Study of cell death and resistance mechanisms in cancer cells for the development of new therapeutic approaches : melanoma model

Restaurer la capacité des cellules tumorales à mourir est une stratégiethérapeutique qui permettrait de lutter contre le cancer. Il est donc important d’identifier denouvelles cibles au sein de la signalisation apoptotique et de tester de nouvelles molécules.La mitochondrie, intégrateur central des signaux de mort cellulaire et actrice de l’exécution de l’apoptose, est une cible de choix pour développer des thérapies anti-tumorales.L’ANT (Adenine Nucleotide Translocase) est la protéine majoritaire de la membrane internemitochondriale. Elle est possède une fonction de transporteur ATP/ADP, en conditionphysiologique, et suite à un stimulus apoptotique, acquiert une activité de pore létal. Ainsi, ilest intéressant d’inhiber la fonction transporteur et d’activer la fonction pore d’ANT pourinduire l’apoptose. Il existe quatre isoformes d’ANT : ANT1, 2, 3 et 4. Nous avons étudié lerôle d’ANT4, récemment identifiée, dans la signalisation apoptotique. Notre étude montre lerôle anti-apoptotique d’ANT4 dans des cellules cancéreuses et l’intérêt d’ANT comme ciblethérapeutique anti-cancéreuse.Une augmentation de l’expression de protéines anti-apoptotiques, l’adaptation auxstress cellulaires et l’activation de voies de survie sont les mécanismes les plus fréquemmentdécrits pour expliquer la chimiorésistance du mélanome. A l’aide de modèles cellulaires, nousavons étudié la capacité de deux nouvelles molécules : Withaférine A (WFA) et Plumbagine(PBG) à stimuler l’apoptose et déterminé les mécanismes moléculaires impliqués. Nous avonsmontré la capacité de WFA à induire spécifiquement la voie mitochondriale de l’apoptose decellules de mélanome par un mécanisme dépendant de la production d’espèces activées del’oxygène (EAO) qui déclenchent la voie mitochondriale et de la diminution du niveaud’expression de la protéine anti-apoptotique Bcl-2. En revanche, PBG induit une mortapoptotique et une mort nécrotique des cellules de mélanomes. Dans les deux cas, PBG agitpar l’augmentation des EAO suite au déclenchement d’un stress du reticulum endoplasmique.WFA et PBG sont donc deux molécules pro-oxydantes capables d’induire la mort des cellulesde mélanome en tirant partie de leur vulnérabilité au stress oxydant.Nos travaux ont participé à la mise en évidence d’une cible thérapeutique anticancéreusepotentielle et de deux agents capables d’induire la mort cellulaire dans un contextede chimiorésistance.Apoptosis is a programmed cell death process necessary for tissue homeostasis duringdevelopment. Cancer cells acquire the capacity to evade apoptosis. Restoring tumor cellsability to die is a therapeutic strategy against cancer. It is therefore important to identify newtherapeutic targets within the apoptotic signaling and to test new molecules.Mitochondrion being a central integrator of cell death signals and a key player inapoptosis execution, it is a target of choice to develop new anticancer therapies. ANT(Adenine Nucleotide Translocase) is the main protein of the inner mitochondrial membrane. Itpresents a ADP/ATP transporter function in physiological conditions and acquire a lethal poreactivity upon apoptotic stimulus. It is thus interesting to inhibit the transporter function and- 6 -activate ANT pore function in order to induce apoptosis. There are four isoforms: ANT1, 2, 3and 4. We studied the role of the recently discovered ANT4 in apoptotic signaling. Our studyemphasize ANT4 anti-apoptotic role in cancer cells and ANT potential as an anticancertherapeutic target.Increase in anti-apoptotic proteins, adaptation to cellular stress and activation ofsurvival pathways are the main mechanisms responsible for chemoresistance. Using cellularmodels we studied the ability of two molecules: Withaferin A (WFA) and Plumbagin (PBG)to stimulate apoptosis and determined the molecular mechanisms involved. We showed WFAcapacity to specifically induce the mitochondrial pathway of apoptosis in melanoma cellsthrough reactive oxygen species (ROS) generation leading to mitochondrial pathwayactivation and the decrease in anti-apoptotic protein Bcl-2 expression level. However, PBG isresponsible for apoptosis and necrosis induction in melanoma cells. In both cases PBG actsthrough an increase in ROS following endoplasmic reticulum stress. WFA and PBG are thustwo pro-oxidant molecules able to induce the death of melanoma cells by taking advantage oftheir vulnerability to oxidative stress.Our work took part in the demonstration of a potential anticancer target and two agentsable to induce cell death in a context of chemoresistance
Going Beyond Counting First Authors in Author Co-citation Analysis

counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings
are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that
only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into
account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed. Reasons for these effects are discussed

Get PDF“Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship
Appropriate Similarity Measures for Author Cocitation Analysis
Get PDFWe provide a number of new insights into the methodological discussion about author cocitation analysis. We first argue that the use of the Pearson correlation for measuring the similarity between authors’ cocitation profiles is not very satisfactory. We then discuss what kind of similarity measures may be used as an alternative to the Pearson correlation. We consider three similarity measures in particular. One is the well-known cosine. The other two similarity measures have not been used before in the bibliometric literature. Note that the objective for the hut study was to evaluate the IRS treatment applications for residual efficacy against Anopheles mosquitoes, including the local An. coluzzii mosquito population. Cows were only used to attract mosquitoes into the huts and no tests were carried out directly on the cows. The author checklist is intended for use with studies where experiments are carried out on animals, which is why we have had such difficulty in completing this for the hut study, as many of the questions do not relate to how the cows were used
