Genética y biología molecular de la anemia de Faconi
Consultable des del TDXTítol obtingut de la portada digitalitzadaLa anemia de Fanconi (FA) es un desorden genético de naturaleza autosómica recesiva y caracterizado por una serie de malformaciones congénitas, fallo de médula ósea y una elevada predisposición a adquirir tumores sólidos y leucemia mieloide aguda. Las células FA, poseen una elevada inestabilidad cromosómica tanto espontánea como inducida por agents inductors de enlaces cruzados. Otros rasgos celulares son la sensibilidad frente al daño oxidativo, fallos en el ciclo cellular, niveles elevados de apoptosis regulación deficiente de la transcripción y disfunción telomérica. Hasta el momento, se ha descrito que existen 12 genes diferentes que pueden causar esta enfermedad, aunque no todos ellos han sido clonados y caracterizados. De entre ellos, FANCA es el que más frecuentemente se halla mutado entre la población y presenta un amplio espectro de mutaciones, siendo las grandes deleciones una de las principales. Un conocimiento más profundo de la biología de esta enfermedad nos va a permitir adquirir una visión más global de la reparación del daño en el DNA en respuesta a determinados agentes así como un mejor entendimiento de las interacciones moleculares que se establecen entre distintos síndromes genéticos y que tienen un indiscutible valor diagnóstico y terapéutico. En este trabajo de tesis se han planteado cuatro objetivos principales y generales, que son: - Establecer potenciales relaciones entre las características hematológicas de la enfermedad y algunas características celulares tales como el acortamiento telomérico y las anomalías cromosómicas numéricas o estructurales. - Caracterizar genéticamente a la población afectada por la anemia de Fanconi española. - Explorar el papel de la ruta FA en la biología y el mantenimiento telomérico. - Estudiar la biología molecular de la ruta FA en respuesta al daño genómico. Con los estudios llevados a cabo, se ha observado un acortamiento telomérico acelerado en los pacientes Fanconi acompañado de un incremento en el número de fragmentos extrateloméricos y fusiones, aunque este exceso de fusiones terminales ocurre de manera independiente a la presencia de TRF2 en el telómero. Además, este acortamiento no está directamente relacionado con la severidad de la enfermedad a nivel hematológico. Si que se relaciona, sin embargo con la enfermedad hematológica la frecuencia espontánea de roturas cromosómicas. Por otra parte, se ha estudiado y caracterizado exhaustivamente la población Fanconi española, haciendo especial énfasis en los portadores de mutaciones en el gen FANCA y en la población de etnia gitana, poseedores de la mayor frecuencia de portadores de mutaciones en FANCA a nivel mundial. Un dato importante extraído de estos estudios es la participación de la histona H2AX dentro de la ruta Fanconi en respuesta a la irradiación con UV-C.Fanconi anemia (FA) is an autosomic recessive genetic disorder characterized by a subset of congenital malformations, bone marrow failure and a high predisposition to solid tumours and acute myeloid leukaemia acquisition. FA cells harbour a high spontaneous and induced by interstrand crosslinking agents chromosomic instability. Some other cellular features are the sensibility in front of oxidative damage, cell cycle failure, increased levels of apoptosis, impaired transcription regulation and telomeric dysfunction. Up to date, there have been described 12 genes that cause this disease though not all of them have been cloned and characterized so far. Among them, FANCA is the most prevalent gene in the affected population and it can be affected by a wide spectrum of mutations, being large deletions one of the most common. Acquire a sight about the biology of this illness will allow us having a wider view about/on DNA damage repair in response to some agents. At the same time, we can have a better knowledge about the molecular interactions established among different genetic syndromes with an indisputable diagnostic and therapeutic value. Within the present thesis four main objectives have been raised: - Establishment of potential relationship between the haematological disease and telomere shortening, and numerical or structural chromosomal abnormalities - Genetic characterization of Fanconi anemia Spanish population - Explore the role of the FA pathway on telomeric biology and maintenance - Study the molecular biology of the FA pathway in response to genomic damage As a conclusion of these studies, an accelerated telomeric shortening together with and increased number of extratelomeric fragments and end-to-end fusions has been observed in the FA patients compared to age-matched controls. The excess of terminal fusions is not related with an impaired role of TRF2. This telomeric shortening is not related with the hematologic disease observed in these patients. However, the spontaneous frequency of chromosomal breaks seems to correlate with the haematological severity. Spanish FA population has been extensively studied and characterized, especially those belonging to FA-A complementation group and the gypsy population, which has come out to be the group with the highest frequency of FANCA mutated carriers worldwide. An important point unraveled from our studies is the role of histone H2AX in the FA pathway in response to UV-C irradiation
