Long-Term Spatiotemporal Reconfiguration of Neuronal Activity Revealed by Voltage-Sensitive Dye Imaging in the Cerebellar Granular Layer
Understanding the spatiotemporal organization of long-term synaptic plasticity in neuronal networks demands techniques capable of monitoring changes in synaptic responsiveness over extended multineuronal structures. Among these techniques, voltage-sensitive dye imaging (VSD imaging) is of particular interest due to its good spatial resolution. However, improvements of the technique are needed in order to overcome limits imposed by its low signal-to-noise ratio. Here, we show that VSD imaging can detect long-term potentiation (LTP) and long-term depression (LTD) in acute cerebellar slices. Combined VSD imaging and patch-clamp recordings revealed that the most excited regions were predominantly associated with granule cells (GrCs) generating EPSP-spike complexes, while poorly responding regions were associated with GrCs generating EPSPs only. The correspondence with cellular changes occurring during LTP and LTD was highlighted by a vector representation obtained by combining amplitude with time-to-peak of VSD signals. This showed that LTP occurred in the most excited regions lying in the core of activated areas and increased the number of EPSP-spike complexes, while LTD occurred in the less excited regions lying in the surround. VSD imaging appears to be an efficient tool for investigating how synaptic plasticity contributes to the reorganization of multineuronal activity in neuronal circuits
When training becomes incentive for generative living and working together in organizations
This article has come about from two considerations: on one hand, living together in a work environment is becoming more and more challenging; on the other, training professionals at work represents a used and relevant action incentive in order to support and improve individual, group, and organizational development. In light of the fact that organizations are asking more and more complex ques- tions, this work aims at developing a re ection on how adopting a certain perspec- tive and educational method is particularly suitable to support the organization to achieve substantial outcomes, such as developing generative living and working together in organizations. In this article, we will analyze two illustrative cases, underlining evolution and learning process with a particular focus on the conse- quences of living and working together in organizations
Training-research paths to support a developing professional identity: youth soccer sector coaches
Nowadays social background is characterized by some phenomena such as fragmentation, shakiness, flexibility and globalization (Sennet, 1999, Bauman, 2002). In this context even sport organizations under go deep changes, as any kind of organizations. They are characterized by an increasing complexity. The research works that we are carring out in the Italian context, but also some international literatures (Cassidy, Jones, Potrac, 2004), point out the central role of coaches in sport organizations, especially in youth sport context. Coaches are deeply involved in a complex net of relationships (children, parents, club manager), in which everyone has different motivations; they work in a situation of ongoing tension between performance requirements and educational demands. For these reasons the coach professional figure seems be passing through a crucial period of crisis and changing his mind about his role, his efforts and the resources of his own professional profile.
This paper analyses a temporary training - research organization which uses the ideas of learning with people, focusing and sharing problems (Gozzoli, Gorli, D’Angelo, FEPSAC 2007) in order to support a developing professional identity. Approximately 60 soccer youth sector coaches attended three training-research devices characterized by the same aims: a) knowing closely youth sector coaches’ practices, professional efforts and capabilities ; b) developing a challenging process and a professional development, throught a more complex representations of problems and throught interchange between participants.
