Two Distinct Pathways Support Gene Correction by Single-Stranded Donors at DNA Nicks

No full text
SummaryNicks are the most common form of DNA damage. The mechanisms of their repair are fundamental to genomic stability and of practical importance for genome engineering. We define two pathways that support homology-directed repair by single-stranded DNA donors. One depends upon annealing-driven strand synthesis and acts at both nicks and double-strand breaks. The other depends upon annealing-driven heteroduplex correction and acts at nicks. Homology-directed repair via these pathways, as well as mutagenic end joining, are inhibited by RAD51 at nicks but largely independent of RAD51 at double-strand breaks. Guidelines for coordinated design of targets and donors for gene correction emerge from definition of these pathways. This analysis further suggests that naturally occurring nicks may have significant recombinogenic and mutagenic potential that is normally inhibited by RAD51 loading onto DNA, thereby identifying a function for RAD51 in maintenance of genomic stability

The major goal of the study has been to explain theoretical issues connected with the onomastic characterization of names used by internet users, i.e., the so called nicks. Can they be regarded as traditional pseudonyms, or do they constitute a unique anthroponymic category? The author suggests that nicks are a subcategory of pseudonyms, but they have certain properties that become especially salient in connection with the communicative medium. Theoretical and descriptive considerations have been supplemented with nicks used on the film.web website. An attempt has been made to show the diversity of nicks but without unnecessary detail as regards the motivation for individual naming decisions. Instead, onus is put on various examples of onymic license, which has a multidimensional nature in computer-mediated communication. For example, it becomes noticeable through its associations on the appellative language plane, through interonymic relations, or through various neo-geographical forms. This license also underlies the processes of extending the boundaries of a particular language (e.g. English elements are used in Polish nicks)

Quasiregular maps are a natural generalisation of holomorphic maps to higher dimensions. Recently, there is an increasing interest in studying the dynamics of quasiregular maps on the dd-dimensional Euclidian space. Until now, not much is known about the dynamical behaviour of general quasiregular maps. However, quasiregular analogues of the complex exponential map as well as of trigonometric functions have been constructed and turned out to behave quite similarly to their counterparts in the plane. In this thesis, we examine the dynamical behaviour of certain quasiregular maps on the dd-dimensional Euclidian space which can be seen as quasiregular analogues of complex trigonometric functions. This class was introduced by Bergweiler and Eremenko in 2011. We will denote a member of this class by \Sin(x). We are interested in the iteration of these maps, in particular in the escaping set consisting of all points that tend to infinity under iteration.\\ We prove the following results: Theorem 1 is an analogue of a well known result by McMullen from 1987. We show that for f(x)=\lambda\Sin(x), where λ>0\lambda>0, the escaping set I(f)I(f) has positive Lebesgue measure. In fact, this is also true for the fast escaping set A(f)A(f) consisting of all points that essentially escape as fast as possible.\\ Theorem 2 states that the measure of the non-escaping set of ff in vertical square beams is finite. This corresponds to a result in the plane by Schubert from 2008 concerning the escaping set of sin(z)\sin(z) in vertical strips. Again, this is also true for the fast escaping set.\\ We also use the map \Sin(x) to construct a quasiregular power mapping PmP_m which is similar to a quasiregular power mapping introduced by Mayer based on Zorich maps. We discuss some properties of the map PmP_m. Theorem 3 then states that for md+1m\geq d+1, the composition of the power mapping PmP_m with a suitable quasiregular analogue of the trigonometric functions yields a quasiregular map with non-escaping set of finite measure. This generalises a result by Hemke from 2005. In the last section, we discuss the sharpness of Theorem 3.Quasireguläre Abbildungen sind eine natürliche Verallgemeinerung holomorpher Abbildungen auf höhere Dimensionen.

A transcendental entire function is a holomorphic function on the whole complex plane which is not a polynomial. The Julia set of an entire function ff denotes the set of all points in \C where the iterates fkf^k of ff do not form a normal family in the sense of Montel. Many mathematicians investigated the iteration of these functions. Even the simplest example of a transcendental entire function, namely the exponential map λez\lambda e^z with \lambda \in \C\setminus\{0\}, is of rich structure. Devaney and Krych showed in 1984 that for the exponential family λez\lambda e^z, where 0<λ<1/e0<\lambda <1/e, the Julia set consists of uncountably many pairwise disjoint simple curves tending to \infty, which they called hairs. Recently, the iteration of a natural higher-dimensional analogue of holomorphic maps, the so-called quasiregular maps, became of great interest. A special family of quasiregular functions, the so-called Zorich maps, was introduced by Zorich in 1969 and can be consi-dered as a higher-dimensional counterpart of the exponential family. Bergweiler showed in 2010 that for several Zorich maps hairs also exist. In this thesis we want to focus on the differentiability of hairs for transcendental entire functions and Zorich maps. A first result was given by Viana in 1988, who proved that the hairs of the exponential family are smooth. In the first part of this thesis, we show that his result generalizes to the Zorich maps considered by Bergweiler under some mild additional regularity assumptions. At first glance it seems that the regularity of the function transfers to the regularity of the hairs. In the second part of this thesis we construct in contrast to the first result a transcendental entire function of the so-called Eremenko-Lyubich class, where every hair is nowhere differentiable.

Titel: Comparative study between traditional seminars and webinars for marketing purpose. Level: Final assignment for Bachelor Degree in Business Administration Author: Peter Nicks and Alexander Widell Supervisor: Aihie Osarenkhoe Date: 2011 – January Aim: We want to examine how businesses can use webinars to supplement or replace seminars in its industrial marketing. When should businesses use seminars, when should they use webinars and when should they use a combination of the two? Method: We have chosen to do a combination of quantitative and qualitative research. We see the benefits of both methods for our research. Many issues raised in the aim are difficult to respond to by only using one method. The quantitative part is the questionnaires completed by participants and the qualitative part consists of two interviews. All investigated events are made at a large international IT company. Result & Findings: The results of our studied seminars shows that the larger events the greater the need for a physical seminar. We have therefore concluded that we must choose different types of seminar for various events. For minor events, it is better to just use webinars. For the big events, you always should have a traditional seminar but it is possible to combine with a webinar to reach out to even more customers. Suggestions for future research: What are the results of similar studies in other industries? Make repeated studies on a wider basis than the three events that we implemented. Further research into how seminar and webinar affect sales. It was something we set as a target at the beginning of this study, but we were not able to measure. Contribution of the thesis: With this paper, we hope to provide guidance to organizations that want to make use of webinars for marketing purposes. Key words: marketing, webinar, workshop, customers, I

In response to a request from the Mine Safety and Health Administration, an investigation was begun into the prevalence of silicosis at specific ground silica (14808607) operations by conducting a medical evaluation of active and retired miners. This investigation centered around Nicks Silica Company (SIC-1446), Jackson, Tennessee. The study population included workers with 1 year or greater cumulative tenure subsequent to the opening of the mill in the grinding area or in areas downstream of the grinding process. A medical survey was conducted which included a questionnaire, spirometry, and a single view posterior/anterior (PA) chest X-ray. None of the 12 current and former workers participating in the survey had changes on their chest X-ray consistent with silicosis. One participant showed an abnormal spirometry pattern. Three reported chronic respiratory symptoms. All four participants with positive findings were current cigarette smokers. Because of the long latency usually associated with chronic nodular silicosis, the finding is not sufficient to conclude that current silica dust exposures are without any adverse effect. The author recommends that the medical monitoring program be modified to include classification of chest X-rays by a B-reader
