Modello in vitro per lo studio dello sviluppo e delle caratteristiche fitochimiche dei noduli fogliari di Hypericum perforatum L.
Hypericum perforatum L. (erba di S. Giovanni) è una pianta officinale appartenente alla famiglia delle Clusiaceae, spontanea in Europa, Asia, Nord Africa e naturalizzata in Nord America e Australia. E’ una pianta erbacea perenne, alta 25-60 cm, possiede fiori gialli a cinque petali e foglie opposte, che viste in trasparenza, danno l’impressione di essere perforate, poiché dotate di numerose tasche traslucide di natura schizogena. L’Hypericum perforatum, oltre a ghiandole traslucide e canali di secrezione presenti su foglie e fiori contenenti olii essenziali, possiede anche noduli ghiandolari diffusi su foglie, petali, fusto e stami (1). Tali strutture, presenti per lo più lungo i margini fogliari, hanno differenti stadi di sviluppo; infatti si distinguono noduli allo stadio giovanile, di colore rosa, che virano verso un colore rosso scuro-nero a maturità assumendo un aspetto rigonfio e un diametro di circa 0,1 mm (2-3).
La maggior parte degli studi condotti e pubblicati su questa pianta riguarda le modalità di azione dei principi attivi ritenuti responsabili dell’azione farmacologica, mentre sono tuttora scarse le conoscenze relative alla biosintesi e all’accumulo di tali molecole nei tessuti della pianta. Fra i principi attivi contenuti in H. perforatum vi sono i naftodiantroni ipericina, pseudoipericina e derivati, che determinano il titolo dell’estratto commerciale e l’iperforina, un derivato del floroglucinolo; sono inoltre presenti metaboliti secondari più comuni come flavonoidi, xantoni, terpeni e olii essenziali (4). L’ipericina e le sostanze ipericino-simili sono fotodinamiche e la loro biosintesi è connessa con la morfogenesi e lo sviluppo dei noduli rosso scuro-neri. Tradizionalmente, le ipericine sono state considerate le principali responsabili delle proprietà antidepressive di H. perforatum. Tuttavia, da poco il loro ruolo è diventato secondario, sopravanzato da quello della iperforina, che studi più recenti indicano come l’autentica, anche se non l’unica, sostanza antidepressiva della droga. Inoltre sono oggetto di studio l’attività inibitoria dell’Iperico nei confronti di alcuni retrovirus (HIV) e quella citotossica verso alcune forme tumorali (5).
In questo lavoro si è valutata la sintesi e l’accumulo di principi attivi quali ipericina, pseudoipericina e iperforina in un modello sperimentale in vitro di Hypericum perforatum. Gli obiettivi erano:
- indagare le caratteristiche fitochimiche e istologiche dei noduli fogliari di H. perforatum presenti nel modello in vitro mettendole a confronto con analoghe strutture presenti nelle foglie in vivo.
La presenza di ipericine per lo più all’interno dei noduli è stata dimostrata con l’analisi all’HPLC su campioni di foglie provenienti da colture idroponiche; infatti, all’interno dei noduli è stato localizzato l’96% delle ipericine, mentre nel tessuto fogliare extra nodulare solo il restante 4 %.
Nelle colture in vitro si è verificato il differenziamento di strutture nodulari che mostrano gli stessi stadi di sviluppo dei noduli fogliari, almeno all’osservazione al microscopio stereoscopico e fino a 40 gg. dalla semina. Dopo i 40 gg. non sono più visibili i classici noduli, mentre si possono solo osservare degli ammassi cellulari rosso-vivo che sembrano non possedere una struttura ben organizzata. Inoltre, il numero e lo sviluppo dei noduli stessi appaiono influenzati dalla luce; infatti i noduli differenziati nel callo cresciuto al buio sono meno numerosi e si trovano soltanto allo stadio iniziale di sviluppo (stadio giovanile). La presenza di ipericina e pseudoipericina è stata confermata dalle analisi con HPLC di estratti ottenuti dai calli. L’iperforina è stata trovata in tutte le colture cellulari alla luce , ma mai in quelle al buio.
