    The Sleeping Beauty (SB) transposon is a non-viral integrating system with proven efficacy for gene transfer and functional genomics. To optimize the SB transposon machinery, a transcriptionally regulated hyperactive transposase (SB100X) and T2-based transposon are employed. Typically, the transposase and transposon are provided transiently by plasmid transfection and SB100X expression is driven by a constitutive promoter. Here, we describe an efficient method to deliver the SB components to human cells that are resistant to several physical and chemical transfection methods, to control SB100X expression and stably integrate a gene of interest (GOI) through a "cut and paste" SB mechanism. The expression of hyperactive transposase is tightly controlled by the Tet-ON system, widely used to control gene expression since 1992. The gene of interest is flanked by inverted repeats (IR) of the T2 transposon. Both SB components are packaged in integration defective lentiviral vectors transiently produced in HEK293T cells. Human cells, either cell lines or primary cells from human tissue, are in vitro transiently transduced with viral vectors. Upon addition of doxycycline (dox, tetracycline analog) into the culture medium, a fine-tuning of transposase expression is measured and results in a long-lasting integration of the gene of interest in the genome of the treated cells. This method is efficient and applicable to the cell line (e.g., HeLa cells) and primary cells (e.g., human primary keratinocytes), and thus represents a valuable tool for genetic engineering and therapeutic gene transfer

They were especially found in Daunia and they are generally considered places of worship. Each of them has specific features, but it seems that they form part of a common “hypogeic worship”.     Il contributo introduce un numero monografico della Rivista, dedicato all’approfondimento sullo stato dell’arte e sulle tendenze delle tecniche di tutela nel contrasto alle discriminazioni, partendo dalla tradizionale distinzione tra rimedi giudiziali e vie alternative di prevenzione, attivazione e soluzione dei conflitti. Se il diritto antidiscriminatorio è divenuto negli ultimi anni oggetto di attenzioni particolari, se non inedite, certamente singolari per la loro scala, si tratta comunque di un diritto complesso e dinamico, di cui è necessario saggiare concretamente e a più riprese il grado di efficacia, affidato com’è non solo al suo solido impianto teorico, ma soprattutto alla sua interpretazione e alla sua pratica.The contribution introduces a monographic issue of the Journal, dedicated to an in-depth examination of the state of the art and trends in protection techniques in the fight against discrimination, starting from the traditional distinction between judicial remedies and alternative ways of preventing, activating and resolving conflicts. If anti-discrimination law has become the object of special attention in recent years, if not unprecedented, certainly singular in its scale, it is nonetheless a complex and dynamic law, whose degree of effectiveness must be concretely and repeatedly tested, entrusted as it is not only to its solid theoretical framework, but above all to its interpretation and practice

    The finding of human remains inside the Bronze Age settlements of the Italian peninsula is not so frequent, but their presence in some of them is an element of great interest. It can be variously interpreted. The ways of treatment of the human remains are different, also inside the same settlement: there are real burials, also with grave goods (for example inside the posterns of the walls at Coppa Nevigata - Foggia), whole skeletons to link to fortuitous death because of their context (for examples the individuals unearthed inside a postern of the walls of Roca Vecchia - Lecce) and sporadic bones in specific areas of the settlement. The differences are to relate to the conditions of death, to the funerary rituals and, first of all, to the categories of individuals (as ancestors, «heroes», enemies) that were variously treated on the occasion of their burial. Author aims at outlining the different patterns of behaviour (to link to specific meanings) in the archaeological contexts taken into consideration

    Se la verità gode del lusso dell'immediatezza nei Vangeli, essa si fa invece problematica nel diritto, dove pretende di essere pensata ed espressa con maggiore profusione di argomenti. Se guardato dal punto di vista del diritto, quell'interrogativo non è né sciocco (secondo Kierkegaard, la domanda di Pilato “non poteva essere più sciocca: non perché egli chieda cos'è la verità, ma perché lo domanda a Cristo”), né giocoso (Francis Bacon liquiderà altrettanto decisamente la questione, parafrasando quel passo evangelico attraverso l'espressione “disse scherzando” -“said jesting Pilate”-). Ai nostri fini, torna più utile isolare la riflessione di Kelsen, che commenta il dialogo tra Pilato e Gesù in questi termini : “Poiché egli (Pilato), relativista scettico, non sapeva cosa fosse la verità, la verità assoluta in cui quell'uomo credeva, agì in modo democratico -con assoluta coerenza- rimettendo la decisione del caso al voto del popolo

Clayton County, the Supreme Court held that Title VII of the Civil Rights Act of 1964, which prohibits sex discrimination in employment, covers discrimination on the basis of sexual orientation and gender identity. The Author tracks the judicial route that led to the decision, the textual approach adopted by the Court in the interpretation of the expression “because of sex” and offers some offers some remarks on the prospects for anti-discriminatory law, also at European level.Nella sentenza Bostock v. Clayton County, la Corte Suprema degli Stati Uniti ha stabilito che il Titolo VII del Civil Rights Act del 1964, che nei rapporti di lavoro vieta la discriminazione a causa del sesso, è invocabile anche nei casi di discriminazione per orientamento sessuale e identità di genere.