Optimizaciones metodológicas del ensayo del cometa y su aplicación en biomonitorización humana
Descripció del recurs: el 23 de novembre de 2010La integridad del DNA es un aspecto fundamental para la salud y el buen funcionamiento de nuestro organismo. Sin embargo, sabemos que el material genético es susceptible de ser dañado por numerosos agentes y/o procesos. Aunque la mayoría del daño que se produce en nuestro material genético es separado eficientemente, parte de él escapa a este proceso constituyendo uno de los sustratos para el potencial desarrollo de un proceso carcinogénico. En los últimos años, se han desarrollado con éxito nuevas metodologías capaces de evaluar el daño en el DNA, diseñándose técnicas que permiten medir directamente roturas de cadena simple (SSB) y de doble cadena (DSB) en el DNA. Entre ellas, el ensayo del cometa se ha convertido en una de las más populares para la evaluación de este tipo de daño. Aunque el ensayo del cometa se ha propuesto como una herramienta apropiada, todavía existen muchos aspectos que deben reconsiderarse, por lo que el objetivo principal de este trabajo de Tesis fue la optimización del ensayo del cometa como biomarcador de daño en el DNA y su aplicación tanto en radiobiología, mediante la construcción de una curva de calibración, como en la biomonitorización de dos cohortes de madres e hijos de dos localidades de Barcelona. Las metodologías del ensayo del cometa que se aplican en los diferentes laboratorios son variadas pero, sin duda alguna, la versión alcalina es la más utilizada. A pH >13, la técnica resulta más sensible para la detección de niveles bajos de daño, por lo que la hemos elegido como versión estándar. Se desarrollaron numerosos experimentos con modificaciones que optimizaron el ensayo en aspectos como: el soporte utilizado, que permitió la manipulación de un elevado número de muestras en un mismo ensayo; la utilización de enzimas de reparación para la detección de daño oxidativo, y la criopreservación para el almacenamiento masivo de muestras valorables mediante el ensayo. En cuanto a las aplicaciones, la construcción de una curva de calibración resulto ser un gran aporte ya que con ella se pudo establecer el daño genético basal de una población, en términos de roturas/109 Da de DNA. Además, se ha puesto de manifiesto que la radiosensibilidad individual puede ser medida con el ensayo de cometa, lo que respalda a la técnica como un buen indicador de inestabilidad genómica individual. Finalmente, en el área de la biomonitorización humana, se encontró una fuerte correlación, tanto en el daño basal como oxidativo, entre madres e hijos; aunque, no se pudo establecer fehacientemente una influencia significativa de ninguno de los factores de confusión considerados. Esto pone en evidencia los problemas asociados con la reciente inclusión del ensayo del cometa en los estudios de biomonitorización, por lo que surge la necesidad de desarrollar estudios específicos que tengan como objetivo principal la determinación de aquellos factores que puedan incidir significativamente en los niveles de daño en el DNA de poblaciones humanas.The DNA integrity is a fundamental aspect for human health and for the suitable functions of our organism. However, the genetic material is susceptible to be damaged by many agents and processes. Although the majority of damage induced in our genetic material is repaired efficiently, a part of this damage escapes from this process and becomes one of the substrates for the potential development of a carcinogenic process. During the last years new methodologies have been developed to assess the DNA damage by performing techniques which allow to measure single strand breaks (SSB) and double strand breaks (DSB) directly. The comet assay has become one of the most popular approaches to evaluate this type of damage. Although this technique has been proposed as an appropriate tool, many aspects must be still reconsidered. For this reason, the aim of this Thesis was to contribute to the optimization of the comet assay as a biomarker of DNA damage and its application in radiology, by means of the construction of a calibration curve and, also, biomonitoring of two cohorts composed by mothers and newborns recruited from two hospitals of the Barcelona province.The methodologies of the comet assay, which are applied in the different laboratories, are variable but, with no doubt, the alkaline version is the most used. This technique is more sensitive to detect lower levels of damage at pH>13. For this reason, we have chosen it as the standard version to be used in our study. We have developed numerous experiments with modifications which have optimized the assay in three aspects: the type of support utilized, which allows the manipulation of many samples in the same experiment; the use of repair enzymes in order to detect the oxidative damage; and the cryopreservation to keep many samples which can be properly evaluated in the assay.As regards the applications, the construction of a calibration curve was an