Ten work sessions and group discussions, participants’ products and interviews have been conducted. Analysis of the results has been made with the help of Atlas.it software in order to grasp the challenging process of coaches’ representations and feelings relating to their professional role and identity
RPSAP52 lncRNA inhibits p21Waf1/CIP expression by interacting with the RNA binding protein HuR
Long-non coding RNAs have been recently demonstrated to have an important role in fundamental biological processes, and their deregulated expression has been found in several human neoplasias. Our group has recently reported a drastic overexpression of the lncRNA RPSAP52 in pituitary adenomas. We have shown that this lncRNA increased cell proliferation by upregulating the expression of the chromatinic proteins HMGA1 and HMGA2, functioning as a ceRNA through competitively binding to miR-15a, miR-15b and miR-16. The aim of this work has been to identify further mechanisms by which RPSAP52 overexpression could contribute to the development of pituitary adenomas. We investigated the involvement of RPSAP52 in the modulation of the expression of cell cycle-related genes, such as p21Waf1/Cip, whose deregulation plays a critical role in pituitary cell transformation. We report that RPSAP52, interacting with the RNA binding protein HuR, favors the delocalization of miR-15a, miR-15b and miR-16on the cyclin45 dependent kinase inhibitor p21Waf1/Cip1 that, accordingly, results downregulated in pituitary adenomas. A RIPseq analysis performed on cells overexpressing RPSAP52 identified 40 mRNAs enriched in AGO2 immunoprecipitated samples. Among them we focused on GAS8 (Growth Arrest-Specific Protein 8) gene. Consistently, GAS8 expression was downregulated in all the analyzed pituitary adenomas with respect to normal pituitary and in RPSAP52-overepressing cells, supporting the role of RPSAP52 in addressing genes involved in growth inhibition and cell cycle arrest to miRNA-induced degradation. This study unveils another RPSAP52-mediated molecular mechanism in pituitary tumorigenesis
The Different Roles in Youth Soccer Sector Between Cooperation and Conflict
Today's (Baumann, 2002; Sennett, 1999) sports associations are defined by an increasing complexity that requires the management of varied roles and functions on the part of players, coaches, and others simultaneously (technical, social, cultural, educational and promoting frames). The actors responsible are connected in a web of complicated relationships, and have a fundamental need for open communicational channels. Being able to navigate the inevitable differences is a result of parties' skills in addressing cooperation and conflict (Gozzoli, 2004) in order to mantain and promote the wealth and health of a sport club. The Knowing differents aspects and be able to manage the differences is due to the skill of recognizing and facing off cooperation and conflict.(Gozzoli, 2004) in order to maintain and promote the links that form the basis of wealth and good working of a sport club. The present research was developed by Catholic University of Milan in collaboration with FIGC(Federazione Italiana Giuoco Calcio). Its aim was to bring to light the differences between sports roles and the contraddiction and tensions between them in order to make raccomandation for improvement. This research focused on the youth sector, and paid particular attention to the delicate relationship beetween adults and teen in sport settings. The work was based on ethnographic observations, individual interviews with parents and coachs and directors, and focus-groups with the kids. An analysis was conducted (with the supoport of ATLAS.ti) in order to graso individual feelings related to sport, soccer, being on a team, rules and their defence, and relationships among different roles. This reserahc revealed how teens face crisis and conflicts and how they deal with teammates during competition. The project also analyzed different experiences (toil and resources) that denotes sport.
Results suggested fundamental recommendations for reorganizing youth sport clubs for the benefits of all parties involve
Collecting large-scale publication data at the level of individual researchers: A practical proposal for author name disambiguation
The disambiguation of author names is an important and challenging task in bibliometrics. We propose an approach that relies on an external source of information for selecting and validating clusters of publications identified through an unsupervised author name disambiguation method. The application of the proposed approach to a random sample of Italian scholars shows encouraging results, with an overall precision, recall, and F-measure of over 96%. The proposed approach can serve as a starting point for large-scale census of publication portfolios for bibliometric analyses at the level of individual researchers.Merit, Expertise and Measuremen
Teachers’ Wellbeing/Malaise: Which Resources and Efforts at Individual, Group and Organizational Levels?
The social productive environment has changed rapidly generating new opportunities and challenges for individuals, groups, and organizations. The school-world is affected by these general changes and also subject to constant specific pressure (e.g. reforms, resources reallocation, precariousness, increasinglycomplex demands). In this scenario teachers play a crucial role. The research aims at investigating which are resources and efforts perceived by teachers and how these influence their wellbeing or malaise. We explored teachers’ professional histories and their perception of their work group and work context(s).A phenomenological approach was adopted;50 high-school teachers were involved; semi-structured interviews and the “Professional Life-Space Drawing” were used; content analysis and a descriptive-phenomenological analysis of Drawings were conducted. Results permit the description of different professional situations
