Human and murine granzyme B exhibit divergent substrate preferences
The cytotoxic lymphocyte protease granzyme B (GzmB) can promote apoptosis through direct processing and activation of members of the caspase family. GzmB can also cleave the BH3-only protein, BID, to promote caspase-independent mitochondrial permeabilization. Although human and mouse forms of GzmB exhibit extensive homology, these proteases diverge at residues predicted to influence substrate binding. We show that human and mouse GzmB exhibit radical differences in their ability to cleave BID, as well as several other key substrates, such as ICAD and caspase-8. Moreover, pharmacological inhibition of caspases clonogenically rescued human and mouse target cells from apoptosis initiated by mouse GzmB, but failed to do so in response to human GzmB. These data demonstrate that human and murine GzmB are distinct enzymes with different substrate preferences. Our observations also illustrate how subtle differences in enzyme structure can radically affect substrate selection
Appropriations of Irish drama by modern Korean nationalist theatre : a focus on the influence of Sean O’Casey in a colonial context
My thesis explores how a translated author on the periphery of the host culture’s
translated repertoire can be at once subversive and innovative on the colonial scene,
using as an example the case of Sean O’Casey in colonial Korea. It explores the
importation of Irish drama in modern Korean theatre during the colonial period and
examines the appropriations of O’Casey’s plays by a central Korean playwright, Yu
Chi-jin, in creating his own plays. Under Japanese colonial rule in the early twentieth
century, intellectuals perceived the supreme task for the Korean people to be the
recovery of national sovereignty and independence. The modern Korean theatre
movement which rose among Korean intellectuals and dramatists during the colonial
period was to play a major part in this task. The ultimate goal of this movement was
to establish a modern national theatre promoting Korean culture and educating the
people, thereby recovering national independence. As their modernised dramatic
polysystem was still "young", Korean intellectuals and dramatists who were
involved in the theatre movement had to borrow dramatic models from other
countries. One of the models they chose was Irish playwrights, especially those who
were involved in the Irish dramatic movement. They published or staged the works
of W.B. Yeats, Lord Dunsany [Edward John Moreton Drax Plunkett], Augusta
Gregory, J.M. Synge, St. J. Ervine, T.C. Murray and Sean O'Casey. Although
O'Casey was considered an important dramatist in the Irish dramatic movement, he
was a playwright on the periphery in the list of translated Irish dramatists in Korea
due to the colonisers’ censorship. However, he remained as a subversive and
innovative playwright on the colonial scene by virtue of being appropriated by Yu
Chi-jin who used O’Casey’s plays as models when creating his own works. In
discussing the subject matter of my thesis, I use Even Zohar’s polysystems theory as
a starting point in looking at ideological issues surrounding translation and extend
the discussion to offer a postcolonial perspective. While most translation in a
colonial context was considered as "an expression of the cultural power of the
colonisers," my thesis shifts the focus to translation as an expression of the cultural
power of the colonised. I explore how the colonised uses another colonised culture to
subvert the colonisers’ power
The Influence of Policy Implementation Service Standards, and Information Systems on User Satisfaction of the MELATI Application at Badan Pemeriksa Keuangan Republik Indonesia
The increase in the number of requests for IT services provided by the IT Bureau is not accompanied by an increase in user satisfaction of IT services, this is indicated by a decrease in the satisfaction index for IT services by several work units at the BPK. In order to increase user satisfaction with IT services, the Author is interested in conducting research on the Effect of Implementation of Service Standards and Information Systems Policies on User Satisfaction of the Melati Application at the Audit Board of the Republic of Indonesia. This study uses a quantitative method with primary data collection techniques by distributing questionnaires to 275 IT service users at the BPK Head Office. The collected data were processed using the Covariance Based - Structural Equation Model (CB-SEM) analysis technique using Smart-PLS software. The results of the study showed that user satisfaction of the MELATI application (Y) was influenced by the implementation of service standard policies (X1) and information systems (X2) by 67.5% and the remaining 32.5% was influenced by other variables not used in this study. The variable of IT service standard policy implementation (X1) has a large and significant effect on user satisfaction of MELATI application in BPK with a path coefficient value of 0.495, t-value of 8.378> t-table 1.96 and p-value of 0.000 <0.05. Information system (X2) has a large and significant effect on user satisfaction of MELATI application in BPK with a path coefficient value of 0.465 or 46.5%, t-value of 8.199> t-table 1.96, and p-value of 0.000 <0.05. Efforts that can be made by the BPK IT Bureau are to improve the implementation of service standard policies and information systems by increasing the quantity of resources, communication, evaluation and updating of IT service standard policies and SOPs and implementing a survey of IT service user satisfaction on the MELATI application
Learning theories and interprofessional education: a user's guide
There is increasing interest in the theoretical underpinning of interprofessional education (IPE) and writers in this field are drawing on a wide range of disciplines for theories that have utility in IPE. While this has undoubtedly enriched the research literature, for the educational practitioner, whose aim is to develop and deliver an IPE curriculum that has sound theoretical underpinnings, this plethora of theories has become a confusing, and un-navigable quagmire. This article aims to provide a compass for those educational practitioners by presenting a framework that summarizes key learning theories used in IPE and the relationship between them. The study reviews key contemporary learning theories from the wider field of education used in IPE and the explicit applications of these theories in the IPE literature to either curriculum design or programme evaluation. Through presenting a broad overview and summary framework, the study clarifies the way in which learning theories can aid IPE curriculum development and evaluation. It also highlights areas where future theoretical development in the IPE field is required