important contribution because, with its use we have been able to establish the basal genetic damage of the population, in terms of breaks /109 Da DNA. Moreover, it has been proposed that the individual radiosensitivity can be measured with the comet assay. This supportsthat this technique is a satisfactory indicator of individual genomic instability.Finally, in the area of human biomonitoring, we have found a strong correlation for both the DNA basal damage and oxidative damage between mothers and children; but we couldn't clearly establish a significant influence of any of the confusion factors that has been considered in this study. This evidence has revealed related problems with the recent use of comet assay in biomonitoring studies, and has resulted in the necessity to develop specific studies in order to determine these factors, which can influence significantly on the levels of DNA damage in human populations
Riesgo genotóxico de los subproductos de la desinfección del agua
Descripció del recurs: el 24 març 2011BibliografiaLos subproductos de la desinfección del agua de consumo son compuestos químicos que se generan por la reacción entre el desinfecante usado, cloro en la mayoría de los casos, y la materia orgánica e inorgánica presente en el agua a desinfectar. El perfil de estos subproductos es diferente entre las distintas plantas de tratamiento, debido a varios factores involucrados en su formación como son el tipo de desinfectante usado, el desinfectante secundario, el pH, la temperatura y la materia orgánica e inorgánica presente, entre otros factores. Además de la exposición a los SPD mediante el agua de consumo, otra forma de exposición la constituye el uso recreacional del agua de las piscinas. En estas aguas se pueden llegar a encontrarse niveles mayores de SPD, en comparación a los del agua de consumo; este incremento en los niveles de SPD se debe a factores como la recirculación del agua, un mayor tiempo de contacto con el desinfectante y la presencia de precursores orgánicos provenientes de los bañistas. Numerosos estudios epidemiológicos han investigado los efectos adversos que la exposición a estos SPD puede tener en la salud humana, encontrando una posible asociación entre el incremento en el riesgo de cáncer de vejiga y la exposición a estos subproductos. Por lo tanto, con la finalidad de incrementar la información sobre el potencial genotóxico de algunos SPD, se seleccionaron 11 SPD (bromonitrometano, tricloronitrometano, tribromoacetaldehido, hidrato de cloral, ácido mucobrómico, ácido mucoclórico, nitrosodimetilamina, nitrosodietilamina, ácido yodoacético, ácido bromoacético, ácido cloroacético), los cuales se evaluaron en los ensayos del cometa, de micronúcleos y de linfoma de ratón. En función de los resultados obtenidos, la mayoría de los SPD seleccionados pueden ser considerados como genotóxicos (ensayo del cometa), aunque tan sólo uno ha inducido daño clastogénico/aneugénico (ensayo de micronúcleos) y otro se ha mostrado como mutagénico (ensayo de linfoma de ratón). Además, la evaluación del efecto genotóxico del agua de piscina ha corroborado que el proceso de desinfección adicional incrementa su potencial genotóxico, al tiempo que se ha encontrado una posible asociación entre los niveles de ciertos trihalometanos en el aire exhalado de usuarios de piscina y los incrementos en los marcadores de daño genotóxico (frecuencia de micronúcleos).Disinfection by-products are chemical compounds generated from the reaction of the disinfectant, usually chlorine, with natural organic and inorganic matter present in the source water during the disinfection process. Each water treatment plant has a unique profile of disinfection by-products (DBPs) depending on many factors such as the disinfectant used, secondary disinfectant, pH, temperature, natural organic and inorganic matter content, etc. In addition to the ingestion exposure to DBPs by the consumption of water, the recreational use of swimming pools is another important type of exposure to DBPs. In this water, the levels of DBPs are greater than the ones detect in tap water; this increase in DBPs levels is due to factors like the recirculation of water, the greater time of contact with the disinfectant and the presence of many organic precursors originating from the swimmers. Several epidemiologic studies have investigated the adverse effects of the DBP exposure in the human health, finding a possible association between the increase in the risk of urinary bladder cancer and the exposure of these by-products. Therefore, with the purpose of increasing the information of the genotoxic potential of some DBPs, we selected 11 DBPs (bromonitromethane, trichloronitromethane, tribromoacetaldehyde, chloral hydrate, mucobromic acid, mucochloric acid, nitrosodimethylamine, nitrosodiethylamine, iodoacetic acid, bromoacetic acid, chloroacetic acid) which were evaluated in the comet assay, the micronucleus test and the mouse lymphoma assay. According to our results, almost all of the selected DBPs can be considered as genotoxic agents (comet assay), but only one DBP induced clastogenic/aneugenic damage (micronucleus test) and another DBP was considered as mutagenic (mouse lymphoma assay). The evaluation of the genotoxic effect of swimming pool water has corroborated that the additional disinfection process increased the genotoxic potential of swimming pool water, and also it has been found a possible association between the presence of some trihalomethanes levels in the exhalated air of swimming pools users, and the increase in the biomarkers of genotoxic damage (frequency of micronuclei)
