    Chromatin-like structures in polyoma virus and simian virus 10 lytic cycle

    Nucleoprotein complexes containing viral DNA and cellular histones were extracted from nuclei of permissive cells infected with polyoma virus or simian virus 40 (SV40) and examined by electron microscopy. Polyoma and SV40 nucleoprotein complexes are almost identical. They appear as relaxed circular molecules consisting of 20 to 21 globular particles interconnected by thin filaments. Their contour length in 0.02 M salt is 2.7 times shorter than that of viral DNA form I obtained after dissociation of the proteins in 1 M NaCl. The nucleosomes have an average diameter of 12.5 nm. Each nucleosome contains 175 to 205 DNA base pairs condensed fivefold in length. The nucleosomes are regularly spaced on the circular molecule. The internucleosomal filaments are made of naked DNA, and each filament contains about 55 base pairs. The partial sensitivity of the nucleoprotein complex to cleavage by EcoR1 endonuclease suggests that the nucleosomes are not formed at specific sites on the viral genome. Faster sedimenting nucleoprotein complexes containing replicative intermediates were studied. Isopycnic centrifugation in metrizamide gradients in the absence of aldehyde fixation showed that these molecules conserved the same DNA-to-protein ratio as the form I DNA-containing complexes

    “Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship

    Going Beyond Counting First Authors in Author Co-citation Analysis

    The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed. Reasons for these effects are discussed

    Mestrado - BioquímicaO presente trabalho aborda a conceção de um plano baseado no sistema Hazard Analysis and Critical Control Points (HACCP) para implementação na produção do croissant tradicional salgado, realizado em ambiente empresarial na Pastelaria Central Lda (Pastelaria Latina). Em primeiro lugar é efetuada uma revisão bibliográfica referente às matérias-primas utilizadas na produção dos produtos de panificação, em que se inclui o croissant. É abordada também a aplicação da metodologia do sistema HACCP, descrevendo-se esta metodologia através da implementação dos pré-requisitos e dos sete princípios que permitem analisar os potenciais perigos que podem ocorrer ao longo da cadeia alimentar, identificar os passos de produção em que realmente esses perigos constituem um risco alimentar (determinação dos PCCs) e estabelecer um controlo eficaz desses PCCs. O croissant tradicional salgado, tal como outros produtos de panificação, pode apresentar problemas de segurança alimentar, identificados em três categorias de perigos: biológicos, químicos ou físicos. Para além destes, podem ocorrer defeitos de produção, nomeadamente alterações a nível de cor, sabor ou aparência, que podem levar também à rejeição do produto. Neste trabalho identificou-se a etapa de cozedura como um PCC no processo de fabrico do croissant tradicional salgado. Para prevenção da ocorrência de defeitos de produção, para a obtenção de um croissant com as características desejadas: miolo leve e fofo e crosta de cor marron e estaladiça, é fundamental controlar o binómio temperatura/tempo dos passos de levedação (23 ºC, 2h45 a 3h00) e cozedura (200 °C, 1 h).The present work proposes a plan based on the Hazard Analysis and Critical Control Points system (HACCP) to implement on the production of baked salty croissant. A bibliographic review of the state of the art concerning the raw materials used for baked products, which included the croissant is perfomed. Also, the implementation of the HACCP methodology is described. This includes the implementation of the prerequisites and the seven principles which allow to analyze 1) the potential hazards that may occur along the food chain, 2) the identification of the production steps where these hazards constitute a real risk (determination of CCPs), and 3) the establishment of an effective monitoring of these CCPs. The salty croissant, such as other baked products, can present food safety problems, identified in three hazard categories: microbiological, chemical, or physical. Beyond these, production defects, namely, changes in color, flavor or appearance, may also cause a rejection of the product. In this work, the cooking phase was identified as a CCP for the production of the salty croissant. To prevent the occurrence of production defects, in order to obtain a croissant with the desired characteristics: soft and fluffy crumb and a brown and crisp crust, a tight control of the temperature/time of fermentation (23 ºC, 2h45 a 3h00) and cooking steps (200 °C, 1 h) is required

    Dispelling the Myths Behind First-author Citation Counts

    We conducted a full-scale evaluative citation analysis study of scholars in the XML research field to explore just how different from each other author rankings resulting from different citation counting methods actually are, and to demonstrate the capability of emerging data and tools on the Web in supporting more realistic citation counting methods. Our results contest some common arguments for the continued use of first-author citation counts in the evaluation of scholars, such as high correlations between author rankings by first-author citation counts and other citation counting methods, and high costs of using more realistic citation counting methods that are not well-supported by the ISI databases. It is argued that increasingly available digital full text research papers make it possible for citation analysis studies to go beyond what the ISI databases have directly supported and to employ more sophisticated methods

    Étude des parlers du Croissant via la cartographie informatisée.

    International audienceL’aire que l’on nomme le Croissant linguistique – terme venant de Ronjat (1913) – correspond à la frange nord du Massif Central (Brun-Trigaud, 1990). Plus précisément, d’ouest en est, cette zone traverse les départements de la Charente, la Vienne, la Haute- Vienne, l’Indre, la Creuse, le Cher, l’Allier et le Puy-de-Dôme. Ces parlers ont la particularité d’être en contact avec des variétés d’occitan (limousin et auvergnat), des langues d’oïl (poitevin, berrichon et bourbonnais), dans une moindre mesure le francoprovençal, ainsi qu’avec la langue nationale, le français. Les parlers du Croissant présentent donc simultanément des traits caractéristiques des différentes formes de gallo-roman mentionnées ci-dessus. Les premiers à s’intéresser à cette zone et à essayer d’en déterminer les limites sont les chercheurs C. Tourtoulon et O. Bringuier (1876), mais leur étude n’a pu être achevée. Le projet « Les parlers du Croissant : une approche multidisciplinaire du contact oc-oïl » (http://parlersducroissant.huma-num.fr/) a pour but de développer les connaissances disponibles sur les parlers du Croissant, en combinant l’apport des travaux passés à de nouvelles recherches (enquêtes de terrain, études descriptives, comparatives et multidisciplinaires). Dans le cadre de cette communication, je présenterai dans une première partie le projet « Les parlers du Croissant » et son action. Puis, dans une seconde partie, je présenterai quelques caractéristiques marquantes de la zone du Croissant. Dans une troisième partie, je me centrerai sur ma propre recherche doctorale qui, dans le cadre dudit projet, vise notamment à sélectionner des traits caractéristiques des parlers du Croissant présentant un intérêt au niveau (i) du lexique (ii) de la morphologie, (iii) de la sémantique, (iv) de la phonologie et de la phonétique. Je montrerai, en m’appuyant sur des exemples précis, comment ces traits peuvent donner lieu à l’élaboration de représentations cartographiques (conçues grâce à des outils informatiques contemporains) afin d’illustrer leur variation dans l’espace. Je conclurai ensuite sur l’intérêt de ces expériences cartographiques pour l’analyse de la variation dialectale, dans le Croissant et ailleurs
