstream of EphB2 is mediated by the stemness factor Sox2 through N-cadherin relocalization to Schwann cell-cell contacts. In vivo, loss of EphB2 signaling impaired organized migration of Schwann cells, resulting in misdirected axonal regrowth. Our results identify a link between Ephs and Sox proteins, providing a mechanism by which progenitor cells can translate environmental cues to orchestrate the formation of new tissue.PaperFlic
Organism–environment interactions in a changing world: a mechanistic approach
Understanding the interactions of an organism and its environment is essential for us to integrate ultimate and proximate causation on a global scale. Organism-environment interaction includes all organisms including animals, plants, and non-eukaryotes, etc. because all of them are responsive to environmental change including those that are human-induced. A mechanistic approach is important for us to understand why some organisms can cope with change and others cannot. Here, we present three examples of environments ( the three poles ) that are changing rapidly and how avian species typical of these ecosystems are responding. These examples include apparently adaptive responses to change in climate (i.e. the predictable environment) in one species in which a lengthened breeding season now allows multiple breeding attempts. Why other species are unable to respond in a similar way remains unclear. A second example describes how changing weather (i.e. the unpredictable) may have disastrous results for breeding success in a species adapted to an extreme cold environment. Implications for climate change in which weather extremes will become more common again suggest a mechanistic approach will be important to understand how organisms may respond. The third example outlines a scenario in which multiple human-induced rapid changes (a combination of predictable and unpredictable such as development, habitat change, introduction of invasive species and climate change) may influence indigenous species in different ways. Organism-environment interaction is a fundamental concept that may unify ultimate and proximate causation and point the way for future investigations striving to understand coping mechanisms in a world where both predictable and unpredictable components of the environment are changing. © 2011 The Author(s)
Representations of Voodoo : the history and influence of Haitian Vodou within the cultural productions of Britain and America since 1850
This thesis is the first major investigation into the representations of Vodou
within the cultural productions of Britain and America. It also opens up
opportunities for further research to be undertaken in the representations of
Vodou, Haiti and the culture and religions of other Caribbean countries.
This thesis explores the representations of 'Voodoo,' the widely accepted and
recognised term for the re-imagined religion, in Britain and America since 1850.
The history of the Caribbean and Haiti is examined before considering the
influence that the religion of Haitian Vodou has had on cultural production.
Through a historical perspective the thesis will consider the evolution of Vodou
during the horrors of slavery. The historiographic representations form the basis
of the productions and are explored to contextualise Vodou in the British and
American imagination. All genres of literature are examined, from the first
mention of Vodou in the eighteenth century through to the present day. This is
followed by an examination of the cultural reproductions of Vodou in film,
animation, theatre and television to explore the diversity of the representations.
The wider societal influences are considered throughout this work to
contextualise the productions of 'Voodoo'.
This thesis argues that the cultural reproductions of Vodou since 1850 have not
changed greatly, despite various efforts to redress the misrepresentations, they
remain rooted in colonialism. It will argue that many of the cultural productions
are reliant on previous representations. They do not in the majority introduce
authenticity, instead opting for the more sensational approach. Many of the
representations will be shown to be derogatory to the religion, culture and people
of Haiti and the diaspora. This is despite Vodou as a religion having survived,
gained strength and continuing to thrive in the twenty-first century

Evaluation of tuberculosis diagnostics in children: 2. Methodological issues for conducting and reporting research evaluations of tuberculosis diagnostics for intrathoracic tuberculosis in children. Consensus from an expert panel
Confirming the diagnosis of childhood tuberculosis is a major challenge. However, research on childhood tuberculosis as it relates to better diagnostics is often neglected because of technical difficulties, such as the slow growth in culture, the difficulty of obtaining specimens, and the diverse and relatively nonspecific clinical presentation of tuberculosis in this age group. Researchers often use individually designed criteria for enrollment, diagnostic classifications, and reference standards, thereby hindering the interpretation and comparability of their findings. The development of standardized research approaches and definitions is therefore needed to strengthen the evaluation of new diagnostics for detection and confirmation of tuberculosis in children.In this article we present consensus statements on methodological issues for conducting research of Tuberculosis diagnostics among children, with a focus on intrathoracic tuberculosis. The statements are complementary to a clinical research case definition presented in an accompanying publication and suggest a phased approach to diagnostics evaluation; entry criteria for enrollment; methods for classification of disease certainty, including the rational use of culture within the case definition; age categories and comorbidities for reporting results; and the need to use standard operating procedures. Special consideration is given to the performance of microbiological culture in children and we also recommend for alternative methodological approaches to report findings in a standardized manner to overcome these limitations are made. This consensus statement is an important step toward ensuring greater rigor and comparability of pediatric tuberculosis diagnostic research, with the aim of realizing the full potential of better tests for children. © 2012 The Author
Embroidered rhetoric: the social, religious and political functions of elite women's needlework, c.1560-1630
This thesis focuses on the Elizabethan and Jacobean aristocracy and upper gentry to yield the first detailed study of the elite needleworking woman as fashioner of her social personage, and of the objects she produced as indices of social persona, religious conscience and political agency.