Evaluación genotóxica de algunos metales pesados en Drosophila melanogaster mediante los ensayos SMART de alas y Cometa
Descripció del recurs: el 19 d'abril de 2010El presente estudio tiene como objetivo analizar la genotoxicidad de los metales mercurio, plomo y níquel en D. melanogaster. Para este propósito, se han utilizado y desarrollado dos ensayos in vivo: 1) el ensayo de mutación y recombinación somáticas (SMART) en alas de D. melanogaster; y 2) el ensayo del cometa en hemocitos de Drosophila. Los resultados obtenidos con el ensayo SMART en alas de D. melanogaster indican que ninguno de los tres metales muestra una actividad genotóxica detectable. Sin embargo, al analizar la interacción de estos metales frente al daño genotóxico inducido por la radiación gamma, se observa que únicamente los compuestos de níquel muestran una interacción de tipo sinérgica, lo cual indicaría una interferencia con los mecanismos involucrados en la reparación. Por otro lado, los resultados obtenidos con el ensayo del cometa en hemocitos aislados de larvas previamente tratadas con metales, muestran un daño genético significativo con el metilmercurio, el nitrato de plomo y el sulfato de níquel, indicando una mayor sensibilidad de este ensayo en comparación con el ensayo SMART de alas. Como conclusión final de este trabajo de tesis doctoral se desprende que la genotoxicidad de los metales pesados evaluados en D. melanogaster podría se considerada débil y dependiente de su formulación química.The present study aims at analysing the genotoxicity of mercury, lead and nickel metals in D. Melanogaster. Two in vivo assays have been employed and developed in order to achieve this: 1) the somatic mutation and recombination test (SMART) in D. melanogaster wings, and, 2) the comet test in Drosophila haemocytes. Our results with the SMART test in D. melanogaster indicate that none of the three metals show a detectable genotoxic activity. However, when analysing the interaction of these metals with the genotoxic damage induced by gamma irradiation, only nickel compounds show a synergic interaction, indicating an interference with the repair mechanisms. On the other hand, the results obtained in the comet assay with isolated haemocytes from larvae previously treated with the metals show a significant genetic damage under methylmercury, lead nitrate, and nickel sulphate, indicating that this test is more sensitive than the SMART assay. Finally, through this doctoral dissertation we have been able to conclude that the genotoxicity of heavy metals on D. melanogaster can be considered weak and dependant of its chemical formulation
Biomonitorización citogenética de cuatro poblaciones agrícolas europeas, expuestas a plaguicidas, mediante el ensayo de micronúcleos
Consultable des del TDXTítol obtingut de la portada digitalitzadaLa importancia de los plaguicidas recae tanto en los numerosos beneficios que tienen para la sociedad (protectores de plagas de las cosechas, erradicación de vectores que propagan enfermedades, etc.); como en los efectos negativos que representan para la salud (problemas reproductivos, sarcomas, linfomas, etc.). Los colectivos en contacto más directo con estos productos, como son los agricultores, tienen un mayor riesgo de sufrir los efectos adversos que se han asociado con los plaguicidas. Esta Tesis comprende una serie de trabajos sobre la evaluación del daño genotóxico de los plaguicidas en cuatro poblaciones agrícolas europeas, mediante la técnica de micronúcleos (MN) en dos tipos celulares: linfocitos de sangre periférica y células de descamación de la mucosa bucal. Paralelamente, también se determinó el índice de proliferación celular (CBPI). Las poblaciones que se han estudiado pertenecen a colectivos agrícolas con una continua exposición a plaguicidas, procedentes de Almería (España), Malopolska (Polonia), Nea Makri (Grecia) y del sudeste de Hungría. Cada población ha sido evaluada individualmente y finalmente se han agrupado y analizado los datos conjuntos, para obtener una visión general de los efectos de la exposición. Además, en la población almeriense se realizó un estudio longitudinal, para evaluar la existencia de diferencias estacionales en dos periodos de distinta aplicación de plaguicidas. Asimismo, en esta población se evaluó la posible influencia de los genotipos GSTM1 y GSTT1 sobre la susceptibilidad individual en al respuesta genotóxica a la exposición a plaguicidas. Únicamente se observó una relación entre GSTM1 y CBPI disminuyendo este último en presencia del producto del gen GSTT1 en los agricultores. A pesar de que en alguna población se observa una tendencia a un mayor número de células binucleadas con micronúcleos (BNMN), en los linfocitos de los individuos expuestos a plaguicidas, en ninguna de las poblaciones evaluadas individualmente