    L'intérêt pour les applications cliniques des lysats plaquettaires humains (HPL) en médecine régénérative est en hausse. Le contrôle de qualité et la documentation du matériel de départ et du mode de préparation sont importants pour une utilisation clinique directe. Les avantages cliniques de ces préparations de plaquettes sont considérés comme étant liés à leur contenu en nombreux facteurs de croissance. Le HPL purifié riche en facteurs neurotrophiques a montré qu'il fournissait une neuroprotection et une neuro-restauration aux neurones dopaminergiques dans des modèles de maladie de Parkinson (PD) et de traumatisme crânien (TBI) in vitro et in vivo. La sécurité virale est également une préoccupation, en particulier avec les HPL préparés à partir de produits sanguins et produits à plus grande échelle pour l'administration cérébrale. Nos études préliminaires montrent que le matériau initial, le concentré plaquettaire (CP), et les HPL contiennent des vésicules extracellulaires (EV), soulevant des questions sur la contribution potentielle de ces PEV à leur activité fonctionnelle.Nos objectifs étaient d'évaluer (a) la composition et l'activité fonctionnelle de divers HPL, y compris celle apportée par les EV, destinés à des indications cliniques spécifiques; (b) l'efficacité de l'élimination des virus par nanofiltration et l'impact sur les compositions chauffées-HPL et leurs fonctions thérapeutiques de neuroprotection et de neuro-régénération; et (c) le profil de caractérisation bio-physique et de fonction biologique des préparations PEV sélectionnées, ainsi que leurs. Nous avons obtenu des concentrés de plaquettes humaines auprès du Centre du sang de Taipei et les avons transformés en différents lysats. Les lysats ont été quantifiés pour leur teneur en facteurs de croissance, testés pour leur activité fonctionnelle et leurs propriétés anti-oxydantes. Les HPL neuroprotecteurs chauffés ont été purifiés à l'aide de la nanofiltration Planova-20N pour éliminer les virus. Leur efficacité a été évaluée in vitro et in vivo. Nous avons également évalué l'activité neuroprotectrice exercée par certaines préparations de PEVs et le NHPPL en utilisant des modèles de cellules. Les PEVs ont montré un effet anti-inflammatoire sur les cellules microgliales BV-2. Nous avons mené une étude in vivo pour confirmer leur capacité à exercer une activité fonctionnelle lorsqu'ils sont administrés par voie intranasale, en particulier leur potentiel à moduler les cytokines inflammatoires, et pour évaluer leur capacité à protéger les cellules dopaminergiques dans un modèle animal de la maladie de Parkinson.Nos résultats ont révélé que toutes les cinq préparations de HPL contenaient des niveaux supraphysiologiques détectables de tous les facteurs de croissance évalués, sauf le facteur de croissance analogue à l'insuline-1 détecté uniquement dans le HPL contenant du plasma. Des différences significatives ont été observées parmi ces HPL en termes de teneur en protéines totales, en fibrinogène, en composants du complément C3 et C4, en albumine et en immunoglobuline G, et surtout en termes de leurs activités coagulantes et procoagulantes fonctionnelles et de leurs capacités antioxydantes. Le HPPL nanofiltré, dépourvu de PEVs, a préservé une gamme de facteurs neurotrophiques et leur activité fonctionnelle. Les PEVs isolés du surnageant de concentré plaquettaire ont exercé une activité neuroprotectrice dans le modèle de neurones dopaminergiques LUHMES de la maladie de Parkinson, ont stimulé la différenciation des cellules neuronales SH-SY5Y et ont montré une fonction anti-inflammatoire préservée lors d'une administration intranasale dans un modèle murin de traumatisme cérébral, ainsi qu'une protection des marqueurs positifs de la tyrosine hydroxylase contre l'intoxication par le MPTP chez les souris.Interest in clinical applications of human platelet lysates (HPLs) in regenerative medicine is increasing. The virus removal efficacy was evaluated by spiking small virus of mice-mock particles and immunoquantitative PCR analysis. The nanofiltration impact to the HPPL compositions and functionals was evaluated in vitro and in vivo. We assessed the neuroprotective activity exerted by selected PEVs preparations and the nanofiltered HPPL (NHPPL) using LUHMES cells model of PD. Their neuro-regenerative potentials were assessed using SH-SY5Y neuroblastoma cells. BV-2 microglia cells induced LPS was used to evaluate PEVs anti-inflammatory effect. Moreover, we conducted an in vivo study in a controlled cortical impact (CCI), traumatic brain injury (TBI) model to confirm their capacity to exert a functional activity when administered intranasally, in particular, the potential to modulate inflammatory cytokines. We also evaluated the PEVs capacity to protect tyrosine hydroxylase (TH), a marker of dopaminergic cells in MPTP mice model of PD.Our results revealed that all five HPL preparations contained detectable supraphysiological levels of all growth factors assessed, except insulin-like growth factor-1 detected only in HPL containing plasma. There were significant differences observed among these HPLs in total protein content, fibrinogen, complement components C3 and C4, albumin, and immunoglobulin G, and, most importantly, in their functional coagulant and procoagulant activities and antioxidative capacities. The nanofiltered HPPL depleted of EVs, preserved a range of neurotrophic factors and their functional activity.     Going Beyond Counting First Authors in Author Co-citation Analysis

    The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed. Reasons for these effects are discussed

    “Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship

    Appropriate Similarity Measures for Author Cocitation Analysis

    We provide a number of new insights into the methodological discussion about author cocitation analysis. We first argue that the use of the Pearson correlation for measuring the similarity between authors’ cocitation profiles is not very satisfactory. We then discuss what kind of similarity measures may be used as an alternative to the Pearson correlation. We consider three similarity measures in particular. One is the well-known cosine. The other two similarity measures have not been used before in the bibliometric literature.     Dispelling the Myths Behind First-author Citation Counts

    We conducted a full-scale evaluative citation analysis study of scholars in the XML research field to explore just how different from each other author rankings resulting from different citation counting methods actually are, and to demonstrate the capability of emerging data and tools on the Web in supporting more realistic citation counting methods. Our results contest some common arguments for the continued use of first-author citation counts in the evaluation of scholars, such as high correlations between author rankings by first-author citation counts and other citation counting methods, and high costs of using more realistic citation counting methods that are not well-supported by the ISI databases. It is argued that increasingly available digital full text research papers make it possible for citation analysis studies to go beyond what the ISI databases have directly supported and to employ more sophisticated methods

    We have done our best to complete the author checklist relating to the use of animals in the hut study. Note that the objective for the hut study was to evaluate the IRS treatment applications for residual efficacy against Anopheles mosquitoes, including the local An. coluzzii mosquito population. Cows were only used to attract mosquitoes into the huts and no tests were carried out directly on the cows. The author checklist is intended for use with studies where experiments are carried out on animals, which is why we have had such difficulty in completing this for the hut study, as many of the questions do not relate to how the cows were used
