Characterization and analysis of D-amino acids.
The need to screen a large number of natural extracts, with the aim of detecting D-amino acids or isolating and characterizing peptides containing them, has stimulated the development of novel and improved procedures for the analysis of amino acid enantiomeric mixtures, with special attention paid to automation. Different methods for the analysis of D-amino acids are described and discussed
O amor, o sorriso e o povo brasileiro: Vinícius de Moraes e os movimentos culturais do Brasil contemporâneo
Dissertação (mestrado) - Universidade Federal de Santa Catarina, Centro de Filosofia e Ciências Humanas, Programa de Pós-Graduação em Sociologia Política, Florianópolis, 2006.Este estudo propõe uma relação entre produção cultural e contexto social através da análise das composições de Vinicius de Moraes, tendo com referência duas de suas principais obras: Chega de Saudade e Gente Humilde. Para a realização desta pesquisa, inseriu-se o poeta no contexto histórico da época em que as letras foram feitas, além de se verificar também o quadro político vivido por Moraes em toda sua trajetória (1913 até 1980). Em um breve histórico, foram organizados momentos políticos, fazendo-se um levantamento dos presidentes do Brasil e as características de seus governos. Em seguida, apresenta-se um relato a respeito da vida de Vinicius de Moraes, suas profissões, suas viagens, suas parcerias e seus ideais políticos. Abordou-se ainda o contexto musical no qual as duas letras foram transformadas em músicas, descrevendo a Bossa Nova e as Músicas de Protesto. Ao final, analise-se as letras de Chega de Saudade e Gente Humilde, do ponto de vista da relação do poeta com suas parcerias, com o público e consigo.This study it considers a relation between cultural production and social context through the analysis of the production of Vinicius de Moraes having with reference two of its main compositions: #Chega de Saudade# and #Gente Humilde#. To build this research it was necessary to insert the poet into the historical context of the time the letters had been made and also verify the political context that Moraes lived during his entire trajectory (1913 up to 1980). In a brief description, the political setting had been rebuilt, including Brazilian Presidents and their governments# characteristics. After that, it was made a narrative regarding Vinicius de Moraes life, his professions, his trips, his partnerships and his political ideals. The musical context in which the two lyrics were transformed into music was also considered, describing the #Bossa Nova# and the Music of Protest. At last it was carried out an analysis of the songs #Chega de Saudade#and #Gente Humilde# considering the relation of the poet with his partnerships, with the public and with himself

Ruptura e continuidade na MPB: a questão da linha evolutiva
Dissertação (mestrado) - Universidade Federal de Santa Catarina, Centro de Filosofia e Ciências Humanas, Programa de Pós-Graduação em Antropologia, Florianópolis, 1992.O objetivo central deste trabalho está em interpretar o sentido da discursividade nativa Bossa Nova (BN) e Tropicalista (TROPI) no que tange à evolução musical popular associado à ruptura estética moderna. Para obter este objetivo utilizo-me de duas frentes: a primeira, examina a topologia discursiva, enquanto a segunda mergulha na feitura das canções. A obra divide-se em quatro partes. Na primeira, faço um apanhado de umas obras musicológicas para demonstrar o atual estágio deste assunto em ciências humanas. Na segunda verifico a discursividade em seus nexos com a ideologia de construção nacional do Brasil através da música. Na terceira parte realizo um exame semântico de algumas canções do movimento BN-TROPI, tentanto apontar para os momentos de entrelaçamento destes com esquemas estilísticos tradicionais. Por fim, a quarta parte é a conclusão, onde faço um rápido arremate de algumas idéias levantadas nas partes anteriores
ANTIMICROBIAL PEPTIDES FROM SKIN SECRETIONS OF RANA ESCULENTA. MOLECULAR CLONING OF cDNAs ENCODING ESCULENTIN AND BREVININS AND ISOLATION OF NEW ACTIVE PEPTIDES