ni en el análisis global de la población, las diferencias observadas llegan a alcanzar un nivel estadísticamente significativo. Lo mismo ocurre con las células bucales con MN (CBMN). Respecto del CBPI, en la mayoría de poblaciones se observan descensos, significativos o no, de los valores del mismo en el colectivo agrícola. Al valorar toda la población en global estas variaciones se traducen en un descenso significativo del CBPI en los individuos expuestos a plaguicidas, lo que refleja el efecto citotóxico de los mismos. En el análisis global, observamos que el hecho de pertenecer a un país u otro confiere una serie de características particulares a cada grupo, que se reflejan en una mayor frecuencia de un determinado tipo de daño genético según el sistema celular estudiado. Otro de los objetivos de la Tesis era valorar los factores higiénicos, de salud, de exposición a plaguicidas, de protección laboral, de alimentación, etc., que podían influir en las variaciones de la frecuencia de micronúcleos y en la proliferación celular. Entre los factores que han mostrado tener un efecto significativo en la variación de las frecuencias de las variables de estudio están la edad, el hecho de ser mujer, el tabaco, la exposición a rayos X por diagnóstico médico, así como factores de la dieta (consumo de vegetales crudos, pescado y aceite de oliva). Hay que destacar el incremento significativo en el porcentaje de abortos encontrados en la población agrícola de Polonia, lo que junto con el elevado número de intoxicaciones y el descenso del CBPI en este colectivo nos indica que, a pesar de no observarse un incremento significativo de daño genético detectable mediante el ensayo de MN, los plaguicidas están ejerciendo efectos biológicos adversos sobre los agricultores.The importance of pesticides can be reflected as much on the benefits that they represent for society (crops pest prevention, vector erradication); as on the negative effects they have on health (reproductive problems, sarcomas, lymphomas, etc.). These adverse effects mainly affect occupationally exposed populations, such as farmers. This PhD Thesis comprises a series of works on the evaluation of genotoxic damage caused by pesticide exposure of four agricultural European populations, using the micronucleus assay (MN). Both lymphocytes of peripheral blood and exfoliated cells of the oral mucosa were used. The cytokinesis-block proliferation index (CBPI) was also calculated to detect possible variations in the proliferative kinetics of lymphocytes (due to the pesticide exposure). Populations from Greece, Spain, Poland and Hungary, all with an intense and continuous agriculture activity, were evaluated individually. Finally, in order to obtain a general effect of the exposure to pesticides, all the data was pooled and analysed together. Furthermore, a follow-up study was carried out in the Spanish population to determine, at two different periods of time and different pesticide intensity application, if there were season-related differences in the MN frequency. In this population, to look for relationship between the genotypes and the cytogenetic response, each donor was assessed for the presence or absence of glutathione S-transferase M1 and GSTT1. Only in the exposed individuals, the CBPI appeared to be affected, decreasing in the presence of GSTT1. Although in some populations we observed a trend to a high number of BNMN (binucleated cells with micronucleus) in the lymphocytes of farmers exposed to pesticides, none of the statistical analysis showed significant differences between controls and farmers, for neither lymphocytes nor buccal cells with MN. Regarding the CBPI, decreases were observed in most of the pesticide-exposed populations. In the evaluation of all the populations, the group occupationally exposed to pesticides showed a significantly lower CBPI level, indicating some cytotoxicity due to the exposure. The global analysis showed us that the increases in genetic damage seemed to be related to different cellular systems for each country, revealing the particular characteristics of the populations evaluated. One of the aims was to assess the hygienic and health factors, exposure to pesticides, work protective measures, diet, etc., that could influence on the frequency variations of MN and cell proliferation. From the studied variables, those with a significant effect on the frequencies studied were age, the fact of being woman, tobacco, X-ray medical exposure and some diet factors (consuming raw vegetables, fish and olive oil). The agricultural Polish population showed a significant increase in the percentage of miscarriages, which together with the high number of intoxications and the CBPI decrease in this group, showed us that pesticides do have biological adverse effects on the farmers despite the lack of a significant increase of cytogenetic damage evaluated by the MN assay
Biomonitorización genotóxica de poblaciones humanas expuestas ambientalmente al arsénico