The first chapter explores how needlework mediates between wtiwomeann d their social context. It surveys the way in which needlework, both as practice and as object, functioned as a vehicle for projecting persona and personage into a social context which interpreted needlework according to complex value systems of personal virtue and the husbandries of conspicuous wealth. The chapter explores needlework as a site for intellectual expression. The theories developed in the first chapter are tested in a case study of Bess of Hardwick, whose textiles show her construction of a virtuous aristocratic persona proclaiming its self-assured place in the social hierarchy.
Chapter Two is the first study to consider the needlework of Elizabethan and Jacobean Catholics in the light of the Protestant proscription of iconic vestments. It recovers the history of lost needlework from English convents on the Continent, and of the English recusants' covert provision of vestments to Jesuit missioners. The first detailed case studs' of Helena Wintour's vestments reads Wintour's Jesuit-influenced Marian floral emblems and iconography alongside Hawkins's meditation handbook Partheneia Sacra to theorise Wintour's devotion to the Immaculate Conception, and explores the vestments' relationship to the liturgy and their iconographical importance to the Mass.
Chapter Three considers needlework gifts as political currency within patronage structures at the Elizabethan and Jacobean courts. Narrated with a contemporary vocabulary of grace, needlework gifts contribute to the construction of court-crown relations, symbolised by needlework gifts in Jacobean court masques. Through needlework gifts a `feminine commonwealth' availed itself of power structures at the court of James's consort that parallel his departments, and the women's political agency in a female political hierarchy is seen encoded within gifts of needlework in the Queen's Courts final masque. The case study uses Mary's needlework gifts to Elizabeth as an index of changes in their relationship. Mary's needlework joins parallel texts such as poetry, portraiture and planned masques in developing an iconographical vocabulary centring on the Judgement of Paris, with which diplomatic negotiations sought to clarify the Queens' relative positions
Transient observations : the textualizing of St Helena through five hundred years of colonial discourse
This thesis explores the textualizing of the South Atlantic island of St Helena (a
British Overseas Territory) through an analysis of the relationship between
colonizing practices and the changing representations of the island and its
inhabitants in a range of colonial 'texts', including historiography, travel writing,
government papers, creative writing, and the fine arts.
Part I situates this thesis within a critical engagement with post-colonial
theory and colonial discourse analysis primarily, as well as with the recent
'linguistic turn' in anthropology and history. In place of post-colonialism's rather
monolithic approach to colonial experiences, I argue for a localised approach to
colonisation, which takes greater account of colonial praxis and of the continuous
re-negotiation and re-constitution of particular colonial situations.
Part II focuses on a number of literary issues by reviewing St Helena's
historiography and literature, and by investigating the range of narrative tropes
employed (largely by travellers) in the textualizing of St Helena, in particular
with respect to recurrent imaginings of the island in terms of an earthly Eden.
Part III examines the nature of colonial 'possession' by tracing the island's
gradual appropriation by the Portuguese, Dutch and English in the sixteenth and
early seventeenth century and the settlement policies pursued by the English
East India Company in the late seventeenth and early eighteenth century.
Part IV provides an account of the changing perceptions, by visitors and
colonial officials alike, of the character of the island's inhabitants (from the late
eighteenth to the early twentieth century) and assesses the influence that these
perceptions have had on the administration of the island and the political status of
its inhabitants (in the mid- to late twentieth century).
Part V, the conclusion, reviews the principal arguments of my thesis by
addressing the political implications of post-colonial theory and of my own
research, while also indicating avenues for further research.
A localised and detailed exploration of colonial discourse over a period of
nearly five hundred years, and a close analysis of a consequently wide range of
colonial 'texts', has confirmed that although colonising practices and
representations are far from monolithic, in the case of St Helena their continuities
are of as much significance as their discontinuities