An integrated biomarker approach for assessing exposure and effects of endocrine disruptors and other contaminants in killifish (Fundulus heteroclitus) from the New York-New Jersey harbor estuary:
A multi-tiered approach was used to evaluate killifish (Fundulus heteroclitus) health by examining a suite of biomarkers in killifish inhabiting the heavily industrialized Newark Bay and a reference population in Tuckerton, NJ. The general hypothesis of this study was that Newark Bay killifish would exhibit biomarker responses indicative of impaired health when compared to a reference population from Tuckerton, NJ. The biomarkers investigated included classical endpoints (histopathology, morphometrics, gonad maturation), hepatic mRNA expression (CYP1A, metallothionein, vitellogenin I), gonadal aromatase mRNA expression, hepatic protein levels (CYP1A and vitellogenin I) and chemical exposure analyses (bile PAHs). Newark Bay fish had significantly higher levels of bile PAHs compared to reference fish. Females had significantly higher concentrations of naphthalene, pyrene and benzo[a]pyrene (3, 6 and 4 fold higher, respectively, p<0.05). Males had significantly higher concentrations of pyrene (7-fold higher, p<0.05), and higher concentrations of naphthalene (4-fold higher, p=0.06) and benzo[a]pyrene (9-fold higher, p=0.07). Histological lesions of the liver and pancreas in Newark Bay fish were similar to reference fish. Newark Bay fish had significantly higher expression of hepatic CYP1A for both males and females (7 and 3 fold higher, respectively, p<0.05) and CYP1A protein levels for both males and females (5 and 8 fold higher, p<0.05). Endocrine disruption in male gonads is demonstrated by a decreased gonad weight, altered testis development and upregulated aromatase expression (3-fold, p<0.05), which indicates exposure to endocrine active compounds. Efforts to strip spawn Newark Bay females produced few viable eggs, while at the same time Tuckerton females produced large numbers of viable eggs. There was a significant increase in the percent of pre-vitellogenic follicles (43% at Tuckerton, 64% at Newark Bay) and a significantly decreased percent of follicles at the mid-vitellogenic and mature stages (25% at Tuckerton, 3% at Newark Bay) in Newark Bay females (p<0.05). Vitellogenin mRNA and protein (egg yolk-protein) was significantly decreased in Newark Bay females (6-fold lower mRNA, 27-fold lower protein, p<0.05) while gonadal aromatase (produces 17β-estradiol) was significantly increased (210-fold higher, p<0.05). Killifish in the NY-NJ Harbor Estuary are exposed to high amounts of PAHs and aryl hydrocarbon agonists and their reproductive health is impaired. Bicaudal is a conserved substrate for Drosophila and mammalian caspases and is essential for cell survival
Members of the caspase family of cysteine proteases coordinate cell death through restricted proteolysis of diverse protein substrates and play a conserved role in apoptosis from nematodes to man. However, while numerous substrates for the mammalian cell death-associated caspases have now been described, few caspase substrates have been identified in other organisms. Here, we have utilized a proteomics-based approach to identify proteins that are cleaved by caspases during apoptosis in Drosophila D-Mel2 cells, a subline of the Schneider S2 cell line. This approach identified multiple novel substrates for the fly caspases and revealed that bicaudal/betaNAC is a conserved substrate for Drosophila and mammalian caspases. RNAi-mediated silencing of bicaudal expression in Drosophila D-Mel2 cells resulted in a block to proliferation, followed by spontaneous apoptosis. Similarly, silencing of expression of the mammalian bicaudal homologue, betaNAC, in HeLa, HEK293T, MCF-7 and MRC5 cells also resulted in spontaneous apoptosis. These data suggest that bicaudal/betaNAC is essential for cell survival and is a conserved target of caspases from flies to man
The Author Naming Task: A newly-proposed measure for assessing print exposure
A common measure of print exposure (i.e., how much a person reads) is the Author Recognition Test (ART). This however is a blunt tool with concerns about its reliability and validity (McCarron & Kuperman, 2021). This project will develop and test a new method for capturing people’s reading experience. In line with measures of verbal fluency, (e.g. “How many animals can you name beginning with the letter S?”) an “Author Naming Task” (ANT) is proposed. Adult native English-speaking participants will be given 3 minutes to produce as many author names as possible from memory. An English-language ART will also be administered, but with author and foil names presented serially, following the format of a lexical decision task (LDT). Participants will also complete an LDT which presents a series of words and pseudowords and asks participants to decide if each presentation is a word or not. Words selected for this LDT are those most representative of fiction books, as fiction is generally associated with reading for leisure and thus is understood to index additional reading practice (Mol & Bus, 2011). Fiction keywords were selected from a large-scale corpus analysis comparing fiction and non-fiction writing, using a metric of keyness. Response accuracy and reaction times from participants are recorded. We test whether reading experience (as assessed by ANT and ART) is associated with individual differences in word reading and vocabulary knowledge (as indicated by the LDT). Participants will also be asked about the frequency and nature of their reading experience. As an exploratory step, participants will additionally be asked about their media use, about the ease with which mental images come to mind when reading, their views on the essential nature of other people, and their general disposition
Absorbing new subjects: holography as an analog of photography
I discuss the early history of holography and explore how perceptions, applications, and forecasts of the subject were shaped by prior experience. I focus on the work of Dennis Gabor (1900–1979) in England,Yury N. Denisyuk (b. 1924) in the Soviet Union, and Emmett N. Leith (1927–2005) and Juris Upatnieks (b. 1936) in the United States. I show that the evolution of holography was simultaneously promoted and constrained by its identification as an analog of photography, an association that influenced its assessment by successive audiences of practitioners, entrepreneurs, and consumers. One consequence is that holography can be seen as an example of a modern technical subject that has been shaped by cultural influences more powerfully than generally appreciated.

to explain why the cultural effects of holography have been more muted than anticipated by forecasters
between the 1960s and 1990s