Consultable des del TDXTítol obtingut de la portada digitalitzadaEl Arsénico (As) es un elemento natural de amplia y heterogénea distribución en la naturaleza, siendo altamente tóxico para el organismo humano. El As se encuentra a concentraciones mucho más altas de lo que se considera seguro en el agua de consumo de millones de personas en distintos países. La exposición ambiental al As constituye un factor de riesgo para distintos tipos de cáncer; sin embargo, el mecanismo por el cual el As ejerce su acción toxicológica permanece incierto. El objetivo de esta Tesis Doctoral ha sido el de evaluar el posible daño genotóxico de una población expuesta a altas concentraciones de As en el agua de consumo. Se ha utilizado la concentración de As en orina y en uñas como biomarcadores de exposición interna. Para determinar el nivel de daño citogenético, se ha utilizado el ensayo de micronúcleos en linfocitos de sangre periférica y en células de descamación de la mucosa bucal. Teniendo en cuenta que el daño genético puede estar modulado por las características genéticas individuales, también se ha realizado un análisis de genotipos para los polimorfismos de los genes GSTM1, GSTT1 y GSTP1 en la población de estudio, lo que se ha utilizado como biomarcador de sensibilidad individual. La población expuesta ha sido seleccionada en el norte de Chile donde, por las características geológicas de la región, se presentan altas concentraciones de As en el agua de consumo, llegando a superar los 0,75 mg As/L. Esta población cuenta con 106 individuos de los cuales 50 son de etnia atacameña. La población indígena habita en la región bajo similares condiciones desde hace unos 11.000 años. Se ha propuesto que los atacameños presentan algún tipo de resistencia frente a la exposición al As debido a que no tienen los problemas de salud descritos en otras poblaciones expuestas bajo condiciones similares. La población control cuenta con 111 individuos y se ha seleccionado en la ciudad de Concepción, ubicada unos 2.000 km al sur del primer lugar de muestreo, presentando niveles de As en agua de 0,0002 mg As/L. En este trabajo se ha evidenciado que la exposición al As por medio del agua de consumo supone un riesgo genotóxico, induciendo un aumento significativo en la frecuencia de MN en linfocitos en la población expuesta. Sin embargo, no se ha detectado daño en las células de la mucosa bucal. En relación con la supuesta resistencia de la etnia atacameña a los altos niveles de As ambiental, no se han detectado diferencias significativas entre los atacameños y los no atacameños en cuanto al daño citogenético evaluado mediante el ensayo de MN, ni en linfocitos ni en células de la mucosa bucal, a pesar de que la población atacameña presenta una concentración mayor de As en las uñas. En cuanto a los polimorfismos genéticos, sólo se han encontrado diferencias significativas para el gen GSTP1, en que el genotipo val/val (homocigoto mutante) es más frecuente en la población que presenta un mayor nivel de daño citogenético, medido en ambos tipos celulares. También se ha encontrado con mayor frecuencia en la población atacameña.The arsenic (As) is a ubiquitous element in the environment and its distribution is heterogeneous in nature. In some locations, the As occurs naturally in drinking water in higher concentrations to those considered as safe, and affects millions of people. Environmental exposure to As is considered as a cancer risk factor. Nowadays, no general agreement has been reached about its mechanism of action related to the genotoxic potential of arsenic. Taking this into account, the objective of this Doctoral Thesis has been the assessment of genotoxic damage of a population environmentally exposed to As in drinking water. Individual measures of As exposure were determined in fingernails and urine as internal biomarkers of exposure. The micronucleus assay has been used to determine the possible genotoxic damage, using peripheral blood lymphocytes and epithelial cells of buccal mucosa. Considering that cytogenetic damage can be modulated by individual genetic characteristics, the polymorphisms for the genes GSTM1, GSTT1 and GSTP1 in the population studied have been analyzed and they have been used as biomarkers of individual susceptibility. The exposed population has been selected form different villages from northern Chile. In this region As occurs naturally in water in connection with geological characteristics. We have determined concentrations over 0.75 mg As/L. The exposed population consisted in 106 people; taking into account their ethnicity they were classified in 50 atacameño and 56 non-atacameño. The atacameño people inhabits this region under similar conditions for almost 11,000 years, and it has been proposed that this people have some kind of resistance to As effects because they don't present classical problems of arsenicism shown in populations under similar conditions of exposure. The control group consisted in 111 healthy people from Concepción city, located almost 2,000 km south, where As levels in drinking water were as low as 0.0002 mg As/L. The results obtained in this study indicated that exposition to As in drinking water involved a genotoxic risk, by the induction of a significant high frequency of micronucleus in peripheral blood lymphocytes. In this work there is no evidence of cytogenetic damage in oral mucosa cells. In relation with the supposed resistance to As exposure of atacameño people, we did not found significant differences in the frequency of genetic damage between the atacameño group and the non-atacameño exposed group in both cellular types. However, the atacameño group shows high concentrations of As in nails. With respect to the genotype analysis, we found significant differences in the frequency distribution polymorphisms of gene GSTP1. The homozygous genotype mutant (val/val) presents a higher frequency in the population with a high level of cytogenetic damage in both cellular types. The results show that it occurs frequently in atacameño people when compared to the non-atacameño group
