    The undrained shear strength of remoulded soils is of great concern in geotechnical engineering applications. This study aims to develop a reliable approach for determining the undrained shear strength of remoulded fine-grained soils, through the use of index test results, at both the plastic and semi-solid states of consistency. Experimental investigation and subsequent analysis involving a number of fine-grained soils of widely varying plasticity and geological origin have led to a two-parameter linear model of the relationship between logarithm of remoulded undrained shear strength and liquidity index. The numerical values of the parameters are found to be dependent to a lesser extent on the soil group and to a greater extent on the soil state. Based on the values of regression coefficient, ranking index and ranking distance, it seems that the relationship represents the experimental results well. It may be pointed out that the possibility of such a relationship in the semi-solid state of a soil has not been explored in the past. It is also shown that the shear strength at the plastic limit is about 32–34 times that at the liquid limit

    Improved insulin sensitivity associated with reduced mitochondrial complex IV assembly and activity

    Mice lacking Surf1, a complex IV assembly protein, have 50-70% reduction in cytochrome c oxidase activity in all tissues yet a paradoxical increase in lifespan. Here we report that Surf1-/- mice have lower body (15%) and fat (20%) mass, in association with reduced lipid storage, smaller adipocytes, and elevated indicators of fatty acid oxidation in white adipose tissue (WAT) compared with control mice. The respiratory quotient in the Surf1-/- mice was significantly lower than in the control animals (0.83- 0.93 vs. 0.90-0.98), consistent with enhanced fat utilization in Surf1-/- mice. Elevated fat utilization was associated with increased insulin sensitivity measured as insulinstimulated glucose uptake, as well as an increase in insulin receptor levels (2-fold) and glucose transporter type 4 (GLUT4; 1.3-fold) levels in WAT in the Surf1-/- mice. The expression of peroxisome proliferator- activated receptor β-coactivator 1 (PGC-1α) mRNA and protein was up-regulated by 2.5- and 1.9- fold, respectively, in WAT from Surf1-/- mice, and the expression of PGC-1α target genes and markers of mitochondrial biogenesis was elevated. Together, these findings point to a novel and unexpected link between reduced mitochondrial complex IV activity, enhanced insulin sensitivity, and increased mitochondrial biogenesis that may contribute to the increased longevity in the Surf1-/- mice.-Deepa, S.     J. M. Coetzee: Ethics, Subalternity, and the Critique of Humanism

    In the era of globalization, postcolonial studies is again confronted with the question of Western humanism and its attendant project of universalizing Eurocentric assumptions about the human. My dissertation argues that J. M. Coetzee’s postmodern novels deliberately reconstruct a literary genealogy of this project from the colonial through the postcolonial periods in order to disrupt it. My thesis addresses four novels of Coetzee that cover his entire oeuvre: the early novel Waiting for the Barbarians (1980), the novel of the middle phase Foe (1986), and the later novels— Disgrace (1999) and Diary of a Bad Year (2007). Each chapter elaborates on the intertextual nature of Coetzee’s novels and explores the rich dialogue between him and the canonical Western writers—Daniel Defoe, Franz Kafka, and Michel de Montaigne. I argue that through metafictional literary forms such as allegory, parody, and the essay, Coetzee’s postmodern novels simultaneously construct and interrupt the Eurocentric humanist universals of progress, reason, rights, the sovereign subject, democracy, and universal history. The totalizing and humanizing impulses of these metanarratives have shaped much of the history of apartheid and colonialism, and continue to do so in our era of neo-imperial globalism. Coetzee’s fiction, with the exception of Dusklands (1974), exclusively stages the relationship between the figure of the liberal humanist and the subaltern, a self-other dynamic based on hegemony rather than pure domination. My analysis demonstrates that, in the figure of the representative humanist, Coetzee’s oeuvre stages the tendency of the humanizing discourses to produce gendered and racialized subaltern bodies. The novels also dramatize the resistance of the subalterns to the epistemic violence of these narratives, resulting in the eventual failure of the intellectual to grant voice to them. Thus, I propose that Coetzee’s work gestures towards an ethics of the other, in which the Eurocentric self is called into question by the other of history. In the tradition of postmodernism, the relationship between fiction and history is unstable and insecure. His fiction does not stand witness to or represent history, but ceaselessly re-presents the relationship between fiction and fiction, fiction and history, and fiction and the self-of-writing

    Investigating the roles of Hsp40s and an E3 ubiquitin ligase in the biogenesis of Apolipoprotein B

    Apolipoprotein B (ApoB) is the primary component of atherogenic lipoproteins and is responsible for the transport of dietary and endogenous fats and cholesterol in the blood stream. Elevated levels of ApoB-containing particles lead to the development of Coronary Artery Disease, which kills millions of people every year. As it is synthesized, ApoB is co-translationally translocated into the endoplasmic reticulum (ER) through the Sec61 translocon and is lipidated, leading to the formation of a lipoprotein particle that is eventually secreted into the plasma. However, in the absence of lipidation, ApoB is retrotranslocated to the cytosol and destroyed via ER Associated Degradation (ERAD), a regulated pathway that maintains proteostasis by normally degrading misfolded proteins in the early secretory pathway. In this thesis, I investigated the contributions of select cytosolic Hsp40s on ApoB biogenesis. To this end, I used a rodent cell line that endogenously synthesizes and secretes ApoB. By performing siRNA mediated knockdown and metabolic pulse chase experiments, I discovered that a class A Hsp40, DNAJA1, associates with and facilitates ApoB degradation. Consistent with these data, Ydj1 (the yeast homolog of DNAJA1) also facilitates ApoB degradation in a heterologous yeast expression system. The ablation of DNAJA1 also leads to the accumulation of ubiquitinated ApoB, suggesting that DNAJA1 functions prior to degradation. In stark contrast to DNAJA1, DNAJB1, a class B member, stabilizes ApoB. Because knockdown decreases the levels of ubiquitinated ApoB, DNAJB1 might act prior to DNAJA1 as ApoB enters the ER. Finally, because ERAD substrates must be ubiquitinated after chaperone-dependent selection, I examined the contributions of E3 ubiquitin ligases on ApoB stability and discovered that Hrd1 targets ApoB for degradation in the rodent cell line. Together, my findings highlight the unexpected functional diversity of Hsp40s during the biogenesis of ApoB

    Open innovation, product portfolio innovativeness and firm performance: the dual role of new product development capabilities

    Despite a growing interest in the phenomenon of open innovation (OI), empirical evidence documenting the link between new product development capabilities, OI practices, and new product innovativeness is scarce. Eminent scholars have called for large-scale studies that systematically investigate the OI paradigm. Drawing on the knowledge-based view of the firm, new product development, and NPD capabilities literature streams, we conceptualize a framework in which OI practices are disentangled according to the stage of the new product development process in which they occur (development stage or commercialization stage). We identify two major types of OI practices: development-centric OI (which occurs in the development stage) and commercialization-centric OI (which occurs in the commercialization stage). Specific types of NPD capabilities R&D, market information management, and launch are expected to both influence the extent to which each OI practice is implemented and moderate the effect of each OI practice on product portfolio innovativeness and firm performance. The empirical analysis combines primary data from a survey of 239 firms with secondary data on innovation and financial outcomes. Our results support our hypotheses and indicate a need to differentiate among the different kinds of OI practices while elaborating on the complex role played by NPD capabilities in influencing OI practices