Three cytolytic peptides, termed brevinin-1E, brevinin-2E, and esculentin, were isolated from skin secretions of the European frog Rana esculenta (Simmaco, M., Mignogna, G., Barra, D., and Bossa, F. (1993) FEBS Lett. 324, 159-161). Nucleotide sequence analysis of cDNAs coding for the corresponding precursors revealed that in all of them a single copy of the sequence of the mature peptide is present preceded by a dibasic cleavage site and followed by a stop codon. The signal peptides of these precursors show a clear homology to the corresponding region of the precursor of dermorphin, a neuropeptide occurring in the skin of amphibians of the subfamily Phyllomedusinae. Ten new peptides, ranging in size from 24 to 46 residues, all possessing an intramolecular disulfide bridge located at the carboxyl-terminal end, were isolated from skin secretions of R. esculenta. These peptides can be grouped into four subfamilies on the basis of their distinctive structural and/or functional properties. All of these new peptides have antimicrobial and/or hemolytic activities typical for the respective subfamily. In addition, we demonstrate that esculentin-1 also inhibits the growth of Pseudomonas aeruginosa, Candida albicans, and Saccharomyces cerevisiae
CHEMISTRY OF THE INACTIVATION OF 4-AMINOBUTYRATE AMINOTRANSFERASE BY THE ANTIEPILEPTIC DRUG VIGABATRIN
The chemical modification of pig liver 4-aminobutyrate aminotransferase by the antiepileptic drug 4-aminohex-5-enoate (Vigabatrin) has been studied. After inactivation by C-14-labeled Vigabatrin, the enzyme was digested with trypsin, and automated Edman degradation of the purified labeled peptide gave the sequence FWAHEHWGLDDPADVMTFSKK. Chymotryptic digestion of the tryptic peptide and sequencing of a resulting tripeptide identified the penultimate lysine residue of this peptide as the site of covalent modification. This lysine normally binds the coenzyme. Absorption spectroscopy demonstrated the absence of coenzyme from the tryptic peptide, and mass spectrometry showed its mass/charge ratio to be increased by 128. All of the bound coenzyme released after denaturation of the inactivated enzyme was as pyridoxamine phosphate. The structural nature of the modification is deduced, and mechanisms for its occurrence identified. Initially, 1 mol of radiolabeled inhibitor was bound per mol of monomer of the enzyme, although approximately half was released during denaturation and digestion, while the remainder was irreversibly bound. Coenzyme not released as pyridoxamine phosphate retained the absorbance characteristics of the aldimine, although the enzyme was completely inactive. Mass spectrometry of the sample of purified radiolabeled tryptic peptide revealed the presence of an approximately equal amount of a second fragment that contained no modification and from which the second lysine was absent, indicating that at the time of proteolysis the active site lysine was unaltered in 50% of the enzyme molecules

Pig kidney 3,4-dihydroxyphenylalanine (dopa) decarboxylase. Primary structure and relationships to other amino acid decarboxylases
The complete amino acid sequence of pig kidney 3,4-dihydroxyphenylalanine (Dopa) decarboxylase is reported. The sequence was determined from analysis of peptides isolated after tryptic and cyanogen bromide cleavages of the enzyme. Each subunit is made up of 485 residues, corresponding to a molecular mass of 53858 Da. The N-terminus of the polypeptide chain is an acetylated methionyl residue. A number of structural features, previously shown to be important for the structure and function of the enzyme, could be localized along the polypeptide chain. Comparison of the primary structure with the known cDNA-deduced sequences of other Dopa decarboxylases (i.e. the human, bovine, rat, guinea-pig and Drosophila enzymes) reveals 50% identity. The identity increases to 73%, if the comparison is restricted to the mammalian sequences. Comparison with other aromatic and non-aromatic decarboxylases allows some consideration to be made in terms of structure/function and evolutionary relationships in this class of enzymes