Utilidad del ensayo de linfoma de ratón en la determinación de los mecanismos de genotoxicidad del arsénico : espectro mutacional del trióxido de arsénico

Está ampliamente aceptado su potencial carcinógenico aunque, a diferencia de otros carcinógenos, diversos estudios de genotoxicidad han puesto de manifiesto que el arsénico no es un mutágeno muy potente. Hasta ahora, su mecanismo de acción en el desarrollo del cáncer continúa siendo un misterio, y existe cierta controversia respecto al tipo de daño genotóxico que induce, y el tipo de compuesto arsenical que más influye en su carcinogenicidad. En ausencia de buenos modelos animales para estudiar el mecanismo carcinogénico del arsénico, los estudios in vitro se han convertido en herramientas útiles para dilucidar estos mecanismos. Por lo tanto, con la finalidad de evaluar el potencial genotóxico de seis compuestos de arsénico (arsenito sódico, trióxido de arsénico, MMAV, DMAV, arsenobetaína y cloruro de tetrafenilarsonio), se llevó a cabo inicialmente la puesta a punto del ensayo MLA. Este ensayo se considera como el ensayo in vitro de mutación génica en células de mamífero más sensible y con mayor capacidad de detección de un amplio espectro de mutaciones, tanto puntuales como cromosómicas. De esta manera, se pretendía obtener más información respecto al potencial genotóxico y los mecanismos de acción de distintos compuestos de arsénico. Cuatro de los seis compuestos arsenicales evaluados han presentado efectos genotóxicos, consecuencia de su actividad clastogénica: el arsenito sódico, el trióxido de arsénico, el MMAV y el DMAV. Paradójicamente, en la actualidad el trióxido de arsénico (TOA) se emplea con éxito en el tratamiento de la leucemia promielocítica aguda y existe un especial interés en evaluar su efectividad terapéutica en otros tipos de cáncer. En este contexto, con motivo de estudiar más profundamente el mecanismo de acción de este compuesto, se analizó el espectro mutacional inducido por el TOA empleando los mutantes inducidos en el MLA y técnicas moleculares y citogenéticas. Nuestros resultados revelan que el principal tipo de cambio presente en estos mutantes es la pérdida de heterocigosidad (LOH), pudiendo ser generada tanto por deleción como por recombinación.Arsenic is an important environmental element causing a world-wide health problem due to large amount of people chronically exposed and the seriousness of its effects, including increase of different types of cancer. Its carcinogenicity is generally accepted, according to epidemiological evidences, and many studies have been carried out to determine its genotoxicity, showing that different from other carcinogens, it is not a potent mutagen. Nevertheless, despite the large amount of literature dealing with both the genotoxic and carcinogenic effects of arsenic, it still remains unclear how arsenic causes cancer (mode of action) and what types of arsenic compounds are most likely associated with carcinogenicity. The lack of appropriate animal models to study arsenic carcinogenicity makes in vitro studies particularly useful tools to figure out its carcinogenic mechanisms. Therefore, we evaluated the genotoxicity potential of six arsenic compounds (sodium arsenite, arsenic trioxide (ATO), MMAV, DMAV, tetraphenylarsenium and arsenobetaine) by the mouse lymphoma assay (MLA), which was optimized in our group. This test is considered the most sensitive and widely applied among the various in vitro mammalian cell gene-mutation assays, and detects a broad spectrum of mutational events, from point mutations to chromosome alterations. This way, we wanted to provide more information about the genotoxic potential and mode of action of several arsenic compounds. Our results show that four of the six arsenic compounds evaluated, sodium arsenite, arsenic trioxide, MMAV and DMAV are genotoxic according to the MLA. Paradoxically, ATO has currently been shown to be effective in inducing complete remission during treatment of patients with acute promyelocytic leukaemia (APL) and it exists a big interest in evaluating its therapeutic effectiveness against other types of cancers. This, together with the open discussion about how arsenic compounds induce genotoxic damage, prompted us to evaluate the mutational spectrum induced by ATO in mouse lymphoma cells. We studied both small and large colonies, using molecular and cytogenetical techniques to clarify the nature of the ATO induced mutations. Our results show that ATO is a strong clastogenic compound mainly inducing loss of heterozygosity (LOH), as a result of deletions and recombinational events