L’indagine ultrastrutturale dei noduli provenienti dai modelli in vivo e in vitro ha condotto alle seguenti osservazioni: i noduli delle foglie sono strutture cellularizzate contenenti al loro interno la secrezione. Essi sono formati da una fascia periferica di cellule appiattite e una zona centrale di cellule isodiametriche. Nei noduli giovani, in entrambi i tipi di cellule, sono evidenti organuli ben strutturati e si riscontra la presenza di vescicole contenenti materiale elettrondenso disposte nello spazio periplasmatico, alle estremità tra le cellule e lungo le pareti cellulari. Durante lo sviluppo del nodulo si assiste ad un ispessimento della parete delle cellule che lo compongono, accompagnato dalla degenerazione degli organuli cellulari. In questo stadio le cellule del nodulo rimangono intatte e non formano nessun lume; infatti non appare nessuna cavità ghiandolare, ma il materiale si accumula progressivamente nello spazio intracellulare che risulta, quindi, pieno di materiale elettrondenso. Nella parete cellulare del nodulo maturo non è presente nessuna sostanza osmiofilica, come suberina o lignina (6). Analogamente al nodulo fogliare, il nodulo prelevato dalla coltura in vitro alla luce e ad uno stadio avanzato dello sviluppo (stadio maturo) possiede delle cellule più esterne che circondano quelle interne. Le cellule esterne sono caratterizzate dalla presenza di grandi vacuoli che non mostrano inclusi; il citoplasma e gli organuli sono poco riconoscibili per la notevole presenza di materiale elettrondenso. Le cellule più interne, invece, non mostrano differenze dalle altre cellule del callo: sono ben strutturate e possiedono grandi vacuoli, che anche in questo caso sono privi di inclusi.
Dai risultati ottenuti si può affermare che, nonostante nei noduli fogliari sia presente la maggior parte delle ipericine, queste sono state trovate anche nel mesofillo, con una ripartizione tale da far ipotizzare una sintesi nodulare e un successivo trasporto o diffusione extra nodulare dei principi attivi suddetti.
Il modello in vitro messo a punto si presta ad indagini relative alla sintesi e all’accumulo di questi metaboliti secondari, oltre allo studio del differenziamento delle strutture nodulari. Inoltre nelle colture cellulari, la luce è un fattore che influisce notevolmente sia sul differenziamento dei noduli sia sulla sintesi di iperforina.

Going Beyond Counting First Authors in Author Co-citation Analysis
The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation
counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings
are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that
only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into
account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed. Reasons for these effects are discussed
Variations on the Author
“Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship
Appropriate Similarity Measures for Author Cocitation Analysis
We provide a number of new insights into the methodological discussion about author cocitation analysis. We first argue that the use of the Pearson correlation for measuring the similarity between authors’ cocitation profiles is not very satisfactory. We then discuss what kind of similarity measures may be used as an alternative to the Pearson correlation. We consider three similarity measures in particular. One is the well-known cosine. The other two similarity measures have not been used before in the bibliometric literature. Dispelling the Myths Behind First-author Citation Counts
We conducted a full-scale evaluative citation analysis study of scholars in the XML research field to explore just how different from each other author rankings resulting from different citation counting methods actually are, and to demonstrate the capability of emerging data and tools on the Web in supporting more realistic citation counting methods. Our results contest some common arguments for the continued
use of first-author citation counts in the evaluation of scholars, such as high correlations between author rankings by first-author citation counts and other citation
counting methods, and high costs of using more realistic citation counting methods that are not well-supported by the ISI databases. We have done our best to complete the author checklist relating to the use of animals in the hut study. Note that the objective for the hut study was to evaluate the IRS treatment applications for residual efficacy against Anopheles mosquitoes, including the local An. coluzzii mosquito population. Cows were only used to attract mosquitoes into the huts and no tests were carried out directly on the cows. The author checklist is intended for use with studies where experiments are carried out on animals, which is why we have had such difficulty in completing this for the hut study, as many of the questions do not relate to how the cows were used