Estudio de biomonitorización de una población de trabajadores expuestos al arsénico y caracterización de los posibles factores moduladores del daño genotóxico

Aunque los mecanismos de acción del arsénico no están del todo dilucidados, está ampliamente aceptado su potencial citotóxico, genotóxico y carcinógeno. En este trabajo se pretende estudiar sus efectos genotóxicos, entender los mecanismos implicados en el proceso de su metabolización e investigar posibles biomarcadores de susceptibilidad asociados a la exposición. Inicialmente, se llevó a cabo un estudio de biomonitorización para averiguar las posibles correlaciones entre efectos genotóxicos y exposición ocupacional al arsénico. Se utilizaron los niveles de arsénico y de sus metabolitos en orina como biomarcador de exposición y los ensayos de MN y de SCE como medida de daño citogenético. Dado que recientemente se ha propuesto que los dos miembros funcionales de la nueva clase Omega, GSTO1 y GSTO2, juegan un papel importante en la homeostasis celular, estando también involucradas en la biotransformación del arsénico, hemos postulado que, polimorfismos en estos genes podrían estar correlacionados con cambios en la actividad de estas proteínas que lleven a diferencias en el perfil de excreción del arsénico y, consecuentemente, a diferencias de respuesta frente a una exposición. La segunda parte del trabajo ha consistido en el análisis de los polimorfismos de GSTO1 y de GSTO2 entre tres grupos étnicos y en la caracterización enzimática de dos de las variantes de GSTO1. Además, los polimorfismos de GSTO1 y de GSTO2 también se han analizado en la población de trabajadores chilenos, estableciéndose correlaciones entre genotipos, perfil de excreción y daño citogenético, integrando así las dos partes del estudio. A pesar de los avances en los estudios de expresión de GTSO1, el papel que ejerce esta enzima en el metabolismo y en la toxicidad del arsénico sigue en discusión. Así, se han utilizado dos líneas celulares con diferentes niveles de expresión de GSTO1, junto con la reducción de sus niveles de expresión mediante la técnica de RNAi, para evaluar las diferencias de respuesta frente a distintos compuestos de arsénico. No se han detectado diferencias en la toxicidad del arsénico relacionadas con los diferentes niveles de expresión. En contraste, la reducción del estrés oxidativo por la NAC indica que diferencias en los niveles de GSTO1 pueden estar modulando las diferencias de respuesta al estrés oxidativo inducido por los compuestos de arsénico. Nuestros resultados no excluyen la participación de otras enzimas en el proceso de biotransformación y toxicidad del arsénico.Arsenic has become a major world health problem, due to the size of the exposed population and the seriousness of its effects, which are mainly related with the increase of cancer cases among the chronically exposed populations. Even though the mechanisms of action of this element are not fully understood, it has been clearly reported that it is cytotoxic, genotoxic and carcinogenic in humans. In this context, we planned to study the genotoxic effect of this metal, to understand the metabolic mechanisms involved in this process, and to find biomarkers of susceptibility to arsenic exposure. Initially, a biomonitoring study was carried out to investigate the correlation between arsenic occupational exposure and genotoxic effects in smelting plant workers, using the levels of arsenic and its metabolites excreted in urine as an exposure biomarker and the SCE and the MN assays as measures of cytogenetic damage. Two members of the recently identified Omega class glutathione S-transferase enzymes (GSTO1 and GSTO2) have been proposed to play an important role in cellular homeostasis and in the arsenic reduction pathway. Therefore, polymorphisms in these genes could be related with variations in the protein activity leading to changes in the arsenic excretion profile, and consequently with the response to chronic exposures. The second part of this study was based on the analysis of hGSTO1/2 polymorphisms among three ethnical groups and the enzymatic characterization of two GSTO1 variants. Moreover, GSTO1 and GSTO2 polymorphisms were also analyzed in the Chilean workers. Then, the first and the second part of this work have been linked, establishing correlations among genotypes, variations of urine arsenic profile and cytogenetic damage. Despite the recent progress in the study of GSTOs expression and pharmacokinetics, the role of these enzymes in the arsenic metabolism and toxicity is under discussion. To clarify this, two cell lines with different expression levels of GSTO1 have been used to evaluate the cytotoxic response of various arsenic compounds. In addition, depletion of the GSTO1 levels with siRNA has also been used. Results do not shown significant variations in arsenic toxicity, according to the different expression levels. In contrast, reduction of oxidative stress by NAC indicates that differences at GSTO1 expression levels could modulate the response to the oxidative stress induced by arsenic compounds. These results do not rule out others enzymes participating in the arsenic metabolism and toxicity
